포유동물의 생식을 조절하는 다수의 호르몬 가운데 가장 중요한 것들로 난소로부터의 estrogen과 progesterone을 들 수 있다. 반면 다양한 스트레스 인자들은 암컷의 성 반응 행동과 번식을 억제함이 잘 알려졌다. 이러한 스트레스가 가해지는 동안 부신에서는 카테콜아민(catecholamine)이 다량 분비되어 위기 상황에 대처하며 이 과정에서 생식 현상의 억제가 일어나는 것으로 추정된다. 본 연구에서는 생식호르몬 분비와 성 행동 양식에 광범위한 영향을 미침이 알려진 카테콜아민 중 특히 부신의 norepinephrine(NE)과 epinephrine(E) 합성ㆍ분비 양상과 발정주기 간의 상관관계를 조사하였다. 이를 위해 HPLC-ECD를 사용하여 주기 중인 흰쥐 부신 수질내 NE와 E함량과 체외 배양한 부신으로부터의 분비를 조사하였다. NE 함량은 proestrus에서 증가하기 시작하여 diestrus I에서 최고에 도달하였고, diestrus II에 최소치로 감소하였다. 부신 내 E 함량의 최고치는 proestrus, 그리고 최저치는 diestrus II에서 관찰되었다. 흰쥐 부신 내 NE : E ratio는 diestrus I에서 1 : 4.81로 가장 낮았고 기타 시기에는 1 : 6.13~7.02였다. 체외 배양한 흰쥐 부신으로부터의 NE 분비는 diestrus II에서 가장 낮았으며 estrus에서 최고에 도달하였고, proestrus에서의 분비 역시 diestrus II 때보다 유의성있게 높았다. E분비의 최고치는 estrus에서, 그리고 최저치는 diestrus 떼서 II 관찰되었다. 한편 배양액 중 Ne : E ratio는 estrus에서 1 : 3.32로 가장 높았고 기타 시기에는 1 : 2.34~2.65였다. 본 연구 결과는 (1) 흰쥐 부신에서 카테콜아민 생성과 분비 양상이 발정주기 중 역동적으로 변화하며, (2) NE로부터 E로의 전환이 발정주기 중 stage-specific하게 일어남을 나타내는 것으로서, 이는 카테콜아민 합성율을 결정하는 rate limiting enzyme인 tyrosine hydroxylase(TH)와 NE에서 E로의 전환 과정을 매개하는 phenylethanolamine-N-methyltransferase(PNMT)의 발현과 활성이 중추신경계에서와 유사하게 생식호르몬, 특히 estrogen and/or progesterone의 영향을 받음을 시사한다.
The objective of this study was to investigate the relationship between changes in body temperature, adiposity and sympathoadrenal activity in normal weight female college students, eating an instant-noodle lunch in a laboratory setting at ambient temperatures of 22-24.8$^{\circ}C$. Preprandial epinephrine(EPI) concentration, as an indicator of adrenal activity, was inversely and significantly correlated with body weight, body mass index(BMI), and waist girth. Changes in pre- and postprandial EPI concentrations showed positive correlations with % body fat, fat mass, waist girth, hip girth, and waist/hip girth ratio(WHR). The preprandial norepinephrine (NE) concentration was negatively correlated with recovery time from the peak postprandial core temperature to the meal-start core temperature(RTST). However, the NE concentration, an indicator of sympathetic neural activity, was not related to anthropometric measurements in normal weight young women. In conclusion, adrenal activity was negatively associated with adiposity and central body fat distribution. Sympathetic activity was related to body temperature regulation capacity after a meal, but was not related to adiposity in normal weight young women. (Korean J Nutrition 31(7) 1130-1138, 1998)
Magnesium ($Mg^{2+}$) transport across the plasma membrane of cardiac myocytes appears to be under hormonal control. Repeated stimulations with adrenergic or histaminergic agonist produced a progressive decrease in $Mg^{2+}$ efflux from hearts. Thus we hypothesized that the $Mg^{2+}$ efflux may be resulted from a down-regulation of receptors or from a depletion of $Mg^{2+}$ from intracellular pool(s) in the hearts. In the present study, the regulation of $Mg^{2+}$ homeostasis by receptor stimulation was studied in perfused rat and guinea pig hearts. The successive short addition of norepinephrine (NE) to rat and guinea pig, and of histamine (HT) to perfused guinea pig hearts induced a progressive decrease in $Mg^{2+}$ efflux. These $Mg^{2+}$ effluxes were blocked by propranolol or ranitidine, respectively. These decrease in $Mg^{2+}$ efflux were inhibited by sodium cyanide (NaCN), which increases intracellular $Mg^{2+}$ ($[Mg^{2+}]_i$) levels. When NE (or HT) was added after HT (or NE), this efflux was also decreased in the guinea pig hearts. In the rat hearts and myocytes, HT did not stimulate $Mg^{2+}$ efflux. But NE produced a large $Mg^{2+}$ efflux after stimulation with HT. 8-(4-Chlorophenylthio)-adenosine cAMP (cAMP), like NE and HT, also induced a progressive decrease in $Mg^{2+}$ efflux in guinea pig hearts. This effect was inhibited by NaCN. These data provide evidence that the progressive decrease in receptor-stimulated $Mg^{2+}$ efflux is considered to be due to a decrease in $[Mg^{2+}]_i$ levels rather than receptor down-regulation.
Objectives : The Meridians and acupuncture points are the fundamental theories for acupuncture therapy. They have been associated with nervous system, but It is not well defined. We investigated that the effects of acupuncture at the river points(LU8, HT4, PC5, SP5, KI7, LR4) on the changes in the expression of nNOS, iNOS, eNOS, and NE in rats. Methods : The Male Sprague-Dawley rats were divided into 6 groups each non-acupuncture and acupuncture group. We inserted needle and retained for 5 minutes on both left and right sides of LU8, HT4, PC5, SP5, KI7, LR4 which were the river points of five transport points for 6 yin meridian vessels. After that, blood was drawn via cardiac puncture, and tissues for each point near meridian vessels were extracted to examine the changes in the changes of nNOS, eNOS, iNOS and NE. Results : The LU8 and HT4 group showed a significant decrease on nNOS. In terms of eNOS and iNOS, the LU8 group decreased significantly while the KI7 group increased significantly. However, the experimental groups didn't show any significant changes on the plasma and tissue norepinephrine without plama NE in SP5 group. Conclusions : The effect on the nNOS, iNOS, eNOS of acupuncture at LU8 and KI7 could be observed, and it is considered that the effect of acupuncture related with on nervous system could be studied by additional researches based on this one.
The purpose of this study was to analyze the Rhizoma on the blood pressure, heart rate and to define the mechanism of Notopterygii Rhizoma-induced relaxation in rabbit common carotid arterial contracted by agonists. Method : In order to explore the effect of Notopterygii Rhizoma on the blood pressure and heart rate, Notopterygii Rhizoma extract was injected in vein of rabbit ear. In order to investigate the effect of Notopterygii Rhizoma on norepinephrine(NE)-induced contracted rabbit carotid arterial strips, transverse strips with intact or damaged endothelium were used for the experiment using organ bath. To analyze the mechanism of Notopterygii Rhizoma-induced relaxation, Notopterygii Rhizoma extract infused into NE-induced contracted strips induced by agonists after treatment of methylene blue, propranolol, ouabain and it infused into serotonin, potassium chloride-induced contracted strips. Result : The blood pressure was significantly decreased by Notopterygii Rhizoma, but heart rate was insignificantly. In addition, Notopterygii Rhizoma significantly relaxed the norepinephrine, serotonin, potassium-induced contracted strips with intact endothelium or damaged endothelium. The relaxing effect of Notopterygii Rhizoma In NE-induced contracted strips with damaged endothelium by pretreatment of methylene blue, propranolol was not changed, but Ouabain was significantly decreased. Conclusion : These results were shown that Notopterygii Rhizoma affected the NE -induced contracted smooth muscle without the participation of endothelium, and demonstrated that the mechanism of NotoDtervgii Rhizoma-induced relaxation was the obstruction of receptor-operated Ca2+ channel.
Previous studies have suggested that brain stem noradrenergic inputs differentially modulate neurons in the paraventricular nucleus (PVN). Here, we compared the effects of norepinephrine (NE) on spontaneous GABAergic inhibitory postsynaptic currents (sIPSCs) in identified PVN neurons using slice patch technique. In 17 of 18 type I neurons, NE $(30{\sim}100{\mu}M)$ reversibly decreased sIPSC frequency to $41{\pm}7%$ of the baseline value $(4.4{\pm}0.8\;Hz,\;p<0.001).$ This effect was blocked by yohimbine $(2{\sim}20{\mu}M),$ an ${\alpha}_2-adrenoceptor$ antagonist and mimicked by clonidine $(50{\mu}M),$ an ${\alpha}_2-adrenoceptor$ agonist. In contrast, NE increased sIPSC frequency to $248{\pm}32%$ of the control $(3.06{\pm}0.37\;Hz,\;p<0.001)$ in 31 of 54 type II neurons, but decreased the frequency to $41{\pm}7$ of the control $(5.5{\pm}1.3\;Hz)$ in the rest of type II neurons (p<0.001). In both types of PVN neurons, NE did not affect the mean amplitude and decay time constant of sIPSCs. In addition, membrane input resistance and amplitude of sIPSC of type I neurons were larger than those of type II neurons tested (1209 vs. 736 $M{\Omega},$ p<0.001; 110 vs. 81 pS, p<0.001). The results suggest that noradrenergic modulation of inhibitory synaptic transmission in the PVN decreases the neuronal excitability in most type I neurons via ${\alpha}_2-adrenoceptor,$ however, either increases in about 60% or decreases in 40% of type II neurons.
The $Ca^{2+}-substitutional$ roles of strontium for the contractile processes were investigated in the rabbit renal artery. The contractions induced by either norepinephrine or high $K^+$ in the condition which intra- and extracellular $Ca^{2+}$ were replaced by $Sr^{2+}$, i.e. $Sr^{2+}-mediated$ contractions, were dose-dependent. And then the maximal amplitude of contraction, as compared with $Ca^{2+}-mediated$ contraction, was about 50% in norepinephrine and about 70% in high $K^+$. The $Sr^{2+}-mediated$ contractions were independent in the contraction by norepinephrine $(10^{-5}M)$ but dependent in those by high $K^+(100\;mM)$ on the extracellular $Sr^{2+}$ concentration. Also $Sr^{2+}-mediated$ contractions induced by norepinephrine were observed in the $Sr^{2+}-free$ Tyrode's solution. The $Sr^{2+}-mediated$ contractions induced by either norepinephrine or high $K^+$ were suppressed by verapamil, a $Ca^{2+}-channel$ blocker. By extracellular addition of $Sr^{2+}$, the $Ca^{2+}-mediated$ contractions induced by norepinephrine $(10^{-5}M)$ or 40 mM $K^+$ were inhibited but those by high $K^+(100\;mM)$ were increased. And the $Sr^{2+}-mediated$ contractions were increased by extracellular addition of $Ca^{2+}$ but did not reach the level of $Ca^{2+}-mediated$ contraction. Therfore it is suggested that in the vascular smooth muscle of rabbit renal artery $Sr^{2+}$ could enter the smooth muscle cells easily through the potential-operated calcium channel (POC) but not easily through the receptor-operated calcium channel (ROG), and $Sr^{2+}$ might be stored in the intracellular $Ca^{2+}-binding$ site and released by NE and induced the contraction by a way of activating directly the contractile apparatus.
스트레스는 여러 가지 정신질환의 병태생리와 관련되는 것으로 알려졌다. 최근 여러 가지 동물 모델이 제시되고 뇌에 대한 연구가 활발해지면서 스트레스의 선경생물학적 기전에 대해 많은 사실이 밝혀지고 있다. 저자들은 동물과 사람을 대상으로 스트레스가 지각되고 대뇌에서 처리되고 신경내분비적 반응으로 전환되는 경로를 밝히고자 했던 최근의 연구들을 고찰하였다. 과거 변연계-시상하부-뇌하수체-부신 축(LHPA axis)과 자율신경계가 스트레스반응의 신경생물학적 담당자로 가장 많이 연구되어 왔으나, 최근에는 노르에피네프린(NE), 세로토닌, GABA/Glutamate, 도파민, 아세틸콜린 등의 신경전달물질과 부신피질자극호르몬방출인자(CRF), arginine vasopressin. glucocorticoid 등의 신경호르몬이 상호작용을 하면서 스트레스반응에 관계되는 것으로 알려지고 있다. 이러한 대뇌의 신경전달체계는 LHPA축과 유기적으로 연관되면서 스트레스반응을 매개하며, 구조적으로도 LHPA축은 해마, 편도 등 다양한 대뇌 부위와 연결된다. LHPA축은 이렇게 중층적으로 조절되는데, 여기에 생기는 이상은 만성 스트레스나 우울증 등 병적 상태와 관련된다. CRF는 LHPA축의 호르몬 역할 이외에 대뇌의 광범위한 부위에 분포하면서 신경전달물질로서 기능하며 다양한 스트레스반응을 매개한다. 스트레스를 주변 자율신경계가 활성화되는데, 청색반점에서 기시하는 NE계가 직접 자극되어 카테콜아민을 분비하기도 하지만, CRF나 다른 신경전달계가 먼저 자극되면서 간접적으로 활성화되기도 한다. 특히 CRF와 NE계는 서로 자극시키는 feed-forward 상호작용을 하며, 이것이 생체가 외부환경의 도전에 맞서 내분비계 뿐만 아니라 중추신경계를 동원하는 데 중요한 역할을 할것으로 보인다. 또한 CRF-NE 상호작용은 불안이나 우울 등 비정상적 스트레스반응의 병태생리를 이해하는 데 중요한 역할을 할 것으로 시사된다. 스트레스반응은 구조적, 신경화학적, 유전적 수준의 다양한 신경생물학적 작용을 통해 일어나며, 이에 대한 연구는 스트레스반응의 병태생리를 밝히고 불안장애, 기분장애 등 정신질환의 원인 규명과 치료에도 크게 기여할 것으로 보인다.
수영-스트레스로 나타나는 뇌 catecholamine대사 및 혈장 corticosterone 함량의 변동에 대한 clonidine(500 ug/kg)의 억제작용을 propylthiouracil(0.01% 용액으로 5주간 마시게함) 및 1-thyrorine(4 mg/kg/day로 5일간 복강내에 주사)로 처치한 웅성-마우스에서 실험관찰하여 다음과 같은 성적을 얻었다. 마우스의 일과성 자발운동량의 변동은 갑상선-홀몬의 변동에 영향을 받지 않았고, 수영스트레스(SS)로 나타나는 혈장 corticosterone(CS)의 증가가 propylthiouracil 전처치 (PTU) 및 1-thyroxine 전처치 (T4)로 각각 다소의 감소 및 증강됨을 보였으나, SS에 의한 혈장 CS증가에 대한 clonidine의 억제작용은 PTU 및 T4의 영향을 받지 않았다. SS부하로 뇌 3-methoxy-4-hydroxyphenylglycol 함량(MHPG)가 유의하게 증가되고 clonidine에 의하여는 MHPG가 현저히 감소되었으나 뇌 norepinephrine 함량(NE)은 별 변동을 보이지 않아서 MHPG/NE비는 SS와 clonidine에 의하여 각각 현저한 증가 및 감소를 나타내었다. 아울러, PTU및 T4은 각각 뇌NE을 유의하게 감소 또는 증가시켰으나 뇌 MHPG에는 별 영향을 미치지 않았다. Clonidine은 SS에 의한 뇌 MHPG 및 MHPG/NE비의 증가를 모두 현저히 억제하였으며 그 억제작용은 PTU 및 T4에 의하여 별 영향을 받지 않았다. 이상의 성적으로 미루어서, 마우스의 일과성 자발운동양상 및 스트레스반응으로 나타나는 혈장 corticosterone의 증가현상등이 갑상선-홀몬의 변동에 별 영향을 받지 않으며, 시상하부-뇌하수체-부신계의 활성화가 시상하부의 norepinephrine성 신경-흥분에 매게되어 나타나는 바, 스트레스성 혈장 corticosterone 증가에 대한 clonidine의 억제작용이 그의 절전-${\alpha}_2-adrenoceptor\;agonist$ 작용에 기인되는 것으로 사료된다.
