Introduction of Modifying Solvents to Carbon Dioxide in Supercritical Extractions

  • A simple apparatus for adding a modifying solvent to supercritical CO2 extractant was described. Small, fixed volumes (typically 100 μL) of liquid modifying solvents were delivered during the extraction process by use of an in-line high pressure loop injector and an air pump. Without disconnecting the extraction cell from the supercritical fluid extraction system, the modifying solvent was repeatedly delivered. The solvent modification device was optimized during the extraction of carbaryl and bis(acetylacetonato) copper(Ⅱ). Extraction recoveries from spiked filter paper and soil samples ranged between 22% and 109%, depending on the analyte and matrix components. The addition of polar modifying solvents were necessary to improve the extractability of the nonpolar CO2.

  • In this study, we presented comparative discrimination methods to identify various line and planar defects observed in nonpolar a-GaN epilayers on r-sapphire substrates. Unlike the case of conventional c-GaN, which is dominated by perfect threading dislocations, systematic identification of undistinguishable defects using transmission electron microscopy (TEM) is necessary to suppress the propagation of defects in nonpolar GaN epilayers. Cross-sectional TEM images near the [0001] zone axis revealed that perfect mixed and pure screw type dislocations are visible, while pure edge, partial dislocations, and basal stacking faults (BSFs) are not discernible. In tilted cross-sectional TEM images along the [$1\bar{2}10$] zone axis, the dominant defects were BSFs and partial dislocations for the $g=10\bar{1}0$ and 0002 two-beam images, respectively. From plan view TEM images taken along the [$11\bar{2}0$] axis, it was found that the dominantpartial and perfect dislocations were Frank-Shockley with b=${\pm}1/6$<$20\bar{2}3$> and mixed type without an 1 component including b=${\pm}1/3$<$1\bar{2}10$> and ${\pm}1/3$<$\bar{2}110$>, respectively. Prismatic stacking faults were observed as inclined line contrast near the [0001] zone axis and were visible as band contrast in the two-beam images along the [$1\bar{2}10$] and [$11\bar{2}0$] zone axes.

  • The purpose of this study is to separate nitrocellulose from 175 mm ammunition propellant by environmentally responsible recovery method. Recovery of nitrocellulose from obsolete ammunition is much more desirable than destruction since it can be reused in the several purpose. In spite of durable demilitarization of obsolete ammunition, the current holding amount of obsolete ammunition is gradually increasing and accumulated. The existing demilitarization methods, such as incineration and priming are strictly restricted for the noise, dust, oscillation and air and soil pollution. This study is focused on the separation of nitrocellulose by environmentally responsible recovery method by the solubility difference of nitrocellulose in nonpolar solvent. The purity of an extracted nitrocellulose was analysed by IR and TLC method and was clarified as very high.

THE COMBINATION OF CHEMOMETRICS AND 2D NIR CORRELATION SPECTROSCOPY IN THE ANALYSIS OF DENATURATION PROCESS

  • Despite extensive theoretical and experimental studies the structure of the protein-solvent interface is subject of many controversy. Understanding the processes that occur in aqueous solution requires understanding of the solvent influence on the structure of protein. The aim of this study is to investigate the applicability of NIR methods in the study of hydration phenomena in protein solutions. Temperature-induced changes in NIR spectra of -lactoglobulin (BLG) in aqueous solutions have been investigated by means of two-dimensional correlation spectroscopy (2DCOS) and principal component analysis (PCA). With the temperature increase the balance of forces between the BLG's interaction with itself and the BLGs interaction with its environment is disrupted leading to BLG unfolding. Significant differences of 2D signals and distinct discrepancies of loading on PC1 and PC2 were observed as a result of temperature increase. In the native folded conformation of BLC, most of the nonpolar amino acids are hidden in the centre of the structure, out of contact with water molecules, while charged groups are outside, in the contact with water. The polar groups promote low density Ih-type structure in the water outside this first hydration shell. When BLG unfolds it assumes a more extended configuration on which the previously buried nonpolar groups are exposed to water and promote the higher density II-type structure outside its first shell. Detailed assignments of bands attributed to the bulk water, different states of the hydrated water and the changed conformation of BLG are proposed.

  • Metal distribution in the reaction products and system of reactions between organo chloro acid or ester and zinc, silicon, magnesium, and tin under acetonitrile, dioxane, and toluene solvent were determined by means of radioactive tracer prepared by means of a (n, ${\gamma}$) reaction. It was found that the solubility of the organo halogen metal complex was markedly increased in a hydrophilic solvent and was decreased in a nonpolar solvent which resulted in an increased metal distribution in the recovered metal or water washing of the recovered metal mixture. This was also true in the case of the reaction conducted in the presence of a carbonyl compound. The relative increase of the solubility of the metal complex in a hydrophilic solvent was in order of zinc, silicon, tin and magnesium, and in a nonpolar solvent, it was in order of silicon, tin, magnesium, and zinc. There was no formation of organo metallics throughout the reaction sequence. The result was discussed and the observed solvent influence on the present reaction path was criticized.

Combined Toxic Effects of Polar and Nonpolar Chemicals on Human Hepatocytes (HepG2) Cells by Quantitative Property - Activity Relationship Modeling

  • We determined the toxicity of mixtures of ethyl acetate (EA), isopropyl alcohol (IPA), methyl ethyl ketone (MEK), toluene (TOL) and xylene (XYL) with half-maximal effective concentration ($EC_{50}$) values obtained using human hepatocytes cells. According to these data, quantitative property-activity relationships (QPAR) models were successfully proposed to predict the toxicity of mixtures by multiple linear regressions (MLR). The leave-one-out cross validation method was used to find the best subsets of descriptors in the learning methods. Significant differences in physico-chemical properties such as boiling point (BP), specific gravity (SG), Reid vapor pressure (rVP) and flash point (FP) were observed between the single substances and the mixtures. The $EC_{50}$ of the mixture of EA and IPA was significantly lower than that of contained TOL and XYL. The mixture toxicity was related to the mixing ratio of MEK, TOL and XYL (MLR equation $EC_{50}=3.3081-2.5018{\times}TOL-3.2595{\times}XYL-12.6596{\times}MEK{\times}XYL$), as well as to BP, SG, VP and FP (MLR equation $EC_{50}=1.3424+6.2250{\times}FP-7.1198{\times}SG{\times}FP-0.03013{\times}rVP{\times}FP$). These results suggest that QPAR-based models could accurately predict the toxicity of polar and nonpolar mixtures used in rotogravure printing industries.

Studies on the Production of Lipid by Microorganism (균체 지질 생산에 관한 연구)

  • A potential lipid producing strain of Penicillium sp. was isolated from natural source. Penicillium sp. was cultivated in an appropriate medium containing 6% of glucose as a corbon source, ammonium nitrate as a nitrogen source, C:N ratio 200, pH 4.0 for a period of 17 days at $35^{\circ}C$. Under the condition, the lipid content was 64.2% of dry cell weight. The total lipid produced was 13.7g/100g of glucose consumed. The proportion of nonpolar and polar lipid fractions was 92.2% and 7.8%, respectively. The nonpolar lipid compositions of lipid produced under optimum condition were 5.3% of free fatty acids, 6.8% of free sterols, 9.3% of partial glycerides and 72.0% of triglycerides. The major fatty acids of total lipid were 20.1% of palmitic acid, 21.6% of linoleic acid and 53.3% of oleic acid.

  • For selective purification of protein in Pleurotus cornucopiae (Per.) Rolland, affinity chromatography was performed by benzoyl-AH-Sepharose 4B gel synthesized using AH-Sepharose 4B with starting materials. Ligand capacity of benzoyl group was 9.3 micromole per milliliter of gel. Total apparent molecular weight of affinity protein was 255KD, which were protein complex of 29.5, 31.5 34.0, 71.0 and 89.0KD, respectively. The contents of nonpolar, polar, positively charged, and negatively charged amino acid were 45.68%, 26.93%, 11.81% and 15.58%, respectively. The nonpolar protein was selectively purified by hydrophobic ligand of benzoyl group of gel.

Effects of Mixed Characteristics of Oily Soil on Detergency of PET Fabric in Oily/Particulate Soil Mixed System (지용성/고형오구의 혼합오염계에서 지용성오구의 혼합특성에 따른 PET직물의 세척성)

  • This study investigates the effect of mixed characteristics of oily soil such as mixed ratio, polarity of oily soil on contact angle of fabric, removal of oily and particulate soil from PET fabric in oily/particulate soil mixed system. The contact angle of fabric in the surfactant solution with suspended oily soil was examined as a fundamental environment of detergency of soil from fabrics. Detergency was investigated as function of mixed ratios of oily/ particulate soil, type of oily soil, surfactants concentration, surfactant type and temperature of detergency in surfactant solution. The contact angle of fabric in surfactant solution sharply increased with mixing nonpolar oily soil; in addition, the contact angle slightly increased with increasing contents of oily soil and decreased with increasing surfactant concentration. The removal of oily and particulate soil from fabric was higher in the solution mixed with polar versus nonpolar oily soil. The detergency increased with increasing surfactant concentration and the increased temperature of surfactants solution that were relatively improved in NPE compared to DBS solutions, The results indicated that the detergency of oily and particulate soil showed a similar trend in oily/ particulate mixed soil systems. The general contact angle of fabric was well related with the detergency of oily and particulate soil in oily/particulate mixed soil system, therefore, the primary factor determining the detergency of soil in oily/particulate mixed soil system may be the contact angle of fabric caused by wettability.

Effect of Polar Components on Phase Inversion Temperatures in Systems Containing Nonionic Surfactants and Nonpolar Oils (비이온성 계면활성제, 비극성 오일을 포함한 계에서의 극성 성분의 Phase Inversion Temperature에 대한 영향)

  • Phase Inversion Temperature (PIT) measurements showed that the addition of polar components such as oleyl alcohol and oleic acid to the system comprising n-dodecylpentaoxyethylene monoether ($C_{12}E_5$), nonpolar oil (n-hexadecane) and water produced large reductions in the PIT. The PIT was lowered as the additive-to-surfactant ratio in the surfactant films in the microemulsion phase was increased. Another dramatic effect of additive was the manner in which it affects the volume of the microemulsion phase at the PIT of the oil and water solubilization characteristics. Microemulsion phase volume was increased rapidly with decreasing PIT, i.e., with increasing amounts of additive in the system. Also with a decrease in PIT, the solubilization parameters of both oil and water in the microemulsion phase were strikingly increased. Even though soil removal data were not available for the conditions where our results obtained, PIT measurement seems a useful starting point for estimating conditions when middle-phase microemulsion formation and its associated high solubilization of oil can be expected.
