8q24 rs4242382 Polymorphism is a Risk Factor for Prostate Cancer among Multi-Ethnic Populations: Evidence from Clinical Detection in China and a Meta-analysis

  • Background: Evidence supporting an association between the 8q24 rs4242382-A polymorphism and prostate cancer (PCa) risk has been reported in North American and Europe populations, though data from Asian populations remain limited. We therefore investigated this association by clinical detection in China, and meta-analysis in Asian, Caucasian and African-American populations. Materials and Methods: Blood samples and clinical information were collected from ethnically Chinese men from Northern China with histologically-confirmed PCa (n=335) and from age-matched normal controls (n=347). The 8q24 (rs4242382) gene polymorphism was genotyped by polymerase chain reaction-high-resolution melting analysis. We initially analyzed the associations between the risk allele and PCa and clinical covariates. A meta-analysis was then performed using genotyping data from a total of 1,793 PCa cases and 1,864 controls from our study and previously published studies in American and European populations, to determine the association between PCa and risk genotype. Results: The incidence of the risk allele was higher in PCa cases than controls (0.222 vs 0.140, $P=7.3{\times}10^{-5}$), suggesting that the 8q24 rs4242382-A polymorphism was associated with PCa risk in Chinese men. The genotypes in subjects were in accordance with a dominant genetic model (ORadj=2.03, 95%CI: 1.42-2.91, $Padj=1.1{\times}10^{-4}$). Presence of the risk allele rs4242382-A at 8q24 was also associated with clinical covariates including age at diagnosis ${\geq}65$ years, prostate specific antigen >10 ng/ml, Gleason score <8, tumor stage and aggressive PCa, compared with the non-risk genotype ($P=4.6{\times}10^{-5}-3.0{\times}10^{-2}$). Meta-analysis confirmed the association between 8q24 rs4242382-A polymorphism and PCa risk (OR=1.62, 95%CI: 1.39-1.88, $P=1.0{\times}10^{-5}$) across Asian, Caucasian and African American populations. Conclusions: The replicated data suggest that the 8q24 rs4242382-A variation might be associated with increased PCa susceptibility in Asian, Caucasian and African American populations. These results imply that this polymorphism may be a useful risk biomarker for PCa in multi-ethnic populations.

새만금간척지에서 뚱딴지(Helianthus tuberosus L.) 재배시 염류 특성 변화 및 적정 자원 선발 (The Selection of Proper Resource and Change of Salinity in Helianthus tuberosus L. Cultivated in Saemangeum Reclaimed Tidal Land)

  • 2016년 한국에서 재배 및 유통되고 있는 뚱딴지 12점을 수집하여, 새만금간척지에 재배하였다. 간척지 적응성을 검토한 결과 다음과 같은 결론을 얻을 수 있었다. 1. 뚱딴지를 간척지에 재배하였을 때, 간척지 특성상 0.3% 이상일 때 밭작물 재배가 어려운데, 개화 전 8월 가뭄으로 인하여 토양 염농도가 전체적으로 0.3%이상 올라갔음에도 초장 및 수량으로 보았을 때 높은 염농도에 대한 적응성이 있는 것으로 판단된다. 2. 개화기 무렵 꽃이 50%이상 피었을 때 뚱딴지 초장을 조사한 결과 WY3와 WI1가 207 cm, 202 cm로 가장 컸으며 WG1, WH1, PS1 144 cm, 139 cm, 131 cm로 가장 작았다. 3. 11월 뚱딴지를 수확하여 수량을 확인한 결과 흰색 뚱딴지는 WG1이 549 kg/10 a, WH1이 477 kg/10 a로 가장 많은 수량을 나타내었으며, 자색 뚱딴지에서는 PY4와 PY1이 각각 10 a당 615 kg, 584 kg로 우수한 수량을 보였다. 4. 수집한 뚱딴지 12점의 이눌린 함량을 분석한 결과 PY1이 33.9 g/100 g으로 가장 많은 이눌린 함량을 보였으며, 다음으로 D2가 33.7 g/100 g으로 많았다 결론적으로 내염성이 우수한 뚱딴지가 간척지에서도 적응할 수 있는 것으로 판단되며, 수집 뚱딴지 중 흰색은 수량 및 이눌린함량을 고려하여 G1이 간척지에 적합할 것으로 보이며, 자색은 Y1이 이눌린함량이 두 번째로 많지만, 수량과의 관계를 고려할 때 간척지에서 재배하는데 더 적합한 것으로 판단된다. 각각 색깔별로 우수한 뚱딴지 2점을 가지고 내염성 및 간척지 재배법에 대한 연구가 더욱 필요할 것으로 보인다.

난지도 지역의 대기수은 지화학 (Geochemistry of Total Gaseous Mercury in Nan-Ji-Do, Seoul, Korea)

  • 대규모 면오염원으로 간주되는 난지도지역을 중심으로 2000년 봄 기간중 대기-지표간 수은의 교환율을 측정하고, 측정자료를 기초로 수은의 교환현상을 분석하였다. 측정결과에 의하면 난지도지역은 대규모 면오염원으로 대량의 수은을 배출할 뿐아니라 동시에 대규모의 침적을 경험하는 것으로 나타났다. 일주기별로 농도, 농도구배, 플럭스의 절대치를 분석한 결과 대규모 배출과 침적은 오후시간대에 왕성하게 진행되는 것으로 나타났다. 그러나 이들의 발생빈도에 대한 분석결과는 배출이 새벽 또는 저녁에 빈번하게 발생하는데 비해, 침적은 주로 오후시간대에 집중된다는 것을 확인하였다. 풍향대별로 발생빈도와 교환율을 비교한 바에 따르면, 대규모 침적을 유도하는 외부배출원이 동쪽지역에 위치해 있다는 것을 추정할 수 있었다. 특정 풍향에서 침적이 집중된 점을 감안하여, 침적이 집중된 방향의 자료군과 배출이 지배적으로 발견된 전체자료를 이용하여 조건별 환경인자의 변화동향을 배출/침적비로 환산하여 비교하였다. 그 결과 대부분의 오염인자들의 농도는 외부여건에 따라 규칙적인 변화양상을 보여주었다. 상관분석의 결과는 이들의 거동이 교환현상의 수직적인 방향-침적 또는 배출-의영향을 민감하게 반영하는 것으로 나타났다. 요인분석의 결과는 배출시에 기온과 같은 요인에 의해 유도되는 부분이 그리고 침적시에는 기온, 오존, 비메탄계 탄화수소와 같이 외부인자의 영향을 반영하는 요인들이 복합적으로 중요하게 작용할 수 있다는 점을 시사하였다. 난지도지역을 중심으로 한 수은의 자료를 추정한 결과 연간규모로 약 6kg의 수은이 배출되는 것으로 나타났다.

수원 지역 사과원에서 사과응애와 점박이응애 개체군의 역사적 변천과정 및 해석 (Historical Change of Population Abundances of Panonychus ulmi and Tetranychus urticae (Acari: Tetranychidae) in Selected Apple Orchards in Suwon and Its Hypothetical Explanation)

  • 수원시 이목동에 있는 원예연구소의 사과원에서 조사한 1958년부터 1998년까지 자료를 바탕으로 사과응애와 점박이응애의 역사적 발생변동 과정을 추적하였다. 1970년까지는 사과응애가 우점하였고 1980년 이후에는 점박이응애로 우점종이 바뀌었으며, 그 변화는 1970년 중반을 전후로 나타났다. 이러한 변화를 설명하기 위하여 3가지 가정, 즉 첫째, 두 종의 경쟁에서 점박이응애가 승리한다. 둘째, 과원의 초생은 점박이응애의 수상이동 여부를 결정하는 중요한 요소이다, 즉 초생이 파괴되었을 때 점박이응애의 수상이동이 촉진된다. 셋째, 과원생태계가 교란되지 않을 때 응애류는 천적에 의하여 개체군 밀도가 조절된다 하에 약제살포(고독성 비선택적 약제 처리 및 선택적 저독성 약제처리)와 초생관리(초생재배 및 청경재배=제초제 살포)에 따른 두 종의 장기적 상호작용 결과를 추정하였다. 고독성 농약/초생재배 시스템에서는 점박이응애의 수상이동이 제한되어 사과응애는 경쟁을 피할 수 있고, 또한 천적상이 파괴되기 때문에 전 생육기간 동안 사과응애가 우점한다. 고독성 농약/청경재배 시스템에서는 계절초기부터 점박이응애가 수상으로 이동하여 사과응애를 경쟁적으로 배제시킴으로써 점박이응애가 우점한다. 저독성 농약/초생재배 시스템에서는 점박이응애의 수상이동이 제한되어 사과응애가 우점하지만 천적의 작용으로 높은 밀도를 형성하지 못한다. 저독성 농약/청경재배 시스템에서는 점박이응애가 수상으로 이동하여 사과응애와 경쟁하지만 천적의 작용으로 밀도가 낮아져서 경쟁압력이 낮아지므로 생육후기 사과응애의 발생이 가능하다. 따라서 계절초기 사과응애, 계절후기 점박이응애가 우점하는 형태로 공존할 수 있다. 본 자료에서 제시한 점박이응애 우점화 현상은 부분적으로 고독성 농약/청경재배 시스템 상태에서 두 종의 장기적 상호작용의 결과로 설명될 수 있었다.

금강 지류 백곡천의 어류군집 및 천연기념물 미호종개 Cobitis choii의 서식양상 (Community Structure of Fish and Inhabiting Status of Natural Monument Cobitis choii in the Baekgok Stream, a Tributary of the Geum River Drainage Systrem of Korea)

  • 금강 지류인 충북 진천군 백곡천의 어류군집 및 멸종위기종 미호종개의 서식양상을 밝히기 위하여 2012년 조사를 실시하였다. 상류 (St. 1~3)는 하천형이 계곡형인 Aa 또는 Aa-Bb형이고 하상은 주로 큰돌과 돌로 이루어져 있었으며 10~14종의 어류가 서식하고 있었고, 중류 (St. 4~5)는 중류형인 Bb형 또는 Bb-Bc형으로 하상은 돌과 큰돌, 자갈로 이루어져 있었으며 15~21종의 어류가 서식하고 있었다. St. 6은 백곡저수지로 하상은 대부분 펄이었고 9종이 서식하고 있었으며, St. 7은 백곡저수지 유출지점으로 하천형이 Aa-Bb형이고 하상은 주로 큰돌과 돌로 이루어져 있었으며 7종이 채집되었다. 하류(St. 8~9)는 중류형인 Bb형으로 하상은 주로 돌과 모래로 이루어져 있었으며 20~23종의 어류가 서식하고 있었다. 백곡천에 출현한 종수는 모두 10과 35종이었으며, 우점종은 피라미(34.7%)였고, 아우점종은 돌고기(9.4%)와 돌마자(9.0%)였으며, 그 다음으로 민물검정망둑(8.2%), 모래무지(5.9%), 붕어(4.6%), 참몰개(3.7%), 밀어(3.0%) 순으로 우세하게 출현하였다. 출현종 중 고유종은 10종 (28.6%)이었으며, 외래어종은 떡붕어와 배스 2종이었다. 천연기념물이며 멸종위기종인 미호종개는 St. 5의 유속이 느리고 수심 0.3~1.0 m이며 하상이 모래인 곳에 주로 서식하고 있었다. 이들의 생태적 특징을 조사한 결과 서식개체수는 $7,838{\pm}6,290$개체가, 연령은 당년생 전장 38~45 mm, 1년생 46~60 mm, 2년생 61~80 mm, 3년생 이상은 81~93 mm로 추정되었고, 비만도지수 (${\times}10^5$)는 암컷 0.36~0.39, 수컷 0.35~0.38이었으며, 기생충의 일종인 이생충(digenea)이 13.5%가 감염되어 있었다. 그 밖에 백곡천 미호종개의 건강성과 위협요인, 보전에 대해 논의하였다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N.
