Inter-laboratory Comparison of Stable Carbon and Nitrogen Isotopic Composition Data Using Elemental Analyzer-isotope Ratio Mass Spectrometers

  • In this study, inter-laboratory comparison was done using elemental analyzer-isotope ratio mass spectrometers (EA-IRMSs) to determine carbon and nitrogen contents as well as stable carbon and nitrogen isotopic compositions (${\delta}^{13}C$ and ${\delta}^{15}N$) of five environmental samples containing lake and marine sediments, higher plant leaves, and fish muscle, and one organic analytical standard (Protein (Casein) Standard OAS). Five national laboratories participated in this comparison study, and each laboratory analyzed all five samples and the analytical standard. Results showed that variations in total organic carbon (TOC) and total nitrogen (TN) contents as well as ${\delta}^{13}C_{TOC}$ and ${\delta}^{15}N_{TN}$ values among the laboratories were large compared to the analytical uncertainties. The results highlighted the inhomogeneity of the test samples and thus, the need to select suitable standard reference materials for future inter-laboratory studies. Further inter-laboratory comparison exercises could promote good measurement practices in the acquisition of stable carbon and nitrogen isotopic composition data.

Geochemical Characteristics and Assesment of Nitrate Nitrogen in Groundwater in Yanggu-Gun, Gangwon-Do in Korea

  • An analysis of groundwater quality is significant for monitoring and managing water contamination and groundwater system. For the purpose of those, the geochemical characteristics of groundwater were studied over the concern for water quality, water type and origin of nitrate nitrogen. Total colony counts were detected in 11 out of 20 samples, and the average value was 31.73 CFU/ml. Range and average of NO3-N concentrations were 0.9~24.0 mg/L and 8.3 mg/L. All groundwater types were found to be Ca2+-HCO3-. The range and average of NO3-N were 0.2~17.4 mg/L and 8.7 mg/L, and those of δ15N were 1.7~8.9‰, and 5.0‰. Careful consideration is required for evaluating the origin of nitrogen when NO3-N concentration is low. In general, noticeable difference between rockbed and alluvial water was not found. The ranges of nitrate origins by chemical fertilizer, livestock manure and domestic sewage, and natural soil were 29.6~76.4%, 14.2~58.9% and 2.6~7.0%, and the average values of those were 57.4%, 37.4%, and 5.3%, respectively. Origin of nitrate was affected by more chemical fertilizer than the other parameters. Rockbed water was more affected by chemical fertilizer than alluvial water.

TDS Removal using Bio-sorption with AGS and High Concentration Nitrogen Removal (AGS의 생물흡착을 이용한 TDS 제거 및 고농도 질소제거에 관한 연구)

  • This study aimed to assay the biological removal of TDS (total dissolved solids) from RO (reverse osmosis) rejected water. Following bio-sorption of TDS with AGS (aerobic granular sludge), the effects of TDS on biological nitrogen removal were examined. The bio-sorption of TDS after AGS treatment was confirmed by checking for TDS removal efficiency and surface analysis of microorganisms with SEM and EDS. Then, the effects of TDS on biological nitrogen removal and the denitrification efficiency were evaluated using the MBR reactor. According to the results, the bio-sorption of TDS with AGS was 0.1 mg TDS/mg AGS, and we confirmed that the microorganism surfaces had adsorbed the TDS. Biological nitrogen removal efficiency was measured at inhibiting denitrification at 4,000 mg/L of TDS-injected material. Based on this study, it is necessary to pretreat TDS-containing RO rejected water and to maintain TDS concentration lower than a specific value (≤4,000 mg/L), when considering biological nitrogen removal.

The Effect of Serum Chemical Values in Recipients on Pregnancy Rate following Embryo Transfer (수란우의 혈청화학치의 농도가 수정란이식 후 수태율에 미치는 영향)

  • This study was done to examine the relationship between serum chemical values (urea nitrogen, glucose, total protein and cholesterol) of recipients and pregnancy rate following embryo transfer. Blood samples were taken from 184 Holstein heifers or cows on Day 6 or 7 (Day 0=day of estrus) to analysis for serum urea nitrogen, glucose, total protein and cholesterol concentrations. After selection of recipients, frozen Holstein embryos were thawed and directly transferred to recipients non-surgically. The average serum concentrations of urea nitrogen, glucose, total protein and cholesterol were 13.8 mg/dl, 56.5 mg/dl, 7.2 mg/dl, 124.8 mg/dl, respectively. The average concentrations of serum urea nitrogen and cholesterol were lower (P<0.05) in pregnant recipients (10.7 mg/dl, 99.2 mg/dl) than in non-pregnant recipients (13.0 mg/dl, 122.2 mg/dl), respectively, although the concentrations of glucose and total protein were not different. These results show serum urea nitrogen and cholesterol concentrations can be used important factors for selection of recipients in Holstein.

  • A biuret method by Johnson and Craney (1971) was slightly modified and applied to the multiple analysis of nitrogen in brown rice. The results were compared with those by the dye-binding method to see if the former be applicable to the determination of higher nitrogen content in rice. The nitrogen content of rice samples ranged from $1.2{\sim}2.0%$. The correlation between biuret absorbance and nitrogen content was highly significant; its correlation coefficient being 0.841. The biuret method, however, showed rather lower correlation coefficient in case of high nitrogen samples.

  • Nitrogen (N) loading from domestic, agricultural and industrial sources can lead to excessive growth of macrophytes or phytoplankton in aquatic environment. Many studies have used nitrogen stable isotope ratios to identify anthropogenic nitrogen in aquatic systems as a useful method for studying nitrogen cycle. In this study to evaluate the precision and accuracy of Kjeldahl processes, two reference materials (IAEA-NO-3, N-1) were analyzed repeatedly. Measured the ${\delta}^{15}N-NO_3$ and ${\delta}^{15}N-NH_4$ values of IAEA-NO-3 and IAEA-N-1 were $4.7{\pm}0.2$‰ and $0.4{\pm}0.3$‰, respectively, which are within recommended values of analytical uncertainties. Also, we investigated spatial patterns of ${\delta}^{15}N-NO_3$ and ${\delta}^{15}N-NH_4$ in effluent plumes from a waste water treatment plant in Han River, Korea. ${\delta}^{15}N-NO_3$ and ${\delta}^{15}N-NH_4$ values are enriched at downstream areas of water treatment plant suggesting that dissolved nitrogen in effluent plumes should be one of the main N sources in those areas. The current study clarifies the reliability of Kjeldahl analytical method and the usefulness of stable isotopic techniques to trace the contamination source of dissolved nitrogen such as nitrate and ammonia.

Long-term Variations of Water Quality Parameters in Lake Kyoungpo (경포호에서 수질변수들의 장기적인 변화)

  • In order to identify long-term trends of water quality parameters in Lake Kyeongpo, Mann-Kendall test, Sen's slope estimator and linear regression were applied on data, with 15 parameters from three different sites and rainfall, monitored once in every two months from March to November during 1998~2013. Seasonal variation analysis only used Mann-Kendall test and Sen's slope estimator. Analysis result showed that salinity, transparency and nutrient variables (total phosphorus, dissolved inorganic phosphorus, total nitrogen, nitrate nitrogen, ammonia nitrogen) were only parameters having statistically significant trend. In linear regression analysis, salinity (surface and bottom layer of all sites) and transparency (only at site 1), were figured out with statistically significant increasing trend, while in non-parametric statistical method, salinity and transparency in all sites (surface, middle, deep) were figured out with statistically significant increasing trend. Water quality parameters showing statistically significant decreasing trends were dissolved oxygen (surface layer of site 1 and bottom layer of sites 2 and 3), total phosphorus (sites 1 and 2), dissolved inorganic phosphorus, total nitrogen, nitrate nitrogen and ammonia nitrogen in the linear regression analysis and, dissolved oxygen (bottom layer of all sites), total phosphorus, dissolved inorganic phosphorus, total nitrogen, nitrate nitrogen and ammonia nitrogen in the non-parametric method. Seasonal trend analysis result showed that salinity, turbidity, transparency and suspended solids in spring, salinity, transparency, nitrate nitrogen and suspended solids in summer and temperature, salinity, transparency and suspended solids in fall were the variables depending on the season with increasing trends. In general, rainfall during the research period showed decreasing trend. The significant reduction trends of nutrients in Lake Kyeongpo were believed to be related to lagoon restoration and water management project run by Gangneung city and under-water wear removal, but further detailed studies are needed to know the exact causes.

Assessment of Water Quality in the Miho Stream Using Multivariate Statistics (다변량 통계기법을 이용한 미호천 본류 수질특성 평가)

  • In The study, is to investigate the spatial characteristics of the Miho stream, which is the main tributary of the Geum River system, and to identify the main factors influencing the water quality using water quality analysis and multivariate analysis. The survey subjects were selected as 7 main sites in the Miho stream water system, From 2012 to 2017, 16 items including weather temperature and weather data were used for multivariate analysis. As a result of the water quality analysis, the average concentration of BOD and COD for 6 years was 3grade (normal) compared with the water quality environmental standard (river) of conditions. The concentrations of nitrogen and phosphorus were highest at th upstream site, then decreased and then increased again by the hydrogeological and geomorphological effect. Cluster analysis of spatial and water quality characteristics, it was evaluated as three clusters and the pollution sources is the greatest impact. As a result of principal component analysis and factor analysis on each cluster and mainstream, three to four major components were extracted. Main stream and the Cluster 1, Cluster 3 first principal factor included nitrogen and seasonal factors,first factor of Cluster 2 included nitrogen and water temperature. Nitrogen is the principal factor which affects water quality in Miho stream.

  • Nowadays, It is required human body-friendly, good mechanical properties, and economical efficiency material, simultaneously. The material to meet above requirement condition rear up high nitrogen stainless steel(HNS). However, HNS have a lot of problem such as poor workability, hot crack sensitivity. So, It is needed the condition of plastic working to overcome above many problem. In this study, VIM ingot with 100kg was made by pressurized vacuum induction melting. And then, The slab perform for hot rolling was prepared by open-die forging. Hot rolling process was performed by computer simulation according to change of height reduction, rolling temperature, heating numbers, rolling pass and so forth. The results of analysis were investigated between analysis and lab-scale rolling product.

Correlation Analysis of the Dielectric Breakdown Voltage of Liquid Nitrogen (액체질소 절연파괴전압의 상관 분석)

  • We analyzed the correlation between breakdown voltage(BDV) of liquid nitrogen(LN2) and factors. The chosen factors affecting the breakdown are the diameter of electrode, gap length, temperature of LN2, and pressure of LN2. The BDV of LN2 was increased with increasing the diameter, the gap length and the pressure. And The BDV of LN2 was increased with decreasing the temperature. However, correlation coefficient was different from each other depending on the situation. The BDV exhibited a very high correlation coefficient of 0.92227 to dependence on the diameter. And a very high correlation coefficient of 0.94980 to dependence on the pressure under sphere(D 7.5 mm)-plane electrode. When the pressure is applied, sphere-plane electrode is the correlation coefficient was higher than that of the needle-plane electrode. It shows the dependence of a temperature coefficient of -0.758290 ~ -0.39946 under needle-plane electrode.
