Antioxidative Activities of Different Part Extracts of Physalis alkekengi var. francheti (Winter Cherry) (부위별 꽈리(Physalis alkekengi var. francheti) 추출물의 항산화효과)

  • The total phenolic contents and the antioxidant activities of methanolic extracts of different parts (fruit, calyx, leaf, stem, and root) of Physalis alkekengi var. francheti were investigated using established in vitro systems including DPPH radical-scavenging, nitrite-scavenging, superoxide anion radical-scavenging activity, measurement of reducing power, and assessment of the metal-chelating effect. The highest extraction yield was from fruit (52.55%), whereas the lowest levels were obtained from root (10.49%) and stem (12.88%).The leaf extract had the highest total phenolic (58.47 mg/g) and total flavonoid (4.83 mg/g) contents, plus the greatest antioxidant activity, as shown by the DPPH radical-scavenging assay, and the highest levels of reducing power at concentrations of 1 and 5 mg/mL. In addition, the calyx also showed good antioxidant activity.These findings indicate that methanolic extracts of leaf and calyx may be useful in the food manufacturing and nutraceutical industries.

Effect of Ethanol Extracts in Pinus densiflora, Lithospermum erythrorhizon on the Lipid Oxidation of Oil Emulsion (식물체(솔잎, 자초)의 에탄올 추출물이 유탁액의 지방산화에 미치는 영향)

  • This study was carried out to investigate the effects of ethanol extracts on lipid oxidation of oil emulsion. The results are as follows; The scavenging ability of plant extracts for hydroxyl radical was found, and plant extracts played an important role as a strong chelating agents to bind iron if Fe2+ ion exists in oil emulsion. Pinus densiflora(PD), Lithospermum erythrorhizon(LE) and PD+LE acted as strong chelating agents to bind iron to reduce lipid oxidation in oil emulsion. The content of Fe2+ ion in ethanol extracts from LE and PD+LE were significantly higher(p<0.05) than that of ethanol extracts from PD. The content of total iron has same tendency. The ascorbic acid content of PD(16.36ppm) was slightly higher than those of LE(13.08ppm). Electron donating ability of PD was significantly higher(p<0.05) than those of LE. However, the superoxide(SOD) like ability of LE showed a little higher than those of LE and PD+LE, which means the strong antioxidant activity of LE. The nitrite scavenging effects were dependent on pH value, however, they decreased as pH value increased. Especially, they almost didn't show the nitrite scavenging effect in pH 6.0. In conclusion, the PD and LE extracts may be used as natural antioxidant sources to reduce lipid oxidation in oil emulsion.

Physiological and Functional Properties of Salicornia herbacea (Tungtungmadi) Leaf Extracts

  • The physiologically relevant functional properties of various solvent extracts from Salicornia herbacea leaves were investigated by measuring lipid peroxidation, DPPH radical scavenging, nitrite scavenging, and xanthine oxidase inhibition. Ethyl ether, chloroform, ethyl acetate and n-butanol fractions obtained from the 80% aqueous ethanol extracts of Salicornia herbacea leaves showed strong antioxidative activities in linoleic acid methyl esters. Peroxide values (POV) were not significantly different among the samples treated with the different fractions; the incubation time required to reach a peroxide value of 80 meq/kg was about 40 hrs. However, control linoleic acid methyl esters had POV of more than 480 meq/kg after 40 hrs. The DPPH radical scavenging activity of the ethyl acetate fraction was much more effective than diethyl ether, n-butanol, chloroform and water fractions, with an $IC_{50}$/ of 279 $\mu\textrm{g}$/mL, but less effective than ascorbic acid ($IC_{50}$/ : 67 $\mu\textrm{g}$/mL). The nitrite scavenging activities of all fractions increased as pH decreased. Among the fractions, nitrite scavenging activities of diethyl ether and ethyl acetate fractions at pH 1.2 were highest at 59.0 and 56.2%, respectively. The diethyl ether fraction obtained from the 80% aqueous ethanol extract of Salicornia herbacea loaves was the most effective inhibitor of xanthine oxidase of all the solvent extracts at 84% inhibition for a 1 mg/mL concentration. These results suggest that Salicornia herbacea leaf extracts may be effective antioxidants, not only in food stability, but also in human health.

  • The natural sources extracted from crab shell Korean tagnrine peel, sesame meals were utilized to investigate the effects of extracts on free radical reaction, lipid oxidation and nitrite scavenging ability. The recovery percentage of extracts from waste resources (crab shell, sesame meal, dry korean tangrine peel) were chitosan 11.6%, crude sesamol 2.2% and ascorbic acid 2.8%, respectively. The antioxidants were tended to have a lower TBARS value than those of control. The nitrite scavenging and electron donating ability of crude sesamol were tended to be the most effective extract among all extracts. However, In case of chitosan, the superoxide dismutase (SOD)-like activity was the most highest, compared to other extracts.

Antioxidant and Antimicrobial Properties of Various Solvent Extracts from Robus idaeus (추출 용매에 따른 산딸기 추출물의 항산화 및 항균 활성)

  • The objective of the present study was to determine the total phenol and flavonoid contents and to evaluate the antioxidant potential, of different solvent extracts (ethyl acetate, n-butanol, chloroform, and water) from Robus idaeus in various radical scavenging models (DPPH activity, superoxide dismutase (SOD) activity, reducing power, and nitrite scavenging activity), along with their antimicrobial potential. Measurement of total phenol and flavonoid content of the ethyl acetate extract of R. idaeus was found to be significantly higher than those of the other extracts. The ethyl acetate extract (at a concentration of $1,000{\mu}g/mL$) showed significantly higher reducing power and DPPH radical scavenging activity as compared to the other extracts. Results were dose-dependent. Moreover, the ethyl acetate extract of R. idaeus ($1,000{\mu}g/mL$) showed potent antioxidant efficacy ($85.5{\pm}1.18%$) as evidenced by nitrite scavenging ability at pH 1.2. All solvent extracts of R. idaeus showed lower SOD-like activity (13.72~20.54%). In addition, the antimicrobial activity of all solvent extracts except water extract showed strong inhibition (inhibitory zones in mm) of Staphylococcus aureus ($19.40{\pm}1.00mm$) and Bacillus cereus ($20.50{\pm}0.21mm$) growth. In particular, ethyl acetate extracts (100 mg/mL) showed antimicrobial activity comparable to that of tetracycline (0.01 mg/mL), which was used as a positive control. The results of this study indicate that the ethyl acetate extract of R. idaeus is a natural antioxidant and antimicrobial, with enriched phenols and flavonoids concentration, that has potential in the development of health-enhancing food products.

Comparison of the Antioxidant Activities and Nitrite Scavenging Activity of Black Chokeberry (Aronia melanocarpa) Extracts (블랙 초크베리 추출물의 항산화 활성 및 아질산염 소거 활성)

  • The purpose of this study was to measure the antioxidant activity and bioactivity of black chokeberry (Aronia melanocarpa). The black chokeberry was known to contain many physiologically active substance, such as ascorbic acid, anthocyanin, quercetin. We extracted black chokeberry powder from four solvents such as chloroform:methanol(CM, 2:1, v/v), distilled water (DW), 70% ethanol, 70% methanol. After that, we determined anthocyanin, total phenol, flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power, reducing power and Nitrite scavenging activity of black chokeberry extracts and as results of comparing each extract. respectively. From the above results shows that antioxidant activity and bioactivity of black chokeberry extracts was the highest in 70% methanol (p<0.05). The results suggest that black chokeberry can be used as nutraceutical foods and natural antioxidant.

Antioxidant Activities of Extracts of Solanum nigrum L. (용규 추출물의 항산화 활성 탐색)

  • In this study, the antioxidant activities of fractions (methanol, hexane, methylene chloride, butanol, ethyl acetate and water) of Solanum nigrum L. extract was investigated. The contents of total phenolic compounds of each fractions of methanol, butanol, methylene chloride, ethyl acetate, hexane and water are $4.41{\pm}0.23%$, $5.57{\pm}0.35%$, $9.89{\pm}0.19%$, $9.86{\pm}0.19%$, $1.89{\pm}0.04%$, and $3.18{\pm}0.06%$, respectively. For assay of antioxidant activity, 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity, reducing power and nitrite scavenging activity are evaluated. In DPPH radical scavenging activity, the highest effect was obtained from the fraction of ethyl acetate. Reducing power is ordered as ethyl acetate > methylene chloride > methanol. In nitrite scavenging activity, the highest activity was 5.5% (butanol fraction), whereas hexane fraction did not detected. Overall, antioxidant activities are closely related the content of phenolic compound in extracts of S. nigrum L.

  • This study was performed to investigate the effect of types of microorganisms on the antioxidant activity of soybean yogurt by a single or mixed culture of Bacillus subtilis and Lactobacillus plantarum isolated from chunggukjang and kimchi. The fermented soybean milk by Bacillus subtilis exhibited the highest values of total polyphenol, DPPH radical scavenging activity, reducing power and nitrite scavenging activity compared to those of Lactobacillus plantarum or mixed culture of Bacillus subtilis/Lactobacillus plantarum. As the amount of tomato extract was added to the soybean milk, various antioxidant parameters, such as total polyphenol, DPPH radical scavenging activity, reducing power and nitrite scavenging activity, were linearly increased.

Antioxidant Activity of Black Panax ginseng (흑삼의 항산화 활성)

  • This study was performed to investigate the antioxidant activity of extracts of black Panax ginseng (BGE) and its crude saponin (BGECS). The antioxidant activities of BGE and BGECS were evaluated for free radical scavenging activity against stable free radical (1,1-diphenyl-2-picrylhydrazyl) DPPH, nitrite, hydrogen peroxide and superoxide. In addition, the antioxidant activity of BGE and BGECS against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. As a result, BGE and BGECS were found to have a strong inhibitory activity with >90% against the DPPH radical at $1000{\mu}g/m{\ell}$ concentrations. Also, BGE and BGECS exhibited strong inhibitory activity with >80% against hydrogen peroxide at lower concentration ($125{\mu}g/m{\ell}$). Moreover, specific TOSC values (405 and 473 TOSC/mM) of BGE and BGECS against peroxynitrites were higher than GSH (347 TOSC/mM) used a positive control. These results suggest that BGE and BGECS could be useful to develop functional foods against disease related oxidative stress.

Effects of Harvesting Time, Aging Period and Extracting Temperature of Wild Green Tea (Camell sinesis) Leaves on Physiological Activity of Don Tea (찻잎의 수확시기, 돈차의 숙성기간 및 추출온도가 돈차의 생리활성에 미치는 영향)

  • This study was conducted to determine the potent physiological activity of traditional wild tea ("Don tea"; coin-shaped tea) as affected by different harvesting times, aging periods and extracting temperatures, No difference in anti-oxidative activities in the harvesting time and extracting temperature of tea leaves was observed. However, short aging periods of Don tea showed high ABTS {2-2-azonobis (3-ethylbenzothiazolin-6-sulphonic acid)} activity, ranging from 71.52 to 79.96%. DPPH (${\alpha},\;{\alpha}-diphenyl-{\beta}-picryl-hydrazyl$) radical scavenging activity of Don tea was 71.10 to 91.40%. Especially, longer aging period and an extracting temperature of $100^{\circ}C$ showed higher DPPH radical scavenging activity. With longer aging periods and an extraction at $90^{\circ}C$, nitrite radical scavenging activity of Don tea ranged from 74.04 to 94.92%. On the other hand, angiotensin 1-converting enzyme (ACE) inhibition activity of Don tea was 59.77-81.97%. It showed higher activity when harvested in June and August, aged for longer periods, and extracted at $100^{\circ}C$. These results suggested Korean traditional Don tea exhibited the highest physiological activity when aged over 8 months.
