Phellinus pini (Hymenocaetaceae) has been used for the immunomodulating activity hypolipidemic effect, gastric cancer non-insulin dependant diabetes, diarrhea, and menstrual irregularity. From the screening of each fraction for the inhibitory activity of NO production in lipopolysaccaride (LPS) activated RAW 264.7 cells, methanol extract of Phellinus pini and hexane soluble fraction exhibited inhibition of NO production compared with LPS control without toxicity. The hexane soluble fraction showed dose dependent inhibition of NO production. According to activity guided fractionation, the active hexane fr. was repeatedly chromatographed over silica gel, ergosta-4,6,8(14),22- tetraen-3-one was isolated. The compound inhibited NOS activation (IC$_{50}$ = 29.7 uM) and NO production of activated macrophage at 30 uM.
본 연구는 바지락이 흔들림 스트레스에 노출될 경우 나타나는 생리적 변화를 측정하기 위하여 일산화질소량 (NO) 을 측정하였다. 이를 위하여 흔들림 방지 장치가 없는 그룹, NAME (NO 저해제) 를 주사한 그룹, 나일론 섬유 충전재(밀도 $1kg/m^3$) 로 흔들림이 방지된 그룹 등 총 3개 그룹으로 나눈 후 교반기에서 100 rpm으로 6시간동안 교반 한 후 DAF assay와 Griess assay를 이용하여 바지락 혈림프액의 NO 농도를 측정하였다. 조사결과 흔들림 스트레스에 노출된 바지락에서 NO 농도가 급격히 증가하였고 반면 NO 저해제가 주입된 바지락에서는 NO 농도가 감소하였다. 또한 흔들림 방지 장치가 들어간 그룹에서 NO 농도가 감소함이 확인되었다. 이러한 결과는 DAF assay와 Griess assay 실험에서 모두 동일하게 나타났다. 결론적으로 NO 측정은 바지락의 생리적 스트레스를 측정하는데 유용한 방법임이 확인되었으며, 형망에 의한 바지락 채취시나 수하식으로 양식할 경우 흔들림에 의한 스트레스를 방지할 수단이 필요함을 시사하였다.
Sprague-Dawley(SD) rats were orally administered with $N^G$-nitro-L-arginine methyl ester(L-NAME) which inhibits or blocks the production of nitric oxide from L-arginine in vascular endothelial cells and vessel tissue to statistically examine the effects of nitric oxide on some physiological changes such as blood pressure and heart rate, and to confirm the apoptosis induced by the suppressed nitric oxide activity in some related organs under light microscope. Systolic blood pressure significantly increased 28.5% by the chronic treatment of L-NAME for 8 weeks (P<0.001), no significant difference, however, was observed in heart rate between the control group and the L-NAME-treated group regardless of their age. Hematoxylin-eosin staining showed some histological alterations only in kidney among the examined organs; heart, liver, pancreas, and adrenal gland from the L-NAME-treated group. TUNEL (terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling) test showed a strong positive reaction, representing that the chronic treatment of L-NAME facilitates apoptosis, in the cortex and medulla of kidney, but not any significance detectable in the other organs. These results conclude that chronic treatment of L-NAME significantly increases blood pressure, and that the followed inhibition of nitric oxide synthesis occurs a typical inducement of apoptosis in kidney.
The effect of water extract from Thesium chinese Turczaninow (TCTW) on proliferation of human breast cancer cells, nitric oxide production, nitric oxide synthase expression, and ornithine decarboxylase activity was tested. TCTW inhibited the growth of both estrogen-independent MDA-MB-231 and estrogen-dependent MCF-7 human breast cancer cells. Lipopolysaccharide-induced nitric oxide (NO) production was significantly reduced by TCTW at the concentrations of 1.0 (p<0.05) and 5.0 mg/ml (p<0.005). Expression of inducible nitric oxide synthase (iNOS) was also suppressed with the treatment of TCTW in Western blot analysis. TCTW inhibited induction of ornithine decarboxylase by 12-O-tetradecanoylphorbol-13-acetate (TPA), a key enzyme of polyamine biosynthesis, which is enhanced in tumor promotion. Therefore, TCTW is worth further investigation with respect to breast cancer chernoprevention or therapy.
Background: The role of nitric oxide(NO) in analgesia from opioids is controversial. On the one hand, IV morphine analgesia is enhanced by IV injection of NO synthase inhibitors. On the other hand, IV morphine results in increased release of NO in the spinal cord. There have been no behavioral studies examining the interaction between IV morphine and intrathecal injection of drugs which affect NO synthesis. Method: Rats were prepared with chronic lumbar intrathecal catheters and were tested withdrawal latency on the hot plate after 3~5 days of surgery. Antinociception was determinined in response to a heat stimulus to the hind paw before and after IV injection of morphine, 2.5 mg/kg. Twenty minutes after morphine injection, rats received intrathecal injection of saline or the NO synthase inhibitors, L-NMMA or TRIM, the NO scavenger, PTIO, or the NO synthase substrate, L-Arginine. Intrathecal injections, separated by 15 min, were made in each rats and measurements were obtained every 5 min. Result: Mophine produced a 60~70% maximal antinociceptive response to a heat stimulus in all animals for 60 min in control experiments. Intrathecal injection of idazoxane decreased antinociception of IV morphine. The NO synthase inhibitors and the NO scavenger produced dose-dependent decreases in antinociceptive effect of morphine, whereas saline as a control group and L-Arginine as the NO substrate had no effect on antinociception of morphine. Conclusion: The present study supports the evidences that systemic morphine increase the nitrite in cerebrospinal fluid and dorsal horn. These data suggest that the synthesis of NO in the spinal cord may be important to the analgesic effect of IV morphine and increased NO in spinal cord has different action from the supraspinal NO.
신성고혈압백서모델인 one-kidney, one-clip Goldblatt 고혈압백서에서 혈압과 NO생성에 미치는 고려홍삼 사포닌과 비사포닌의 영향을 규명하고자, 무마취 무제한상태에서 혈압을 측정하고, 혈중 NO와 NO합성효노에 대한 활성 및 단백량을 실시한 결과이다. 고려홍삼 사포닌은 정상혈압백서 및 신성고혈압백서에서 모두 혈압을 강하시키는 효과가 현저하였는데 혈압강하는 초기 2-4분 동안 최대를 보였으며 혈압강하와 동시에 심박수의 증가 현상이 관찰되었다. 그러나 고려홍삼 비사포닌은 혈압강하효과와 심박수의 증가현상은 사포닌에 비하여 미미 하였다. 고려홍삼 사포닌에 의한 혈압강하 현상과 NO와의 상관관계를 규명하기 위하여 혈중 NO를 측정한 결과 홍삼사포닌은 혈중 NO를 상승시켰다. NO합성효소의 활성도를 측정하기 위하여 $[^{3}H]-L-arginine$의 $[^{3}H]-L-cioulline$으로의 변환률을 측정한 결과 고려홍삼 사포닌에 의하여 NO합성효소의 활성도는 증가하였다. 이상의 결과를 종합해 볼 때 고려홍삼은 혈압을 강하시키는 효과가 있으며, 이러한 혈압강하는 주로 사포닌 성분에 의한 것으로 생각된다. 이러한 고려홍삼 혈압강하 현상은 혈중의 NO량을 증가시키는 것에 기인하며 NO의 증가는 NO합성효소의 활성도의 증가에 기인하는것으로 사료된다.
Recent investigations have shown that certain flavonoids, especially flavone derivatives, inhibit nitric oxide (NO) production by inducible NO synthase (iNOS) in macrophages, which contrib-ute their anti-inflammatory action. For the purpose of finding the optimized chemical structures of flavonoids that inhibit NO production, various A- and B-ring substituted flavones were syn-thesized and evaluated for their inhibitory activity using lipopolysaccharide-treated RAW 264.7 cells. It was found that the optimal chemical structures were A-ring 5,7-dihydroxyflavones hav-ing the B-ring 2',3'-dihydroxy or 3',4'-dihydroxy or 3',4'-hydroxy/methoxy (methoxy/hydroxy) groups. These structurally optimized compounds were revealed to be down-regulators of iNOS induction, but not direct iNOS inhibitors. Of these derivatives that were evaluated, 2',3',5,7-tet-rahydroxyflavone and 3',4',5,7-tetrahydroxyflavone (Iuteolin) showed the strongest inhibition. The $IC_{50}$/ values for these compounds were 19.7 and 17.1 11M, respectively. Therefore, these compounds may have a potential as new anti-inflammatory agents.
To search for immunoactive natural products exerting anti-inflammatory activity, we have evaluated the effects of the ethanol extracts of Rubus coreanus Miq. (ERC) on lipopolysaccharide-induced nitric oxide (NO), tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$, and $Interferon-{\gamma}\;(IFN-{\gamma})$ production by RAW 264.7 macrophage cell line. Our data indicate that this extract is a potent inhibitor of NO production and it also significantly decreased $IFN-{\gamma}\;and\;TNF-{\alpha}$ production. Consistent with these results, the protein level of inducible Nitric Oxide Synthase (iNOS) and cyclooxygenase-2 (COX-2) was inhibited by ethanol extracts of ERC in a dose-dependent manner. These results suggest that ERC may exert anti-inflammatory and analgesic effects possibly by suppressing the inducible NO synthase and COX-2 expressions.

Microglia are the major inflammatory cells in the central nervous system and become activated in response to brain injuries such as ischemia, trauma, and neurodegenerative diseases including Alzheimer's disease (AD). Moreover, activated microglia are known to release a variety of proinflammatory cytokines and oxidants such as nitric oxide (NO). Minocycline is a semi-synthetic second-generation tetracycline that exerts anti-inflammatory effects that are completely distinct form its antimicrobial action. In this study, the inhibitory effects of minocycline on NO and prostaglandin E$_2$ (PGE$_2$) release was examined in lipopolysaccharides (LPS)-challenged BV2 murine microglial cells. Further, effects of minocycline on inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression levels were also determined. The results showed that minocycline significantly inhibited NO and PGE$_2$ production and iNOS and COX-2 expression in BV2 microglial cells. These findings suggest that minocycline should be evaluated as potential therapeutic agent for various pathological conditions due to the excessive activation of microglia.
The present study was conducted to evaluate the effect of Yongdamsagantang (YST) on the regulatory mechanism of cytokines and nitric oxide (NO) for the immunological activities in RAW 264.7 cells. After the treatment of YST water extract, cell viability was measured by MTT assay, and NO production was monitored by measuring the nitrite content in culture medium. Inducible nitric oxide synthase (iNOS) and phospholylation of inhibitor of nuclear factor kappa B alpha ($p-I{\kappa}B{\alpha}$) were determined by Immunoblot analysis, and levels of cytokine were analyzed by sandwich immunoassays. Results provided evidences that YST inhibited the production of NO. iNOS, and interleukin-6, and the activation of $p-I{\kappa}B{\alpha}$ in RAW 264.7 cells activated with lipopolysaccharide. These findings showed that YST could have some anti-inflammatory effects which might play a role in therapy in Gram-negative bacterial infections.
