담배 (Nicotiana tabacum L.) 현탁배양세포의 Elicitor 유도성 5-epi-Aristolochene Hydroxylase (Elicitor-Inducible 5-epi-Aristolochene Hydroxylase in Suspension Cultures of Tobacco (Nicotiana tabacum L.))

  • [$^3$H] 5-epi-aristolochene (5-EAS)를 담배 현탁배양세포에 투여하여 elicitor에 의해 유도된 세포가 생합성하여 배지로 방출하는 [$^3$H]-capsidiol의 량을 측정함으로써 5-EAS hydroxylase의 활성을 검정하였고, 이 반응의 전 단계에 관여하는 효소인 sesquiterpene cyclase의 발현특성과 비교하였다. 5-EAS hydroxylase는 정상세포에는 전혀 활성을 보이지 않으나 elicitor로써 cellulase를 처리한 세포는 9시간 후부터 유도를 시작하여 18시간 후에 최대 활성을 보였는데 동일한 세포내에서 유도되는 cyclase와 유사한 패턴을 보였다. Cyt P450계 효소의 특이적 억제제로 알려진 ancymidol과 ketoconazole에 의해 5-EAS hydroxylase의 활성은 강한 억제를 보인 반면 cyclase의 활성은 억제를 보이지 않아 5-EAS hydroxylase가 P450계 효소임이 시사되었다.

연초의 세균성마름병 저항성 유전에 관한 연구 (Studies on the Inheritance of Resistance to Bacterial Wilt(Ralstonia solanacearum) in Tobacco(Nicotiana tabacum L.))

  • Bacterial wilt(Ralstonia solanacearum) is one of the major diseases of flue-cured tobacco (Nicotiana tabacum L.) in the world. This study was conducted to investigate degree of dominance, selection, and correlation between leaf shape and degree of bacterial wilt resistance in flue-cured tobacco. The degree of disease caused by bacterial wilt was evaluated in parents, F$_1$, F$_2$ and F$_3$ populations of two crosses, BY 4 x NC 95 and BY 4 x Coker 86, in the infected field. The leaf shape index was also measured in parents and F$_2$ population of BY 4 x NC 95. The incidence of bacterial wilt was observed in the middle of June and peaked in late July, when the highest value of pathogen density reached 1.0 x 10$^{6}$ colony forming unit per gram. It was concluded that the inheritance mode of risestance to bacterial wilt in the above two crosses of susceptible and resistant varieties was recessive and polygenic. The resistance to bacterial wilt was significantly correlated with leaf shape in F2 generation of BY 4 x NC 95. But certain plants having narrower leaves were also resistant to bacterial wilt. It is considered that the bacterial wilt resistant lines having narrower leaves could be selected. The selection for bacterial wilt resistance in the F$_2$ population might be effective.

연초 버어리종의 자연교잡율과 종자생산에 관련된 몇가지 요인 (Some Management Practices Affecting Outcrossing and Seed Production in Burley Tobacco (Nicotiana tabacum L.))

  • In this study effects of isolation distance, transplanting time of maternal plants, and bagging of flower head with the gauze-cloth bag on the outcrossing of burley tobacco (Nicotiana tabacum L.) were investigated. Also the effect of fertilizer level and control of the number of capsules per plant on seed production and quality were examined. A male sterile line. produced 0.3 to 3.8 capsules Per plant when it was Planted with normally flowering tobacco with the average outcrossing of 7.2 plants, ranging from 2 to 18 out of 20 plants. With the farther the isolation distance between maternal plants and pollen donor plant, the lower the outcrossing occurred. Outcrossing occurred even at the isolation distance of 312 m. When the maternal plants were transplanted 35 days after transplanting the pollen donor ones, the outcrossed plants were not decreased significantly. The bagging of the flower head with the gauze-cloth bas (#0.9∼ 1.0 mm) decreased the outcrossed plants significantly, but couldn't prevent the outcrossing completely. The seed amount per plant was higher in the highly fertilized cultivation. The number of seed capsules per plant affected significantly on seed yield and quality. When the seed capsules was controlled by 30 or 50 capsules per plant, the weight of 1,000 seeds and germination rate were higher than those with 70 or 90 capsules per plant. 연초 버어리종의 저온단일에 따른 품종간 개화일수 및 엽수 반응 (Varietal Response to Days to Flower and Number of Leaves under Cool Temperature and Short-day in Burley Tobacco (Nicotiana tabacum L.))

  • This experiment was conducted to investigate varietal responses to cool temperature (18℃) and short photoperiod (8 hours a day) in a controlled room of phytotron. Five burley tobacco varieties (Nicotiana tabacum L.), Awelius (very early flowering type), Burley 21 and KB 103 (medium), Banket Al and MB 1 (slightly late) and TC 681 (late) were compared in number of leaves per plant and day to flower in different treatment periods (10, 15, 20, 25 and 30 days). The days to flower decreased significantly than the control (in the phytoron with 14hr photoperiods and a minimum temperature 20'c) when the treatment periods were over 15 days for the varieties Aurelius, Burley 21 and KB 103, and 20 days for TC 681, and 30 days for Banket Al, respectively. And also the number of leaves decreased significantly than that of the control when the periods of treatment were over 10 days for Burley 21 and Aurelius, 15 days form 103, and 20 days for TC 681 and Banket Al, and 30 days for MB 1, respectively. The decreasing ratio in the number of leaves per plant under cool temperature and short photoperiod treatments was the lowest in Banket Al and MB 1. 연초(Nicotiana tabacum cv BY4) 이배체 및 반수체 식물의 캘러스로부터 식물체 재생 관련 효소의 변화 (Enzyme Activity in Plant Regeneration from Diploid and Haploid Calli of Nicotiana tabacum cv BY4)

  • 연초의 약배양에 의해서 반수체를 유기하고 형성된 반수체와 이배체의 캘러스로부터 식물체 분화과정에서 일어나는 생리적 차이점을 비교하고자 효소의 활성도와 페놀화합물의 함량을 비교 조사하였다. 반수체의 형성은 화아가 중간정도된 크기를 이용하여 IAA가 1.0 mg/L kietin이 0.5mg/L 조합 처리되고 활성탄이 3 mg/L 함유된 배지에서 가장 양호하였다. 캘러스의 유도는 이배체 및 반수체 공히 2,4-D의 농도가 0.5 mg/L 첨가된 배지에서 양호하였으며, 캘러스로부터 재분화는 BAP 농도 2.0 mg/L에서 효과적이었다. 재분화시 변화되는 효소의 활성도와 페놀화합물의 함량은 처리된 BAP의 농도와 사용한 식물체에 따라 차이가 있었다. Peroxidase의 활성도는 이배체 및 반수체 공히 BAP 농도가 2.0 mg/L일때 가장 높았으며, catalase의 활성도는 BAP의 농도가 1 mg/L일때 가장 높은 경향을 보였다. IAA oxidase 및 catalase의 활성도는 이배체에 비해 반수체에서 더 높은 경향을 보였으나 peroxidase의 활성도는 이배체가 더 높은 경향을 나타내었다.

Variegated 담배 (Nicotiana tabacum L. cv. BY-4)의 잎 절편 배양에 따른 재생 식물체의 특성 (Characterization of Plants Induced by in vitro Culture of Leaf Blade-segments in a Variegated Tobacco (Nicotiana tabacum L. cv. BY-4))

  • 수분 후 담배의 배에 중이온 ($^{14}$ N) beam을 조사하여 유도한 variegated담배의 잎 조직을 기내배양하여 재생된 식물체의 특성을 조사하였다. NAA 0.1mg/L 와 BAP 1/0mg/L를 첨가한 MS배지에서 variegated담배의 잎을 배양하면 백색부위에서는 백색 식물체만이 유도되었으나 녹색부위에서는 녹색 식물체가 47.2%, 백색 식물체가 37.4%, variegated식물체가 15.4% 유도되었다. 재생된 녹색 식물체는 자가수분한 후대 (F$_1$)에서 녹색 식물체 1,651 개:백색 식물체 54개로 분리되었다. 또한 variegated 잎에서 재생된 녹색 식물체와 정상주를 정역교배 한 결과, 전부 녹색체가 분리되었다. 이 결과는 variegated잎의 표현형 유전은 최소한 모성 유전이 아님을 보여준다. Variegated잎의 녹색부위를 배양하여 재생된 variegated식물체의 잎을 이용하여 DNA gel blot한 결과, 엽록체 유전자인 psbA, rbcL, 165 rDNA, 23S rDNA의 양은 흰색부위 잎에서 정상주인 녹색 잎과 variegated식물체 잎의 녹색부위 보다 많았다.

  • 버어리種 담배에 있어서 細胞質的 雄性不稔系統을 利用한 F1雜種 利用 可能性을 究明하고자 MS Burley21$\times$Burley37外 20個 F1雜種을 育成하여 生産力 檢定試驗을 實施한 結果 MS L8$\times$Burley49, MS Kentucky12$\times$kentucky10等 4個의 F1雜種에서 現 栽培品種인 Burley21보다 수량은 2 ~11%, 품질은 1 ~5%의 증가를 보였다.

  • 담배 (Nicotiana tabacum cv. BY4)의 이배체 식물과 약배양으로부터 유도된 반수체 식물의 잎절편을 사용하여 0.5mg/L 2.4-D 단독처리구에서 캘러스를 유도한 후, 2.0 mg/L 2.4-D, 1.0 mg/L Kinetin 및 0.1 mg/L BAP를 조합처리하여 계대배양을 하였다. 이배체에서는 2.0 mg/L 2.4-D와 1.0 mg/L 카이네틴의 조합처리구에서, 반수체는 2.0 mg/L2.4-D 와 0.1 mg/L BAP 조합처리구에서 세포증식이 양호하였으므로, 현탁배양도 동일조건에서 실시하였다. 세포괴를 균일하게 한 현탁배양세포를 p-Fluorophenylalanine (PFP)이 5~100 $\mu$M의 농도로 단독처리한 배지상에 2.0 mL씩 평판배양하여, 10, 20, 30일 후 PFP의 처리에 대한 저항성 colony를 조사하였다. 배양 30일 후 세포주의 생중량을 측정한 결과 PFP의 농도가 높아질수록 감소하였다. 선발된 캘러스는 이배체서 peroxidase의 활성도가 5 $\mu$M PFP 처리시에 가장 높았는데 100 $\mu$M PFP 처리까지는 대조구보다는 높았다. Catalase의 활성도는 5 $\mu$M PFP 처리시 가장 높았으나 100 $\mu$M PFP 처리시 가장 낮았다. 한편 반수체에서는 peroxidase와 catalase의 활성도는 50 $\mu$M PFP 처리시 가장 높게 나타나서 이배체와 반수체 사이에 효소활성의 차이는 뚜렷했다.

황색종 연초에서 mammoth gene과 흰꽃의 유전 (Inheritance of Mammoth Gene and White Flower in Flue-cured Tobacco(Nicotiana tabacum L.))

  • The genetic makeup could be the most important among many factors affecting yield and quality of tobacco(Nicotiana tabacum L.). The mammoth gene found in N. tabacum is associated with greater leaf number and poor leaf quality. This study was carried out to obtain the basic information about the inheritance of mammoth gene and white flower color. Two flue-cured breeding lines, KF 9373-2 and KF 8832-85, F$_1$, F$_2$, two parents backcrossed with F$_1$, and F$_3$ lines derived from cross of above two lines were investigated for flowering type(mammoth gene) and flower color. All plants of F$_1$ population revealed normal flowering type and pink flower color. The progeny of F$_2$ generation was segregated into the phenotypic ratio of 9 : 3 : 3 : 1 with normal flowering type and pink flower color, normal and white, non flowering type(NF) and pink, and NF and white, respectively. Among the progenies of back-crossing populations, the flowering type showed a segregation ratio of 1 : 1 as normal and NF in BP$_1$ and flower color did also 1 : 1 as pink and white in BP$_2$. All lines have the mammoth gene in F$_3$. that were selected in F$_2$ progeny as non flowering. But 9 lines among 14 were segregated with 3 : 1 as pink and white flower in F$_3$. which were selected in F$_2$ as pink flower color. These results indicated that the characters of mammoth gene and white flower were controlled by a pair of recessive genes, respectively.

연초(Nicotiana tabacum L.)의 반수체 육종법 1주1계통법, 집단육종에 의한 육성계통의 특성비교 (Comparison of Lines from Anther and Maternally-derived dihaploids, Single-seed Descent and Bulk Breeding Method in Flue-cured Tobacco (Nicotiana tabacum L.))

  • The present study was conducted to compare the relative efficiency of four different breeding methods in tobacco varietal development. A single Fl hybrid plant from cross of two flue-cured cultivars of Nicotiana tabacum L Bright Yellow 4(BY4) and NC95, was selfed. F2population above cross was screened for resistance against bacterial wilt caused by Pseudommonas solanacearum E.F.Smith under the naturally infested field conditions, and the 30 lines were developed from F2 individual plant by anther culture (ADH), maternal method utilizing Nicotiana afpicana (MDH), single- seed descent(SSD) and Bulk breeding method, respectively. All characters except content of total alkaloids of ADH and MDH which wore bred by haploid methods reduced more than that of lines bred by conventional methods(SSD & Bulk) : however, the yields were 8% lower than other lines. The total alkaloid content of ADH was higher than that of MDH, and yield was reduced about 4n even though the number of leaf was identical with the MDH. All other characters of ADH were also reduced significantly. In the lines bred by conventional methods, population developed by SSD showed significantly wider leaf width, shorter plant height, later days to flower, and lower in percent reducing sugar than those by Bulk. The populations derived from haploid method showed greater phenotypic variance and wider range of variation than conventionally developed ones. The results obtained indicate that selection will provide a significantly greater genetic gain for leaf number and leaf length in the ADU and MDH populations, and for plant height and days to flower in the SSD and Bulk populations.
