Perfusion Cultivation of Transgenic Nicotiana tabacum Suspensions in Bioreactor for Recombinant Protein Production

  • A perfusion culture of transgenic Nicotiana tabacum cell suspensions, transformed to express recombinant glucuronidase (GUS), was successfully performed in a 5-1 stirred tank bioreactor. With 0.1 $day^{-1}$ of perfusion rate, the maximum dry cell weight (DCW) reached to 29.5 g/l in 16 days, which was 2.1-fold higher than the obtained in batch culture (14.3 g/l). In terms of the production of GUS, the volumetric activity could be increased up to 12.8 U/ml by using perfusion, compared with 4.9 U/ml in batch culture. The specific GUS activities in both perfusion and batch cultures were maintained at similar levels, 200-400 U/g DCW. Consequently, a perfusion culture could be a good strategy for the enhanced production of recombinant proteins in a plant cell culture system.

  • Six flue-cured cultivars of Nicotiana tabacum L., 15 possible $F_1$ hybrids and 15 $F_2$ populations among them, and 15 haploid populations of $F_1$ hybrids and 6 haploid from parents, were evaluated. The variances of general combining ability were predominant for all characters studied in $F_1$ hybrid and $F_2$ population, and in haploid populations of $F_1$ hybrids except leaf width, value and reducing sugar. There were significant correlations between generations in variances and ranks of parental GCA effects estimated from general combining ability analysis of $F_1,\;F_2$ and haploid generation of $F_1$ hybrids for yield, stem height, days to flower. leaves per plant, value and total alkaloids.

Varietal Difference of Sucker producing Characteristics in Burley Tobacco(Nicotiana tabacum L. var. burley) (버어리종 연초 곁순발생의 품종간 차이)

  • This study was carried out to obtain the basic information about the sucker producing characteristics of certain burley tobacco varieties(Nicotiana tabacum L. var. burley) at Suwon and Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute in 1999. The number and weight of suckers for 12 varieties or breeding lines were investigated three times, 45 days after transplanting for ground sucker, 10 days after topping, and 10 days after maleic hydrazide(MH) applying for upstalk sucker. KB 103 and KB 9416-1 produced less ground suckers and upstalk suckers than other entries did. KB 9210-8 produced less ground suckers and (Male sterile TC 612 x KB 108)F$_1$ also produced less upstalk suckers than other entries did. There was no differences among entries in sucker production at 10 days after MH treatment. Between the weight of total sucker and days to flower revealed the negative correlation, but not significant. The low sucker producing entries described above could be used as a parental line in the breeding program for the low sucker producing variety.

The Effect of Growth Regulators and Light Quality on the Changes in Protein Pattern of Callus from Intergeneric Protoplast Fusion between Nicotiana tabacum and Solanum nigrum (Nicotiana tabacum과 Solanum nigrum의 속간 원형질체 융합에서 유도된 캘러스의 단백질 양태변화에 미치는 생장조절제 및 광선의 효과)

  • The effect of growth regulators (NAA, BA and $ extrm{GA}_3$) and light (blue, red and far-red) on the changes in total protein and thylakoid membrane protein pattern of callus from intergeneric protoplast fusion between Nicotiana tabacum and Solanum nigrw were investigated. When the callus were irradiated with different wavelengths of light, blue and red light accelerated the synthesis of total proteins and thylakoid membrane proteins. Particularly, red light led to an increase in the protein synthesis compared to blue light. When the callus were subjected to various combinations of growth regulators, NAA+$ extrm{GA}_3$ and NAA+BA treatments induced remarkable increase of total proteins and thylakoid membrane proteins accumulation, particularly in the combination of NAA+$ extrm{GA}_3$. NAA.$ extrm{GA}_3$ treatment with irradiation of red ligh showed highest value in the accumulation of total proteins and thylakoid membrane proteins. We conclude that simultaneous application of red light and NAA+$ extrm{GA}_3$ treatment may induce synergistic effect in the synthesis of total proteins and thylakoid membrane Proteins.

  • Production of therapeutic proteins by transgenic plant cell suspension cultures is an attractive system alternative to the other expression system. However, plant cell cultures have shown low expression level of foreign proteins and decreased cell viability by the changes of culture conditions. Therefore, it is necessary to enhance cell viability during the culture period. In this study, a quantitative analysis technique was designed to measure relative cell viability for plant suspension cells which have cell wall and aggregates. It was found that the programmed cell death of plant cells by apoptosis was essentially linked with the apoptotic pathway of animal cells. Therefore, effects of nicotinamide, 3-aminobenzamide and antioxidants on cell viability and apoptosis were examined in transgenic Nicotiana tabacum cells producing hGM-CSF. With those additives, cell viability could be maintained and apoptosis could be redued. In the result, the extracellular production of hGM-CSF could be enhanced 2.5 fold. It was also found that the supplementation of glutathione and ascorbic acid suppressed both the cold stress-induced decrease in cell viability and the increase of total genomic DNA fragmentation.

  • This study was conducted to identify useful germ plasm sources of resistance to black shank, Phytophthora. parasitica, in Nicotiana tabacum and to characterize the interaction between host plant resistance to black shank and pathogen variability. The six internationally represen-tative set of black shank resistant and susceptible tobacco genotypes and four Korean lines were tested for black shank on the field infested with P. parasitica at Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute during the 1992~1997 growing season. Almost 100% of the plants of the most susceptible genotype, Ky 14, did not survive. Complete survival of (MS Ky 14 x L8)F$_1$, which is resistant to reee 0 and ausceptible to reace 1, suggests that Race 0 is the only race of the P. parasitica in this bield. Coker 371 Gold and Beinhart 1000-1 having excellent resistance to black shank could be recommended as the parental cultivars for improving black shank resistance in Korea. Data of the diseased plants showed that KB 101 and KB 110 were medium high resistant and KB 103 and KB 108 being low resistant to black shank.

  • After topping, axillary buds of haploid plants derived from cultured anthers were treated with 0.4% aqueous solution of colchicine. Due to the high temperature and dry air at the time of treatment, most of the buds perished. A few months after the colchicine application, however, several shoots arose from the places where the dead buds were originally located. These shoots were mostly diploid. Induction of adventive shoots from the colchicine-treatedaxils was supposed to be rather effective method of obtaining diploid shoots from haploid plants. The diploid plants had larger floral organs than the haploid plants, and had good pollen fertility and seed setting. 24 bivalent chromosomes were observed at MI of the PMC's.

Effect of Agitation and Aeration Rate on Nicotiana tabacum Suspension Cell Culture in Bioreactors (Bioreactor를 이용한 담배세포 현탁배양에서 교반형태와 통기량이 미치는 영향)

  • For the optimization of operating conditions for plant cell suspension culture in bioreactors, effects of bioreactor types, various kinds of impellers, and aeration rates were examined using Nicotiana tabacum cells as a model system. Stirred tank bioreactor and airlift bioreactor were used for the comparison of bioreactor type. Growth rates in both bioreactors were lower than in shake flasks. In terms of final cell concentration, stirred tank bioreactor supported a little bit better growth compared to airlift bioreactor. Impeller type did not affect cell growth significantly, but it was apparent that cell size index decreased in the case of using hollowed paddle impeller. When the aeration rate was maintained at 0.3 vvm, cell growth was the best. At above 1.0 vvm, growth inhibition as well a browning was noticed. In addition, it was found that cell size index reduced proportionally to the increased of aeration rate.

  • The effect of phytohormones on in vitro production of ubiquinone 10 from the callus cultures of Nicotiana tabacum cv Xanthi was investigated. The growth of callus cultures of Xanthi was in proved by addition of NAA and 2,4-D, especially NAA 0.5 mg/L alone, at the light condition. Ubiquinone 10 was detected by HPLC, and confirmed from Xanthi callus cultured on the all of uppermedia. The ubiquinone 10 content in Xanthi tobacco callus cultured on the medium with NAA 0.5 mg/L only was higher than that of other mixed medium with NAA and 2,4-D. However addition of IBA 1 mg/L and NAA 0.5 mg/L to the medium was more effective in promoting ubiquinone 10 formation than that of NAA 0.5 mg/L only As the callus growth of Xanthi was considerabley restrained at concentration of kinetin, Content and production of ubiquinone low as the highest at kinetin 0.5mg/L and 2,4-D 0.5mg/L in the light.
