토양중(土壤中)의 Phytophthora spp. 방제(防除)를 위한 길항식물(桔抗植物)의 탐색 (Screening for Antagonistic Plants for Control of Phytophthora spp. in Soil)

  • 54과(科) 100종(種)의 식물 추출액(抽出液)을 공시(供試)하여 Phytophthora spp.에 대한 항균성(抗菌性)을 검정(檢定)한 바 균사생장(菌絲生長) 억제효과(抑制效果)가 있는 것은 양파, 마늘, 등배나무, 호장근 및 당대황이었고 이중에서 특히 마늘과 등배나무가 가장 뚜렷하게 나타냈다. 유주자낭(遊走子囊) 발아억제효과(發芽抑制效果)는 P. capsici에서 마늘, 등배나무 및 당대황이 뚜렷했고 P. nicotiana 와 P. infestans에서는 등배나무가 뚜렷했다. 또한 등배나무 추출액(抽出液)이 유묘(幼苗)에서 강(强)한 발병억제효과(發病抑制效果)를 나타냈다. 이상의 결과로 보아 항균물질(抗菌物質)은 분비(分泌)하는 능력을 가진 길항식물(拮抗植物)을 이용하여 토양전래성(土壤傳來性) 병원균(病原菌)을 저지(沮止)할 가능성이 있다고 믿는다.

한국에서 분리한 Tobacco rattle virus(TRV-K)의 특성 (Characterization of Tobacco rattle virus(TRV-K) isolated in Korea)

  • 대구시와 경상북도 지역에서 Tobacco rattle virus(TRV) 감염에 의한 증상으로 보이는 글라디올러스(Gladiolus hybridus), 크로커스(Crocus spp.), 수선화(Narcissus spp.)를 채집하였다. 톱니무늬(notch), 줄무늬(stripe), 변형 (malformation)등의 증상의 잎 조직을 시료로 하여 direct negative staining method(DN) 및 immunosorbent electron microscopy(ISEM)법에 의해 투과전자현미경으로 바이러스 입자를 관찰한 결과, 각각의 시료에서 긴 입자는 170~200 X 22nm, 짧은 입자는 40~114$\times$22nm 크기의 막대모양 입자가 다수 확인되었다. 글라디올러스에서 분리한 tobacco rattle virus(TRV-K)를 담배(Nicotiana tabacum)에 즙액 접종, 증식시킨 후, 정제하여 항혈청을 제작하였다. TRV-K 의 coat protein(CP)유전자를 특이적인 올리고뉴클레오타 이드 프라이머를 이용하여 역전사-중합효소연쇄반응(RT-PCR)으로 증폭시킨 후에 염기서열분석법으로 확인하였다. 그 결과 TRV-ORY의 CP와 99.5%의 상동성을 나타냈다.

Ralstonia solanacearum Type III Effectors with Predicted Nuclear Localization Signal Localize to Various Cell Compartments and Modulate Immune Responses in Nicotiana spp.

  • Ralstonia solanacearum (Rso) is a causal agent of bacterial wilt in Solanaceae crops worldwide including Republic of Korea. Rso virulence predominantly relies on type III secreted effectors (T3Es). However, only a handful of Rso T3Es have been characterized. In this study, we investigated subcellular localization of and manipulation of plant immunity by 8 Rso T3Es predicted to harbor a nuclear localization signal (NLS). While 2 of these T3Es elicited cell death in both Nicotiana benthamiana and N. tabacum, only one was dependent on suppressor of G2 allele of skp1 (SGT1), a molecular chaperone of nucleotide-binding and leucine-rich repeat immune receptors. We also identified T3Es that differentially regulate flg22-induced reactive oxygen species production and gene expression. Interestingly, several of the NLS-containing T3Es translationally fused with yellow fluorescent protein accumulated in subcellular compartments other than the cell nucleus. Our findings bring new clues to decipher Rso T3E function in planta.

당근 절편을 이용한 Agrobacterium spp. 분리, 동정과 선발균주에 의한 연초의 형질전환 (Isolation and identification of Agrobacterium spp. using carrot disc and transformation of Nicotiana gluca by selected strains)

  • These studies were carred out to obtain the transformant from tobacco cells by Agrobacterium spp. from crown gall and soil at the natural field in Korea, and identified their virulence. Kodo's and Clark's selective media were used for isolation of Agrobacterium spp. In these media, total of 99 strains were characterized based on the morphological characteristics of colonies. Among them 34 strains were able to induce on carrot discs. And hypervirulent strains C23-1 and K29-1 were identified as Agrobacterium tumefaciens biotype 1 and biotype 2, respectively. These strains formed fast growing, larger gall as compared to those induced by other strains on the carrot discs. Transformed tobacco callus was initiated on the phytohormone free MS medium with 250$\mu\textrm{g}$/ml carbenicillin after co-cultivation of tobacco stem explants and Agrobacteria. On the phytohormone free media, shoot was rarely formed from transformed callus. However, these shoot were teratoma shoots which were not grown as normal shoot, and teratoma shoot from transformant by C23-1 was smaller than that of K29-1.

Isolation and Characterization of Fungal Diversity from Crop Field Soils of Nigeria

  • In order to find indigenous beneficial fungal species from crop field soils of Nigeria, 23 soil samples were collected from various places of Nigeria in June, 2013 and fungi were isolated through serial dilution technique. Isolated fungi were purified and differentiated according to their morphological and microscopic characteristics. In total, 38 different representative isolates were recovered and the genomic DNA of each isolates was extracted using QIAGEN$^{(R)}$ Plasmid Mini Kit (QIAGEN Sciences, USA) and the identification of fungi was carried out by sequence analysis of internal transcribed spacer (ITS) region of the 18S ribosomal DNA (18S rDNA). Recovered isolates belonged to 9 fungal genera comprising Fusarium, Aspergillus, Chaetomium, Coniothyrium, Dipodascaceae, Myrothecium, Neosartorya, Penicillium and Trichoderma. Aspergillus spp., Penicillium spp. and Trichoderma spp. were the most dominant taxa in this study. The antagonistic potentiality of species belonged to Trichoderma against 10 phytopathogenic fungi (F. oxysporum, C. gloesporoides, P. cytrophthora, A. alternata, A. solani, S. rolfsii, F. solani, R. solani, S. sclerotiorum and P. nicotiana) was assessed in vitro using dual culture assay. The dual culture assay results showed varied degree of antagonism against the tested phytopathogens. The potential Trichoderma spp. will be further evaluated for their antagonistic and plant growth promotion potentiality under in vivo conditions.

담배, 토마토 및 고추에서 분리한 TMV 계통의 추가적인 생물학적 특성 (Additional Biological Characteristics of TMV Strains Isolated from Tobacco, Tomato and Pepper Plants)

  • Tobacco (Nicotiana tabacum) cultivars including NC 82 and KF 114, and Datura stramonium, Physalis floridana, Gomphrena globosa, and Chenopodium spp. were added to the previous host plants tested for the further examination on the biological characteristics of tobacco mosaic virus (TMV) strains isolated from tobacco (TMV-Common), tomato (TMV-Tomato), and pepper (TMV-Pepper), In TMV-Tomato and TMV-Pepper, different symptoms were noted in P. floridana (no symptom development), and NC 82 (local lesion production on the inoculated leaves) from TMV-Common with which systemic mosaic symptoms were developed. Local lesions were developed in KF 114, D. stramonium, G. globosa, and Chenopodium spp by TMV-Common and TVM-Tomato, while no symptom was observed in KF114 and G. globosa. Also the number and size of local lesions were smaller in KF 114 than Xanthi-nc tobacco (local lesion host) infected with TMV-Tomato. Systemic necrosis was induced in Xanthi-nc and KF 114 when infected with TMV-C at high temperature, but not with the other strains.

Survey of the Incidence of Viral Infections in Calanthe spp. and Characterization of a GW Isolate of Cymbidium mosaic virus in Korea

  • Cymbidium mosaic virus (CymMV) is a major virus infecting orchid plants and causing economic loss. In this study, the incidence of viral infection in Calanthe spp. at the Korean Institute of Calanthe was investigated using reverse transcription polymerase chain reaction. The CymMV infection rate was 42%, and the two viruses Odontoglossum ringspot virus and Cucumber mosaic virus had frequencies of 8% and 2%, respectively. Additionally, we characterized an isolate of CymMV, CymMV-GW, using biological tests and examined the nucleotide sequence properties of its complete genome. CymMV-GW induced chlorotic ringspots and chlorotic spot symptoms in inoculated leaves of Chenopodium amaranticolor and Nicotiana benthamiana, respectively. In this study, we have for the first complete genome sequence of CymMV-GW in Korea. The CymMV-GW genome was 6,225 nucleotides in length, excluding the poly-(A) tail, and showed whole-genome nucleotide and amino acid sequence identities of 97.7% and 100%, respectively, with the NJ-1 isolate of CymMV. Here, we report the complete genome sequence of the CymMV-GW isolate and viral infection rates for Calanthe spp. in Korea.

Three Different Viruses Isolated from Typical Weed Plants that Grown Adjacent to Common Crop Fields

  • Weeds are widely grown in the field and are infected by many viruses. A survey was conducted to identify viruses infecting weeds in Korea. Virus-infected weed samples including Rorippa indica (L.) Hiern, R. islandica (Oed.) Bord, Crepidiastrum denticulatum (Houtt.) Pak & Kawanno, Achyranthes japonica (Miq.) Nakai, and Chrysanthemum boreale (Makino) Makino were collected in Kyonggi Province. These weeds were grown in the greenhouse and were isolated on 10 test plants. Several virus isolates were isolated fron infected tissues and were further studied by host range assay, serological test, electron microscopy (EM), reverse transcription-polymerase chain reaction (RT-PCR) and sequencing. Each isolated virus strain was mechanically transmitted to weeds and various hosts including Nicotiana spp., Brassica spp., Vigna unguiculata, Capsicum annuum, and Cucumis sativus and showed systemic mosaic, vein clearing, necrosis, mottle, malformation, chlorosis, and/or death of host plants in some cases. Each virus was then purified using infected leaves and observed by EM. From these results three viruses were isolated and identified as Turnip mosaic virus (TuMV), Broad bean wilt virus (BBWV), and Cucumber mosaic virus (CMV). RT-PCR using virus-specific oligonucleotide primers and the cloning were conducted to determine the nucleotide sequences of coat proteins of the three viruses their amino acid sequence were deduced. The amino acid sequence homologies were about 92.7 to 99.7%, 96.2 to 97.7%, and 93.9 to 98.6% to other reported TuMV, BBWV, and CMV strains, respectively. These results suggest that many weeds may serve as primary inoculum source of diseases caused by TuMV, BBWV, CMV and that the management of these viral diseases can be achieved through weed control.

한국에서 CMV에 항바이러스 효과를 나타내는 Serratia spp. Gsm01 균주의 분리 동정 및 효과 검정 (Isolation and Evaluation of an Antiviral Producing Serratia spp. Strain Gsm01 against Cucumber mosaic virus in Korea)

  • 항바이러스 효과를 나타내는 세균은 강원도 홍천 인삼재배지의 수삼으로부터 분리하였다. 분리세균은 생리 생화학 테스트와 16s rRNA 유전자 분석을 통해 동정한 결과, Serratia 속으로 동정 되어져 본 세균을 Gsm01으로 명명하였다. Gsm01 균주를 MGY 액체배지에 증식시켜 배양 여액을 0.45 ${\mu}m$ filter에 통과시킨 Culture filtrate (CF)를 명아주 (Chenopodium amaranticolor)에 반엽법으로 처리한 결과, 오이모자이크 바이러스 (CMV)에 대한 억제율이 98%로 매우 높은 것을 확인하였다. 또한 전신유도저항성을 확인하기 위하여 담배 (Nicotiana tabacum cv. Xanthi-nc)의 하엽에 CF와 CMV-Y를 처리하였을 때, CF를 처리한 식물에서 접종15일 후까지 바이러스 증상이 관찰되지 않았다. 고추의 포장시험에서 무처리 식물과 비교해 보았을 때 바이러스 증상은 52.9% 감소한 것으로 보아 포장 시험에서 CF를 처리한 농작물의 산출량이 무처리 식물에 비해 14% 증가함을 확인 할 수 있었다. 한편, Gsm01 균주의 CF는 고추에 어떠한 약해도 나타내지 않아 매우 안전한 것으로 판단되었다.
