Tobacco Transformation Using Expansin Genes (Expansin 유전자를 이용한 담배의 형질전환)

  • Tobacco (Nicotiana tabacum L.) cells were transformed with rice expansin genes, OsEXPA4, OsEXPB3, OsEXPB4, and OsEXPB6, to elucidate the function of the genes in tobacco cells. The transformation increased the mass of the callus by $36\%-65 \%$, and the cell length by $12\%-28\%$. The cell width was decreased by $3\%$ for OsEXPB3, not changed for OsEXPB4, increased by $25\%\;and\;20\%$ for OsEXPA4 and OsEXPB6, respectively. From database search, seven expansin genes were found and six of them belong to EXPA group and one of them belongs to EXPB group. EXLA and EXLB were not found. All tobacco expansin genes were evenly distributed in the phylogenetic tree of rice and Arabidopsis expansin genes.

Molecular Characterization of Tomato Yellow Leaf Curl Virus in Korea and the Construction of an Infectious Clone

  • Several tomato production regions in Korea were surveyed for tomato yellow leaf curl disease (TYLCD). Tomato leaf samples showing TYLCD-like symptoms were collected from Tongyeong (To), Geoje (Gi), and Gimhae (Gh) cities of the southern part of Korea. Tomato yellow leaf curl virus (TYLCV) was detected and the full-length genomes of the isolates were sequenced. The TYLCV isolates found in Korea shared high sequence identity (> 99%) with TYLCV-IL [JR:Omu:Ng] (AB110217). Phylogenetic relationship analysis revealed that they formed two groups (with little genetic variability), and the To, Gj, and Gh isolates belonged to the TYLCV-IL group. An infectious clone of TYLCV-To (JQ013089) was constructed and agroinoculated into Nicotiana benthamiana, Nicotiana tabacum var. Xanthi, Petunia hybrida, Capsicum annuum, and Lycopersicon esculentum cv. Hausumomotaro. Agroinfection with a dimeric infectious clone of TYLCV-To induced severe leaf curling and stunting symptoms in these plants, excluding C. annuum. Tomato plants then developed typical yellow leaf curl symptoms.

Theobroxide Treatment Inhibits Wild Fire Disease Occurrence in Nicotiana benthamiana by the Overexpression of Defense-related Genes

  • Theobroxide, a novel compound isolated from a fungus Lasiodiplodia theobromae, stimulates potato tuber formation and induces flowering of morning glory by initiating the jasmonic acid synthesis pathway. To elucidate the effect of theobroxide on pathogen resistance in plants, Nicotiana benthamiana plants treated with theobroxide were immediately infiltrated with Pseudomonas syringae pv. tabaci. Exogenous application of theobroxide inhibited development of lesion symptoms, and growth of the bacterial cells was significantly retarded. Semiquantitative RT-PCRs using the primers of 18 defense-related genes were performed to investigate the molecular mechanisms of resistance. Among the genes, the theobroxide treatment increased the expression of patho-genesis-related protein 1a (PR1a), pathogenesis-related protein 1b (PR1b), glutathione S-transferase (GST), allen oxide cyclase (AOC), and lipoxyganase (LOX). All these data strongly indicate that theobroxide treatment inhibits disease development by faster induction of defense responses, which can be possible by the induction of defense-related genes including PR1a, PR1b, and GST triggered by the elevated jasmonic acid.

The Cause of Calcium Deficient leaf of Burley Tobacco (Nicotiana tabacum L.) (Burley종 연초의 칼슘결핍엽 발생원인)

  • Field experiment was conducted to find out the cause of calcium deficiency of burley tobacco (Nicotiana tabacum L.), Liming materials and application rate were CaSO4 : Ca 35kg/10a, CaCO3 : 35kg/10a and agricultural lime : Ca 142kg/10a. The breakdown in tissues at tips and margins of upper leaves was developed at maximum growth stage. If complete breakdown and death does not occur and growth takes place later, giving the leaf a scalloped appearance. Upper leaves contained lower calcium content than other leaves. The stem and midvein of calcium deficient plants contained lower calcium and calcium minus oxalic acid, but higher oxalic acid contents than those of normal plant. Fresh leaves of limed plot contained higher calcium and oxalic acid, but not significant increment of calcium minus oxalic acid than those of unlimed plot. Since calcium oxalate is insoluble, it could precipitate within the culls if the calcium and oxalic acid are accessible to each other. It suggest that high level of oxalic acid in stem and midvein could be interfering with translocation of calcium to upper leaves.

Physiological Function of NbRanBP1 in Nicotiana benthamiana

  • This study addresses the physiological functions of the Ran-binding protein homolog NbRanBP1 in Nicotiana benthamiana. Virus-induced gene silencing (VIGS) of NbRanBP1 caused stunted growth, leaf yellowing, and abnormal leaf morphology. The NbRanBP1 gene was constitutively expressed in diverse tissues and an NbRanBP1:GFP fusion protein was primarily localized to the nuclear rim and the cytosol. BiFC analysis revealed in vivo interaction between NbRanBP1 and NbRan1 in the nuclear envelope and the cytosol. Depletion of NbRanBP1 or NbRan1 reduced nuclear accumulation of a NbBTF3:GFP marker protein. In the later stages of development, NbRanBP1 VIGS plants showed stress responses such as reduced mitochondrial membrane potential, excessive production of reactive oxygen species, and induction of defense-related genes. The molecular role of RanBP1 in plants is discussed in comparison with RanBP1 function in yeast and mammals.

  • This study was conducted to determine the inheritance of duvatrienediol(DVTs) and cis-abienol concentration from flesh tobacco leaves. NC744, which has a very low levels of DVTs and no cis-abienol was used as a parent in cross to TI 1068 producing a normal amount of DVTs and cis-abienol. Presence of DVTs and cis-abienol on the leaf surface was determined using thin layer chromatography(TLC). Segregation pattern from F$_2$ and BC$_2$[ (TI 1068 $\times$ NC744) $\times$ NC744J generations revealed that TI 1068 have a single dominant gene controlling DVTs and cis-abienol production. And DVTs production was inherited independently of the ability to produce cis-abienol.

Fertilizer Placement Effect on Agronomic Characteristics of Burley Tobacco(N. tabacum L.) (시비방법이 버어리종 잎담배의 농경적 특성에 미치는 영향)

  • Recently the method of basal compound fertilizer (N : P : K = 18.2 : 9.8 : 35.0) Placement has been changed from banding to broadcasting in tobacco cultivation. The effects of the compound fertilizer placement, level of fertilizer and additional urea application on the agronomic characteristics, two chemical compositions and physical Properties of burley tobacco(Nicotiana tabacum L. cv. KB 108) were investigated at Chonju Experiment Station, Korea Ginseng and Tobacco Research Institute in 1995-1996. Six treatments consisted of (1) band (method of fertilizer Placement) + In kg/10a(compound fertilizer) + 0(additional urea applied), (2) broadcast+140+0, (3) broadcast+140+25, (4) band+180+0, (5) broadcast+180+0, and (6) broadcast +180+25. The additional urea was applied at hilling. No significant differences were detected between banding and broadcasting method of compound fertilizer Placement in field 9rowth, wield, organoleptic qualify(price Per kilogram), chemical composition and Physical properties of cured leaf, The increased compound fertilizer by 30%(40k/10a) or the additional urea application by 25kg per 10a produced slightly higher yield than the recommended amount of basal compound fertilizer without additional urea application did. It also increased the total alkaloid content of cured leaf. It is recommended that no more basal fertilizer above the recommended amount and no additional urea application are needed in burley tobacco fertilization, even though the method of basal fertilizer placement being chanced from banding to broadcasting.   • he study was conducted to produce an antiserum of potato virus S for identification and screening of seed-potatoes. Potato virus S was isolated from infected plants and identified by means of indicator plants and electro microscopy. Isolated potato virus S was multiplied in Nicotiana deebneyii and the virus was purified by a modified method that was developed through this study. The purity of potato virus S was 1.18mg/ml. Purified potato virus S was injected into rabbit intravenously once a week for 5 weeks. Antiserum was collected 10 days after the last injection. The produced antiserum was determined to have a titer of, 1/2048 by means of microprecipitin tests.

Varietal Difference of Leaf Breakdown in Field of Flue-Cured Tobacco (Nicotiana tabacum L.) (황색종 연초(Nicotiana tabacum L.)에서 엽탈락의 품종간 차이)

  • This study was conducted to obtain basic information for varietal difference of leaf breakdown in field of flue-cured tobacco at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute in 1987. The experiment was designed in randomized block with 3 replications, data ware analysed as Split split-plot design. Main plots were varieties, sub-plots were leaf positions, 4, 5 and 6th from bottom, and each sub-plot was divided into 3 parts with distance of midrib, 7 10 and 13em from stalk. Four varieties, NC 95, NC 2326, NC 82 and BY 4 were transplanted in 15 April, and measured the weight of leaf breakdown by artificially weighted in 5 June. The results obtained are as follows : 1. Weights of leaf breakdown according to leaf position, NC 95 were lower as 358-5799 than those of other varieties as 555-597g, were not significantly different regardless of varieties. 2. Weights of leaf breakdown in relation to distance of midrib from stack, NC 95 were lower as 309-419g than those of other varieties as 472-710g. 3. Weights of leaf breakdown were significantly different according to distance of midrib from stalk, and not significantly different according to leaf position under the same distance of midrib from stalk regardless of varieties.

  • Structures of the adventitious root meristem induced from callus culture of tobaco (Nicotiana tabacum cv. NC 82) leaves were investigated by light and transmission electron microscopy. Structural differences between in vitro root and callus cells were also examined by the microscopy. The submicroscopic features of the in vitro root cells were as follows. Intercellular spaces were not developed and nuclei with two nucleoli were observed occasionally. Plasmodesmate were found in groups or sing1y on transverse and longitudinal walls. Amyloplast solely filled with starch grains, with one to five electron - dense bands, was surrounded by single membrane. in the callus cells, vacuolization of central part in the cytoplasm having mitochondria with swollen cristae and starch grains like those of in vitro root cells was a distinct feature. Vesicles which were found between cell wall and plasma membrane may be arisen by a process of protoplasmic invagination. By comparing of ultrastructures between the cells of callus and in vitro roots we found that the distinct differences lied on thickened cell walls and hypertrophed vacuoles in the former, and less thickened cell walls and several small vacuoles in the later.
