항정신병 약물에 의한 혈중 프로락틴 변화와 도파민 전달체 유전자 다형성 (Prolactin Response to Antipsychotic Drug and Dopamine Transporter Gene Polymorphisms)

  • Object:We investigated the relationship between prolactin response to antipsychotics and clinical courses of psychotic symptoms and DAT gene polymorphisms. Method:Twenty-four acute psychotic inpatients completed the 12-week trial of risperidone. Serum prolactin, BPRS, ESRS and hyperprolactinemia-related symptoms were measured at baseline, 2, 4, 8 and 12 weeks after medication. The DAT gene polymorphisms were analyzed. Results:The serum prolactin was significantly increased over time. According to the prolactin level at 2-week, the subjects were divided into the severe group(serum prolactin>60ng/mL, N=15) and the mild group (serum prolactin<60ng/mL, N=9). The prolactin levels of the mild group didn't increase beyond 60ng/mL throughout 12 weeks. Severe group had slower decrement of BPRS scores than those of mild group. Six females in severe group complained of irregular menstruations, but no female in mild group. Most patients had 10 allele of DAT gene. Conclusion:This study suggests that the magnitude of prolactin elevation at the 2-week of risperidone medication is correlated with severity of hyperprolactinemia throughout treatments. Our results did not show the relationship between prolactin responses and DAT gene polymorphisms.

Kinetics and Biological Function of Transforming Growth Factor-$\beta$ Isoforms in Bovine and Human Colostrum

  • Colostrum contains various kinds of cytokines including TGF-$\beta$ that has potent regulatory effects on cells of the immune system. We compared the levels of TGF-$\beta$1 and TGF-$\beta$2 in bovine and human colostrum. Based on the isoform-specific ELISA, bovine colostrum collected on day 1 post-delivery retained $53.71{\pm}29.55\;ng/ml$ of TGF-$\beta$1 and $40.41{\pm}21.78\;{\mu}g/ml$ of TGF-$\beta$2 (n=4), while in human, $381.45{\pm}158.24\;ng/ml$ of TGF-$\beta$1 and $41.47{\pm}9.63\;ng/ml$ of TGF-$\beta$2 (n=5). Thus, dominant TGF-$\beta$ isoforms were completely opposite between human and bovine colostrum samples. The concentrations of both isoforms declined as lactation proceeded. Biological activities of the colostrum samples were determined using an MV1LU cell line. Consistent with the result from the immunoassay, TGF-$\beta$1 in human and TGF-$\beta$2 in bovine colostrum were responsible for the anti proliferative activity against MV1LU cells. Furthermore, bovine colostrum increased IgA secretion by LPS-stimulated mesenteric lymph node (MLN) cells, and this effect was abrogated by either anti­TGF-$\beta$2 antibody or combined anti-TGF-$\beta$1/$\beta$2 antibody, but not by anti- TGF-$\beta$1 antibody alone. Similarly, TGF-$\beta$2 in bovine colostrum enhanced the Ig germ line (GL) promoter activity, which is the earliest event toward IgA isotype switching. Taken together, these results suggest that TGF-$\beta$ isoforms, differentially expressed in human and bovine colostrum, may promote IgA isotype production in the neonatal intestine.

CEA and CA 19-9 are Still Valuable Markers for the Prognosis of Colorectal and Gastric Cancer Patients

  • Background: The purpose of this study was to assess the predictive effect of preoperative CEA and CA 19-9 levels on the prognosis of colorectal and gastric cancer patients. Materials and Methods: CEA and CA 19-9 were evaluated preoperatively in patients undergoing surgery for colorectal cancer (n=116) and gastric cancer (n=49). Patients with CEA levels <5 ng/mL were classified as CEA Group 1, 5-30 ng/mL as CEA Group 2 and >30 ng/mL were classified as CEA Group 3. Similarly the patients with a CA 19-9 level <35 U/mL were classified as CA 19-9 Group 1, with 35-100 U/mL as Group 2 and with >100 U/mL as Group and 3. TNM stages and histologic grades were noted according to histopathological reports. Patients with a TNM grade 0 or 1 were classified as Group A, TNM grade 2 patients constituted Group B and TNM grade 3 and 4 patients constituted Group C. Demographic characteristics, tumor locations and blood types of the patients were all recorded and these data were compared with the preoperative CEA and CA19-9 values. Results: A significant correlation between CA 19-9 levels (>100 U/mL) and TNM stage (in advanced stages) was determined. We also determined a significant correlation between TNM stages and positive vlaues for both CEA and CA 19-9 in colorectal and gastric cancer patients. In comparison between CEA and CA 19-9 levels and age, gender, tumor location, ABO blood group, and tumor histologic grade, no significant correlation was found. Conclusions: Positive levels of both CEA and CA 19-9 can be considered to indicate an advanced stage in colorectal and gastric cancer patients.

이차 미분 형광 분광광도법에 의한 울산만 해양 저질토양 중의 다환 방향족 탄화수소(PAHs)의 동시 분석 (Synchronous determination of polycyclic aromatic hydrocarbons(PAHs) in sediment of Ulsan Bay by synchronous 2nd derivative fluorescence spectrophotometry)

  • 현이차 미분 형광 분광광도법을 이용하여 울산만 해양 저질토양중의 PAHs를 n-hexane용매로 추출하여 11종의 PAHs를 동시 정량분석하였다. Acenaphthene (Ace), anthracene (Anth), benzo(a)pyrene (BaP), benz(a)anthracene (BaA), benzo(b)fluoranthene (BbFt), benzo(k)fluoranthene (BkFt), chrysene (Chry), perylene (Per), phenanthrene (Phen), pyrene (Pyr) 및 fluoranthrene (Ft) 등을 정량분석 하였다. 이들 성분들의 검정선은 대략 0.15~166 ppb의 농도범위에서 직선관계를 보였으며, 0.999이상의 좋은 직선 상관계수를 보였다. 울산만의 해양 저질 토양 (sediment)에 함유된 11종의 PAHs 총량은 68.8 ng/g ~ 324.4 ng/g의 농도범위로 함유되어 있었다. 또한 PAHs의 총량은 울산만의 안쪽으로 갈수록 증가하는 경향을 보였으며, 특히 Pyr과 BaA 등과 같은 4고리화합물의 함량비가 높았다.

유아체육프로그램이 정상체중유아와 비만유아의 체력과 혈중지질 및 성장관련인자에 미치는 영향 (The Effects of Physical Activity Programs on Physical Fitness, Blood Lipids, and Growth-Related Factors among Young Normal Weight and Obese Children)

  • 본 연구는 비만유아 7명과 정상체중유아 7명을 대상으로 유아체육프로그램이 신체조성, 체력, 혈중지질 및 성장관련인자의 변화를 분석하기 위해 실시되었다. 본 연구에서 실시한 유아체육프로그램이 체지방량을 감량시키는데 효과적인 결과를 얻지 못하였지만 비만군과 정상체중군 모두 체력은 향상되는 것으로 나타났다. 그리고 정상체중유아들의 경우 HDL-C와 제지방량의 증가는 본 연구에서 실시한 유아체육프로그램이 성인기의 비만과 성인병 예방 측면에서 정상체중유아들에게 권장될 수 있는 것으로 사료된다. 또한 성장관련인자 IGF-I은 성장을 촉진하는 활성 물질로 성장호르몬 분비의 증가에 따라 IGF-I 농도 역시 상승한다. 본 연구에서도 정상체중군에서 긍정적인 효과가 나타나 유아체육프로그램이 성장기 유아의 성장호르몬의 분비를 활성화시키고 성장을 촉진시키며 신체발달을 증진시키는데 도움을 줄 수 있을 것으로 생각한다.

Occupational Exposure during Intraperitoneal Pressurized Aerosol Chemotherapy Using Doxorubicin in a Pig Model

  • Background: This study evaluated occupational exposure levels of doxorubicin in healthcare workers performing rotational intraperitoneal pressurized aerosol chemotherapy (PIPAC) procedures. Methods: All samples were collected during PIPAC procedures applying doxorubicin to an experimental animal model (pigs). All procedures were applied to seven pigs, each for approximately 44 min. Surface samples (n = 51) were obtained from substances contaminating the PIPAC devices, surrounding objects, and protective equipment. Airborne samples were also collected around the operating table (n = 39). All samples were analyzed using ultra-high performance liquid chromatography-mass spectrometry. Results: Among the surface samples, doxorubicin was detected in only five samples (9.8%) that were directly exposed to antineoplastic drug aerosols in the abdominal cavity originating from PIPAC devices. The telescopes showed concentrations of 0.48-5.44 ng/cm2 and the trocar showed 0.98 ng/cm2 in the region where the spraying nozzles were inserted. The syringe line connector showed a maximum concentration of 181.07 ng/cm2, following a leakage. Contamination was not detected on the surgeons' gloves or shoes. Objects surrounding the operating table, including tables, operating lights, entrance doors, and trocar holders, were found to be uncontaminated. All air samples collected at locations where healthcare workers performed procedures were found to be uncontaminated. Conclusions: Most air and surface samples were uncontaminated or showed very low doxorubicin concentrations during PIPAC procedures. However, there remains a potential for leakage, in which case dermal exposure may occur. Safety protocols related to leakage accidents, selection of appropriate protective equipment, and the use of disposable devices are necessary to prevent occupational exposure.

HF-LPME를 이용한 수용액 시료중의 카페인 분석 (Analysis of caffeine in aqueous sample by hollow fiber-liquid microextraction (HF-LPME))

  • 속빈 섬유-액상 미량추출법(hollow fiber-liquid phase microextraction, HF-LPME)과 기체 크로마토그래프/질소-인 검출기(GC-NPD)를 사용하여 사람의 뇨와 여러 가지 음료 중에서 극미량의 카페인을 분석하는 방법을 확립하였다. 수용액 시료로부터 카페인의 양을 측정하기 위해서 속빈 섬유의 길이, 추출용매의 종류, 교반효과, pH 및 염석효과 등 여러 가지 실험적인 파라미터를 변화시켜서 최적의 조건을 확립하고자 하였다. 검정곡선의 회귀계수($r^2$)는 0.9994이상이었으며, 평균 상대 회수율은 102%(n=3)이었으며, 기기검출한계는 2.5 ng/mL, 정량한계는 10 ng/mL이었다. 확립된 HF-LPME 방법은 생물학적 시료, 음식물, 환경시료 중에서 카페인의 농도를 측정하기에 편리하고 정확한 방법이 될 것이다.

Establishment of Efficient Microinjection System in the Porcine Embryos

  • Transcription activator like effector nucleases (TALENs) are artificial restriction enzymes generated by fusing a TALE DNA binding domain to a DNA cleavage domain which remove and introduce specific genes to produce transgenic animals. To investigate the efficient laboratory techniques for the injection of TALEN mRNA, pEGFP-N1 commercial plasmid were microinjected into porcine parthenogenetic and in vitro fertilization (IVF). In Experiment 1, to investigate injection time, compared 4 different time durations (2 hr, 4 hrs, 6 hrs & 8 hrs) after post activation of parthenogenetic embryos and after 6 hrs of co-incubation with sperms in IVF embryos. There were significant difference (P<0.05) in development to the blastocysts (4.4, 8.9, 3.9, 0.6%), GFP expression in blastocysts (1.3, 5.7, 2.3, 0.0%) which injected after post activation of 4 hrs compared with other 3 groups. IVF embryos after 2 hrs and 4 hrs injected were expressed GFP significantly higher than rest of two groups (P<0.05). In Experiment 2, compared development of 2 different concentrations ($20ng/{\mu}l$ and $50ng/{\mu}l$) of EGFP injection. There were significant difference (P<0.05) between two treatments which has higher cleavage (58.8 vs 41.9%), blastocysts development rate (13.0 vs 11.1%) and GFP expressed blastocysts (5.7 vs 0.0%) in $20ng/{\mu}l$ than the $50ng/{\mu}l$ in parthenogenetic embryos. In IVF embryos, only $20ng/{\mu}l$ injected embryos were expressed GFP (4.2%) after 7 days of incubation and 77.3 vs 64.7% of cleavage, 26.4 vs 23.5% development to blastocysts. In Experiment 3, three different volumes (5, 10 and 20 pl) were microinjected into porcine embryos to determine the most appropriate volume. Out of 3 groups, significantly higher development rates of cleavage (68.3, 58.0, 29.3%), blastocysts (11.7, 12.7, 0.5%) and GFP expressed blastocysts (2.9, 7.8, 0.0%) were shown in the 10 pl group (P<0.05). In conclusion, these results imply that $20ng/{\mu}l$ concentration, 10 pl of volume and injection at 4 hrs after post activation for parthenogenetic and 2~4 hrs after IVF, $20ng/{\mu}l$ concentration and 10 pl volume for IVF embryos were more effective microinjection conditions.

낙동강 하천 및 호소 퇴적물에서의 PAHs, OCPs, Co-PCBs 농도 분포와 발생원 평가 (Assessment of Distribution Patterns and Sources for PAHs, OCPs, and Co-PCBs in the Surface Sediments from the Nakdong River Basin, Korea)

  • 본 연구에서는 낙동강 수계 내 하천 및 호소의 퇴적물을 대상으로 16종의 다환방향족탄화수소(PAHs), 다염화바이페닐(PCBs), 유기염소계 농약(OCPs)류를 분석하였다. 하천 퇴적물에 존재하는 16종의 PAHs와 12종의 Co-PCBs 농도 수준은 각각 N.D.~969.3 ng/g-dry, 4.2~7716.5 pg/g-dry(0.0~10.1 pg-TEQ/g-dry)이었다. 호소 퇴적물에 존재하는 16종의 PAHs류는 5.8~2987.2 ng/g-dry, 12종의 Co-PCBs는 4.3~461.1 pg/g-dry(0.0~0.6 pg-TEQ/g-dry), OCPs의 농도 수준은 N.D.~1.5 ng/g-dry이었으며, OCPs 중 DDT류만 검출되었다. 이는 환경질 평가를 위한 가이드 라인보다 훨씬 낮은 수준이었다. 낙동강 수계 내 하천 퇴적물에 존재하는 16종의 PAHs 중 저분자 PAHs의 비율이 높은 반면, 호소에서는 중 고분자 PAHs의 비율이 높았다. 그리고 PCBs는 모든 퇴적물 시료에서 PCB-118과 PCB-105의 비율이 가장 높게 나타났으며, 이는 이전의 연구와도 일치하는 결과이다. 검출된 PAHs와 Co-PCBs의 배울원 추정 결과, PAHs의 경우 일부 지점들을 제외한 대부분의 지점들이 석탄 및 바이오매스의 연소와 관련 있었으며, Co-PCBs의 경우 상업적인 PCBs와 관련이 있음을 알 수 있었다.
