Glucosamine Hydrochloride and N-Acetylglucosamine Influence the Response of Bovine Chondrocytes to TGF-β3 and IGF in Monolayer and Three-Dimensional Tissue Culture

  • BACKGROUND: Glucosamine hydrochloride (GlcN HCl) has been shown to inhibit cell growth and matrix synthesis, but not with N-acetyl-glucosamine (GlcNAc) supplementation. This effect might be related to an inhibition of critical growth factors (GF), or to a different metabolization of the two glucosamine derivatives. The aim of the present study was to evaluate the synergy between GlcN HCl, GlcNAc, and GF on proliferation and cartilage matrix synthesis. METHOD: Bovine chondrocytes were cultivated in monolayers for 48 h and in three-dimensional (3D) chitosan scaffolds for 30 days in perfusion bioreactors. Serum-free (SF) medium was supplemented with either growth factors (GF) $TGF-{\beta}$ ($5ng\;mL^{-1}$) and IGF-I ($10ng\;mL^{-1}$), GlcN HCl or GlcNAc at 1mM each or both. Six groups were compared according to medium supplementation: (a) SF control; (b) SF + GlcN HCl; (c) SF + GlcNAc; (d) SF + GF; (e) SF + GF + GlcN HCl; and (f) SF + GF + GlcNAc. Cell proliferation, proteoglycan, collagen I (COL1), and collagen II (COL2) synthesis were evaluated. RESULTS: The two glucosamines showed opposite effects in monolayer culture: GlcN HCl significantly reduced proliferation and GlcNAc significantly augmented cellular metabolism. In the 30 days 3D culture, the GlcN HCl added to GF stimulated cell proliferation more than when compared to GF only, but the proteoglycan synthesis was smaller than GF. However, GlcNAc added to GF improved the cell proliferation and proteoglycan synthesis more than when compared to GF and GF/GlcN HCl. The synthesis of COL1 and COL2 was observed in all groups containing GF. CONCLUSION: GlcN HCl and GlcNAc increased cell growth and stimulated COL2 synthesis in long-time 3D culture. However, only GlcNAc added to GF improved proteoglycan synthesis.

  • A simple and sensitive high performance liquid chromatographic method to quantitate ursodeoxycholic acid in crude drug pharmaceuticals was investigated. Ursodeoxycholic acid react with 2-bromoacetyl-6-methoxynaphthalene (Br-AMN) in the presence of triethylamine to form highly fluorescent derivative. The derivatization procedure was performed at 7$0^{\circ}C$ and completed within 30 min. The optimal wavelength of the fluorescence detector are λ$_{ex}$=300 nm and λ$_{em}$ = 460 nm. The LOD of the ursodeoxycholic acid was 25 ng/mι based on the S/N =3, and the LOQ was 80 ng/mι based on S/N = 10. Crude drug pharmaceuticals pretreated by solid phase extraction (Sep-pak $C_{18}$ cartridge) which were shown very good separation and recovery values for the compound.d.

  • 지난 및 년간 정부에서는 소각시설에서 배출되는 다이옥신 등 대기오염물질을 저감하기 위하여 많은 노력을 기울여 왔다. 먼저, '97년 7월 17일, 폐기물관리법 시행규칙을 개정하여 생활폐기물 소각시설에 대한 다이옥신 규제기준$^{1)}$ 을 '99년 7월 1일부터 기존 소각시설에 대해서는 0.5ng-TEQ/N㎥을 신설 소각시설에 대해서는 0.1ng-TEQ/N㎥으로 제정했으며, 또한 소각시설의 광역화, 신규 소각시설의 설치비용 및 기존 소각설비의 개ㆍ보수비용의 일부에 대한 국고지원, 소각처리 기술지원단에 의한 기술평가 및 자문활동, 다이옥신 공인측정ㆍ분석기관의 지정ㆍ고시, 생활폐기물 소각시설 설치ㆍ운영지침의 제정 등 짧은 기간임에도 불구하고 소각시설에서 배출되는 다이옥신을 저감하기 위하여 각종 정책이나 제도를 시행 또는 개선해왔다.   • A investigation of electrochemical analysis of antibiotics Neomycin ($C_{23}H_{46}N_6O_{13}$) was searched using electrochemical square wave (SW) stripping and cyclic voltammetry (CV) using working sensor of the modified carbon nanotube combination electrodes, optimum diagnostic parameters were searched by anodic stripping, final conditions were attained to working range of 1.0-14.0 ng/L, detection limit (S/N) was found to be 0.6 ng/L. The developed method was discovered to be fitting in quality control in the food, pharmaceutical and other manufacturing sectors.

  • In 1995 we licensed pyrolysis gas melting technology of indirect heating type (using kiln) from Siemens AG, and built its demonstration facility in 1998 at Clean-Park-East of Fukuoka City to demonstrate the technology for municipal solid waste (MSW). In 1997 we were awarded an order from Kanemura Co., Ltd. to build a pyrolysis gas melting and power generation plant, specifically for treating residue from car shredder. The latter was launched in 1998, and is currently in commercial operation. The operation of these plants have proven the following facts. (1) The system is capable for performing a stable operation with a wide variety of waste. (2) Pyrolysis is achieved steadily regardless of the variation in the quality of waste. (3) The system can be operated under low excess air ratio (1.2∼1.3). (4) The concentration of dioxins at the furnace outlet is 0.062ng-TEQ/㎥$\_$N/, and 0.002ng-TEQ/㎥$\_$N/, at the stack. (the value is corrected to dryO$_2$ 12%) (5) The purity of recovered metals exceeds 90%.

50세 이상 한국 성인에서 비만과 부갑상샘호르몬의 관련성 -2011 국민건강영양조사에 근거하여 (The association of abdominal obesity, obesity and parathyroid hormone in Korean adults (aged≥50 years): The Korea National Health and Nutrition Survey, 2011)

  • 본 연구는 국가자료인 2011년도 국민건강영양조사 자료를 이용하여 50세 이상 성인에서 비만이 PTH에 미치는 영향을 평가하고자 실시하였다. 조사 참여자수는 8,518명이었고, 이중 50세 이상 성인 3,305명을 최종분석 대상자로 하였다. 본 연구의 주요결과는 PTH 평균값에 영향을 주는 관련변수(연령, 성별, SBP, DBP, 25(OH)D)를 보정 후, 정상군 (BMI, < $23.0kg/m^2$; $66.25{\pm}0.89ng/dL$)에 비하여 과체중군(BMI, $23.0-24.9kg/m^2$; $67.83{\pm}1.07 ng/dL$), 비만군(BMI,${\geq}25.0kg/m^2$; $70.71{\pm}0.94ng/dL$)의 PTH 평균값이 유의하게 증가하였고(p=0.003), Non-abdominal obesity군($66.63{\pm}0.74ng/dL$)에 비하여 Abdominal obesity군($69.30{\pm}0.80ng/dL$)의 PTH 평균값이 증가하였다(p=0.017). 결론적으로 비만과 복부비만은 PTH의 증가와 관련이 있다.

소나무잎을 Passive Air Sampler로 이용하여 지역별 대기 중 다환방향족 탄화수소의 농도 및 발생원 추정 (Estimation of Atmospheric PAH Concentrations and Sources at Several Sites Using Pine Needles as a Passive Air Sampler)

  • Objectives: This study was carried out in order to estimate atmospheric polycyclic aromatic hydrocarbon (PAH) concentrations and sources using pine needles as a passive air sampler (PAS) in urban (Pyeongtaek), semirural (Anseong) and rural (Jincheon) sites. Methods: One-year-old pine needles were collected for analysis of their PAH concentrations ($C_{p,n}g/g$ dry) at the end of December. PAHs concentrations in the ambient air ($C_a$, $ng/m^3$) were calculated with a $Log(C_p/C_a)-LogK_{oa}$ correlational equation. Results: PAHs concentrations in ambient air ($C_a$) were high, in the order of urban ($114.03ng/m^3$), semirural ($105.17ng/m^3$) and rural ($61.91ng/m^3$) sites. However, distributions of PAH isomer concentrations were very similar. PAHs of which molecular weight is smaller than 228.30 (AcPy, Acp, Flu, Phen, Ant, Flt, Pyr, BaA, Chry) made up most of the PAHs in the ambient air (96.6-98.5%). Conclusion: At urban, semirural and rural sites, it was concluded that the main source of PAHs in the ambient air ratio of each PAH isomer concentration was cars, especially diesel vehicles.

염소화법에 의한 토양 중 Polychlorinated Biphenyls의 잔유분석 (Determination of Polychlorinated Biphenyls in the Soil by Perchlorination)

  • PCBs have been measured using GC-ECD, GC-MS, GC-ELCD, HPLC, TLC, NMR and Immunoassay. The analysis of PCBs using GC-ECD include the peak pattern method as none derivatization and the Perchlorination method as derivatization. This study was conducted to establish the perchlorination method with Sbcls from PCBs to decachlorinated biphenyl(DCB). The aroclor 1242 of PCBs was chlorinated and then, converted into the DCB which showed a single peak in GC-ECD chromatogram. The detection limit of DCB was 2pg. The quantification detection concentration of PCBs extracted with soxhlet was 0.5ng/g in the soil. PCBs were not detected in the suburban soil, but 174ng/g in the soil of industrial complex. Mean PCBs concentration of Shinchun stream at Kumho river and Jinchun stream at Nakdong river was calculated average 낙ngjg in 각e sediment. PCBs concentration in the sediment of Kumho river near 2-7km from conjunction with Nakdong river was average 154ng/g. PCBs concentration in the sediment of Nakdong river near conjunction with Kumho river was average 159ng/g.

Determination of 11 Phenolic Endocrine Disruptors using Gas Chromatography/Mass Spectrometry-Selected Ion Monitoring in Five Selected Wastewater Influents

  • An efficient method for the simultaneous determination of eleven phenolic endocrine-disrupting chemicals (EDCs) present in wastewater influent samples was described. The 11 phenolic EDCs including alkylphenols, chlorophenols, and bisphenol A were determined by gas chromatography/mass spectrometry-selected ion monitoring (GC/MS-SIM) following two work-up methods for comparison; isobutoxycarbonyl (isoBOC) derivatization and tert-butyldimethylsilyl (TBDMS) derivatization. The wastewater influent samples containing the 11 EDCs were adjusted to pH 2 with $H_2SO_4$ and then cleaned up with n-hexane. Next, they were subjected to solid-phase extraction (SPE) with XAD-4 resin and subsequently converted to isoBOC or TBDMS derivatives for sensitivity analysis with gas chromatography/mass spectrometry-selected ion monitoring (GC/MSSIM). Following isoBOC derivatization and TBDMS derivatization, the recoveries were 86.6-105.2% and 97.6-142.7%, the limits of quantitation (LOQ) for the 11 phenolic EDCs for SIM was 0.001-0.050 ng/mL and 0.003-0.050 ng/mL, and the SIM responses were linear with the correlation coefficient varying by 0.9717-0.9995 and 0.9842-0.9980, respectively. When these methods were applied to five selected wastewater influent samples, for isoBOC derivatization and TBDMS derivatization the ranges of concentration detected were 0.2-99.6 ng/mL and 0.4-147.4 ng/mL, respectively.

  • DNA를 고정한 탄소나노튜브 페이스트 전극으로 옥시테트라싸이클린 항생제의 분석조건을 연구하였다. 사각 파형 벗김 전압전류법과 순환 전압 전류법을 사용하여 최적조건에서 1-10 ngL?1의 정량 가능 범위에 도달하였다. 벗김법 0.1 mgL?1 농도에서 15번 반복 측정하여 0.068%의 상대표준편차였으며, 벗김법 최적 분석 조건에서 0.4 ngL?1의 검출 한계를 얻었다. 개발된 결과를 양식어류의 세포에서 응용하였다.
