Multi-epitope vaccine against drug-resistant strains of Mycobacterium tuberculosis: a proteome-wide subtraction and immunoinformatics approach

  • Mycobacterium tuberculosis (Mtb) is the causative agent of tuberculosis, one of the most deadly infections in humans. The emergence of multidrug-resistant and extensively drug-resistant Mtb strains presents a global challenge. Mtb has shown resistance to many frontline antibiotics, including rifampicin, kanamycin, isoniazid, and capreomycin. The only licensed vaccine, Bacille Calmette-Guerin, does not efficiently protect against adult pulmonary tuberculosis. Therefore, it is urgently necessary to develop new vaccines to prevent infections caused by these strains. We used a subtractive proteomics approach on 23 virulent Mtb strains and identified a conserved membrane protein (MmpL4, NP_214964.1) as both a potential drug target and vaccine candidate. MmpL4 is a non-homologous essential protein in the host and is involved in the pathogen-specific pathway. Furthermore, MmpL4 shows no homology with anti-targets and has limited homology to human gut microflora, potentially reducing the likelihood of adverse effects and cross-reactivity if therapeutics specific to this protein are developed. Subsequently, we constructed a highly soluble, safe, antigenic, and stable multi-subunit vaccine from the MmpL4 protein using immunoinformatics. Molecular dynamics simulations revealed the stability of the vaccine-bound Tolllike receptor-4 complex on a nanosecond scale, and immune simulations indicated strong primary and secondary immune responses in the host. Therefore, our study identifies a new target that could expedite the design of effective therapeutics, and the designed vaccine should be validated. Future directions include an extensive molecular interaction analysis, in silico cloning, wet-lab experiments, and evaluation and comparison of the designed candidate as both a DNA vaccine and protein vaccine.

코로나-19 감염증의 확산과 공간적 특성: 숙주, 환경적 요인, 기존 신규 감염병과의 관계 (The Outbreak of COVID-19 and Spatial Characteristics: Relationship between COVID-19 and Host, Environment and Nationally Notifiable Infectious Diseases)

  • 본 연구의 목적은 2020년 1월부터 8월까지 발생한 COVID-19 감염증 발생의 공간적 확산의 패턴을 파악하고 영향요인을 추정하고자 하는 것이다. 우리나라 시·군·구별 COVID-19 확진자를 대상으로 탐색적 공간자료분석과 공간회귀모형을 활용하여 분석하였다. 분석의 결과 COVID-19는 기존에 논의되던 감염병 출현의 요인이었던 숙주와 환경요인이 큰 영향을 미치는 것으로 나타나지 않았으며 공간적으로 인접한 지역과의 관계가 높게 나타났다. 특히 신종 감염병인 4군 감염병의 발생 패턴과 밀접한 관련이 있는 것으로 나타났다. 이 연구의 결과는 COVID-19가 새로운 감염병임에도 불구하고 기존에 발생하였던 신종 감염병의 발생과 전혀 다른 패턴이 아니라는 점을 보여준다. 이는 신종 감염병을 효과적으로 통제하기 위해서 과거 새로운 감염병이 출현하였던 공간의 특성을 지속적으로 관리해야 함을 의미한다.

  • Recent studies for the Milky Way globular clusters (GCs) have reported that most of them host multiple stellar populations. However, only a few GCs have shown abundance variations in heavy elements such as iron and calcium. These GCs, as galaxy building blocks, are important to understand the formation of the Milky Way in hierarchical merging paradigm. In this study, we report our discovery from the Ca narrow-band photometry and low-resolution spectroscopy that NGC 6273 is a new Milky Way building block candidate.

라오스에서 미기록속 Tinocallis Matsumura (노린재목: 진딧물과) 보고 (First Record of the Genus Tinocallis Matsumura (Hemiptera: Aphididae) on Lagerstromia speciosa in Laos)

  • Tinocallis Matsumura 속은 전세계 19종이 포함되는 비교적 큰 속이다. 본 연구에서는 최근 라오스에서 바나바 (부채꽃과: 배롱나무속)에 심각한 피해를 주는 알락진딧물 일종, Tinocallis himalayensis Ghosh, Ghosh & Raychaudhuri, 1971를 처음으로 발견하여 보고한다. 유시형 처녀생식세대의 암컷 형태, 생태 및 피해 사진을 함께 제시하였다.

Explicit Multicast Extension (Xcast+) for Efficient Multicast Packet Delivery

  • In this letter, we propose a new multicast scheme, named Xcast+, which is an extension of Explicit Multicast (Xcast) for an efficient delivery of multicast packets. The mechanism incorporates the host group model and a new control plane into existing Xcast, and not only does it provide the transparency of traditional multicast schemes to senders and receivers, but it also enhances the routing efficiency in networks. Since intermediate routers do not have to maintain any multicast states, it results in a more efficient and scalable mechanism to deliver multicast packets. Our simulation results show distinct performance improvements of our approach compared to Xcast, particularly as the number of receivers in a subnet increases.

  • Address Resolution Protocol (ARP) cache poisoning is a MAC layer attack that can only be carried out when an attacker is connected to the same local network as the target machines. ARP is not a new problem, but wireless network introduces a new attack point and more vulnerable to the attack. The attack on wireless network cannot be detected by current detection tool installed on wired network. In order to detect the ARP poisoning attack, there must be a ARP poisoning detection tool for wireless LAN environment.   • We report on the development of ferronematic liquid crystals, new materials that consist of a dilute suspension of ferroelectric particles in a nematic liquid crystal host. The particles share their ferroelectric properties with the nematic liquid crystal and impose a spontaneous dielectric polarization of about 10 nC/$cm^2$ to the entire medium, typical for many ferroelectric liquid crystals. As expected, these new materials have enhanced dielectric anisotropy and are sensitive to the sign of an applied electric field. The potential of their use in a number of devices are discussed.

New Korean Record of Twenty Eight Species of the Family Ichneumonidae (Hymenoptera)

  • We report twenty eight ichneumonid species new to Korea. These species belong to seven subfamilies. Among them five subfamilies, Diacritinae Townes, 1965, Microleptinae Townes, 1958, Orthocentrinae F$\ddot{o} $rster, 1869, Orthopelmatinae Schmiedeknecht, 1910, Phrudinae Townes and Townes, 1949, are newly introduced to Korean fauna. All specimens are based on the insect collection of animal systematic laboratory at the Yeungnam University Gyeongsan Campus. Photographs of habitus of newly recorded subfamilies, diagnosis of 28 species and host information are provided.

  • Braconid wasps is second largest family of Hymenoptera. It is comprising more than 18,000 described species. In Korea, a total of 910 species have been recorded to date. In the present study, nine species of the family Braconidae are recorded for the first time from Korea: Aneurobracon philippinensis (Musebeck), Cenocoelius japonicus (Watanabe), Rattana sinica He & Chen, Adelius clandestinus (Förster), Blacometeorus brevicauda (Hellen), Centistes minutus Chen & Achterberg, Centistes sylvicola Belokobylskij, Gnamptodon sichotaealinicus Belokobylskij, Cotesia urabae Austin & Allen. Diagnosis, distribution and host information for the each species are provided.

Five Species of Olethreutinae (Lepidoptera, Tortricidae) New to Korea

  • Five species of Olethreutinae: four in Olethreutini, Olethreutes manoi (Kawabe, 1987), Olethreutes exilis Falkovitsh, 1966, Olethreutes komaii (Bae, 2005), and Proschistis marmaropa (Meyrick, 1907), and one in Grapholitini, Dichrorampha canimaculata Komai, 1979, are reported from Korea for the first time. Olethreutes komai is recorded out of Japan for the first time. The genus Proschistis is new to the Korean fauna. Proschistis can be characterized by the shapes of tegumen and juxta in the male genitalia. Brief descriptions and imaginal and genital photos are provided for the species. Distribution, host plants, and taxonomic notes when necessary are given for each species.
