Current status of CRISPR/Cas9 base editor technologies and their applications in crop precision breeding

  • Plant biotechnologists have long dreamed of technologies to manipulate genes in plants at will. This dream has come true partly through the clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9 technology, which now has been used to edit genes in several important crops. However, there are many restrictions in editing a gene precisely using the CRISPR/Cas9 technology because CRISPR/Cas9 may cause deletions or additions in some regions of the target gene. Several other technologies have been developed for gene targeting and precision editing. Among these, base editors might be the most practically and efficiently used compared to others. Base editors are tools which are able to cause a transition from cytosine into thymine, or from adenine into guanine very precisely on specific sequences. Cytosine base editors basically consist of nCas9, cytosine deaminase, and uracil DNA glycosylase inhibitor (UGI). Adenine base editors consist of nCas9 and adenine deaminase. These were first developed for human cells and have since also been applied successfully to crops. Base editors have been successfully applied for productivity improvement, fortification and herbicide resistance of crops. Thus, base editor technologies start to open a new era for precision gene editing or breeding in crops and might result in revolutionary changes in crop breeding and biotechnology.

Effects of the Application of Alternative Agricultural Materials on the Growth of Rice and Water Quality in Paddy Field (벼 생육 및 수질에 대한 대체농자재의 영향)

  • The aim of this study was to apply the new agricultural technique developed for three years($'96{\sim}'98$) by demonstration at an model farm of environmental agriculture located in Bangsan-ri, Dagang-myun, Namwon-city. Rice growth of environmental farming plot, which supplied with oil cake and compost fermented with chicken manure was reduced compared to that of conventional farming plot, which supplied with chemical fertilizers. Weed control by using of Ampullarius insularus in the rice paddy field was effective compared to that of control, which treated with chemical herbicide twice. The yield of rice was decreased of 35% in both environmental and eclectic farming polt compared to that of conventional farming plot. The water quality of environmental not and eclectic farming plot were good compared to conventional farming plot. Density of water flea was high in environmental and eclectic farming plot compared to that of the conventional farming plot.

Herbicidal Activity of Newly Rice Herbicide Tefuryltrione Mixture against Sulfonylurea Resistant Weeds in Korea (설포닐우레아계 제초제 저항성 논잡초에 대한 신규 제초제 Tefuryltrione 합제의 약효 및 선택성)

  • This study was conducted to investigate the herbicidal activity against resistant biotype on sulfonylurea (SU) herbicides such as Scirpus juncoides and Monochoria vaginalis at the glasshouse and paddy rice cultivation area. In order to investigate resistance breakability against SU-resistanted annual weeds, new rice herbicides containing tefuryltrione [p-hydrophenyl pyruvate dioxygenase (HPPD) inhibitor] were tested. In both lab test and field experiment, tefuryltrione mixture, 4-HPPD inhibitor had shown excellent efficacy with a bleaching on the leaves of resistant annual weeds at early time after treatment and showed excellent persistance. Especially, tefuryltrione mixture had shown excellent controlling effect on annual and perennial SU-resistant S. juncoides in the regional field experiment. In phytotoxicity test, this tefuryltrione mixture had shown good selectivity to common rice species.

  • This study was conducted to determine the response of newly developed chlorophyllous cells against photosynthesis inhibitory herbicides in LS medium. Inhibition of the growth of the selected chlorophyllous cells in the LS medium containing sucrose 1%, NAA $10^{-5}$ M and BA $10^{-6}$ M under light condition increased as the concentrations of paraquat increased from $10^{-6}$ M to $10^{-4}$ M. The calli died in $10^{-4}$ M paraquat treatment and the inhibition of calli growth was greater when $CO_2$ was supplied. In the treatment of herbicide diuron, the inhibition of calli growth also increased as the concentrations of diuron increased from $10^{-6}$ M to $10^{-3}$ M and more inhibition was observed at 1% sucrose than 2% sucrose.

  • 3-Chloro-2-(4-chloro-2-fluoro-5-hydroxyphenyl)-4,5,6,7-tetrahydro-2H-indazole, which is the key intermediate of cyclic imide type compounds such as EK 5439 and S-275 series, were practically synthesized by the procedure of hydrolysis of by-products and were produced in the reaction of 2-(2-fluoro-4-chloro-5-hydroxyphenyl)-2,3a,4,5,6,7-hexahydroindazol-3-one with phosgene.

Water Solubilities and Vapor Pressures of Chlorothalonil and Command (농약 Chlorothalonil 과 Command 의 수용성 및 증기압)

  • The water solubilities and vapor pressures of chlorothalonil and Command were measured following the guidelines of the U.S. EPA and OECD. Water solubility of the two compounds is consistent with respective values in the literature. However, the vapor pressures of Chlorothalonil and Command were 5,000 times$(2{\times}10^{-7}\;torr\;at\;25^{\circ}C)$ and 100 times$(<1{\times}10^{-6}\;torr\;at\;45^{\circ}C)$ lower than the literature values, respectively. Courteous use of the vapor pressure values in the handbooks is suggested. With this study, experimental difficulties involved were recognized. Based on the low vapor pressure of Command, the cause of the accidental bleach of non-target plants in the United States might not be attributed to the volatilization of Command, but to the drift during the application of the herbicide. These approaches will be utilized to predict the environmental fate of new chemicals under development, to screen the potential environmental pollutants among chemicals already in use, and to assess measures to minimize the hazards.

2D-QSAR and HQSAR Analysis on the Herbicidal Activity and Reactivity of New O,O-dialkyl-1-phenoxy-acetoxy-1-methylphosphonate Analogues (새로운 O,O-dialkyl-1-phenoxyacetoxy-1-methylphosphonate 유도체들의 반응성과 제초활성에 관한 2D-QSAR 및 HQSAR 분석)

  • Quantitative structure-activity relationships (QSARs) on the pre-emergency herbicidal activity and reactivity of a series of new O,O-dialkyl-1-phenoxyacetoxy-1-methylphosphonates (S) analogues against seed of cucumber (Cucumus Sativa) were discussed quantitatively using 2D-QSAR and HQSAR methods. The statistical values of HQSAR model were better than that of 2D-QSAR model. From the frontier molecular orbital (FMO) interaction between substrate molecule (S) and $BH^+$ ion (I) in PDH enzyme, the electrophilic reaction was superior in reactivity. From the effect of substituents, $R_2$-groups in substrate molecule (S) contributed to electrophilic reaction with carbonyl oxygen atom while X, Y-groups contributed to nucleophilic reaction with carbonyl carbon atom. And the influence of X,Y-groups was more effective than that of $R_2$-groups. As a results of 2D-QSAR model (I & II) and atomic contribution maps with HQSAR model, the more length of X, Y-groups is longer, the more herbicidal activity tends to increased. And also, the optimal ${\epsilon}LUMO$ energy, $({\epsilon}LUMO)_{opt.}$=-0.479 (e.v.) of substrate molecule is important factor in determining the herbicidal activity. It is predicted that the herbicidal activity proceeds through a nucleophilic reaction. From the analytical results of 2D-QSAR and HQSAR model, it is suggested that the structural distinctions and descriptors that contribute to herbicidal activities will be able to applied new herbicide design.

Responses of Tobacco Photomixotrophic Cultured Cells to Various Herbicides (다양한 제초제에 대한 담배 Photomixotrophic 배양세포의 반응)

  • To establish an efficient screening system for new herbicides using plant cultured cells, responses of tobacco photomixotrophic cultured (PH) cells to various herbicides with different modes of action were surveyed by measuring the cell growth and ion conductivity in medium. The cells were cultured in Murashige and Skoog (MS) medium containing 0.7mg/L 2,4-D, 0.3mg/L kinetin and 30 g/L sucrose at $25^{\circ}C$ in the light (100 rpm). Chemicals were treated to suspension cultures of tobacco PH cells at the time of subculture. The cell growth and ion conductivity in the medium were investigated on 12 days after chemical treatment. The ion conductivity assay gave well correlated results to the cell growth inhibition data. The responses of tobacco PM cells were dependent on the modes of action of chemicals tested. Atrazine, an inhibitor of photosynthetic electron transport (PET), strongly inhibited both the cell membrane and cell growth ($IC_{50}$/, about 1 $\mu$M). Butachlor (an inhibitor of cell division), glufosinate (an inhibitor of amino acid biosynthesis), and fluridone (an inhibitor of carotenoid biosynthesis) showed a dose-dependent inhibition. However, Quinclorac, a herbicide with an auxin activity, did not affect the cell growth and ion leakage. These results suggested that tobacco PM cells is suitable materials for the simple screening of new herbicides such as PET, amino acid biosynthesis, ceil division inhibitors by measuring the cell growth and ion conductivity.

Fluorescence Assay for High Efficient Mass Screening of the Herbicides Inducing Rapid Membrane Peroxidation (막과산화를 신속히 유발하는 제초제의 고효율 대량스크리닝을 위한 형광검정법)

  • This study was conducted to establish a fluorescence assay system for high efficient mass screening of the herbicides causing rapid membrane peroxidation, based on the fact that peroxide in cellular leakage could be fluorometrically determined through the fuorescent compounds formed after reacting with homovanillic acid (HVA) and peroxidase (HRP). The assay procesure established in this study was as follows. Only single disc (4 mm diameter) excised from cucumber cotyledon is placed on the well containing test solution ($200{\mu}L$) with 96-well microplate. The plate is shaking-incubated for 8 h under light condition. Then after removing the cucumber disc, HVA and HRP are supplied in the medium buffer and incubated for 5 min at room temperature. Fluorescence values are determined at Ex 320 nm/Ex 425 nm. The higher fluorescence values are obtained in the treatment of chemical having higher herbicidal activity. Using this assay with 96-well microplates, a large number of herbicides inducing rapid membrane peroxidation seemed to be screened more efficiently than spectrophotometric microtiter assay reported previously.

Development of an Official Analytical Method for Determination of Aclonifen in Agricultural Products Using GC-ECD (GC-ECD를 이용한 농산물 중 제초제 aclonifen의 공정분석법 확립)

  • BACKGROUND: Aclonifen is used as a systemic and selective herbicide to control a wide spectrum broad-leaf weeds by inhibition carotenoid biosynthesis, and then its MRLs(Maximum Residue Limits) will be determined in onion and garlic. In this study, a new official method was developed for aclonifen determination in agricultural products to routinely inspect the violation of MRL as well as to evaluate the terminal residue level. METHODS AND RESULTS: Aclonifen was extracted from crop samples with acetone and the extract was partitioned with dichloromethane and then purified by silica solid phase extraction(SPE) cartridge. The purified samples were detected GC using an ECD detector. Limits of detection(LOD) was 0.001 mg/kg and quantification(LOQ) was 0.005 mg/kg, respectively. For validation purposes, recovery studies were carried out at three different concentration levels (LOQ, $10{\times}LOQ$, $50{\times}LOQ$, n=5). The recoveries were ranged from 74.3 to 95.0% with relative standard deviations(RSDs) of less than 8%. All values were consistent with the criteria ranges requested in the Codex guidelines(CAC/GL 40). CONCLUSION: The proposed analytical method was accurate, effective and sensitive for aclonifen determination and it will be used to as an official method in Korea.
