Quantification of Metabolic Alterations of Dorsolateral Pre-Frontal Cortex in Depression SD Rat by MR Spectroscopy

  • Purpose: Contrary to the human study, it has rarely investigated metabolic alterations in the dorsolateral prefrontal cortex (DLPFC) of depressed rats versus age and sex-matched controls using proton magnetic resonance spectroscopy (MRS). Thus, the purpose of this research was to verify the feasibility of metabolic differences between the normal rat and the depression model rat. Materials and Methods: A homogeneous group of 20 SD male rats was used for MRI and in vivo 1H MRS. To induce a depressed status in SD rats, we performed the forced swimming test (FST). Using image-guide, water suppressed in vivo 1H MRS with 4.7 T MRI/MRS system, NAA/Cr and Cho/Cr ratios were mainly measured between depressed rats and normal subjects. Results: In depressed rats, increased Cho/Cr ratio was measured versus control subjects. However, no significant group effect for NAA/Cr was observed between case-control pairs. Discussion and Conclusions: The present 1H MRS study shows significant brain metabolic alterations of dorsolateral prefrontal cortex with experimental depressed status of SD rat induced by FST compared to normal subjects. This result provides new evidence that in vivo 1 H MRS may be a useful modality for detecting localized functional neurochemical markers alterations in left DLPFC in SD rats.

Nutritional Status of the Long-lived Elderly People in Kyungpook Sung-Ju Area(I) -Estimation of Nutrients Intakes- (경북 성주지역 장수노인의 영양상태(I) -영양섭취상태-)

  • The purposes of this study were to estimate nutritional intakes of the long-lived elderly and to obtain the data for establishing dietary guidelines that may be recommended for the general population for the sake of longevity. The subjects of the study were 300 elederly people of age over 85 years living in Kyungpook Sung-Ju area who had no problem in daily living. Four times of food consumption survey were carried out seasonally by the repeated 24-hr recall method for one year. Mean daily energy intakes and RDA percentage of energy intakes of the male and female subjects were estimated as 1222 kcal(67.9%) and 1047 kcal(65.4%) respectively. Mean daily intakes of nutrients were estimated as 38.3g for protein, 287mg for calcium, 5.8mg for iron, 314R.E. for vitamin A, 0.6mg for vitamin B1 and 0.43mg for vitamin B2 The mean RDA percentages of nutrients intake were 64.4% and 59.8% for protein in male and female, 39.8% for Ca, 48.3% for Fe, 44.9% for vitamin A, 60% for vitamin B1, and 35.8% for B2. The average PFC ratio of energy-yielding nutrients throughout the year in male and female were 15.1 : 15.2 : 69.7 and 13.8 : 13.2 : 73.0 respectively. The mean daily intakes of energy and most of nutreints were significantly high in winter season. The contribution of plant food sources to nutrient intakes were over 60% for protein and fat, 50% for calcium, and 70% for iron. Long-lived elderly people an Sung-Ju, Kyungpook showed considerably smaller physiques compared to the avergae Korean elderly, however their average BMI fell in normal range. The subjects were consuming much less energy and nutrients compared to the present Korean RDA for the elderly over age 75. The subject showed relatively good health state in spite of low intakes of energy and nutrients. Therefore it seems to be necessary to establish a set of new RDA for the elderly over age 8.5.

Analysis of Heavy Metals in the Hair of Children with Attention-Deficit Hyperactivity Disorder and Tourette's Syndrome (주의력결핍 과잉행동장애와 뚜렛증후군 아동의 모발 중금속 분석)

  • Objectives : The purpose of this study was to examine the association of exposures to heavy metals with positive diagnosis for attention-deficit hyperactivity disorder (ADHD) and Tourette's syndrome (TS). Methods : Study participants included 27 children diagnosed with ADHD ($9.9{\pm}2.9$ years of age), 21 diagnosed with Tourette's disorder ($10.7{\pm}2.2$ years of age), and 45 normal control children ($9.6{\pm}0.5$ years of age). A Perkin-Elmer mass spectrometer was used to measure the concentrations of 5 heavy metals (Pb, Cd, U, Be, Hg) in hair samples obtained from each participant. Each heavy metal concentration was compared among the groups by use of a Kruskal-Wallis test. Results : The levels of lead (p=.006) and cadmium (p=.037) observed in the hair of children diagnosed with ADHD were significantly higher than those found in the control subjects. There were no significant differences observed for heavy metal levels when comparing TS and control subjects. Conclusion : We confirmed that lead exposure is a risk factor for ADHD. We also identified that cadmium may be a new candidate risk factor for manifestation of ADHD. We did not find an association between heavy metals and manifestation of TS.

A Study on Diagnostic Criteria of Noise-Induced Hearing Loss among Workers in an Iron Foundry (철강공장 근로자를 대상으로 살펴본 소음성 난청 진단기준에 관한 조사)

  • This study was carried out to evaluate diagnostic criteria of noise-induced hearing loss (NIHL) among-workers in an iron foundry. Of 1,093 workers under the observation of noise-specific health examination, 184 workers were selected by way of first and second screening audiometric examination. A questionnaire survey, otological examinations, Rinne test and audiometric test were performed and the results were as follows ; The degree of hearing impairment in the left ear was more severe than in the right ear (p<0.05). The difference between hearing threshold of the first and the second hearing test at 1,000 Hz was about 5 dB with a narrow range of deviations while the difference at 4,000 Hz was about -7 dB with a wide range. Of the total study workers, 84.8% were tested within 15 hours away from noise exposure, and the rest after 16 hours. This study has identified that mean hearing loss at 4,000 Hz showed a significant statistical difference among the two study groups while mean hearing loss by 4-divided classification did not. The same phenomena were observed between the group with and without tinnitus and between the group with and without difficulty in hearing (p<0.05). Among 184 workers, 10 workers (5.4%) diagnosed as NIHL by old diagnostic criteria in contrast to 150 workers diagnosed as NIHL by the new diagnostic criteria. There was a significant difference between the two groups in the average hearing loss at 4,000 Hz and 4-divided classification (p<0.01), but there were no significant differences in age, the duration of employment, blood pressure and the duration wearing the personal hearing protector (p>0.05). If we apply Early Loss Index (ELI) method, some workers in younger age group diagnosed as NIHL by the new diagnostic criteria were fallen into within the normal range. In the mean time older age group show reverse results in contrast to the above finding. It is too early to confirm the value of the usage of the new diagnostic criteria in hearing examination. Further study is called for to verify the value of this criteria.

Gut Microbiota Community and Its Assembly Associated with Age and Diet in Chinese Centenarians

  • Increasing evidence suggests that gut microbiota underpin the development of health and longevity. However, our understanding of what influences the composition of this community of the longevous has not been adequately described. Therefore, illumina sequencing analysis was performed on the gut microbiota of centenarians (aged 100-108 years; RC) and younger elderlies (aged 85-99 years; RE) living in Bama County, Guangxi, China and the elderlies (aged 80-92 years; CE) living in Nanning City, Guangxi, China. In addition, their diet was monitored using a semiquantitative dietary questionary (FFQ 23). The results revealed the abundance of Roseburia and Escherichia was significantly greater, whereas that of Lactobacillus, Faecalibacterium, Parabacteroides, Butyricimonas, Coprococcus, Megamonas, Mitsuokella, Sutterella, and Akkermansia was significantly less in centenarians at the genus level. Both clustering analysis and UniFraq distance analysis showed structural segregation with age and diet among the three populations. Using partial least square discriminate analysis and redundancy analysis, we identified 33 and 34 operational taxonomic units (OTUs) as key OTUs that were significantly associated with age and diet, respectively. Age-related OTUs were characterized as Ruminococcaceae, Clostridiaceae, and Lachnospiraceae, and the former two were increased in the centenarians; diet-related OTUs were classified as Bacteroidales, Lachnospiraceae, and Ruminococcaceae. The former two were deceased, whereas the later one was increased, in the high-fiber diet. The age and high-fiber diet were concomitant with changes in the gut microbiota of centenarians, suggesting that age and high-fiber diet can establish a new structurally balanced architecture of gut microbiota that may benefit the health of centenarians.

  • Objective : Response time plays a prominent part in research on cognitive ability and the aging effect. This study aimed to identify the impact of cognitive ability on information processing by conducting cognitive response time (CRT) using a computer program. Methods : This study was conducted in 30 normal elderly (NE) and 30 elderly with amnestic MCI (aMCI), aged 65-79 years old living in Daegu and Gyeongbuk. The results were analyzed using the statistical analysis program R 4.0.2 (University of Auckland, New Zealand). Results : In the three sub-areas of CRT, the total response time showed a significant difference depending on group or age, and the error rate showed a significant difference depending on age or group in some sub-areas. In the aMCI group, the performance of CRT significantly correlated with that of the overall cognition and memory test. Conclusion : Information processing depending on aging or cognitive ability and the differential performance of processing speed could be observed through CRT. The performance of this test was found to be significantly correlated with that of the overall cognition and memory test. Therefore, CRT could be used meaningfully as a simplified tool to predict the initial cognitive disorder of the elderly in the community.

F-Wave Analysis in Patients with Clinically Diagnosed Carpal Tunnel Syndrome (임상적으로 진단된 수근관증후군 환자에서 F파 분석)

  • Background and Objective : Carpal tunnel syndrome (CTS) is the most common mononeuropathy encountered in clinical practice. No single procedure or group of procedures has demonstrated adequate sensitivity. F-wave study in CTS is very rarely reported. To determine the diagnostic usefulness of new parameters of F-wave and comparative study of F-wave parameters of median and ulnar nerves in patients with CTS. Methods : F-wave responses of median and ulnar nerves were analyzed from 27 patients with clinically diagnosed CTS and 22 age and gender-matched normal control. Conventional F-wave parameters were studied. Also, the usefulness of new parameters such as mean and maximal ulnar-median F-wave latency differences, ulnar-median F-wave persistence and chronodispersion differences, median/ulnar F-wave amplitude ratio, and F-wave conduction velocity (FCV) using mean and maximal latency were assessed. Results : Compared with controls, median F-wave minimal, maximal and mean latencies, mean F-wave amplitude/M-wave amplitude, minimal, mean and maximal ulnar-median F-wave latency differences, and FCVs using minimal, maximal and mean latency were significant (P<0.05~0.001). Median F-wave minimal, maximal and mean latencies, mean ulnar-median F-wave latency difference, and FCVs using minimal, maximal and mean latency showed high sensitivity and specificity. Mean ulnar-median F-wave latency difference and FCVs using maximal and mean latency were new parameters. Conclusion : New F-wave parameter including mean ulnar- median F-wave latency difference and FCVs using maximal and mean latency may be a useful to assess the CTS. Also, median F-wave minimal, maximal and mean latencies, and FCV using minimal latency may be included in routine diagnostic tests in CTS.

Autophagy-related protein LC3 and Beclin-1 in the first trimester of pregnancy

  • Autophagy is a degradation process that acts in response to environmental stressors. Recently, autophagy has been detected in normal term, preeclamptic and intrauterine growth-restricted placentas. The object of this work was to investigate the presence of autophagy in first trimester voluntary interruption of pregnancy placental villi by the expression of autophagy-related proteins, light chain 3 (LC3), and Beclin-1. In first trimester placental villi laser scanning confocal microscopy (LSCM) analysis revealed LC3 and Beclin-1 immunoreactivity prevalently located in villous cytotrophoblasts. Using LSCM, LC3, and Beclin-1 were localized to the cytoplasm of the trophoblast layer in human full-term placentas. Beclin-1 expression and LC3 activation were confirmed by western blotting. These data emphasize that autophagy activation is different among cytotrophoblasts and syncytiotrophoblasts depending on the gestational age and thus we speculate that autophagy might play a prosurvival role throughout human pregnancy.

Development of New Balance Evaluation Index through Biomechanical Verification for Healthy Elderly (운동역학적 검증을 통한 건강한 노인의 새로운 균형평가지표 개발)

  • Objective: The purpose of this study is to develop new balance evaluation index that can discriminate fall risk factors and provide effective interventions for healthy elderly. In order to conduct this study, the balance assessment tools (TUG, mCTSIB, OLST, FRT and BBS) currently used in clinic were re-evaluated using biomechanical analysis. Method: The participants were healthy elderly people over 65 years old, n=26, age: 69.31±3.13 years; height: 154.00±4.12 cm, body weight: 56.13±6.04 kg. The variables are length of CoM-BoS, length of CoP-BoS, range of CoP, mean distance of CoP, mean frequency of CoP, root mean square of CoP, joint angle, ASM (%SL), CoP-CoM angle. Results: As a result of this study, the following items were included in the list of new balance evaluation index for the healthy elderly, showing differences in the biomechanical evaluation based on the clinical evaluation (Inclusion list: TUG, OLST, 8th assessment item of BBS (reaching forward with outstretched arm), 11th item (turning 360 degrees), 13th item (standing with one foot in front), 14th item (standing on one foot)). Conclusion: Based on the results, the new balance evaluation index for the healthy elderly determined through this study can be used to prevent the fall by evaluating the balance ability in various situations that can be experienced in the normal daily life of the healthy elderly.

Development of Prediction Equation of Diffusing Capacity of Lung for Koreans

  • Background: The diffusing capacity of the lung is influenced by multiple factors such as age, sex, height, weight, ethnicity and smoking status. Although a prediction equation for the diffusing capacity of Korea was proposed in the mid-1980s, this equation is not used currently. The aim of this study was to develop a new prediction equation for the diffusing capacity for Koreans. Methods: Using the data of the Korean National Health and Nutrition Examination Survey, a total of 140 nonsmokers with normal chest X-rays were enrolled in this study. Results: Using linear regression analysis, a new predicting equation for diffusing capacity was developed. For men, the following new equations were developed: carbon monoxide diffusing capacity (DLco)=-10.4433-0.1434${\times}$age (year)+0.2482${\times}$heights (cm); DLco/alveolar volume (VA)=6.01507-0.02374${\times}$age (year)-0.00233${\times}$heights (cm). For women the prediction equations were described as followed: DLco=-12.8895-0.0532${\times}$age (year)+0.2145${\times}$heights (cm) and DLco/VA=7.69516-0.02219${\times}$age (year)-0.01377${\times}$heights (cm). All equations were internally validated by k-fold cross validation method. Conclusion: In this study, we developed new prediction equations for the diffusing capacity of the lungs of Koreans. A further study is needed to validate the new predicting equation for diffusing capacity.
