Objectives: The reproductive health of female workers has been extensively investigated in South Korea and other countries worldwide. However, few studies have discussed the reproductive health of male workers. In this study, we reviewed the recent literature that reports on the effects of occupational exposure on the reproductive health of male workers and the health of their children. Methods: In May 2020 we used the PubMed search engine to search the literature over the last 10 years and chose case-control, cohort, and cross-sectional studies and reviews. We selected epidemiological studies that investigated the association between pre-pregnant occupational exposure and the reproductive health of male workers and the health of their children. We excluded case reports, non-epidemiological studies (animal experiments, cellular-level experiments, and similar articles), and studies that described postnatal occupational exposure. Results: We eventually selected 23 studies. The studies that included exclusively male workers reported that those employed in the agricultural sector or those exposed to pesticides showed lower blood levels of reproductive hormones and a high risk of lympho-hematopoietic system cancer in their children. Male workers exposed to complex organic solvents and organic compounds showed a high risk of poor semen quality, increased time to pregnancy, decreased blood levels of reproductive hormones, and a high risk of lympho-hematopoietic system cancer in their children. Male workers employed in occupations that involved significant social contact, or in the leather and livestock industries, and in occupations with high levels of exposure to lead and organic solvents showed a high risk of malignancies, including lympho-hematopoietic system cancer, neuroblastoma, and central nervous system tumors in their children. Studies that investigated both male and female workers reported that children of male smelters showed a high risk of premature birth, and children of male workers exposed to metals showed a high risk of hypospadias and cryptorchidism. Children of male welders and workers employed in the glass, ceramic, and tile industries showed a high risk of premature birth. Conclusion: The findings of this study will serve as basic data for further research on male workers' reproductive health and provide a scientific basis for the development of strategies to protect the reproductive health of males employed in high-risk occupations. Moreover, the results of this study may provide guidelines to improve the understanding of and knowledge on male workers' reproductive health.
Aim: This study investigates the effects of ginsenoside Rb1 (GsRb1) on methamphetamine (METH)-induced toxicity in SH-SY5Y neuroblastoma cells and METH-induced conditioned place preference (CPP) in adult Sprague-Dawley rats. It also examines whether GsRb1 can regulate these effects through the NR2B/ERK/CREB/BDNF signaling pathways. Methods: SH-SY5Y cells were pretreated with GsRb1 (20 mM and 40 mM) for 1 h, followed by METH treatment (2 mM) for 24 h. Rats were treated with METH (2 mg/kg) or saline on alternating days for 10 days to allow CPP to be examined. GsRb1 (5, 10, and 20 mg/kg) was injected intraperitoneally 1 h before METH or saline. Western blot was used to examine the protein expression of NR2B, ERK, P-ERK, CREB, P-CREB, and BDNF in the SH-SY5Y cells and the rats' hippocampus, nucleus accumbens (NAc), and prefrontal cortex (PFC). Results: METH dose-dependently reduced the viability of SH-SY5Y cells. Pretreatment of cells with 40 µM of GsRb1 increased cell viability and reduced the expression of METH-induced NR2B, p-ERK, p-CREB and BDNF. GsRb1 also attenuated the expression of METH CPP in a dose-dependent manner in rats. Further, GsRb1 dose-dependently reduced the expression of METH-induced NR2B, p-ERK, p-CREB, and BDNF in the PFC, hippocampus, and NAc of rats. Conclusion: GsRb1 regulated METH-induced neurotoxicity in vitro and METH-induced CPP through the NR2B/ERK/CREB/BDNF regulatory pathway. GsRb1 could be a therapeutic target for treating METH-induced neurotoxicity or METH addiction.
본 연구에서는 15-20 cm 길이의 더덕순을 70% 에탄올로 추출하여 추출물 (CLBE)을 제조하고, H2O2로 산화적 스트레스를 유발한 SH-SY5Y세포에 전처리하여 신경세포 보호 효과를 평가하였다. 그 결과, CLBE는 H2O2로 자극된 세포에서 세포 손상 및 LDH 방출 억제, ROS 소거를 통하여 세포의 사멸을 막아주었다. 또한 CLBE는 Bcl-2와 Bax 단백질의 발현을 조절함으로써 caspase의 활성을 억제하여 신경세포를 보호하였다. 이상의 연구결과들을 종합할 때, CLBE는 산화적 스트레스에 대하여 신경세포 보호 효과가 있는 것으로 보이며, 향후 신경질환 연구를 위한 치료제 개발 및 고부가가치 식품 소재 개발에 유용하게 사용될 수 있을 것으로 판단된다.
Glycogen synthase kinase-3β (GSK-3β) is an important serine/threonine kinase that implicates in multiple cellular processes and links with the neurodegenerative diseases including Alzheimer's disease (AD). In this study, structure-based virtual screening was performed to search database for compounds targeting GSK-3β from Enamine's screening collection. Of the top-ranked compounds, 7 primary hits underwent a luminescent kinase assay and a cell assay using human neuroblastoma SH-SY5Y cells expressing Tau repeat domain (TauRD) with pro-aggregant mutation ΔK280. In the kinase assay for these 7 compounds, residual GSK-3β activities ranged from 36.1% to 90.0% were detected at the IC50 of SB-216763. In the cell assay, only compounds VB-030 and VB-037 reduced Tau aggregation in SH-SY5Y cells expressing ΔK280 TauRD-DsRed folding reporter. In SH-SY5Y cells expressing ΔK280 TauRD, neither VB-030 nor VB-037 increased expression of GSK-3α Ser21 or GSK-3β Ser9. Among extracellular signal-regulated kinase (ERK), AKT serine/threonine kinase 1 (AKT), mitogen-activated protein kinase 14 (P38) and mitogenactivated protein kinase 8 (JNK) which modulate Tau phosphorylation, VB-037 attenuated active phosphorylation of P38 Thr180/ Tyr182, whereas VB-030 had no effect on the phosphorylation status of ERK, AKT, P38 or JNK. However, both VB-030 and VB-037 reduced endogenous Tau phosphorylation at Ser202, Thr231, Ser396 and Ser404 in neuronally differentiated SH-SY5Y expressing ΔK280 TauRD. In addition, VB-030 and VB-037 further improved neuronal survival and/or neurite length and branch in mouse hippocampal primary culture under Tau cytotoxicity. Overall, through inhibiting GSK-3β kinase activity and/or p-P38 (Thr180/Tyr182), both compounds may serve as promising candidates to reduce Tau aggregation/cytotoxicity for AD treatment.
We recently reported that Gastrodia elata extracts (GEE) had an effects to protect against lipopolysaccharide-induced cognitive impairment in vivo model. In this study, we investigated the neuroprotective effects and the mechanism of action of GEE in hydrogen peroxide (H2O2)-induced cell death of SH-SY5Y human neuroblastoma cell. The SH-SY5Y cells were divided into five groups, including control(non-treated group), 100 μM H2O2, 100, 200, 500 ㎍/㎖ GEE+ 100 μM H2O2 groups. Pre- and co-treatment with GEE prevented cell death induced by 100 μM H2O2 for 24 h in SH-SY5Y cells. Our findings also showed that anti-oxidants enzymes (Cu/Zn superoxide dismutase, Mn superoxide dismutase, catalase) were up-regulated by 100 μM H2O2. But GEE suppressed H2O2-induced anti-oxidants enzymes decrease in a dose-dependent manner. Treatment with GEE also inhibited phosphorylation of eukaryotic initiation factor-2α (eIF-2α) and p38 by H2O2. Taken together, the neuroprotective effects of GEE in terms of recovery of antioxidant enzymes expression, down-regulation of eIF-2α and p38 phosphorylation, and inhibition of cell death are associated with reduced oxidative stress in SH-SY5Y cells.
BACKGROUND/OBJECTIVES: Oxidative stress is a fundamental neurodegenerative disease trigger that damages and decimates nerve cells. Neurodegenerative diseases are chronic central nervous system disorders that progress and result from neuronal degradation and loss. Recent studies have extensively focused on neurodegenerative disease treatment and prevention using dietary compounds. Heseperetin is an aglycone hesperidin form with various physiological activities, such as anti-inflammation, antioxidant, and antitumor. However, few studies have considered hesperetin's neuroprotective effects and mechanisms; thus, our study investigated this in hydrogen peroxide (H2O2)-treated SH-SY5Y cells. MATERIALS/METHODS: SH-SY5Y cells were treated with H2O2 (400 µM) in hesperetin absence or presence (10-40 µM) for 24 h. Three-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide assays detected cell viability, and 4',6-diamidino-2-phenylindole staining allowed us to observe nuclear morphology changes such as chromatin condensation and apoptotic nuclei. Reactive oxygen species (ROS) detection assays measured intracellular ROS production; Griess reaction assays assessed nitric oxide (NO) production. Western blotting and quantitative polymerase chain reactions quantified corresponding mRNA and proteins. RESULTS: Subsequent experiments utilized various non-toxic hesperetin concentrations, establishing that hesperetin notably decreased intracellular ROS and NO production in H2O2-treated SH-SY5Y cells (P < 0.05). Furthermore, hesperetin inhibited H2O2-induced inflammation-related gene expression, including interluekin-6, tumor necrosis factor-α, and nuclear factor kappa B (NF-κB) p65 activation. In addition, hesperetin inhibited NF-κB translocation into H2O2-treated SH-SY5Y cell nuclei and suppressed mitogen-activated protein kinase protein expression, an essential apoptotic cell death regulator. Various apoptosis hallmarks, including shrinkage and nuclear condensation in H2O2-treated cells, were suppressed dose-dependently. Additionally, hesperetin treatment down-regulated Bax/Bcl-2 expression ratios and activated AMP-activated protein kinase-mammalian target of rapamycin autophagy pathways. CONCLUSION: These results substantiate that hesperetin activates autophagy and inhibits apoptosis and inflammation. Hesperetin is a potentially potent dietary agent that reduces neurodegenerative disease onset, progression, and prevention.
목 적: 항산화 효소인 peroxiredoxin (Prx) I과 II가 유해자극에 의해서 유발되는 세포내 반응성 산소족(reactive oxygen species, ROS)에 대한 방어기전에 관여하는지 알아보고자 이 연구를 시행하였다. 대상 및 방법: SK-N-BE2C (신경아세포종세포)와 Rat2 (섬유모세포)에서 PrxI과 PrxII 발현을 보기 위하여 방사선조사, cisplatin 단독투여 및 cisplatin-방사선조사병합투여 후에 PrxI과 PrxII에 대한 western blot을 시행하였다. 또한 N-acetyl-L-cysteine (NAC)에 의하여 PrxI과 PrxII 발현에 미치는 영향과 세포생존율을 함께 조사하였다. 두 종류 세포에 방사선조사, 다양한 농도의 cisplatin을 단독투여 및 방사선조사와 병합투여 시 생존율을 각각 분석하였고 SK-N-BE2C의 각 군에서 시간별 생존율을 관찰하였다. 결 과: PrxI의 발현은 SK-N-BE2C에서 방사선조사 후 60분까지 증가하였으나 NAC 전처치한 경우 방사선조사 후 60분에서는 대조군에 비하여 약간 증가하였다. Rat2에서는 방사선조사 후 NAC 전처치 여부에 관계없이 증가하지 않았다. PrxII의 발현은 두 가지 세포 모두에서 방사선조사와 NAC 전처치 여부에 관계없이 증가하지 않았다. SK-N-BE2C와 Rat2에서 다양한 농도의 cisplatin 단독투여나 cisplatin-방사선조사병합시에는 PrxI 및 PrxII의 발현은 증가하지 않았다. SK-N-BE2C와 Rat2에서 NAC 전처치 여부에 따른 방사선조사군 및 cisplatin-방사선조사병합군의 생존율을 각각 비교한 결과 PrxI의 발현이 증가되었고, NAC 전처치하였으며 cisplatin의 농도가 낮을수록 유의하게 생존율이 높았다. SK-N-BE2C에서 NAC를 전처치한 방사선조사군의 생존율이 NAC 전처치 안한 방사선조사군에 비하여 높은 경향만 보였으나, Rat2에서는 유의한 차이로 NAC를 전처치한 방사선조사군의 생존율이 높았다. SK-N-BE2C에서 시간별 생존율을 측정한 결과는 방사선조사군과 cisplatin-방사선조사병합군을 비교하면 방사선조사군이 빠른 세포생존율의 감소를 보였으며 12시간 때에 최대의 차이를 보였으나 48시간에서는 cisplatin-방사선조사병합군의 세포생존율이 유의하게 낮았다. 결 론: 방사선조사로 반응성 산소족이 증가되면 PrxI 발현이 증가되었으며 반응성 산소족 청소제인 NAC의 전처치에 의하여 PrxI의 발현이 감소되었다. Cisplatin은 PrxI의 발현을 억제하여 반응성 산소족에 의한 세포손상을 증가시키며 방사선으로 인한 세포치사효과를 증가시켰다고 판단된다. 이상의 결과로 PrxI의 발현여부가 방사선조사나 cisplatin의 세포치사기전에 부분적으로 관여하고 있을 것이라고 생각한다.
목 적 : 노로바이러스는 바이러스성 설사의 흔한 원인이며, 면역 저하 환자에서도 심각한 감염증을 유발하는 것으로 알려져 있어, 소아 암환자에서 노로바이러스 감염의 임상 양상을 알아보고자 하였다. 방 법: 2008년 11월부터 2009년 9월까지 삼성 서울병원에 입원한 환아 중 대변에서 노로바이러스 PCR 양성을 보였던 소아 암환자를 대상으로 하였고, 이들의 의무기록을 후향적으로 분석하였다. 결 과:연구에 포함된 환자는 총 10명이었으며, 이들 연령의 중앙값은 0.83세, 남녀의 비율은 1.5대 1이었다. 기저질환은 혈액암 4명, 신경모세포종 4명, 뇌종양 2명이었고, 감염 진단 당시 조혈모세포이식 수여자는 4명이었다. 모든 환자에서 설사 증상을 보였으며, 설사 횟수의 중앙값은 8.5회/일, 바이러스 배출 기간의 중앙값은 72.5일이었다. 장기종 소견을 보인 4명의 환자는 금식 및 총정맥 영양을 투여 받았으며, 노로바이러스 관련 사망은 관찰되지 않았다. 결 론 : 노로바이러스 감염은 소아 암환자에서 다양한 임상 증상을 유발 할 수 있으며, 이들 면역저하 환자에서 바이러스 배출 기간이 매우 길어, 설사가 있는 암환자에서 고려해야 할 바이러스 감염이다.
본 연구에서는 다래 순 아세트산에틸 분획물(ethyl acetate fraction from Actiniadin arguta sprout; EFAS)의 in vitro 산화방지활성 및 고포도당으로 인한 산화적 스트레스로부터의 뇌신경세포 보호 효과를 검증하였다. EFAS은 상대적으로 우수한 총 폴리페놀(246.25 mg GAE/g) 및 총 플라보노이드(571.59 mg RE/g) 함량을 나타내었으며, 이로 인한 ABTS 라디칼 제거 활성($IC_{50}=252.00{\mu}g/mL$) 및 MDA 생성 억제 활성($IC_{50}=42.70{\mu}g/mL$)을 보여주었다. EFAS의 in vitro 항당뇨 활성은 알파글루코사이드 가수분해효소와 최종당화산물 생성 억제 활성을 통해 측정하였으며 각각 99.18과 $85.19{\mu}g/mL$의 $IC_{50}$ 값을 나타냈고, 뇌신경 전달물질인 아세틸콜린의 세포 내 유지 효과를 아세틸콜린 분해효소 억제 활성을 통해 측정하였을 때 $164.20{\mu}g/mL$의 $IC_{50}$ 값을 나타냈다. 또한 인간 유래 MC-IXC 뇌신경세포에 과산화수소와 고포도당을 처리하여 산화적 스트레스를 유발시켜 뇌신경세포 사멸에 대해 우수한 보호효과를 보여주었다. 마지막으로 HPLC를 통하여, EFAS의 주요 생리활성 물질을 분석한 결과는 루틴으로 유추되었으며, 그 함량은 $123.76{\mu}g/mL$ of dried weight로 분석되었다. 이상의 결과로부터 다래 순 분획물(EFAS)이 알파글루코사이드 가수분해효소 및 최종당화산물 생성 억제를 통해 식후 혈당의 급격한 상승을 예방할 수 있는 항당뇨 소재로의 가능성뿐만 아니라, 고포도당에서 유래되는 산화적 스트레스로부터 뇌신경세포 보호 및 뇌신경전달물질 체계 개선을 통한 당뇨성 퇴행성뇌질환 등의 합병증을 예방할 수 있는 고부가가치 식품 소재로의 활용 가능성을 확인하였다.
본 연구는 한국 고유 식용 자원인 황칠나무의 잎을 이용한 항산화 효과 및 고혈당으로 인한 신경/뇌신경세포 보호 효과를 알아보고자 실시하였다. 황칠나무 잎 추출물의 총폴리페놀 함량, ABTS와 DPPH 라디칼 소거 활성을 측정하였으며 활성이 가장 높은 80% 에탄올 추출물을 이용하여 극성 정도에 따라 분획을 하였다. 이들 중 ethyl acetate 분획물이 총 항산화력(FRAP assay)과 지질과산화물(MDA) 생성 억제 활성이 다른 분획물에 비하여 유의적으로 높은 값을 나타냈다. 이를 이용하여 신경세포로서의 PC12 세포와 인간 뇌조직 유래 뇌신경세포로서의 MC-IXC 세포에서 $H_2O_2$와 고혈당에 의한 세포생존율을 측정하였고, ethyl acetate 분획물은 산화적 스트레스로부터의 효과적인 세포 보호 효과를 나타냈다. 또한, 뇌신경말단에서 신경전달물질(ACh)의 분해를 유발하는 효소(AChE)의 저해 효과를 측정하였고, ethyl acetate 분획물은 유의적인 AChE 저해 효과를 보였다. 마지막으로 황칠나무 잎 ethyl acetate 분획물의 생리활성 물질을 확인하고자 HPLC 분석을 하였으며, 주요 생리활성 물질은 rutin으로 추정되었다. 이러한 연구 결과를 바탕으로 고려할 때 황칠나무 잎 추출물은 천연 항산화제의 역할을 가지고 있을 뿐만 아니라 이를 통해 산화적 스트레스로부터 뇌신경세포 등을 효과적으로 보호할 수 있으며 더불어 AChE 저해 활성을 통한 인지기능 개선 가능 소재로서 연구될 수 있을 것으로 판단된다.

제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
