Sewage treatment using microorganisms is affected by multiple factors such as microbial properties, characteristics of sewage and operating conditions, and nanoparticles inflow may cause negative effects on sewage treatment system especially on the system stability and efficiency. It was studied to assess the toxic effects of nanoparticles on microorganism growth. The activated sludge in the sewage treatment plant of university was cultured in the optimized medium for each strain. Bacillus (gram-positive), Pseudomonas and E.coli (gram-negative) in the activated sludge were selected as target microorganisms, and ZnO and $TiO_2$ were chosen as nanoparticles. For same concentration of nanoparticles, average growth inhibition rate of Bacillus was 60% or more, while that of Pseudomonas was less than 10%. The toxicity of nanoparticles was shown to be higher for gram-positive bacteria than gram-negative bacteria because of their differences on structure of cell wall, components of cell wall protein, physiology of cells and metabolism. ZnO affected 3 times more negative on the growth of microorganisms as compared to $TiO_2$. It was assumed that, therefore, toxicity of ZnO was found to be greater than $TiO_2$.

Although crack on concrete exists from its early formation, crack requires attention as it affects stiffness of structure and can lead demolition of structure as it grows. Detecting cracks on concrete is needed to take action prior to performance degradation of structure, and deep learning can be utilized for it. In this study, transfer learning, one of the deep learning techniques, was used to detect the crack, as the amount of crack's image data was limited. Pre-trained Inception-v3 was applied as a base model for the transfer learning. Web scrapping was utilized to fetch images of concrete wall with or without crack from web. In the recognition of crack, image post-process including changing size or removing color were applied. In the visualization of crack, source images divided into 30px, 50px or 100px size were used as input data, and different numbers of input data per category were applied for each case. With the results of visualized crack image, false positive and false negative errors were examined. Highest accuracy for the recognizing crack was achieved when the source images were adjusted into 224px size under gray-scale. In visualization, the result using 50 data per category under 100px interval size showed the smallest error. With regard to the false positive error, the best result was obtained using 400 data per category, and regarding to the false negative error, the case using 50 data per category showed the best result.
Background: In patients with coronary artery disease, dysfunctional hypoperfused myocardium at rest may represent either nonviable or viable hibernating myocardium. Two-dimensional echocardiography can detect regional wall motion abnormalities resulting from myocardial ischemia by dobutamine infusion. The purpose of the present study was to identify the prediction of improvement of regional left ventricular(LV) function after surgical revascularization. Materials and methods: Sixteen patients with chronic regional LV dysfunction underwent dobutamine stress echocardiography(DSE) (dobutamine: baseline, 5, 10, 20$\mu$g/kg/min) before coronary artery bypass grafting(CABG) and underwent echocardiography at least 2 months after CABG. Results: All patients were male with mean age of 58 years ranging from 42 to 73 years. The mean LV ejection fraction was 41.8% with a range from 19% to 55%. During DSE, there were no complications, also, there were no operative morbidities or mortalities. Improvement of wall motion within the dysfunctional myocardium was found in 8(50%) of 16 patients in DSE. Among them, 6 patients(75%) showed functional recovery after CABG. Another 8 patients did not show improvement of wall motion in DSE. But among them, 3 patients(38%) showed functional recovery after CABG. 84 dysfunctional segments were found in 256 segments of 16 patients. Improvement of wall motion was found in 34 of 84 segments in DSE. Among them, 23 segments(74%) showed functional recovery after CABG. Another 53 segments did not show improvement of wall motion in DSE. But among them, 12 segments(23%) showed functional recovery after CABG. The sensitivity and specificity of DSE for the prediction of postoperative improvement of segmental wall motion were 66% and 84%, respectively. The positive and negative predictive value of DSE were 74% and 77%, respectively. In patients with chronic regional LV dysfunction, think that DSE is a good predictor of the improvement of dysfunctional segments after CABG.

Gaegurin 4(GGN 4), an antimicrobial peptide isolated from a Korean frog, is five times more potent against Gram-positive than Gram-negative bacteria, but has little hemolytic activity. To understand the mechanism of such cell selectivity, we examined GGN4-induced $K^+$ efflux from target cells, and membrane conductances in planar lipid bilayers. The $K^+$ efflux from Gram-positive M. luteus(2.5 ${\mu}g/ml$) was faster and larger than that from Gram-negative E. coli(75 ${\mu}g/ml$), while that from RBC was negligible even at higher concentration(100 ${\mu}g/ml$). GGN4 induced larger conductances in the planar bilayers which were formed with lipids extracted from Gram-positive B. subtilis than in those from E. coli(p<0.01), however, the effects of GGN4 were not selective in the bilayers formed with lipids from E. coli and red blood cells. Addition of an acidic phospholipid, phosphatidylserine to planar bilayers increased the GGN4-induced membrane conductance(p<0.05), but addition of phosphatidylcholine or cholesterol reduced it(p<0.05). Transmission electron microscopy revealed that GGN4 induced pore-like damages in M. luteus and dis-layering damages on the outer wall of E. coli. Taken together, the present results indicate that the selectivity of GGN4 toward Gram-positive over Gram-negative bacteria is due to negative surface charges, and interaction of GGN4 with outer walls. The selectivity toward bacteria over RBC is due to the presence of phosphatidylcholine and cholesterol, and the trans-bilayer lipid asymmetry in RBC. The results suggest that design of selective antimicrobial peptides should be based on the composition and topology of membrane lipids in the target cells.

The safe operation of high pressure pipe line systems is of significant importance. Leaks due to faulty operation from the pipelines can lead to considerable product losses and to exposure of community to dangerous gases. There are several leak detection methods, which have been recently suggested on pipeline network. The negative pressure wave detection technology, which has advantages of short time detection availability, accurate leaking location estimate capability and cost effective, is concentrated in this study. Theoretical analysis of the flow characteristics for leaking through a hole on the pipe wall has been performed by using CFD++, commercial CFD package. The results of 3-dimensional analysis near leaking hole confirm the occurrence of negative pressure wave and verify the characteristics of propagation of the wave which travels with speed equal to the speed of sound in the pipeline contents. For the application of long pipe line system. The method of 1-dimensional analysis has been suggested and verified with results of CFD++.

Purpose: Tuberculosis(TB) care unit in public health center should be carefully considered to be re-designed as an infection safety environment for both patient and healthcare workers. So, for the enhancement, this study analyses the facility requirements for co-using the screening clinic as a TB and other respiratory disease care unit. Methods: Not only screening clinic facility guidelines from "A Study for Standard Triage Design and Construction Document" but also the guidelines of TB care and related medical facility are reviewed; KDCA, CDC, ECDC and WHO as a TB care, and FGI and NHS for facility. The facility requirements are summarized space, approach, and mechanical requirement in order. By comparing the summary and screening clinic facility guidelines, supplementations are proposed for TB care unit setting. Results: The result of this study shows that both the space program and mechanical requirement of the screening clinic and that of TB care unit are almost identical and could be share, which include direct airflow or negative air pressure in an exam room. To increase functional and economical efficiency, however, it is necessary to consider a multi-functional negative pressured room, So care process may be re-designed based on a room type; face-to-face room or glass wall inbetween. Implications: The facility guidelines for TB care unit of a public health center should be developed to build a safe environment for infection control by reflecting its medical plan and budget.
It is a common failure type that high-filled embankment slope sideslips. The deformation mechanism and factors influencing the sideslip of embankment slope is the key to reduce the probability of this kind of engineering disaster. Taking Liujiawan high-filled embankment slope as an example, the deformation and failure characteristics of embankment slope and sheet-pile wall are studied, and the factors influencing instability are analyzed, then the correlation of deformation rate of the anti-slide plies and each factor is calculated with multivariate linear regression analysis. The result shows that: (1) The length of anchoring segment is not long enough, and displacement direction of embankment and retaining structure are perpendicular to the trend of the highway; (2) The length of the cantilever segment is so large that the active earth pressures behind the piles are very large. Additionally, the surface drainage is not smooth, which leads to form a potential sliding zone between bottom of the backfill and the primary surface; (3) The thickness of the backfill and the length of the anti-slide pile cantilever segment have positive correlation with the deformation whereas the thickness of anti-slide pile through mudstone has a negative correlation with the deformation. On the other hand the surface water is a little disadvantage on the embankment stability.
Bacterial UDP-N-acetylglucosamine enolpyruvyl transferase (MurA) catalyzes the transfer of enolpyruvate from phosphoenolpyruvate (PEP) to uridine diphospho-N-acetylglucosamine (UNAG), which is the first step of bacterial cell wall synthesis. We identified thimerosal, thiram, and ebselen as effective inhibitors of Haemophilus influenzae MurA by screening a chemical library that consisted of a wide range of bioactive compounds. When MurA was preincubated with these inhibitors, their 50% inhibitory concentrations ($IC_{50}s$) were found to range from 0.1 to $0.7\;{\mu}M$. In particular, thimerosal suppressed the growth of several different Gram-negative bacteria such as Escherichia coli, Pseudomonas aeruginosa, and Salmonella typhimurium at a concentration range of $1-2\;{\mu}g/ml$. These inhibitors covalently modified the cysteine residue near the active site of MurA. This modification changed the open conformation of MurA to a more closed configuration, which may have prevented the necessary conformational change from occurring during the enzyme reaction.
Results of measurements of surface pressure and of velocity field made on a full-scale 6 m cube in natural wind are reported. Comparisons are made with results from boundary-layer wind-tunnel studies reported in the literature. Two flow angles are reported; flow normal to a face of the cube (the $0^{\circ}$ case) and flow at $45^{\circ}$. In most comparisons, the spread of wind-tunnel results of pressure measurements spans the full-scale measurements. The exception to this is for the $0^{\circ}$ case where the roof and side-wall pressures at full-scale are more negative, and as a result of this the leeward wall pressures are also lower. The cause of this difference is postulated to be a Reynolds Number scale effect that affects flow reattachment. Measurements of velocity in the vicinity of the cube have been used to define the mean reattachment point on the roof centre line for the $0^{\circ}$ case, and the ground level reattachment point behind the cube for both $0^{\circ}$ and $45^{\circ}$ flow. Comparisons are reported with another full-scale experiment and also with wind-tunnel experiments that indicate a possible dependency on turbulence levels in the approach flow.
Fibrobacter succinogenes, a Gram-negative, anaerobic ruminal bacterium is a major fibre digesting species in the rumen. It intensively degrades plant cell walls by an erosion type of mechanism, burrowing its way through the complex matrix of cellulose and hemicellulose with the release of digestible and undigested cell wall fragments. The enzymes involved in this process include a combination of glucanases, xylanases, arabinofuranosidase(s) and esterases. The genome of the bacterium has been sequenced and this has revealed in excess of 100 putative glycosyl hydrolase, pectate lyase and carbohydrate esterase genes, which is greater than the numbers reported present in other major cellulolytic organisms for which genomes have been sequenced. Modelling of the amino acid sequences of two glycanases, CedA and EGB, by reference to crystallized homologs has enabled prediction of the major features of their tertiary structures. Two dimensional gel electrophoresis in conjunction with mass spectroscopy has permitted the documentation of proteins over expressed in F. succinogenes grown on cellulose, and analysis of the cell surfaces of mutant strains unable to bind to cellulose has enabled the identification of candidate proteins with roles in adhesion to the plant cell wall substrate, the precursor to cellulose biodegradation.
