Construction of an Oscillator Gene Circuit by Negative and Positive Feedbacks

  • Synthetic oscillators are gene circuits in which the protein expression will change over time. The delay of transcription, translation, and protein folding is used to form this kind of behavior. Here, we tried to design a synthetic oscillator by a negative feedback combined with a positive feedback. With the mutant promoter PLacC repressed by LacIq and PLux activated by AHL-bound LuxR, two gene circuits, Os-LAA and Os-ASV, were constructed and introduced into LacI-deleted E. coli DH5α cells. When glucose was used as the carbon source, a low level of fluorescence was detected in the culture, and the bacteria with Os-ASV showed no oscillation, whereas a small portion of those carrying Os-LAA demonstrated oscillation behavior with a period of about 68.3 ± 20 min. When glycerol was used as the carbon source, bacteria with Os-ASV demonstrated high fluorescence value and oscillation behavior with the period of about 121 ± 21 min.

  • Extracellular signal-regulated kinase (ERK) signaling pathway is one of the mitogen-activated protein kinase (MAPK) signal transduction pathways. This pathway is known as pivotal in many signaling networks that govern proliferation, differentiation and cell survival. The ERK signaling pathway comprises positive and negative feedback loops, depending on whether the terminal kinase stimulates or inhibits the activation of the initial level. In this paper, we attempt to model the ERK pathway by considering both of the positive and negative feedback mechanisms based on Michaelis-Menten kinetics. In addition, we propose a fraction ratio model based on the mass action law. We first develop a mathematical model of the ERK pathway with fraction ratios. Secondly, we analyze the dynamical properties of the fraction ratio model based on simulation studies. Furthermore, we propose a concept of an inhibitor, catalyst, and substrate (ICS) controller which regulates the inhibitor, catalyst, and substrate concentrations of the ERK signal transduction pathway. The ICS controller can be designed through dynamical analysis of the ERK signaling transduction pathway within limited concentration ranges.

VFC type A/D Converter에 관한 연구 (A Study on the VFC type A/D Converter)

  • 종래의 VFC type A/D Converter에서는 출력주파수가 수 100KHz까지는 linearity가 좋았으나 출역주파수가 높아 질수록 linearity error가 증가하였기 때문에 본 연구에서는 출력주파수 영역을 수 MHz까지 넓히기 위해서 tunnel diode voltage controlled oscillator를 사용하였고 입력전압과 출력주파수 사이에 linearity를 개선하기 위하여 negative feedback 회로를 사용하였다. 실험 결과에 의하면 제안된 VFC type A/D converter의 linearity는 최고 출력주파수 3.7MHz까지 0.209%이었다.

Chemical Coupling between Horizontal Cells in the Catfish Retina

  • The effects of GABA and glutamate on the horizontal cells were explored by an intracellular recording method to discern the mechanisms of receptive field formation by chemical coupling in the catfish outer retina. The results suggest that the horizontal cells of the catfish retina might use GABA as their transmitters and that the GABAergic system contributes to the formation of receptive fields of the horizontal cells. GABAC receptors may be involved in a chemical coupling between horizontal cells and concerned with the depolarizing actions by GABA on horizontal cells in the catfish retina. Since the chloride equilibrium potential is more positive than the dark membrane potential in horizontal cells, GABA released from a horizontal cell may depolarize the neighboring horizontal cells. Thus a chemical coupling between horizontal cells may be formed. $GABA_A$ receptors also may be involved in the negative feedback mechanism between photoreceptor and horizontal cell. And glutamate may be involved in connecting positive and negative feedback systems since it potentiated the GABA's actions. Therefore, it is presumed that large receptive fields in the catfish retina are formed not only by electrical coupling but also by chemical coupling between horizontal cells. And information travels laterally by pathways involving both electrical coupling composed of gap junctions and chemical coupling in the retinal network.

주파수잠금회로(FLL)를 이용한 VCO의 위상잡음 개선 해석 (Analysis of the Phase Noise Improvement of a VCO Using Frequency-Locked Loop)

  • FLL(Frequency-Locked-Loop: 주파수 잠금회로)은 주파수-검출기(frequency detector)를 사용하여 VCO의 위상잡음을 개선하는 부-궤환(negative feedback) 시스템이다. 본 논문은 FLL에 의한 VCO의 위상잡음의 이론적 분석을 새로이 제시하였다. 분석 결과, VCO의 위상잡음은 FLL 루프-대역폭 내에서는 주파수검출기와 루프-필터로 결정된 위상잡음을 좇아가며, 반면 루프-대역폭 밖에서는 VCO의 위상잡음이 그대로 나타나게 된다. 따라서 이론적 분석 결과를 바탕으로 VCO의 위상잡음을 최소화 하는 FLL을 설계할 수 있게 된다. 또한 실험을 통하여 이론적으로 분석된 위상잡음 결과는 검증하였다.

  • In this paper, we present the results of the design and fabrication of the VCO(Voltage controlled Oscillator) using RF circuit simulator GENESYS and electromagnetic field simulator EMpower Frequency range is fabricated VCO is 850 MHz ~ 950 MHz, which is used Colpitts Circuit. the fabricated VCO is consisted of resonator, oscillator and MSL(Microstrip Line) is used in LC tuning circuit.(operated by negative feedback) MSL(Microstrip Line), Varactor(Plastic package), low noise TR(SOT-23), chip inductor(1608), chip capacitor(1005), chip resistance(1005). 1005 type is used for sample fabrication of VCO. In the fabrication process, circuit pattern is screen printed on the alumina substrates of over 99.9% purity. Center frequency of the sample VCO is 850MHz at $V_T=1.5V$, while the simulated value was 1.0GHz at $V_T=1.5V$. Variable frequency range of the sample is 860~950MHz in contrast to the 1068~1100MHz of the simulated values.

Prototype Milli Gauss Meter Using Giant Magnetoimpedance Effect in Self Biased Amorphous Ribbon

  • In our present work, we developed a GMI (giant magnetoimpedance) sensor system to detect magnetic fields in the milli gauss range based on the asymmetric magnetoimpedance (AGMI) effect in Co-based amorphous ribbon with self bias field produced by field-annealing in open air. The system comprises magnetoimpedance sensor probe, signal conditioning circuits, A/D converter, USB controller, notebook computer, and program for measurement and display. Sensor probe was constructed by wire-bonding the cobalt based amorphous ribbon with dimensions $10\;mm\;{\times}\;1\;mm\;{\times}\;20\;{\mu}m$ on a printed circuit board. Negative feedback was used to remove the hysteresis and temperature dependence and to increase the linearity of the system. Sensitivity of the milli gauss meter was 0.3 V/Oe and the magnetic field resolution and environmental noise level were less than 0.01 Oe and 2 mOe, respectively, in an unshielded room.

On the properties of six cores in the λ Orionis cloud: triggered or non-triggered star formation?

  • We present preliminary results of 1.1 and 1.3 mm dust continuum and 12CO (J=2-1) line data obtained with the Submillimeter Array toward six cores harboring Class 0/I objects in the λ Orionis cloud. They are located in bright rimmed clouds, which are exposed to the far-ultraviolet radiation field by the O-type star λ Ori. Compact dust continuum emission is observed from all six cores. Among the six cores, only one core G196.92-10.37 shows a signature of binarity with separation of 4000 AU. The numbers of singles and binaries in our sample are five and one, respectively and the derived multiplicity frequency (MF) is 0.17. This value is lower than those found in the binary surveys toward Class 0/I objects, which may be a hint for negative feedback by the nearby massive star, lambda Ori. The derived excitation temperature (Tex) using 12CO emission shows a lower median value (20 K) than those of triggered star-forming regions (30 K). The lower MF and Tex support our previous study that star formation was not triggered in the λ Orionis cloud. We aim to further investigate whether the Class 0/I YSOs in the λ Orionis cloud have less circumstellar materials and smaller accretion rates than in other filamentary clouds (e.g., Orion A & B), which might be attributed to negative feedback from the massive star in limiting accretion of protostars.

Effect of Exogenous Porcine GH Administration on GH Responses to GH-releasing Peptide-2 and GH-releasing Hormone in Swine

  • Negative feedback on GH responses to GH-releasing hormone (GHRH) and GH-releasing peptides (GHRPs) has been reported and this action has been suggested to act through an increase in somatostatin. To determine whether the acute administration of porcine GH (pGH) inhibits GH responsiveness to GHRP-2 and GHRH in swine, swine were intravenously administered with pGH (5${\mu}g$/kg BW) or placebo followed 180 min later by a second intravenous administration of saline, GHRP-2 (30 ${\mu}g$/kg BW), GHRH (1${\mu}g$/kg BW) and a combination of GHRP-2 and GHRH. Plasma GH concentration was measured by radioimmunoassay. Administration of pGH caused a significant increase in GH area under curve and GH peak concentrations (p<0.001) over placebo-treated group. Plasma GH concentrations peaked at 15 min and returned to baseline level within 90 min. Pretreatment of pGH abolished (p<0.01) GH response to GHRH and attenuated (p<0.05) GH response to GHRP-2 and GHRH combined, without affecting GH response to GHRP-2. These results demonstrate that negative feedback action on GH releasing effect of GHRH occurs in swine, and that GHRP-2 has ability to interact in this action.
