Objectives: This study was conducted to evaluate the safety of SU-Eohyeol pharmacopuncture (SUEP) by assessing its potential to cause chromosomal abnormalities in Chinese hamster lung cells (CHL/IC). Methods: A dose-curve was conducted to determine the highest dose of SUEP. Doses of 10, 5, 2.5, 1.25, 0.625, and 0.313% were used, and no cytotoxicity or SUEP precipitation was observed. SUEP doses of 10, 5, and 2.5%, with positive and negative controls, were used in a chromosome aberration test. Results: In this study, the frequency of abnormal chromosomal cells in the SUEP group did not show a statistically significant difference from that of the negative control group in short-term treatments with and without metabolic activation and the continuous treatment without metabolic activation. Compared with the negative control group, the positive control group had a significantly higher frequency of cells with structural chromosomal abnormalities. This test's results satisfied all conditions for determining the results. Conclusion: SUEP did not induce chromosomal aberrations under the conditions of this study. Other toxicity evaluations, safety studies in humans, and various clinical trials are required to evaluate the safety and efficacy of SUEP.
SU-Eohyeol pharmacopuncture(SUEP) was developed by adding Cervi Pantotrichum Cornu to Jungsongouhyul pharmacopuncture. This genotoxicity evaluation was performed to evaluate the mutagenic potential of the test substance SUEP agent using histidine, which requires strains of Salmonella typhimurium (TA98, TA100, TA1535, TA1537), and tryptophan, which requires Escherichia coli (WP2uvrA) strain in the presence and absence of metabolic activation. According to the results of the dose range finding study conducted prior to the main study, the dose levels of the test substance in the main study were determined as 100, 50, 25, 12.5, 6.25%, and positive and negative controls were established. As a result of the main study, the mean number of revertant colonies compared to negative controls was less than 2-fold at all dose levels of SUEP in all strains with and without metabolic activation. In the positive control group, the mean number of revertant colonies for each strain was markedly increased by more than two times compared to the negative control group. Based on the result of this study, the test substance, SUEP did not show any indication of mutagenic potential under the conditions of this study.
The activation energy of dissociative adsorption of oxygen on polycrystalline nickel surface is calculated from adsorption isotherms obtained using X-ray photoelectron spectroscopy. Negative value of this activation energy (-5.9 kJ/mol) indicates that the adsorption takes place through an undissociated precursor state. An adsorption energy for this precursor state is calculated assuming the precursor state as a moleculary physisorbed state ($E_{ad}$ = -7.9 kJ/mol). Finally, an adsorption isotherm equation is derived as a function of the gas exposure, which agrees with the experimental isotherms reasonably good.
Purpose : Bridging exercise was used most frequently of lumbar stabilization exercise. There has been no reserch regarding the bridging exercise according to hip abductor activation or hip adductor activation. The purpose of this study is to determine correlation of hip abductor, adductor and abdominal muscles, lower limb muscle. Methods : Participants who met the criteria for this study (n=45). Participants performs bridging exercise on three types (normal bridging exercise, bridging exercise with hip abductor contraction, bridging exercise with hip adductor contraction) for evaluate correlation of each muscles. Results : There was a significant negative correlation between adductor magnus and gluteus medius, adductor magnus and rectus femoris. And there was a positive correlation between gluteus medius and rectus femoris on normal bridging exercise (p<0.05). There was a significant positive correlation between adductor magnus and gluteus medius, transverse abdominis, and between gulteus medius and transverse abdominis on bridging exercise with adductor magnus activation (p<0.05). There was a significant positive correlation between adductor magnus and gluteus medius, rectus femoris, and between gulteus medius and rectus femoris on bridging exercise with gluteus medius activation (p<0.05). Conclusion : When we perform bridging exercise for transverse abdominis activation and increase pressure in the abdmen, bridging exercise with hip adductor contraction is more effective than normal bridging exercise and bridging exercise with hip abductor contraction.
Glutamate causes neurotoxicity through formation of reactive oxygen species and activation of mitogen-activated protein kinase (MAPK) pathways. MAPK phosphatase-1 (MKP-1) is one of the phosphatases responsible for dephosphorylation/deactivation of three MAPK families: the extracellular signal-regulated kinase-1/2 (ERK-1/2), the c-Jun N-terminal kinase-1/2 (JNK-1/2), and the p38 MAPK. In this report, the potential involvement of MKP-1 in neuroprotective effects of curcumin, the active ingredient of turmeric (Curcuma longa), was examined using HT22 cells. Glutamate caused cell death and activation of ERK-1/2 but not p38 MAPK or JNK-1/2. Blockage of ERK-1/2 by its inhibitor protected HT22 cells against glutamate-induced toxicity. Curcumin attenuated glutamate-induced cell death and ERK-1/2 activation. Interestingly, curcumin induced MKP-1 activation. In HT22 cells transiently transfected with small interfering RNA against MKP-1, curcumin failed to inhibit glutamate-induced ERK-1/2 activation and to protect HT22 cells from glutamate-induced toxicity. These results suggest that curcumin can attenuate glutamate-induced neurotoxicity by activating MKP-1 which acts as the negative regulator of ERK-1/2. This novel pathway may contribute to and explain at least one of the neuroprotective actions of curcumin.

Mycobacterium avium subsp. paratuberculosis (MAP) is the causative agent of Johne's disease, a chronic debilitating disease affecting ruminants worldwide. In the present study, we aimed to determine the major gene networks and pathways underlying the immune response to MAP infection using whole-blood cells, as well as provide the potential transcriptional markers for identifying the status of MAP infection. We analyzed the transcriptional profiles of whole-blood cells of cattle identified and grouped according to the presence of MAP-specific antibodies and the MAP shed by them. The grouping was based on the results obtained by ELISA and PCR analyses as follows: i) Test1 group: MAP-negative results obtained by ELISA and positive results obtained by PCR; ii) Test2 group: MAP-positive results obtained by ELISA and negative results obtained by PCR; iii) Test3 group: MAP-positive results obtained by ELISA and positive results obtained by PCR; iv) uninfected control: MAP-negative results obtained both by ELISA and PCR analysis. The results showed down-regulated production and metabolism of reactive oxygen species in the Test1 group, activation of pathways related to the host-defense response against MAP (LXR/RXR activation and complement system) in the Test2 and Test3 groups, and anti-inflammatory response (activation of IL-10 signaling pathway) only in the Test3 group. Our data indicate a balanced response that serves the immune-limiting mechanism while the host-defense responses are progressing.

Cisplatin (Cis-DDP) is one of the most widely used anti-cancer drugs. It is applicable to many types of cancer, including lung, bladder, and breast cancer. However, its use is now limited because of drug resistance. p90 ribosomal S6 kinase (p90RSK) is one of the downstream effectors in the extracellular signal-regulated protein kinases 1 and 2 (ERK1/2) pathway and high expression of p90RSK is observed in human breast cancer tissues. Therefore, we investigated the role of p90RSK in the Cis-DDP resistance-related signaling pathway and epithelial-mesenchymal transition (EMT) in breast cancer cells. First, we discovered that MDA-MB-231 cells exhibited more Cis-DDP resistance than other breast cancer cells, including MCF-7 and BT549 cells. Cis-DDP increased p90RSK activation, whereas the inactivation of p90RSK using a small interfering RNA (siRNA) or dominant-negative kinase mutant plasmid overexpression significantly reduced Cis-DDP-induced cell proliferation and migration via the inhibition of matrix metallopeptidase (MMP)2 and MMP9 in MDA-MB-231 cells. In addition, p90RSK activation was involved in EMT via the upregulation of mRNA expression, including that of Snail, Twist, ZEB1, N-cadherin, and vimentin. We also investigated NF-κB, the upstream regulator of EMT markers, and discovered that Cis-DDP treatment led to NF-κB translocation in the nucleus as well as its promoter activity. Our results suggest that targeting p90RSK would be a good strategy to increase Cis-DDP sensitivity in triple-negative breast cancers.
A few researchers have reported that major depression may be associated with higher levels of positive acute phase proteins(APPs), such as haptoglobin(Hp), ${\alpha}1$-antitrypsin(${\alpha}1AT$), ceruloplasmin(Cp) and lower levels of negative APPs(visceral proteins), such as albumin(Alb) and transferrin(Tf). Elevated levels of positive APPs and a drop in negative APPs constitute important indicators of immune activation. This study was designed to investigate whether altered serum concentrations of positive APPs and of negative APPs reflect the state of depression. Twenty patients who fulfilled DSM-III-R criteria for major depressive disorder and for dysthymic disorder and twelve normal healthy controls were included. The authors measured positive APPs(Hp, ${\alpha}1AT$, Cp) and negative APPs(Alb, Tf) using rate nephelometry and bromcresol green method. 1) There were significant increases of ${\alpha}1AT$, Cp in major depressed patients as compared with normal controls. Trends towards higher Hp and lower Alb, Tf in major depressed patients were observed. 2) No significant difference of APPs concentrations between dysthymic patients and normal controls was found. 3) Severity of depression(HDRS, BDI score) was related to Hp, Cp, ${\alpha}1AT$ value positively. Our findings are partially compatible with the hypothesis that major depression may be accompanied by acute phase response with higher levels of positive APPs and lower levels of negative APPs.
Introduction : Recent studies Suggested that there is a strong correlation between acupuncture stimulation and its related cortical activation. Anther study showed that either positive or negative BOLD effects could be observed depending on anatomical structure in acupuncture stimulation. In ttis study, we investigated a new acupoint $KI_6$ (照海), which was known as motor-related acupoint and obtained an evidence that the stimulation of $KI_6$ resulted in either negative or positive BOLD response to stimulation. Methods & Results : 1. Subjects and paradigms : Two separate stimulation paradigms were performed on five healthy (aged 22-23 yrs) in this study. First, the paradigm of acupuncture stimulation was that the acupuncture needle was inserted in acupoints $KI_6$, which is located in lateral side of the foot and then continuously twisted(補瀉를 除外한 捻轉法) for 70 seconds for 10 cycles of activation. During rest period (70 seconds), the needle was completed removed from acupoint. Total 60 cycles were performed and 10 images were obtained per cycle. Second, nonacupoint was randomly selected and the same paradigm was performed as acupoint stimulation. The stimulation protocol comprised 10 cycles of alternating. activation and rest (10 images per cycle). Total 60 cycles were performed and each cycle take about 1.5 sec for motor task. Subjects take an at least 15 minutes break before starting anther paradigm. 2. fMRI mapping : Multi-slice functional images were obtained on a 1.5T Magnetom Vision MRI scanner (Simens Medical, Erlangen, Germany) equipped with high performance whole-body gradients. The BOLD T2 * - weighted images were acquired with acho planar imaging sequence (TR = 1.2 sec, TE = 60 msec, and flip angle = $90_{\circ}$). The other sequence parameter are : FOV = 210 mm, matrix=$64{\times}128$ or $64{\times}64$, slice number=10 and slice thickness = 5 or 8 mm. the anatomic images were obtained with Spin-echo T1-weighted images. The resulting images were then anaiyzed with STIMULATE (CMRR, U. of Minnesota) to generate functional maps using a student T-test (p < 0.005) and cluster analysis. Both positive and negative response were evaluated. Conclusions : We have observed the activation of the motor cortex by stimulating motor-related acupoint ($KI_6$). Among five subjects, negative BOLD response was shown in four and positive response in one. All subjects showed positive response to conventional finger flexion-extension task. To understand the detailed mechanisms of correlation between acupuncture stimulation and BOLD fMRI changes and two typs of response, further study strongly required.
본 연구에서는 정부의 포괄적 네거티브 규제 정책에 의해 선정된 부산 블록체인 규제자유특구의 활성화를 위한 정책방안을 제안하고자 한다. 우리나라는 유럽이나 미국과는 달리 포지티브 규제 기반으로, 활성화가 아닌, 억제를 목적으로 하므로 경제·산업 분야에서 지속적인 개혁 대상으로 취급된다. 문재인 정부는 이러한 포지티브 규제의 문제점을 해소하기 위한 획기적인 규제 혁신 정책으로 '포괄적 네거티브 규제'로의 전환을 제시하였다. 이에 대한 실행 방안으로 규제자유특구법에 기초한 신산업 육성을 위한 1단계 규제자유특구 7곳을 우선 선정하였다. 부산시는 '블록체인 규제자유특구'로 최종 선정되었으며, 블록체인을 바탕으로 침체된 지역경제를 활성화할 기회를 확보하였다. 본 연구는 부산 블록체인 규제자유특구의 활성화를 위한 정책 연구로, 규제자유특구에서 사업화할 수 있는 부산 특화 블록체인 서비스 발굴과 함께 산업 생태계 조성을 위한 방안을 제안하고자 하였다.
