One hundred and eighty-seven water samples were collected from 23 of spring water, 2 of ground water, 1 of tap water in Pusan area and 3 of natural mineral waters. Total coliform group, fecal coliform, viable cell count and microflora were investigated to evaluate water quality of drinking water. The results were as follows: range and geometric mean value of total coliform and fecal coliform MPN's of spring water were 0~1,500/100 ml, 85/100 ml and 0~460/100 ml, 24/100 ml but coliform group was not detected in the samples of tap water and natural mineral water. Viable cell count of spring water, ground water and tap water were lower as 100 cell than the criteria for drinking water but that of natural mineral water was higher as 6.5X 10$^2$~7.4X 10$^3$ /ml. Predominant speces among the 219 strains isolated from the samples were 19.6% Aeromonas spp., 19.2% Enterobacteriaceae, 16% Acinetobacter spp. Especially, spring water and vessels were contaminated by Hafnia spp. and Providencia Spp, inhabitant of the oral cavity.
Interest in aspects of industrialization relating to human health has increased. Accordingly, the use of labels such as 'natural foods' and 'organic ingredients' has become more widespread, and greater emphasis is being placed on improving quality of life. Water is an essential element for human life, and water quality has a significant impact on human health. However, technology that can precisely determine the substances present in water is still lacking. This study was conducted to establish a complete mass spectrometry process, from pretreatment to analysis, to measure and characterize natural organic matter (NOM) in Korean spring water samples. Salts and other matrices were removed from the samples using solid-phase extraction (SPE) with two different columns (PPL and C18). After establishing an accurate analysis method, the experimental results were evaluated based on Van Krevelen diagrams and analysis of molar O/C and H/C ratios. The method for characterizing NOM introduced herein should facilitate evaluation of water quality.
This study was designed to detect and measure the concentration of radioactivity of natural radionuclides ($^{238}U$, $^{232}Th$, $^{40}K$) and artificial radionuclides ($^{137}Cs$, $^{60}Co$) present in the drinking water of the city of Busan and surrounding areas in South Korea, and also to measure the absorbed dose of radiation caused by these elements in the residents so as to help better manage the risk that these radionuclides pose in the future. For the purposes of the study, a total of 42 samples of water were collected from three key water sources (19 samples of groundwater, 4 samples of tap water, and 19 samples of surface water) and their contents were analyzed for radioactivity concentration. The results revealed that two natural radionuclides, $^{238}U$ and $^{232}Th$, exist in the groundwater with an average concentration of radioactivity of 3.34 Bq/L and $8.28{\times}10^{-5}Bq/L$ respectively, while the surface water was found to contain the same two radionuclides with mean concentrations of 0.849 Bq/L and $1.103{\times}10^{-4}Bq/L$ respectively. In addition, of the 19 samples of the groundwater, $^{137}Cs$ was found in eight of them and $^{60}Co$ was detected in ten. Of the four samples of the tap water, $^{137}Cs$ was detected in all samples and $^{60}Co$ was detected in three. Both $^{137}Cs$ and $^{60}Co$ were detected in all 12 samples of surface water. As far as $^{40}K$ is concerned, this element was detected in three of the 19 groundwater samples, but was not detected in any surface or tap water sample. In addition, the absorbed dose of $^{238}U$ from the groundwater was $7.94{\times}10^{-8}Sv/y$, while the absorbed dose of $^{232}Th$ from the surface water was $9.33{\times}10^{-13}Sv/y$. The absorbed dose of $^{137}Cs$ from the tap water was $7.33{\times}10^{-5}Sv/y$, while the absorbed dose of $^{60}Co$ from the surface water was the highest at $4.23{\times}10^{-6}Sv/y$.
Tailings fluidization (i.e., tailings behave as being fluidized) under cyclic loading is one concern during the construction of tailings dams, especially in the shallow tailings layers. The primary goal of this study is to evaluate the responses of tailings under cyclic loadings and the tailings potential for fluidization. A series of cyclic triaxial undrained and drained tests were performed on medium and dense tailings samples under various cyclic stress ratios (CSR). The results indicated that axial strain and excess pore water pressure accumulated over time due to cyclic loading. However, the accumulations were dependent on CSR values, densities, and drainage conditions. The fluidization potential analysis in this study was then evaluated based on the obtained cyclic axial strain and excess pore water pressure. As a result, tailings samples were stable (unfluidized) under small CSR values, and the critical CSR values, where the tailings fluidized, varied depending on the density of tailings samples. Tailings fluidization is triggered as cyclic stress ratios reach critical values. In this study, the critical CSR values were found to be 0.15 and 0.40 for medium and dense samples, respectively.
56 samples were analyzed to understand water quality of the natural mineral water of Korea. The geology according to each sample location is grouped into Precambrian metamorphic rocks, Okcheon metamorphic rocks, Jurassic granite, Cretaceous granite, and Jeju volcanic rocks. Average EC and pH values of the water is 150 μS/cm and 7.3, respectively and water type of the water is mainly Ca-Na-HCO3. Fundamentally, there still is no problem for the water quality of the natural mineral water. Nevertheless, nitrate was detected and arsenic and fluoride contents are near the drinking water standards, it is highly necessary to manage the water quality by installment of casing and grouting or by development of another production well.
Difference in recovery behaviors from the circular deformation of natural rubber (NR) composites aged in air and distilled water, respectively were investigated. Recoveries of the samples aged in air were larger than those of the samples aged in distilled water. Recovery rates of the samples reinforced with filler were faster than those of the unreinforced ones. Recovery rates of the carbon black-filled samples were faster than those of the silica-filled ones. Difference in the recovery behaviors according to the aging media can be explained by the crosslinking density changes and the annealing effect.
Human Astrovirus (HuAstV), known as a waterborne virus, is a group IV positive-sense single-stranded RNA that belongs to Astroviridae. The first outbreak of HuAstV was reported in England in 1975. HuAstV can exist not only among clinical patients but also in various water environments, such as water for agriculture and vegetables. For diagnosis of HuAstV from water samples, a polymerase chain reaction (PCR) system has been developed. However, the PCR-based diagnostic method has problems in field application, such as reaction time, sensitivity and specificity. For this reason, in this study we developed the loop-mediated isothermal amplification assay (LAMP) system, aimed specifically at HuAstV. Three prepared LAMP primer sets were tested by specificity, non-specificity and sensitivity; one LAMP primer set was selected with optimum reaction temperature. The developed LAMP primer set reaction conditions were confirmed at 62℃, and detection sensitivity was 1 fg/μL. In addition, restriction enzyme HaeIII (GG/CC) was introduced to confirm that the LAMP reaction was positive. As a result, selected LAMP primer set was 100 - 1000 times more specific, rapid, and sensitive than conventional-nested PCR methods. For verification of the developed LAMP assay, twenty samples of cDNA from groundwater samples were tested. We expect that the developed LAMP assay will be used to diagnose HuAstV from various samples.
To assess possible risks from the consumption of drinking water from various sources, a survey of the microbiological quality of tap water, commercial bottled drinking water which is exploited from natural mineral water, and natural spring water was conducted. A total of 4 different brands of commercial bottled drinking water, and 4 types of spring water from different sources, and tap water from 4 private houses were tested for four index microorganisms, and the microbial quality changes of the water during the storage at room temperature or refrigerated temperature for 7 days. Aerobic plate counts of all of the initial water samples were still within 100 CFU/ml (drinking water standard of Korea). Total coliforms, fecal coliforms, and E. coli were not detected in all of the water samples at initial. However, aerobic plate counts of three types of spring water and three types of bottled drinking water stored at room temperature showed higher levels than the standards in 5 days. Total coliforms were detected in three types of spring water after one day's storage at room temperature, and in one type of bottled drinking water after 5 days' storage. These results indicate that some of the spring water surveyed are not safe to drink, and the spring water and bottled drinking water after opening the lid should not be stored at room temperature, if they are used for drinking.
In the hydrologic and hydrochemical studies of natural waters, oxygen and hydrogen isotope compositions of waters are very important to elucidate the origin and circulation pattern of water in the hydrologic system. The hydrogen isotope analysis of waters usually has been undertaken through the reduction of water to form hydrogen gas using pure metals (in general, zinc and uranium). In 1996, a new apparatus (H/Device) was developed to prepare the water samples (by the reduction with Cr metal) without some intrinsic problems that may yield incorrect and/or inaccurate data, and was installed at 1997 in the Center for Mineral Resources Research (CMR) in Korea University. However, the optimistic conditions of preparation and analysis of samples has not been established. In this paper, we introduce the efficiency of H/Device to obtain accurate hydrogen isotope values of water, and discuss both the optimum conditions including the effective reduction time and the probable mixing (memory) effect between successive samples. We obtained large amounts of a laboratory working standard (KUW; Korea University Water) with the average ${\delta}D_{SMOW}$ value of $-42.1{\pm}1.0$‰ $(1{\sigma})$.
Various types of inorganic and organic colloids are present in natural water including groundwater. Previous studies showed that Fe, Mn and Al are colloid-forming elements and dissolved concentrations can be erroneous for these elements if water samples are not properly filtered. Dissolved concentrations of elements including Ca, Na, Mg, K, Fe, Mn, Si and Al in groundwater from alluvial and bedrock aquifers, and surface water near Nakdong River were determined to evaluate effects of colloids on dissolved concentrations in natural water samples using various pore sizes of filters. Groundwater is mostly anoxic and have elevated concentrations of Fe and Mn, which provides a unique opportunity to observe the effects of colloids on dissolved concentrations of colloid-forming elements. Membrane filters with four kinds of pore sizes of 1000 nm, 450 nm, 100 nm, and 15 nm were used for filtration of water samples. Concentrations of dissolved concentrations in each filtrate did not show significant differences from 1000 nm to 100 nm. However, concentrations of all elements considered were decreased in the filtrates obtained using 15 nm pore size filters by 10 to 15% compared to those using 450 nm except for bedrock groundwater. Al in surface water showed a distinct linear decrease with the decrease of filter pore sizes. These results showed that 100 nm pore size had little effect to remove colloidal particles in alluvial groundwater and surface water in our study. In contrast, significant concentration decreases in 15 nm pore size filtrates indicate that the presence of 15 to 100 nm colloidal particles may affect determination of dissolved concentrations of elements in natural water.
