지반내 자연대류에 대한 수치해석적 논의 (Numerical Discussion on Natural Convection in Soils)

  • 지반의 열적 거동은 대부분 열전도에 국한되어 연구가 진행되었으며, 자연대류 현상에 대한 연구는 매우 미비한 실정이다. 지반의 온도증가는 간극수의 밀도변화에 따른 부력을 유발하여 자연대류를 발생시키게 된다. 유체역학 관점에서 다공질 재료내의 자연대류 해석의 제약조건에 대하여 논의하고, 거시적 관점에서 완전 결합된 열-수리-역학적인 지배방정식을 이용한 대류현상에 대한 수치해석 기법을 제시하였다. 실내 열전도도 측정을 위한 탐침기 실험에 대한 수치실험은 자연대류를 무시하고 평가된 열전도도의 불확실성에 대하여 논의하고, 모델식과의 오류를 최소화하기 위한 적정한 실험조건을 제시하였다. 해저 전력선의 매설은 해저면 0.2m 깊이에서의 온도상승을 $2^{\circ}C$로 제한하고 있으나, 투수성이 큰 지반재료에 대한 수치해석결과는 기준온도를 초과하는 것으로 나타났다. 해저면의 온도와 열-수리-역학적 물성은 전력선의 매설설계에 중요한 설계인자이며 자연대류의 영향을 고려하여야 한다. 특히, 큰 투수성을 갖는 지반내에 열원이 존재하는 경우, 간극수의 밀도변화에 따른 자연대류가 중요한 열전달의 인자가 되므로 이를 고려한 해석을 수행하여야 한다.

  • 시설토마토에서 사전에 해충밀도를 예찰 하지 않고 작물 정식과 동시에 적용한, 기생성/포식성 천적과 그들의 먹이원/서식처가 결합되어 있는, 'Natural Enemy in First (NEF)' 기술의 총채벌레와 진딧물 방제효과를 확인하였다. 처리 후 12주차에 NEF 처리구에서 총채벌레 밀도억제 효과는 천적처리구 및 관행방제구에 비해 각각 32%와 82% 더 높았다. 처리 후 진딧물의 밀도는 모든 처리구에서 낮게 유지되어 처리구간 유의성 있는 차이를 확인할 수 없었다.

Isolation and Characterization of Chondroitin Sulfates from the Byproducts of Marine Organisms

  • By-products of marine organisms including salmon, skate, flatfish, and yellow goosefish were investigated to search for new source of chondroitin sulfate (CS). Agarose gel electrophoresis with chondroitinase depolymerization showed that purified chondroitin sulfate did not contain any other glycosaminoglycans. 1H-nuclear magnetic resonance (NMR) spectra were acquired to confirm the structure and purity. The average molecular weight ranging from 22 to 64 kDa was determined by high performance size exclusion chromatography. Disaccharide compositions and purities were determined by strong anion exchange-high performance liquid chromatography (SAX-HPLC) after chondroitinase ABC depolymerization. SAX-HPLC data exhibited that the purity was from $81.7{\pm}1.3$ to $114.2{\pm}2.5%$ and the yield was from 1.3 to 12.5%. All analytical results indicate that salmon cartilage, skate cartilage, and yellow goosefish bone could be promising sources of CS to substitute shark cartilage CS in commercial neutraceuticals.

Molecular Structure of the PHA Synthesis Gene Cluster from New mcl-PHA Producer Pseudomonas putida KCTC1639

  • Pseudomonas putida KCTC 1639 was newly identified as a potential producer of biodegradable medium chain length polyhydroxyalkanoates. It exhibited a carbon assimilation pattern quite different from other known P. putida strains, but a more similar pattern with P. oleovorans, which assimilates the carbon sources mainly through ${\beta}$-oxidation rather than the fatty acid biosynthesis pathway. The PHA synthesis gene cluster from P. putida KCTC1639 was composed of two gene loci; the PHA synthase gene locus and granule-associated gene locus, which were cloned and deposited in the GenBank under accession numbers AY286491 and AY750858 as a new nucleotide sequence, respectively. The molecular structure and amino acid homology of the new gene cluster were compared with those from Pseudomonas species, including other P. putida strains and P. oleovorans, and a higher than $90\%$ homology was observed.

Nutritional Value of a Heterotrichous Ciliate, Fabrea salina with Emphasis on Its Fatty Acid Profile

  • Fabrea salina is a hypersaline ciliate having importance as a live food source for juvenile stages of aquatic animals including smaller invertebrates. The analysis of this ciliate for proximate and biochemical composition was carried out. The moisture, protein, fat, carbohydrate and ash content of F. salina from natural sources were 86.66$\pm$0.380, 56.66$\pm$0.494%, 36.66$\pm$0.614%, 1$\pm$0.073% and 4$\pm$0.182%, respectively. Gas chromatographic analysis (percent area below the curve) revealed that the presence of oleic acid was higher over other fatty acids in both natural and cultured F. salina. The absolute content of oleic acid was higher in natural (18.91% area) than in the cultured (10.74% area) F. salina. Linoleic and linolenic acids were also among major fatty acids with the percentage area of 16.29 and 14.58, respectively. The number of fatty acids in cultured Fabrea was less as compared to the natural ones and the oleic acid was followed by palmitic acid, palmitoleic acid, linoleic acid and stearic acid.

Opitimization of culture conditions involved in dissolved oxygen for production of pullulan by Aureosidium pullulan

  • Effect of carbon source and culture conditions involved in the concentration of dissolved oxygen on cell growth and the production of pullulan by A. pullulans HP2001 were investigated. Among those carbon sources, glucose was found to be the best carbon source for the production of pullulan by A. pullulans HP2001. Maximal production of pullulan by A. pullulans HP2001 was 26.6 g/ f when concentrations of glucose and yeast extract were 8% (w/v) and 0.25% (w/v), respectively. It was found that aeration rate, agitation speed and inner pressure of a bioreactor, which were some of physiological factors involved in the dissolved oxygen in the medium may affect cell growth and the production of pullulan by A. pullulans HP2001.

Effects of Insect Crude Drugs on Blood Coagulation and Fibrinolysis System

  • The in vitro anticoagulant and fibrinolytic activities of crude extracts from insects were evaluated in order to find effective therapeutic drugs for the treatment of myocardial and cerebral thrombosis. We prepared three types of extracts (water, methanol and ethylacetate) from 28 insects for use as raw materials for the activity assays. The fibrinolytic activity was tested using the fibrin plate method and the activated partial thromboplastin time and thrombin time were measured for blood clotting activity. With regards to the fibrinolytic system, water extracts of six kinds of insects displayed a remarkable level of activity with a plasmin-like action. The water extracts of [Catharsius molossus, Eupolyphaga sinensis, Huechys sanguinea, Mantidis $o\ddot{o}theca$, Mimela splendens, and Polistes mandarinus (Vespae Nidus)] exhibited the activity. On the other hand, the methanol extracts did not display any fibrinolytic activity. In terms of the coagulation system, an aqueous extract of silkworm Tongchunghacho (Paecilomyces japonica), Oxya japonica japonica and Buthus martensi (Scorpion) increased the clotting time significantly longer (181 times) than the control. These results suggest that crude drugs from insects are useful sources for the development of new drugs for use in treatments involving blood coagulation and fibrinolysis.

Single-Step Solid-State Synthesis of CeMgAl11O19:Tb Phosphor

  • The green-emitting CeMgAl11O19:Tb (CMAT) phosphor has been prepared at 1200 °C by the simple solid-state reaction using AlF3 as a self-flux. This preparation temperature is much lower than those (1500-1700 °C) for conventional solid-state reaction and spray pyrolysis method. In particular, the complete process to produce high-quality phosphor particles was carried out through the single-step heat treatment of the mixture of corresponding oxide-type metal sources. An addition of AlF3 as a self-flux significantly decreased the crystallization temperature of CMAT with plate-like shape. The particle morphology could be controlled from plate-like to spherical by using H3BO3 as an additional flux. Thus, an optimal morphology and luminescence characteristics of CMAT were achieved when both AlF3 and H3BO3 fluxes were simultaneously used. Compared with conventional solid-state process, which is accompanied by the calcination step(s), and other alternative liquid solution techniques such as sol-gel method and spray pyrolysis, no use of active precursors and liquid media that are harmful to the environment is a distinctive advantage for the industrial purpose.

Molecular Genetic Analysis of Ancient Cattle Bones Excavated from Archaeological Sites in Jeju, Korea

  • Ancient cattle bones were excavated from archaeological sites in Jeju, Korea. We used molecular genetic techniques to identify the species and establish its relationship to extant cattle breeds. Ancient DNA was extracted from four sources: a humerus (Gonae site, A.D. 700-800), two fragments of radius, and a tooth (Kwakji site, A.D. 0-900). The mitochondrial DNA (mtDNA) D-loop regions were cloned, sequenced, and compared with previously reported sequences of various cattle breeds (9 Asian, 8 European, and 3 African). The results revealed that these bones were of the breed, Bos taurus, and a phylogenetic tree indicated that the four cattle bones formed a monophyletic group with Jeju native black cattle. However, the patterns of sequence variation and reports from archaeological sites suggest that a few wild cattle, with a different maternal lineage, may have existed on Jeju Island. Our results will contribute to further studies of the origin of Jeju native cattle and the possible existence of local wild cattle.

천연광물의 양극성 표면개질을 이용한 상수원수 중 중금속제거 특성 (Heavy Metal Removal from Drinking Water using Bipolar Surface Modified Natural Mineral Adsorbents)

  • Objectives: The most commonly detected heavy metals in rocks and soils, including Pb, Cd, Cu, Fe, Mn and As, are representative pollutants discharged from abandoned mines and have been listed as potential sources of contamination in drinking water. This study focused on increasing the removal efficiency of heavy metals from drinking water resources by surface modification of natural adsorbents to reduce potential health risks. Methods: Iron oxide coating and graft polymerization with zeolites and talc was conducted for bipolar surface modification to increase the combining capacity of heavy metals for their removal from water. The removal efficiency of heavy metals was measured before and after the surface modification. Results: The removal efficiency of Pb, Cu, and Cd by surface modified zeolite showed 100, 92, and 61.5%, respectively, increases compared to 64, 64, and 38% for non-modified zeolite. This implies that bipolar surface modified natural adsorbents have a good potential use in heavy metal removal. The more interesting finding is the removal increase for As, which has both cation and anion characteristics showing 27% removal efficiency where as non-modified zeolite showed only 2% removal. Conclusions: Zeolite is one of the most widely used adsorptive materials in water treatment processes and bipolar surface modification of zeolite increases its applicability in the removal of heavy metals, especially As.
