Effect of Epididymal Fluid Fractionated by Chromatography on In Vitro Maturation of Porcine Follicular Oocytes

  • The aim of this study was to investigate what protein(s) of porcine epididymal fluid (pEF) are able to enhance the nuclear maturation of porcine germinal vesicle (GV) oocytes in vitro. Proteins of pEF were fractionated by affinity, ion exchange, and gel filtration chromatography. Porcine cumulus-oocytes complexes (COC) from follicles were cultured in tissue culture medium (TCM 199) containing various fractions obtained by chromatography. Porcine COCs were also cultured in TCM 199 containing various meiosis inhibitors and pEF. After 24 or 48 h culture, oocytes were examined for evidence of GV breakdown, metaphase I, anaphase-telophase I, and metaphase II. When porcine COCs were cultured in the medium with meiosis inhibitor such as, dibutyryl cAMP (dbcAMP) and forskolin (Fo), more than 80% of oocytes were unable to resume meiosis. However, porcine COCs supplemented with pEF were able to overcome the inhibitory effect of dbcAMP and Fo. Maturation rate of oocytes was significantly (p<0.05) increased in the media supplemented with cationic protein(s) during in vitro maturation than in those with anionic protein(s) (44.1% vs 20.0%). When oocytes were cultured in the TCM 199 with fractions obtained by gel filtration, the maturation rate of oocytes was significantly (p<0.05) higher in fraction 11 containing 18 kDa than other fractions. The present study suggests that 1) dbcAMP and Fo prevent the spontaneous maturation of oocyte after isolation from follicles, and that pEF contain a substance(s) that improves meiosis resumption in vitro of porcine COCs, 2) cationic 18 kDa protein(s) are responsible for promotion of Mil stage.

Tissue-Specific Localization NUCB2/nesfatin-1 in the Liver and Heart of Mouse Fetus

  • NUCB2/nesfatin-1 is first known to be expressed in the hypothalamus while controlling appetite and energy metabolism. However, recent studies have shown that NUCB2/nesfatin-1 was expressed in the various organs as well as the hypothalamus. Our previous reports also demonstrated that NUCB2/nesfatin-1 was expressed in the ovary, testis, pituitary gland, lung, kidney, and stomach of fetal and adult mice. However, the role of NUCB2/nesfatin-1 in mouse fetus remains unknown. Thus, the aim of this study was to investigate whether NUCB2/nestatin-1 is expressed in mouse fetus at the developmental stage in which organogenesis begins. To do this, we performed in situ hybridization (ISH) and immunohistochemistry (IHC) staining to examine the distribution of NUCB2 mRNA and nesfatin-1 protein in the mouse fetal organs during early developmental stages, especially at embryonic day (E) 10.5. As a result of ISH, NUCB2 mRNA positive signals were more frequent in the liver, but there were relatively few positive signals in heart. On the other hand, no positive signals were detected in other organs. These ISH results were validated by IHC staining and qRT-PCR analysis. Expression of nesfatin-1 protein detected by IHC staining was similar to that of NUCB2 mRNA detected by ISH in the liver and heart. In addition, the levels of NUCB2 mRNA expression analyzed by qRT-PCR were significantly increased in the liver and heart compared to other organs of the mouse fetus at E13.5, whereas its level was extensively decreased in the liver, but increased in the lung, stomach, and kidney of the mouse fetus at E17.5. These results suggest that NUCB2/nesfatin-1 may play an important role in liver and heart development and physiological functions in the developmental process of mouse fetus. Further studies are needed on the function of NUCB2/nesfatin-1, which is highly expressed in the various organs, including liver and heart during mouse development.

$CO_2$ 처리에 의한 배추 화주 유전자의 특이적 발현 연구 (Differential expression of pistil genes induced by $CO_2$ treatment in chinese cabbage)

  • $CO_2$ gas 처리가 배추 자가불화합성 타파에 미치는 영향을 조사하고자 배추 inbred line의 암술에서 mRNA를 분리하여 DD-PCR을 수행하였다. $CO_2$ gas를 처리한 암술에서 분리한 mRNA와 처리하지 않은 꽃에서 분리한 암술 mRNA 발현과의 특이적인 차이를 DD-PCR로 조사한 결과 세 가지 각기 다른 anchor primer에 대해 모두 18개의 특이적 증폭 DNA fragment를 얻을 수 있었다. 이 결과로 미루어 보아 $CO_2$ gas 처리에 의한 암술 mRNA발현의 변화가 배추 자가불화합성 타파에 영향을 미치는 것으로 생각된다.

총채가시응애(Hypoaspis aculeifer)와 긴털가루응애(Tyrophagus putrescentiae)의 저온저장 조건 구명 (Study on the Cold Storage Condition of Hypoaspis aculeifer(Canestrini)(Acari: Gamasida) and Tyrophagus putrescentiae(Schrank)(Acari: Acaridae))

  • 성공적인 생물학적 방제를 위해서는 적용천적의 사육기준과 규격이 설정되어 있어야 한다. 8, $12^{\circ}C$, 습도 $70{\pm}5%$, 24시간 암조건에서 총채가시응애와 그 먹이응애인 긴털가루응애의 저온 저장 실험을 수행하였다. 긴털가루응애는 소맥피(50%)와 쌀겨(50%)로 조성된 먹이사료에 접종하여 $12^{\circ}C$에서 저온저장이 가능하였다. 긴털가루응애와 총채가시응애의 생존율이 70%되는 조건은 $12^{\circ}C$에서 28일, 70일 저장후이다. 총채가시응애를 $12^{\circ}C$에서 28, 56, 84일 저온 저장후 증식율을 확인한 결과 저온기간에 상관없이 유사한 결과를 나타내었다.

YY1 and CP2c in Unidirectional Spermatogenesis and Stemness

  • Spermatogonial stem cells (SSCs) have stemness characteristics, including germ cell-specific imprints that allow them to form gametes. Spermatogenesis involves changes in gene expression such as a transition from expression of somatic to germ cell-specific genes, global repression of gene expression, meiotic sex chromosome inactivation, highly condensed packing of the nucleus with protamines, and morphogenesis. These step-by-step processes finally generate spermatozoa that are fertilization competent. Dynamic epigenetic modifications also confer totipotency to germ cells after fertilization. Primordial germ cells (PGCs) in embryos do not enter meiosis, remain in the proliferative stage, and are referred to as gonocytes, before entering quiescence. Gonocytes develop into SSCs at about 6 days after birth in rodents. Although chromatin structural modification by Polycomb is essential for gene silencing in mammals, and epigenetic changes are critical in spermatogenesis, a comprehensive understanding of transcriptional regulation is lacking. Recently, we evaluated the expression profiles of Yin Yang 1 (YY1) and CP2c in the gonads of E14.5 and 12-week-old mice. YY1 localizes at the nucleus and/or cytoplasm at specific stages of spermatogenesis, possibly by interaction with CP2c and YY1-interacting transcription factor. In the present article, we discuss the possible roles of YY1 and CP2c in spermatogenesis and stemness based on our results and a review of the relevant literature.

  • Linking Ukraine's education system with the trends of global digitalization is mandatory to ensure the sustainable, long-term development of the country, as well as to increase the sustainability of the education system and the economy as a whole during the crisis period. Now the main problems of the education system in Ukraine are manifested in a complex context caused by Russian armed aggression. In the context of war, problems include differences in adaptation to online learning among educational institutions, limited access to education for vulnerable groups in the zone of active hostilities, the lack of digital educational resources suitable for online learning, and the lack of basic digital skills and competencies among students and teachers necessary to properly conduct online classes. Some of the problems of online learning were solved in the pandemic, but in the context of war Ukrainian society needs a new vision of education and continuous efforts of all social structures in the public and private environment. In the context of war, concerted action is needed to keep education on track and restore it in active zones, adapting to the needs of a dynamic society and an increasingly digitized economy. Among the urgent needs of the education system are a change in the teaching-learning paradigm, which is based on content presentation, memorization, and reproduction, and the adoption of a new, hybrid educational model that will encourage the development of necessary skills and abilities for students and learners in a digitized society and enable citizens close to war zones to learn.

Immersion in sea cucumber's steroid extract to increase male production of juvenile freshwater crayfish

  • One of the ways to increase the production for aquaculture is through the cultivation of monosexuals by ensuring genital reversal from which energy for reproduction is diverted towards growth. Masculinization has been identified as one of the most prominent techniques, where sex development was directed from female to male. This approach only altered the phenotype and not the genotype. The red claw crayfish (Cherax quadricarinatus) was a relatively new commercial commodity, and the males were known to grow faster than females. Hence, it was proposed to use monocultures comprising an all-male population to increase yield using steroid hormone, synthetic 17α-methyltestosterone. However, this technique generated residues that detrimentally affect human health, the environment, and cultivated organisms. Therefore, finding new safe natural steroid sources was essential, and one of which is exploring of natural hormones extracted from the viscera of sea cucumbers (Holothuria scabra Jaeger). This study focused on the determination of male formation and testosterone levels among juvenile crayfish, after immersing in sea cucumber steroid extract (SCSE). A completely random design with factorial was used with two variables, encompassing the varied doses (0, 2, 4 mg/L, 2 mg/L 17α-methyl testosterone as control group) and immersion times of 18 and 30 h. The result showed the dose-dependent ability of SCSE increase the male genital formation and promote the testosterone level of juvenile crayfish. In addition, the testosterone was influenced by dose and immersion duration time, with the highest level of testosterone observed in treatments of 4 mg/L SCSE with 30 h immersion was 0.248 ng/mL, while the male percentage was 77%. In conclusion, the combination of dose and immersion time significantly affected growth and testosterone levels.

종모돈의 정액성상과 번식성적에 미치는 품종, 연령, 계절, 산차 및 교배방법의 영향 (Effect of Breed, Age, Season, Parity and Mating Type on Boar Semen Characteristics and Fertilizing Capacity)

  • 본 연구는 1996년부터 1999년까지 다비 AI 센타에서 사육되는 종모돈 199두와 종빈돈 4,181두를 가지고 종모돈의 정액생산과 번식성적에 미치는 품종, 연령, 계절, 산차 및 교배방법의 영향을 구명하고자 실시한 바, 그 얻어진 결과는 다음과 같았다. 1. 정액량은 Landrace (L) 종이 266.8 $m\ell$로 제일 많았고, Yorkshire (Y) 종이 그 다음으로 많았으며, Duroc (D) 종이 제일 적었다. 정자 운동성은 품종간에 차이가 없었다. 정자농도는 Landrace 종이 4.7$\times$109 sperm/$m\ell$로 제일 낮았고, 정액량이 제일적은 Duroc 종이 5.7$\times$$10^{9}$ sperm/$m\ell$ 로 제일 높았다. 종모돈의 연령에 따른 정액량은 2세 때가 213.3 $m\ell$로 제일 많았고, 4, 3 그리고 l 세의 순서로 정액량이 감소하였다. 2. 종모돈의 계절에 따른 정액량은 겨울철이 228.6 $m\ell$로 제 일 많았고, 가을, 여름, 봄의 순서로 정액량이 감소하였다. 정자농도는 봄철이 5.9$\times$109 sperm/$m\ell$로 제일 높았으며, 여름, 겨울, 그리고 가을의 순서로 낮았다. 3. 교배방법에 따른 복당 생시체중은 품종간 처리구간에 차이점을 인정할 수 없었으나 복당 출생시 생존수에 있어서는 자연종부후 인공수정을 실시한 처리구에서 9.5두로 제일 많았으며, 인공수정을 실시한 처리구에서는 9.2두, 그리고 자연종부를 실시한 처리구에서는 8.9두로 처리구간에 유의성 (P<0.01)이 인정되었다. 4. 교배그룹간에서는 L (우)$\times$Y (♂) 교배조합과 L (♀)$\times$L (♂) 교배조합이 Y (♀)$\times$ L (♂) 교배조합 Y (♀)$\times$Y (♂) 교배조합보다 복당 생시체중이 무거웠으며, 복당 출생시 생존 자돈수에 있어서도 비슷한 경향을 나타내었다. 자연종부와 인공수정간에 복당 생시체중과 복당 출생시 생존자돈수는 유의차가 없었다. 5. 산차간에 있어서는 2~6 산차의 복당 생시체중과 복당 출생시 생존 자돈수가 각각 13.5 kg 과 9.5 두로 1산차와 7~9 산차에 비하여 우수하였다.

생쥐 난소에서 Glucosamine-6-Phosphate Deaminase (GNPDA)의 발현 (Expression of Glucosamine-6-Phosphate Deaminase (GNPDA) in Mouse Ovary)

  • 생쥐 난소에서 glucosamine-6-phosphate deaminase (GNPDA)의 발현을 조사하였다. Western blot상에서 Mr. 31 kDa의 항원을 검출하였으며 출생 후2주에 급격한 발현의 증가가 확인되었다. 난소절편의 면역염색 결과 협막세포와 간충조직에서는 균질한 발현을 보인 반면 난포내 발현양상은 난포의 발달에 따라 다르게 나타났다. 일부 1차 난포의 난자에서 GNPDA의 발현이 관찰되었으나 강소형성 난포의 난자에서는 관찰되지 않았다. 황체화 과립세포에서의 발현은 황체발달에 따라 증가하였으며 황체퇴화 부위에서 강한 신호가 검출되었다. 난포발달에 따른 GNPDA발현의 차이는 GNPDA가 생쥐 난소 조직 재구성에 관여하고 있음을 암시한다.

Sex Ratio and Sex Reversal in Two-year-old Class of Oyster, Crassostrea gigas (Bivalvia: Ostreidae)

  • The sex ratio (F:M) in the same population of oyster, Crassostrea gigas at the commencement of the study (2007) was 1:1.0, but changed to 1:2.8 by the end of the study (2008). The sex reversal rate in two-year-old oysters was 40.2%. Specifically, female to male sex reversal rate was 66.1%, which is higher than the male to female sex reversal rate of 21.1%. The sex reversal pattern of C. gigas appears to go from male ${\Rightarrow}$ female ${\Rightarrow}$ male, and as such is determined to be rhythmical hermaphroditism.
