Modification of Estrogenic Effect of Nonylphenol Combined with DEHP in Yeast-based Bioassay (형질전환효모를 이용한 내분비계장애물질검색과 Nonylphenol의 Estrogen 유사작용에 대한 DEHP의 상협작용)

  • The key targets of endocrine disruptors are nuclear hormone receptors, which bind to steroid hormones and regulate their gene transcription. A yeast-based steroid hormone receptor gene trascription assay was previously developed for the evaluation of chemicals with endocrine modulating activity. The yeast transformants used in this assay contain the human estrogen receptor along with the appropriate steroid response elements upstream of the $\beta$-galactosidase reporter gene. We tried to evaluate several natural and synthetic steroids of their potential to interact directly with the steroid receptor. Some putative endocrine disruptors, including nonylphenol, are weakly estrogenic. But the combined treatment oj these chemicals with di-(2-ethylhexyl)phthalate (DEHP) significantly increased the $\beta$-galactosidase activity in the yeast transformant. These results suggest that we also have to consider the synergistic effects of endocrine disruptors. In this study, we showed that yeast-based bioassay is a valuable tool for screening potential endocrine disruptors and quantitative determination of estrogenicity. And the possibility that the estrogen receptor binds multiple environmental chemicals adds another level of complexity to the interaction between the endocrine disruptors and the human hormone system.

A Study of Subclinical Thyroid Function Disorder and the Risk of Coronary Heart Disease in Women (여성의 무증상 갑상샘 기능이상과 관상동맥질환 발생과의 관련성 연구)

  • Purpose: This paper has attempted to investigate the association between the subclinical thyroid function disorder caused by natural change in thyroid hormones or thyroid remedies and the incidence of CHD. Methods: Data was analyzed of 7,675 women who had undergone health examinations in a hospital in Gyeonggi-do between January 2007 and December 2008. The covariation of the coronary risk factors such as age, BMI, HbA1C, systolic blood pressure, LDL:HDL-cholesterol ratio, CRP, smoking and exercise were analyzed by using logistic regression analysis. Results: A significant increase in serum TSH was observed with higher age (F=26.91, p=.00). In terms of age, the risk of CHD started to gradually increase since the 40s and sharply since the 60s (${\chi}^2$=113.29, p=.00). The serum TSH was the most significant parameter influencing CHD (B=.12, p=.00). The risk of coronary heart disease was 3.12 times higher in the subclinical hypothyroidism group (OR=3.12) while no significant difference was observed in the subclinical hyperthyroidism group. Conclusion: Subclinical hypothyroidism may be an independent risk factors for CHD. A nurse nursing patients with thyroid disorder should be well informed of their state of subclinical thyroid function disorder and make efforts to extend their health expectancy.

  • This study was carried out to investigate the ultrastructural changes of ooplasm and follicular membrane of oocytes, obtained from 150 of 3-year-old female rainbow trout(Oncorhynchus mykiss). All data were collected from March in 1989 to February in 1990, and from August to October in 1991. The size of the nucleoli and number of the yolk granules increased as the oocyte growed. Yolk granules were deposited in the oocyte as crystalline granules. Due to the presence of large early and late maturing oocytes, their ovaires were enlarged, transparent and granular. The lattice was broken down at hydration, leaving the egg transparent. As thepercentages of fish in LPO and EMO stage increased from September to October, Mean GSI values increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Seasonal changes in the microscopic appearance of the ovaries were well correlated with those in bothgonadosomatic index and macroscopic apearance. Under the natural conditions,t he ovarian follicle influences the histological development and periodical secretion of the hormones, sufficient for a oogenesis and gonadal steroid production. The electrophoretic pattern of major band in mature stage was much thicker(70∼110k dalton) than that in previtellogenic phase.

Evidence for Volatile Memory in Plants: Boosting Defence Priming through the Recurrent Application of Plant Volatiles

  • Plant defence responses to various biotic stresses via systemic acquired resistance (SAR) are induced by avirulent pathogens and chemical compounds, including certain plant hormones in volatile form, such as methyl salicylate and methyl jasmonate. SAR refers to the observation that, when a local part of a plant is exposed to elicitors, the entire plant exhibits a resistance response. In the natural environment, plants are continuously exposed to avirulent pathogens that induce SAR and volatile emissions affecting neighbouring plants as well as the plant itself. However, the underlying mechanism has not been intensively studied. In this study, we evaluated whether plants "memorise" the previous activation of plant immunity when exposed repeatedly to plant defensive volatiles such as methyl salicylate and methyl jasmonate. We hypothesised that stronger SAR responses would occur in plants treated with repeated applications of the volatile plant defence compound MeSA than in those exposed to a single or no treatment. Nicotiana benthamiana seedlings subjected to repeated applications of MeSA exhibited greater protection against Pseudomonas syringae pv. tabaci and Pectobacterium carotovorum subsp. carotovorum than the control. The increase in SAR capacity in response to repeated MeSA treatment was confirmed by analysing the defence priming of the expression of N. benthamiana Pathogenesis-Related 1a (NbPR1a) and NbPR2 by quantitative reverse-transcription PCR compared with the control. We propose the concept of plant memory of plant defence volatiles and suggest that SAR is strengthened by the repeated perception of volatile compounds in plants.

A Study on Diagnosis of the Fertility of one Aged Female Gorilla by using the Fecal Sex Hormone Metabolites

  • The aim of this study was to diagnose the fertility of a female western lowland gorilla kept in Seoul Zoo, in accordance with age by analyzing the fecal sex hormone metabolites. The study was conducted in two period of times, when the animal was from 35 to 37 years old and when the animal was from 40 to 42 years old. Non-invasive method by using fecal samples was used for safe and efficient fertility diagnosis. We collected the feces from the enclosure at least three times a week. Then $17{\beta}$-estradiol and progesterone, which are fecal sex hormone metabolites, were measured by time-resolved fluoro-immunoassay to compare the menstruation cycle and the annual reproductive cycle. For the duration of the primary study (when the animal was 35~37 years old), irregular menstruation and high concentrations of estradiol and progesterone were observed. However, menstruation was hardly observed and the concentrations of both hormones were statistically very low in the period of secondary study (when the animal was 40~42 years old). This observed phenomenon in our study was very comparable to menopause in adult women; therefore, it was confirmed that our female gorilla has reached menopause because of the natural aging, as they become older.

Development of Health Assistances for Anti Stress used with Ostreae Concha (모려분을 이용한 한의학적 항스트레스성 기능보조제 개발)

  • Ostreae concha is natural products of sea that haves good function for anti-stress. WE observed the effect of Otreae concha, Yukkwooltang and yukwool-tang add Ostreae concha for catecholamine, GOT, GPT, LDH and TG after immobilization stress, Ostreae concha depressed the stress response. In immunohistochemical analysis, The area of withe pulp of the spleen decreased in the control groups as compared with control ones. At same time, more severe histological changes of spleen such as the decreased diameter of periarterial lymphoid sheath and splenic node were demonstrated in the rats challenged with emotional stress. But these changes were slightly recovered in rats pretreated with Sample I, Sample II and Sample III. Immunohistochemical analysis showed that the expression of the inflammation-related protein tended to increase. But a decline of these expression, especially TNF-${\alpha}$ by pretreated with Sample I, were observed in the red pulp of spleen of the experimental rats. These results suggest that ostreae concha may act as a therapeutic agent for stress disease through a regulation of stress-related hormones.

  • Bone homeostasis is maintained by a balance between activities of osteoblasts(bone forming cells) and osteoclasts (bone resorbing cells). The activities of these cells are closely regulated by multiple factors including hormones and cytokines. The cessation of estrogen at menopause disrupts the balanced regulation and is the main cause of osteoporosis in postmenopausal women. Recent molecular biological studies led to a discovery of tumor necrosis factor(TNF) and TNF receptor families genes that play critical roles in the regulation of osteoclast formation and function. RANKL (receptor activator of nuclear factor kappa B ligand; also called ODF, TRANCE, and OPGL) expressed on cells supporting osteoclast is essential for osteoclast differentiation, activation, and survival. RANK, the counter-receptor for RANKL, is expressed on progenitor and mature osteoclasts. The interaction between RANKL and RANK is requlated by a soluble decoy receptor OPG (osteoprotegerin). Gene knock out studies of these molecules showed profound effects on bone. These results prompted development of new strategies for treatment of bone diseases. Inhibition of osteoclast activity by blocking the RANKL-RANK interaction using OPG is being attempted. Research on the signaling pathways of RANK is also actively carried out. Screening natural products that inhibit the RANKL-RANK interaction or the activity of obteoclasts would be another effective means to a new drug target for bone resorbing diseases.

On the Activity of Phosphatase in the Endometrium of the Rat Uterus During Early Pregnancy (초기 임신 기간중 흰쥐 자궁 내막조직의 Phosphatase 활성에 관하여)

  • The quantitative analyses of the phosphatase activity in the endometrium of the rat ovariectomized on Day 2 of pregnancy was carried out in comparison with the intact one, in order to investigate the hormonal dependency of the uterus prior to the implantation, and to study the phosphatase activity in the endometrial tissues in vitro incubated in different acidity of the medium. The results obtained were as follows: 1. The activity of the total phosphatase was the highest at Day 3 of pregnancy of the intact animals irrespective of acidity of the medium. However, the ovariectomized rat showed its peak somewhat delayed. The time of the highest activity of the enzymes was matched with the time of high secretion of the ovarian hormones. 2. The activity of acid phosphatase in the endometrium was twice or four times as much high as that of neutral or alkaline phosphatase, respectively. 3. The activity of alkaline phosphatase was rather steady in Day 3 through Day 5 of the pregnancy of the rat intact or ovariectomized but with low level compared to those of other phosphatase. 4. The present re~lt indicated more important role by $Mg^{2+}$-dependent phosphatase than by $K^+$-dependent one for the preparation for decidualization.

Endocrinological Studies and Potential Biomedical Uses of Antlers

  • Antlers from deer species are alternative animal by-products. Due to the oriental trade, the velvet antler industry is rapidly emerging in North America. The unique biological property of antler wish a deciduous natural phenomenon offers the valuable model of biomedical research. Growing antlers showed different structures according to cell populations consisting of mesenchymes, chondroblasts, chondrocytes and osseous tissues from distal to proximal portions of main beam. Their structures were different from two tissues, cartilage and bone, in growing antlers. Zone of maturing ants calcifying chondrocytes referred as upper section was invaded by osteoblasts indicating the occurrence of endochondral ossification. The cartilaginous tissues were gradually replaced by osseous tissues downward. The bony tissues referred as the middle and base sections in this thesis contained spongy bone and cortical bone structure in the difference of the degree of mineralization antral the thickness of cortical bony in adjacent to outer velvet layer. In addition, the endocrinological regulators such as testosterone, prolactin, growth hormones and other growth factors are actively involved in the unique deciduous nature shown in the growth and development of antler.

Sex-Biased Molecular Signature for Overall Survival of Liver Cancer Patients

  • Sex/gender disparity has been shown in the incidence and prognosis of many types of diseases, probably due to differences in genes, physiological conditions such as hormones, and lifestyle between the sexes. The mortality and survival rates of many cancers, especially liver cancer, differ between men and women. Due to the pronounced sex/gender disparity, considering sex/gender may be necessary for the diagnosis and treatment of liver cancer. By analyzing research articles through a PubMed literature search, the present review identified 12 genes which showed practical relevance to cancer and sex disparities. Among the 12 sex-specific genes, 7 genes (BAP1, CTNNB1, FOXA1, GSTO1, GSTP1, IL6, and SRPK1) showed sex-biased function in liver cancer. Here we summarized previous findings of cancer molecular signature including our own analysis, and showed that sex-biased molecular signature CTNNB1High, IL6High, RHOAHigh and GLIPR1Low may serve as a female-specific index for prediction and evaluation of OS in liver cancer patients. This review suggests a potential implication of sex-biased molecular signature in liver cancer, providing a useful information on diagnosis and prediction of disease progression based on gender.
