육류 식품중 호르몬 성분의 분석 (Determination of hormonal active compounds in meat)

  • 가축의 생산성 향상을 위해 투여될 수 있는 천연호르몬과 합성 호르몬제 등의 내분비계장애물질에 대하여 기존의 분석 방법을 개선 및 확립하고, 이를 토대로 국내에 유통 중인 육류 및 내장부위 중소간에 대해 호르몬 성분의 분포정도를 확인하였다. Estrogens ($17{\beta}$-estradiol, $17{\alpha}$-estradiol, estrone), androgens ($17{\beta}$-testosterone, $17{\alpha}$-testosterone), gestagens (progesterone)의 천연호르몬 6종과 합성호르몬 3종 DES, RALs (zeranol, taleranol)을 모니터링 대상 호르몬으로 GC-MS로 분석하였으며, 이로부터 호르몬의 background level을 측정하고, 상대적으로 호르몬제 등이 잔류될 수 있는 간 등의 장기에 대한 분포조사를 실시하였다. 전체 150건의 시료 중 합성 호르몬제는 검출되지 않았으며, 천연호르몬제는 내인성으로 존재하는 progesterone이 일반적인 농도 범위를 보였다.

Sensitive Determination of Natural and Synthetic Steroidal Hormones of a Free and Conjugated Form in Surface Water by Gas Chromatography-Mass Spectrometry

  • A method based on the TMS derivatives and acidic hydrolysis was developed for the simultaneous determination of free and conjugated steroidal hormones in surface water. A silylation of five natural and two synthetic steroidal hormones was achieved with N-methyl-N-(trimethylsilyl) trifluoroacetamide/$NH_4I$ (1000:3) under catalysis of dithioerythritol for 60 min at $80^{\circ}C$. TMS derivatives of the steroid hormones containing multifunctional groups offer a single derivative product under this condition. The accuracy of the analytes was in the range of 87 to 110% at a concentration of 20 and 50 ng/L with relative standard deviations of less than 10%. The method detection limit was in the range of 0.01 to 0.02 ng/L for surface water. Natural steroidal hormones were detected in a concentration range of 0 to 1.03 ng/L in free form and 0 to 14.6 ng/L in conjugated form, respectively. We found that most of the natural hormonal steroids exist in conjugate forms (43 to 100%) in river water.

Regulation of gastrointestinal hormones during laxative activity of gallotannin-enriched extract isolated from Galla Rhois in loperamide-induced constipation of SD rats

  • Regulation of gastrointestinal hormones have been reported in animal models for constipation undergoing laxative therapy when administered herbal products. We undertook to investigate whether the laxative activity of gallotannin-enriched extracts isolated from Galla Rhois (GEGR) affects the regulation of gastrointestinal hormones, by examining the concentration of four hormones and the activation of their receptors in the loperamide (Lop)-induced constipation model. Stool parameters, including number, weight and water content, were significantly recovered in the Lop+GEGR treated group, relative to the Lop+ vehicle treated group; however, food intake and water consumption were maintained at a constant level. Also, a similar recovery was detected for thickness of mucosa, muscle and flat luminal surface in the Lop+GEGR treated group. Furthermore, concentration of the four gastrointestinal hormones evaluated, namely, cholecystokinin (CCK), gastrin (GAS), somatostatin (SS) and motilin (MTL), were lower in the Lop+vehicle treated group than the No treated group, but were remarkably enhanced in the Lop+GEGR treated group. Moreover, the downstream signaling pathway of MTL and SS receptors were recovered after GEGR administration. Results of the present study therefore indicate that the laxative effects of GEGR treatment may be tightly related with the regulation of gastrointestinal hormones in the Lopinduced constipation model.

분변내 성호르몬의 분석을 통한 멸종위기 사육 오랑우탄에서의 번식형태 (The Reproductive Patterns of Endangered Captive Orangutans by Analysing the Sex Hormones in Feces)

  • To monitor the reproductive patterns of endangered orangutan, Time-resolved fluorescence immuno assay (TR-FIA) were used to analyze metabolites of sex hormones in feces. Orangutan had long-term interbirth intervals (amenorrhea) during lactation period in which the secretion of sex hormones was repressed. The concentration of progesterone in the serum of pregnant orangutan was 30fold higher than that in non-pregnant orangutan. However, the concentration of hCG during pregnant period was different from the result of other primates. The present study suggested that age is not the important factor in determining the reproduction capability, because it is rather greatly influenced by the combination of various factors. Tracing metabolites of sex hormones in orangutan feces seems to be provide the effective solution for the infertility in orangutan. This study result shows the basic data in operating conservation project for endangered orangutan.

옥수수 유식물의 엽록소-단백질 복합체 형성에 미치는 식물호르몬 및 광선의 효과 (The Effect of Plant Hormones and Light Quality on the Formation of Chlorophyll-Protein Complexes in Maize Seedlings)

  • 자연광 및 여러 파장의 광선과 NAA, GA$_3$, BA 등의 식물호르몬을 옥수수 유식물에 처리하여 엽록소-단백질 복합체 형성에 미치는 효과를 조사하였다. 자연광하에서 3 종류의 호르몬 처리는 전체적으로 엽록소-단백질 복합체, 특히 LHCP-1과 LHCP-3의 형성을 촉진하였으나, 2 종류의 호르몬 조합은 엽록소-단백질 형성에 효과적이지 못하였다. LHCP-1, CPA 및 LHCP-3 등의 광계 II 관련 엽록소 형성에 있어서 백색광은 자연광에 비하여 효과가 적었으나 적색광은 효과적이었다. 적색광하에서 식물호르몬의 단독처리는 엽록체 발달 초기에 광계 II의 엽록소-단백질의 양적 증가를 유도하였다. 한편, 적색광하에서 2 종류의 호르몬 조합 처리도 자연광하에서의 처리와는 대조적으로 광계 II의 엽록소-단백질 형성에 매우 효과적이었다. 이와 같은 결과는 광선처리의 효과가 호르몬처리 효과보다 엽록소-단백질 복합체 형성에 중요한 인자로 작용한다는 것을 시사해 준다.

The role of diuretic hormones (DHs) and their receptors in Drosophila

  • Maintaining internal homeostasis and regulating innate behaviors are essential for animal survival. In various animal species, a highly conserved neuroendocrine system integrates sensory inputs and regulates physiological responses to environmental and internal changes. Diuretic hormones 44 and 31, which are homologs of mammalian corticotropin-releasing factor (CRF) and calcitonin gene-related peptide (CGRP), respectively, control body fluid secretion in Drosophila. These neuropeptides and their receptors have multiple physiological roles, including the regulation of body-fluid secretion, sleep:wake cycle, internal nutrient-sensing, and CO2-dependent response. This review discusses the physiological and behavioral roles of DH44 and DH31 signaling pathways, consisting of neuroendocrine cells that secrete DH44 or DH31 peptides and their receptor-expressing organs. Further research is needed to understand the regulatory mechanisms of the behavioral processes mediated by these neuroendocrine systems.
