This study was to make a effective seed mixture types with Korean native plants by researching te seed timing of suitable woody plants and competitive germination results of various plants for the purpose of cut-slotes revegetation. The results are summarized as follows. 1. In field polystand experiment, comparative among severalcomposition combination for early coverage shows good condition. 2. In combination for early-coverage, mixture of native woody plants only seeded in May and June shows over 90% coverage. It was not inferior to the other mixture types. So mixture of Albizzia Julibrissin, Cymbopogon doeringii, Alnus hirsuta and Indigfera pseudo-tinctoria is able to use to mixture type. 3.In case of combination for scenic beauty, Rhus chinensis shows low germination in the mixture for scenic beauty. But 90 days after seeded in May, it recorded maximum 44 cm height, seeded in June recorede 18 cm height. 4. Lespedeza crytobotrya shows good elongation in the combination for early coverage and scenic beauty, but that effects on elongation of other plants. So, it demands close investigation about suitable mixing rate of plants. 5. Alnus hirsuta, Pinus thunbergii, Betula platyphylla, Spiraea prunifolia var. simpliciflora have been rarely germinated. So, the mixture of these plants wasn't adequate to use for revegatation.

To suggest the native plants combination for restoration of the dissected sloping area, the plant communities of thirty seven quadrats from five localities of mid-southern part of Korean peninsula were surveyed from 2007 to 2008. Their flora, vegetation structure, and dominant species based on the importance value were investigated. And the soil characters (pH, moisture content, water holding capacity, and organic matter) of each dominant species were analyzed. Also, germination tests were performed to check the stability of restored native plants using seed chips. As a result, 79 native plants were suggested for the dissected sloping area : 20 trees and subtrees for the upper layer, 18 shrubs and vines for the middle layer, and 41 herbs for the lower layer, taken together their flora, dominant species, vegetation structure, soil condition, germination test, flowering period.

This study was carried out to develop the environmental friendly restoration technique for various disturbed slopes using native plants seed chips. As the result of the separated seeding using the developed seed chips of native plant combination, average germination rate and survival rate of trees, shrubs, and herbs were higher than those of the direct seeding, as 79~70%, 83~73%, and 59~53% or so, respectively. Also, the germination rates and the survival rates of trees, shrubs, and herbs in the separated seeding by the native plants seed chips were 10~15 times higher than those of the direct seeding. Numbers of plants survived were at least 10 times of that of the simple seeding. The native plants seed chip technique is more efficient in soil slopes compared with stone slopes. The application of the native plants seed chip technique makes the restoration stabilized through the herbs dominant vegetation to the woody dominant 1 within 3 years, and the Landscapes also changed from herbs to variously combined herbs and woody. The technique using native plants seed chips that can be applicable to various disturbed slops is more efficient than conventional restoration techniques in the respect of economics and landscape.

Currently, there are researches about environment-friendly road construction plans led by the Ministry of Construction and Transportation in progress. Therefore, in order to create the revegetation techniques of thin layer-soil combination media hydroseeding measures by actively using native herbs and native woody plants instead of using imported foreign grasses as a concept of sustainable environment-friendly land development, this thesis is going to identify problems that can appear when applying the thin- layer-soil-media hydroseeding measures by the suggested in the "Slope revegetation design and guidelines" proposed by the Ministry of Construction and Transportation, and to propose improvement plans for the problems.To this aim, a seeding mixture selection test was conducted by the goal of slope restoration, and a test group for artificial slope was created. As for a test for June sowing, it was intended to identify appropriate combination quantity by conducting a test that differentiates the combination quantity, and as for a test for September sowing, an artificial slope test was conducted by creating an artificial bank for earth and soil and applying 1~2cm and 3~4cm thickness after differentiating the seed combination volume and slope aspects.

This study was conducted to suggest the ecological restoration and environmentally friendly revegetation technology for the rock-exposed cut-slopes by the Natural Topsoil Restoration Methods (NTRM) with the following restoration objectives; (1) prevention or reduction of wind and water erosion, (2) provision of food and cover for variety of animal species, (3) improvement of the visual or aesthetic quality of the disturbed slopes. On Nov. in 1995, the 5cm thick layer of artificial soil and 2cm thick layer of straw-mulching was attached at rock-exposed cut-slopes by NTRM without using anchor wire and anchor pin. The main results during four years surveying on the ground-coverage effect, plant growth, species diversity and importance values were summarized as follows. 1. Artificial soil attached at rock exposed cut-slopes was not eroded until recovered by tree and herbaceous vegetation in spite of not using anchor wire and anchor pin. Also it shows low soil hardiness and has almost the same amount of bacteria and fungi with in surrounding natural topsoil. 2. In 'combination for the woody vegetation', Lespedeza cyrtobotrya, Albizzia julibrissin, Rhus chinensis, Indigofera pseudo-tinctoria occupied upper layer vegetation. Since three years after seeding, Indigofera pseudo-tinctoria had overwhelmed the other woody plants and cool season foreign grasses, Erigeron canadensis, Taraxacum mongolicum, Commelina communis, Arundinella hirta (Thunberg) and Oenothera erythrosepala grows at lower part of the vegetation, 3. The heights of the Rhus chinensis grows 1.8m, Indigofera pseudo-tinctoria 2.0m, so it seems that the objectives of woody vegetation with native plants could be accomplished. 4. After 4 years later after seeding in 'combination for the herbaceous vegetation', the most dominant plant was Indigofera pseudo-tinctoria, the next was in order of cool-season grasses, Taraxacum mongolicum, Erigeron canadensis, lxeris dentata (Thunberg), Oenothera erythrosepala, Arundinella hirta (Thunberg). The diversity index in 'combination for woody vegetation' was higher than that in 'combination for the herbaceous vegetation'. The tendency of the intrusion of secondary succession plants was more effective in 'combination for the herbaceous vegetation' than in 'combination for the woody vegetation'.
Food poisoning accidents caused by poisonous plants occur every year. As certain poisonous plants are mistaken for edible plants causing food poisoning, accurate species identification of poisonous plants is required. DNA barcodes suitable for species identification of poisonous plants and database that can be used for accurate species identification are necessary for their use in forensic cases. In this study, species identification of 19 poisonous plants native to Jeju Island using seven DNA barcodes (trnH-psbA, trnL-trnF, trnL intron, rbcL, matK, ITS1-ITS4, 18S rRNA) was performed to construct a database containing sequence information and DNA barcode universality. trnL-trnF barcode and ITS1-ITS4 barcode were the easiest markers for PCR amplification and sequence retrieval, and the combination of the two barcodes enabled single species identification in 18 out of 19 plants. Therefore, when an investigation of unknown poisonous plants is requested, combination of trnL-trnF and ITS1-ITS4 barcodes is considered as a primary marker for species identification. The database of recommended DNA barcodes for each poisonous plant presented in this study will be helpful in plants poisoning cases.
Twelve Korean native goats, spontaneously infected with mixed species of Eimeria were used to study the possible direct anticoccidial effect of feeding condensed tannin-containing plants on the production of Eimeria oocysts. The effects of feeding pine (Pinus densifora) needles, oak (Quercus acutissima) leaves and lucerne chaff on coccidia oocyst output were studied for a period of 10 days post-feeding. The results indicate that feeding fresh pine needles (40 g condensed tannins (CT) dry matter (DM)/day/goat) and oak leaves (40 g CT DM/day/goat) in combination with lucerne chaff had rapid anticoccidial activities in goats as demonstrated by a sharp decrease in oocyst production. Two days after feeding, the numbers of oocysts per gram of faeces (OPG) from the goats fed pine needles with lucerne chaff, and from goats fed oak leaves reduced by 40% and 44% compared to pre-feeding, respectively. On the sixth day after commencing feeding pine needles and oak leaves, the reduction was 81% and 72%, respectively. Ten days after feeding pine needles and oak leaves, the OPG was reduced by 93% and 85%, respectively compared to pre-feeding. Statistical analysis showed that feeding pine needles and oak leaves to goats naturally infected with coccidia significantly (p<0.001) reduced the numbers of oocysts compared to the control group fed lucerne chaff only. Four clinically important species of coccidia, Eimeria parva, Eimeria ninakohlyakimovae, Eimeria christenseni and Eimeria arloingi were identified in Korean native goats.
Rosa rugosa is a medicinal, ornamental, and edible plant native to Eastern Asian countries, including Korea, Japan, and China. The aim of this study was to establish a system for biomass production and secondary metabolite accumulation during in vitro culture and acclimatization of Rosa rugosa. The highest rate of multiple shoot proliferation was achieved with $8.8{\mu}M$ benzyladenine (BA) (83.3%). However, the number of shoots (14.4 per explant) at $4.4{\mu}M$ BA was higher than that at $8.8{\mu}M$ BA. Compared to BA, a combination of thidiazuron (TDZ) and indole butyric acid (IBA) exhibited significantly lower shoot induction, with only 50.0~79.2% and 4.2~16.7% relative shoot formation, respectively. During acclimatization, shoots were sampled every week and their total phenolic contents were analyzed. Among various growth factors, fresh weight showed the most dramatic increase from the 3rd week (88.0 mg/plant) to 4th week (132.7 mg/plant). Total phenolics and flavonoids contents were the highest at $1^{st}$ week of acclimatization. Depending on developmental stages, total phenolics and flavonoids contents were higher in 1-yr-old shoots grown ex vitro than in those of older field-grown or in vitro-grown plants. Amongst different ages of field grown plants, 6-year-old plants, the oldest in this study, showed the lowest content in total phenolics.

Droughts are one of the abiotic stresses that hinders the growth and productivity of crop plants. Coping with abiotic stress is necessary to understand the molecular regulatory networks that makes plants respond to adverse environmental conditions. In our experiment to find a combination that can cope with abiotic stress (respond to drought), we screened 5 stress-inducible promoters that are expressed only under stress conditions. This founded 36 cis-elements in stress-inducible promoters. With the result we designed 2 synthetic promoters (BL1, BL2) for fine-controlled regulation by assembling cis-elements from the native promoters, which are expressed only under stress caused by droughts. Analysis of the transgenic plant (BL1-GUS, BL2-GUS) showed that the synthetic promoters increased the expression of β-glucuronidase (GUS) in transgenic plants under desiccation. Also in the transient activation assay demonstrated that synthetic promoters induced the co-transformation of effector DREB1A and DREB2C. These results expect that the synthetic promoter with a combination of drought-specific elements can be used to respond to various abiotic stress and is resistant to stress without causing growth retardation.

Ex-situ conservation of the ornamental and medicinal orchid, Coelogyne stricta, was performed by mass propagation using seed culture. Propagation stages were optimized using full- and half-strength solidified MS medium with different phytohormones. Maximum seed germination (88 ± 0.5% over 6 weeks of culture) was achieved on half-strength MS medium supplemented with 15% coconut water. Maximum shoot numbers were found on full-strength MS medium supplemented with 1 mg/L BAP, 2 mg/L Kinetin, and 10% coconut water, while the longest root was developed on full-strength MS medium with 1.5 mg/L IBA. A 2:1:1 combination of coco-peat, pine bark, and sphagnum moss was found to be a suitable potting mixture resulting in 80% seedling survivability. The cytotoxic activity of extracts of both wild plants and in vitro-developed protocorms was determined using an MTT (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide) assay on a cervical cancer cell line. The wild plant extract inhibited the growth of 41.99% of cells, showing that this extract has moderate cytotoxic activity toward cervical cancer cells.
