Genetic Diversity of mtDNA D-loop Polymorphisms in Laotian Native Fowl Populations

  • Here, we studied the genetic diversity of native fowls in Laos by analyzing a mitochondrial DNA (mtDNA) sequence polymorphism. A 546-bp fragment of the mtDNA D-loop region was sequenced in 129 chickens from the areas of Vientiane, Luang Prabang and Pakse. In total, 29 haplotypes were identified and formed five clades. Haplotype diversity and nucleotide diversity of the native fowls in Laos were $0.85536{\pm}0.0172$ and $0.010158{\pm}0.005555$, respectively. Although the Laotian native fowls were distributed across five clades, most of them were clustered in two main clades (A and B), which were originated in China. The other haplotypes were contained in clades D, F, and I, which originated from continental southeast Asia. These results suggest that multiple maternal lineages were involved in the origin of domestic chicken in Laos. Moreover, there appear to be at least two maternal lineages, one from China and the other from the southeast Asian continent.

Prevalence of Theileria sergenti infection in Korean native cattle by polymerase chain reaction

  • This study was performed to investigate the prevalence of theileriosis and to compare the prevalence of this disease in Korean native cattle reared under different environmental conditions, namely, in a grazing area and a non-grazing area by polymerase chain reaction. Three hundred and one Korean native cattle (276 cows and 25 bulls) that had not received prior treatment or been vaccinated to prevent theileriosis were examined by PCR for Theileria sergenti infection from 2001 to 2002. In our study, the parasitemia range in T. sergenti-positive cattle by microscopy were from 0.1 to 3% (mean 0.8%). In terms of mean prevalence, 204 of the 301 Korean native cattle (67.8%) were positive reaction by PCR. Our results also revealed that the infection rate among cows (70.3%) was significantly higher than that among bulls (40.0%) (p < 0.01). T. sergenti infection among the over 3 year-old-group (75%) had a significant higher prevalence than that among the less than 3 year-old-group (61.8%) (p < 0.05). Our data also showed that grazing areas (76.1%) had the significant higher prevalence than non-grazing areas (51%) (p < 0.001). In conclusion, this study demonstrates that the prevalence of T. sergenti infection is high and that its prevalence in grazing cattle is higher than that in non-grazing cattle, Therefore, life-long treatment and the development of an optimal vaccine are needed to reduce the numbers of bovine theileriosis in both grazing and non-grazing areas.

  • The Anthocyanins in the Korean native Brassica juncea, which were favorably used directly for food or as condiments, were investigated. Two kinds of anthocyanin were separated from Korean native Brassica juncea by T.L chromatography and then Robinson test, spectrophotometry, T.L. chromatography, partial hydrolysis were tested for the structural elucidation of these two anthocyanins. It was known that two pigments were peonidin-3-glucoside and peonidin-3-galactoside.

Genetic Diversity and Population Structure in Native Chicken Populations from Myanmar, Thailand and Laos by Using 102 Indels Markers

  • The genetic diversity of native chicken populations from Myanmar, Thailand, and Laos was examined by using 102 insertion and/or deletion (indels) markers. Most of the indels loci were polymorphic (71% to 96%), and the genetic variability was similar in all populations. The average observed heterozygosities ($H_O$) and expected heterozygosities ($H_E$) ranged from 0.205 to 0.263 and 0.239 to 0.381, respectively. The coefficients of genetic differentiation (Gst) for all cumulated populations was 0.125, and the Thai native chickens showed higher Gst (0.088) than Myanmar (0.041) and Laotian (0.024) populations. The pairwise Fst distances ranged from 0.144 to 0.308 among populations. A neighbor-joining (NJ) tree, using Nei's genetic distance, revealed that Thai and Laotian native chicken populations were genetically close, while Myanmar native chickens were distant from the others. The native chickens from these three countries were thought to be descended from three different origins (K = 3) from STRUCTURE analysis. Genetic admixture was observed in Thai and Laotian native chickens, while admixture was absent in Myanmar native chickens.

The Interlanguage Speech Intelligibility Benefit (ISIB) of English Prosody: The Case of Focal Prominence for Korean Learners of English and Natives

  • This study investigated the speech intelligibility of Korean-accented and native English focus speech for Korean and native English listeners. Three different types of focus in English, broad, narrow and contrastive, were naturally induced in semantically optimal dialogues. Seven high and seven low proficiency Korean speakers and seven native speakers participated in recording the stimuli with another native speaker. Fifteen listeners from each of Korean high & low proficiency and native groups judged audio signals of focus sentences. Results showed that Korean listeners were more accurate at identifying the focal prominence for Korean speakers' narrow focus speech than that of native speakers, and this suggests that the interlanguage speech intelligibility benefit-talker (ISIB-T) held true for narrow focus regardless of Korean speakers' and listeners' proficiency. However, Korean listeners did not outperform native listeners for Korean speakers' production of narrow focus, which did not support for the ISIB-listener (L). Broad and contrastive focus speech did not provide evidence for either the ISIB-T or ISIB-L. These findings are explained by the interlanguage shared by Korean speakers and listeners where they have established more L1-like common phonetic features and phonological representations. Once semantically and syntactically interpreted in a higher level processing in Korean narrow focus speech, the narrow focus was phonetically realized in a more intelligible way to Korean listeners due to the interlanguage. This may elicit ISIB. However, Korean speakers did not appear to make complete semantic/syntactic access to either broad or contrastive focus, which might lead to detrimental effects on lower level phonetic outputs in top-down processing. This is, therefore, attributed to the fact that Korean listeners did not take advantage over native listeners for Korean talkers and vice versa.

Characteristics of Seed Germination and Bulb Dormancy in Korean- Native Allium victorialis L. (韓國産 산마늘 種子의 發芽와 種球의 休眠 特性)

  • The experiments were conducted to investigate the characteristics of the seed germination and the bulb dormancy in Korean-native Allium victorialis L. by the physicochemical treatments. The characteristics of two Allium victorialis seeds was similar in shape, color and threshing but different in other characters. Weight of 1,000 grains in Ullrung-native was 14.8g corresponding to 2.1 times as compared with Odae-native. Seeding immediatly after seed collection of Ullrung-native only germinated and chemicals didn't affect in germination improvement. The germination rate, average days to germination and coefficient of germination of Ullrung-native was 86%, 42 days and 2.05 at the dark condition of control, respectively. The germination rate, average days to germination and coefficient of germination of Odae-native was 70%, 47 days and 1.5 at the dark condition of control and 82%, 47 days, 1.75 at the light condition after 5$^{\circ}C$ prechilling for 30 days, respectively. The bedding days to reduce the physiological dormancy in the bulb of Odae-native assumed to be appropriate up to 45 days after the first prechilling.

Identification of Novel SNPs with Effect on Economic Traits in Uncoupling Protein Gene of Korean Native Chicken

  • The avian uncoupling protein (avUCP) is a member of the mitochondrial transporter superfamily that uncouples proton entry in the mitochondrial matrix from ATP synthesis. The sequencing analysis method was used to identify nucleotide polymorphisms within the avUCP gene in Korean native chicken (KNC). This study identified ten single nucleotide polymorphisms (SNPs) in the avUCP gene. We analyzed the SNPs of the avUCP gene to investigate whether polymorphism in the gene might be responsible for quantitative variations in economic traits in KNC. Three significant polymorphic sites for economic traits were avUCP C+282T (mean body weight, p<0.05), avUCP C+433T (daily percent lay, p<0.05), and avUCP T+1316C (daily percent lay, p<0.05). The frequency of each SNP was 0.125 (C+282T in avUCP gene exon 1 region), 0.150 (C+433T in avUCP gene intron 1 region), and 0.15 (T+1316C in avUCP gene exon 3 region), respectively. Among the identified SNPs, one pair of SNPs (genotype CC, C+282T and TT, avUCP C+433T) showed the highest daily percent lay (p<0.05) and mean body weight (p<0.05) and the frequency was 0.067. This study of the avUCP gene could be useful for genetic studies of this gene and selection on economic traits for KNC.

Effects of Dietary Supplementation with Ptecticus tenebrifer Powder on the Mortality Rate and Meat Quality of Korean Native Chickens (동애등에 분말 사료 급여가 토종닭의 폐사율과 육질에 미치는 영향)

  • This study was conducted to investigate the effects of dietary supplementation with Ptecticus tenebrifer powder on the mortality and meat quality of Korean native chickens. A total of 40,000 Korean native chickens (1 day old, Hanhyup No. 3) were allocated to two dietary treatments (20,000 chickens in each treatment), which were fed the following: basal diet (control) and 1% of Ptecticus tenebrifer powder (T1). Feeding trials were conducted for 12 weeks, and mortality was measured weekly. At the end of the experimental period, 16 chickens (8 chickens in each treatment) were selected and slaughtered to obtain breast meat. The items used to analyze meat quality were pH, TBARS, and meat color. The weekly mortality rate was decreased by around 2 to 3 times in the T1 treatment group compared with the control group. The pH, TBARS, L*, and b* values of Korean native chicken breast were not affected by Ptecticus tenebrifer powder supplementation (p>0.05); however, a* values showed statistical significance (p<0.05). In conclusion, the addition of 1% Ptecticus tenebrifer powder reduced mortality rate and demonstrated its potential in livestock environmental management.

Nucleotide Sequences of β-lactoglobulin Gene 5'Flanking Region in Korean Native Goat (한국재래산양 β-lactoglobulin 유전자 5'flanking 영역의 염기서열 분석)

  • This study was analyzed by PCR technique with specific primer in order to investigate the characteristics of ${\beta}$-lactoglobulin (${\beta}$-LG) gene 5'flanking region in Korean native goat. This work confirmed amplified product of 1,077 bp fragments obtained from the amplification of ${\beta}$-LG promoter from genomic DNA using PCR in Korean native goat. The nucleotide sequence of ${\beta}$-LG gene 5'flanking region in Korean native goat as compared with that of sheep ${\beta}$-LG were different at 46 base of 897 nucleotides, and showed high homology as about 94.9% each breed. Especially we confirmed that the difference of nucleotide sequences between Korean native goat and sheep were consisted of $T{\rightarrow}C$ substitution and $C{\rightarrow}T$ substitution reversely. As a consequences, the sequences of ${\beta}$-LG gene 5'flanking region showed a high homology between Korean native goat and sheep. Furthermore we should be studied that relationships between the control of gene expression and nucleotide sequences of transcription factor in Korean native goat.

Latex agglutination test based prevalence of Toxoplasma gondii in native Korean cattle

  • The prevalence of Toxoplasma (T.) gondii was surveyed using a latex agglutination test (LAT) in native Korean cattle. A blood sample was collected from female 105 cattle in the Daejeon area of Korea. All cattle were asymptomatic and had not received any prophylactic treatment for T. gondii. Blood samples were collected via the caudal vein. The cattle ranged in age from 2~6 years (mean 3.7 years). LAT detected antibody to T. gondii in four of 105 (3.8%) cattle. However, the hazard analysis and critical control point protocol has been applied to cattle farms and beef traceability has been strengthen.
