Comparison of Functional Compounds and Micronutrients of Chicken Breast Meat by Breeds

  • The concentrations of functional compounds and micronutrients of chicken breast from native chickens were compared with those from broiler. Totally 200 male chicks from a commercial native chicken (HH) and three newly bred native chicken strains (2A, 2C, and 2D) were reared for about 2 kg of final live weight up to 12 wk. After slaughtered, antioxidant dipeptides, reducing sugar, free amino acids, vitamins, and minerals of the breast muscles were analyzed with those from broilers with similar live weight. Mostly native chicken strains had higher contents of carnosine, anserine, and reducing sugar than the broiler. Especially HH implied the highest values of carnosine and anserine, and 2C did the highest of reducing sugar (p<0.05). Vitamin A contents between native chickens and broiler were not significantly different (p>0.05). The contents of ${\alpha}-tocopherol$ were significantly higher in 2C than those of HH or broiler (p<0.05). Native chicken strains contained lower cholesterol levels than the broiler. Broiler had higher contents of P, Mg, and Na than native chickens (p<0.05), but it had lower content of Cu than HH or 2A. The savory free amino acids including glutamic acid was highest in 2A than the other native chickens and broiler (p<0.05). This study confirms that certain new strains of native chickens be a good source in terms of functional compounds and micronutrients which can be attractive health promoting nutritional quality factors.

Optimization of Gelatin Extracting Condition from Korean Native Black Goat Skin and Quality Comparison with Commercial Gelatin

  • The practical use of Korean native black goat skin as a source of gelatin extraction is limited. The objective of this study was to optimize the extraction temperature and time of gelatin from Korean native black goat skin, and to compare the quality characteristics of goat skin gelatin and other commercial gelatin products. Response surface methodology was applied to optimize the extraction temperature and time of gelatin obtained from native Korean black goat skin. The effects of temperature (50℃-70℃) and time (2-4 h) on extraction yield and gel strength were investigated using a face-centered central composite design with 13 experiments. Gelatin extraction from Korean native black goat skin was prepared through the serial processes of alkali pre-treatment, bleaching, neutralization, hot-water extraction, and freeze-drying. Using the optimization plot of Minitab software, the optimized conditions for extracting temperature and time of goat skin gelatin were 59.49℃ and 3.03 h, and the optimized values of extraction yield and gel strength were 12.52% and 263.37 g, respectively. Based on a quality comparison of goat skin gelatin with commercial gelatin, the pH value of gelatin extracted from Korean native black goat skin was 5.57. The color of gelatin extracted from Korean native black goat skin was darker than that of commercial gelatin (p<0.05). Higher emulsifying properties and gel strength of goat skin gelatin were observed when compared to those of commercial gelatin (p<0.05). Therefore, the results of this study indicate that Korean native black goat skin may be a valuable source for gelatin extraction.

  • This study attempted to propose a space planning of rural villages having been connected by small-regions units as a plan of regional economic growth through cultivating native resources located in rural areas and then developing them. As research methods, this study carried out literature research and empirical study at the same time, and then was progressed to directions of deducing a basic plan by applying alternatives to actual object sites based on the results. Endogenous developments of areas through native resources' developments have been emphasized in Korea recently, and various policies and strategies are being appeared for making regional economic growth under these recognitions. However, rural village planning has directed to physical maintenances centering on functionality till now, and composed of facility repairs and experience projects of urban-rural interchanges mostly so as to solve living environments between cities and agricultural villages. Therefore, this study intended to pose discrimination with existing rural village planning, and set up a growth plan of rural villages by cultivating and developing tangible and intangible native resources having been kept in the rural villages and then commercializing them centering on indigenous native resources.

The Ethnobotanical Knowledge in Mt. Gayasan National Park

  • Ethnobotanical knowledge on native plants is critical on the conservation and management of biological resources. We investigated traditional knowledge of ethnobotanical uses in the Mt. Gayasan National Park area. Interviews were carried out to 189 residents at 176 places, and verified species and usage information was categorized by taxonomic groups, usage, and used parts. The ethnobotanical species of the regions consisted of a total of 275 taxa in 91 families including 105 cultivars. Sunflower family (Asteraceae) is the most widely used family with 30 taxa (7 cultivars). Rose family (Rosaceae, 25 taxa with 11 cultivars), Bean family (Fabaceae, 15 taxa with 11 cultivars), Grass family (Poaceae, 15 taxa with 10 cultivars), and Lily family (Liliaceae, 14 taxa with 4 cultivars) followed. About 50 taxa belong to the Approved Species for Delivering Overseas designated by Korea Ministry of Environment, and six Korean endemic taxa have been traditionally valuable in the regions. Many cultivated species have been utilized for various purposes accompanied local and native plants. The main usage of the plants are edible (175 taxa) and medicinal (168 taxa). Leaves are most commonly used parts (105 taxa), followed by stems (93 taxa), fruits (73 taxa), roots (55 taxa), and wholes (54 taxa). The study does not only provides examples of traditional uses of native plants, but also facilitates conservation of natural resources and sustainable developments of ethnobotanical knowledge for the contemporary human society.

Non-deep physiological dormancy in seeds of Euphorbia jolkinii Boiss. native to Korea

  • Background: Euphorbia jolkinii Boiss. is a perennial species native to Jeju Island and the southern coastal area of Korea. Particularly on Jeju Island, the yellow flowers of E. jolkinii Boiss. have a high ornamental value because of their contrast with basalt. This study was conducted to investigate the effects of different temperatures (5, 15, 20, and 25 ℃) and gibberellic acid (GA3) concentrations (0, 10, 100, or 1000 mg/L) on seed dormancy and germination of E. jolkinii. In addition, we classified the seed dormancy type and compared types with those of other species in the same genus. Results: The number of seeds with viable embryos and endosperms was approximately 66%. The final germination percentages at 5, 15, 20, and 25 ℃ were 51.7%, 83.5%, 2.6%, and 0.0%, respectively. In GA3 concentration experiments, the final germination percentages of 0, 10, 100, and 1000 mg/L were 83.5%, 91.7%, 79.1%, and 83.4%, respectively, at 15 ℃ conditions, and 0.0%, 6.9%, 13.2%, and 27.3%, respectively, at 25 ℃. Conclusions: Germination improved at temperatures of 15 ℃ or lower. Furthermore, GA3 treatment effectively reduced germination times. Thus, the seeds of E. jolkinni were classified as having non-deep physiological dormancy.

한우와 Holstein종의 κ -casein 유전자의 발현조절부위의 염기서열 분석 (Analysis of Sequence on promoter of κ -casein Genes in Korean Native Cattle and Holstein)

  • 본 연구는 한우와 Holstein종의 $\kappa$ -CN 유전자 발현조절부위, 즉 $\kappa$ -CN gene 5'-flanking region에 대한 특성을 구명하기 위하여 specific primer를 이용하여 PCR(polymerase chain reaction) 방법으로 분석하였다. PCR 증폭 결과, 349bp 크기의 증폭산물을 얻었으며 한우와 Holstein종의 $\kappa$ -CN 유전자의 발현조절부위의 염기서열 분석결과에 있어서는 -82bp에서 -431bp까지 분석되었으며, 종간의 염기서열 비교에서 -386bp에서는 Holstein종에서는 T 염기가 한우에서는 C 염기로 치환되었으며, -241bp와 -192bp에서는 Holstein종에서는 T 및 C 염가가 존재하였으나, 한우에서는 결실되었다. 또한, -183bp에서는 한우에서는 T 염기가 존재하였으나, Holstein종에서는 염기가 결실되어 있음을 알 수 있었다. 또한 한우와 Holstein 종간의 상동성 분석에 있어서는 한우 $\kappa$ -CN 유전자 발현조절부위의 염기서열이 Holstein종과 98.9% 의 상동성을 보였다.

한국 재래흑염소의 계통별 인공수정과 임신진단 키트의 활용 (Application of artificial insemination and pregnancy diagnosis kit for Korea native black goats)

  • 본 연구의 목적은 인공수정 기법을 이용한 한국 재래흑염소 계통별 인공수정 수태율을 알아보고자 정액의 성상, 수태율 및 분만율을 조사하고, 임신진단을 위한 소의 임신진단 키트의 활용 가능성을 알아보는 것이다. 재래흑염소에서 전기자극을 통해 정액을 채취하여 3계통(당진, 장수, 통영)별 정액분석과 발정동기화 된 암컷에 인공수정을 실시하여 계통별 번식능력을 조사하였다. 인공수정된 암컷에서는 소의 임신진단 키트(IDEXX Rapid Visual Pregnancy Test kit)와 초음파진단으로 임신을 확인하였다. 그 결과 개통별 정액채취량은 1~1.5 ml 내외였으며, 정액농도는 $18{\sim}25{\times}10^8/ml$, 정액 채취 직후 정자활력은 97% 이상 높게 나타났다. 그리고 임신진단의 결과에서 소의 임신진단 키트와 초음파진단 모두 동일한 결과를 보였으며, 계통별 인공수정 수태율의 효율은 20~44%로, 통영계통이 44%로 가장 높게 나타났고 장수계통이 20%로 인공수정 수태율이 낮게 나타났다. 본 연구 결과를 통해 한국 재래흑염소의 계통별 인공수정 효율을 알 수 있었고, 조기 임신진단을 위한 소 임신진단 키트의 활용가능성을 확인할 수 있었다. 본 연구 결과는 재래흑염소 증식과 개량에 기여할 수 있을 것으로 사료된다.

절개지 사면의 생태환경 복원을 위한 자생식물 조합 (Native Plants Combination for Ecological Environmental Restoration of the Dissected Sloping Area)

  • To suggest the native plants combination for restoration of the dissected sloping area, the plant communities of thirty seven quadrats from five localities of mid-southern part of Korean peninsula were surveyed from 2007 to 2008. Their flora, vegetation structure, and dominant species based on the importance value were investigated. And the soil characters (pH, moisture content, water holding capacity, and organic matter) of each dominant species were analyzed. Also, germination tests were performed to check the stability of restored native plants using seed chips. As a result, 79 native plants were suggested for the dissected sloping area : 20 trees and subtrees for the upper layer, 18 shrubs and vines for the middle layer, and 41 herbs for the lower layer, taken together their flora, dominant species, vegetation structure, soil condition, germination test, flowering period.

Physicochemical properties of M. longissimus dorsi of Korean native pigs

  • Background: The meat quality of Korean native pigs (KNP) and crossbred pigs (LYD; Landrace ${\times}$ Yorkshire ${\times}$ Duroc) was examined to generate data useful for selecting native pigs for improved pork production. Methods: Fifty Korean native pigs (KNP) and 50 crossbred pigs (LYD) were tested. Loin samples (M. longissimus dorsi) of the two breeds were analyzed to determine meat quality and sensory properties. Result: KNP had a higher moisture content than LYD (p < 0.05); however, it had significantly lower crude fat and ash content than that of LYD (p < 0.001). KNP had significantly higher shear force than LYD (p < 0.01). KNP also showed significantly higher cooking loss than LYD (p < 0.05). KNP had a lower $L^*$ value than LYD (p < 0.05); however, it had a markedly higher $a^*$ and $b^*$ value than LYD (p < 0.001). KNP showed significantly higher linoleic acid, linolenic acid, and arachidonic acid content than LYD (p < 0.05). Although KNP had significantly better flavor and overall palatability than LYD, it was less tender than LYD (p < 0.01). Conclusion: KNP had a markedly higher $a^*$ value than LYD. KNP had significantly higher shear force than LYD. The total unsaturated fatty acid content was higher in KNP than in LYD.
