The objective of this study was to investigate the effect of soaking time on physicochemical properties of waxy rice, waxy rice flour and waxy rice starch. Waxy rice (WR), waxy rice flour (WRF), waxy rice starch (WRS) were soaked at $18^{\circ}C$ for 14 days and dried at $40^{\circ}C$. Dried samples were grounded and sieved using 180 mesh. Physicochemical properties of the three samples, such as swelling power, solubility, moisture sorption isotherms and pasting properties using rapid visco analyzer (RVA) and crystal pattern using X-ray diffractometer were determined. In all samples, soaking time greatly influenced moisture sorption isotherms but no typical pattern was shown. Swelling power was not greatly changed by soaking time in the three samples. Solubility increased with increasing soaking time in all three samples, indicating that some soluble particles were exuded during soaking. In RVA pasting properties, WR and WRF showed a similar pattern, while WRS showed different pasting properties, suggesting that rice protein plays a significant role in pasting properties. X-ray diffraction patterns of all three samples showed typical A-type crystal pattern suggesting that soaking did not affect crystalline region of samples.

Although economically viable, the livestock industry is currently facing a number of challenging environmental problems and highly complex social issues, many of which are related to its size and geographically concentrated nature. Increased emphasis on environmental quality has also placed new demands on livestock producers to ensure that their production practices are in harmony with natural environment. In terms of sustainable agricultural systems, ruminants have served and will continue to serve a valuable role. They are particularly useful in converting vast renewable resources from rangeland, pasture, and crop residues or other by-product into food edible for human. With ruminants, land that is too poor or too erodable to cultivate becomes productive. Also, nutrients in by-products are utilized and do not become a waste-disposal problem. In Korea, however, native and dairy cattle production is not consistent with the advantageous roles of ruminant livestock in sustainable agricultural system because imported feed grains become the main basis for cattle raising. At present the ruminant livestock producers are heavily concentrated in and around the urban areas. About 75% of all the nation's cattle herds are kept on the outskirts of urban areas. As a result, the amount of pasture and forage land available per head of cattle is generally small. Furthermore, animals are raised in a cattle shed with high density. This situation is rather unfavorable for the national economic and environmental points of view As nation income increased, the demand for livestock products grew at an unforeseeable pace. But the pasture area involving in current utilization is tended to decrease during recent years. Based on the above figures more than 250,000 ha of pasture ought to be available for the present herd of cattle. It is obvious that these needs can scarcely be met with arable lands. Lands area for the establishment of new grassland have to be found in the hills and mountains which have not yet been used for crop framing or livestock. The development of extensive grasslands in the hill and woodland areas is now a declared aim. The starting point of the present work is the lack of knowledge of forage production and utilization in hill pasture and woodland in spite of indispensable necessity for livestock production in Korea. The importance of pastoral system in hill region and woodland is particularly emphasized in a standpoint of sustainable livestock production. Main chapter comprises the principle and techniques applicable for improving the utilization of hill pasture and woodland. We finally discussed the problems to solve and future works for a successful livestock production in hill and mountainous area in Korea.
Korean lawn grass, which belongs to Zoysia genus in Gramineae family, is one of the major turfgrass which is widely adapted to its native niches in the temperate region of the north east Asia through tropical regions including Philippine, Thailand and Australia. The Turfgrass Information Center' database provides 5,340 descriptive records including 638 referred papers and 1,370 technical reports concerning 'Zoysia'. The database focused on researches on golf courses and lawn care industries. The researches provide informations on seed technology and use of plant protectant for pest management in turf management industries. The purpose of this study is to analyze and classify the research contents of zoysiagrasses which have been published in Korean journals. The total number of research papers published in Korean journals were 274 including 102, 38, and 134 in the 'Botanical', 'Environmental' and 'Cultural' researches, respectively. Publication in foreign journals by Korean investigators were not counted in this research. Research fields concerning golf course maintenance and plant protectant are believed to be necessary when compared to international trends. Moreover, advanced research efforts for the development of new cultivars with various environmental and disease resistance should be accomplished.

We found an unrecorded species, Carex molestiformis Reznicek & Rothrock, in Byeokparyeong, Daehwa-myeon, Pyeongchang-gun, Gangwon-do, South Korea. This species is native to southern parts of the United States (ca. 11 states, including Arkansas, Missouri, and Oklahoma). These plants are usually distributed densely in stream flood plains, bottomlands, stream banks, and along roadsides. Belonging to section Ovales Kunth, perennial herbs with gynecandrous spikes, C. molestiformis is closely related to C. maackii Maxim. and C. scoparia Willd. and is distinguished from them by the number (2-4) of spikes in each inflorescence and the width (2.63.4 mm) and number (69) of veins of perigynia. A new Korean name of C. molestiformis 'Ga-Neun-Ta-Rae-Sa-Cho' was given, as it has slender overall appearance compared with C. maackii (Ta-Rae-Sa-Cho). We also provide a description, illustrations, photographs and a key of related taxa in Korea. We compared three DNA barcode region (chloroplast DNA matK, ndhF and nuclear ribosomal DNA internal transcribed spacer) sequences from C. molestiformis with those of C. maackii and C. scoparia, determining eight species-specific single nucleotide polymorphism sites for C. molestiformis.
At the beginning of the 21st century with taking a serious view of the harmony between human and nature, and of keeping an ecological balance, China has tried to reduce the gap in economy between rural districts and the urban city, and to carry out the policies for establishing a harmonious socialism through full-scale cooperation and continuous developments. And in order to preserve their traditional culture and to keep the historical legitimacy of People's Republic of China against urgent economical changes since the reform and openness, China has stressed the importance of training patriotic attitude on their cultural heritages. Thus the Chinese museums started to recognize more ad more the importance of social education as well as their functions of preserving the cultural property and of exhibiting it. And with them they have turned their attentions to an Ecomuseum, a new type of a museum. The ecomuseum in China was first established at a remote place between mountains where the ethnic minority inhabited. This was because of the feeling of some crises on culture native to the minority, followed by the process of China's Western Region Development. And it was recognized that they should participate actively in creating the cultural demand of the minority and in establishing a harmonious society with improving economic condition. Therefore in order to activate the ecomuseum it is localized and sinicized little by little through strengthening the management of cultural heritages of the minority.
The novel $\alpha$-amylase purified from locally isolated strain, Bacillus sp. KR-8104, (KRA) (Enzyme Microb Technol; 2005; 36: 666-671) is active in a wide range of pH. The enzyme maximum activity is at pH 4.0 and it retains 90% of activity at pH 3.5. The irreversible thermoinactivation patterns of KRA and the enzyme activity are not changed in the presence and absence of $Ca^{2+}$ and EDTA. Therefore, KRA acts as a $Ca^{2+}$-independent enzyme. Based on circular dichroism (CD) data from thermal unfolding of the enzyme recorded at 222 nm, addition of $Ca^{2+}$ and EDTA similar to its irreversible thermoinactivation, does not influence the thermal denaturation of the enzyme and its Tm. The amino acid sequence of KRA was obtained from the nucleotide sequencing of PCR products of encoding gene. The deduced amino acid sequence of the enzyme revealed a very high sequence homology to Bacillus amyloliquefaciens (BAA) (85% identity, 90% similarity) and Bacillus licheniformis $\alpha$-amylases (BLA) (81% identity, 88% similarity). To elucidate and understand these characteristics of the $\alpha$-amylase, a model of 3D structure of KRA was constructed using the crystal structure of the mutant of BLA as the platform and refined with a molecular dynamics (MD) simulation program. Interestingly enough, there is only one amino acid substitution for KRA in comparison with BLA and BAA in the region involved in the calcium-binding sites. On the other hand, there are many amino acid differences between BLA and KRA at the interface of A and B domains and around the metal triad and active site area. These alterations could have a role in stabilizing the native structure of the loop in the active site cleft and maintenance and stabilization of the putative metal triad-binding site. The amino acid differences at the active site cleft and around the catalytic residues might affect their pKa values and consequently shift its pH profile. In addition, the intrinsic fluorescence intensity of the enzyme at 350 nm does not show considerable change at pH 3.5-7.0.
This study was conducted to examine the utilization of immunohistochemistry using the bovine anti-brucella immunoglobulin G (IgG) antibody in the diagnosis of brucellosis and to develop a functional biomarker relation for the progress of the disease. Anti-brucella IgG antibody was purified from the affected bovine serum using an affinity chromatography. We performed our investigation on 17 cases of brucellosis and 19 control cases with negative Rose-Bengal test results. Our purified anti-brucella IgG antibody showed a positive immunoreactivity in cytoplasmic hepatocytes of the centrilobular region, and glomeruli and tubular epithelium of the kidney. The protein pattern of the affected liver versus control was analyzed by two-dimensional electrophoresis, showing a different expression pattern of proteins between the two. Five protein spots were up-regulated and another were five down-regulated in the brucellosis liver. Significant upregulaton of catalase and 3-hydroxyacyl-CoA dehydrogenase might be due to a compensatory reaction in response to the endotoxic shock of brucella. In conclusion, the anti-brucella IgG antibody may be a good tool for discriminative diagnosis of the affected tissues and proteomics data suggest new target proteins underlying a possible pathogenic mechanism of brucellosis.
Investigating endophyte distribution, naturally occurring in native grasses, is important for understanding endophyte-grass associations and using the beneficial effects of endophytes in cultivated plants. The goal of this study was to investigate endophytes from natural Festuca spp. in Yunnan, Guizhou, and Sichuan Provinces of China, and to study the compatibility between the endophytes and turf type tall fescue (Festuca arundinacea) which is widely used for lawn and athletic fields in that region. Of 628 accessions in F. ovina, 421 had endophytes identified in leaf sheath from on-site microscopic examination. From Festuca spp, three isolates were obtained from the seeds and ninety isolates were obtained from seedlings established from the collected seeds. The isolates from F. ovina and F. stapfii were tentatively identified as Neotyphodium typhinum and Neotyphodiumstarii, respectively. We tested compatibility of the two fungal species with seven tall fescue cultivars, Little Hero, Sub Boy, Eldorado, Arid III, Millennium, Crossfire, and Fawn. N. typhinum or N. starii did not infect 'Fawn' with either seed injection or seed soaking method. The highest infection rate byboth N. typhinum and N. starii was in 'Sun Boy' and 'Eldorado'. There were significant interaction effects between tall fescue cultivar and type of endophyte on infection.

This research has been launched in order to observe the osteological development of sevenband grouper, Epinephelus septemfasciatus larvae and juveniles and will be used for the basic data of phylogenetically systematics research. We got some samples of larvae and juveniles from a adult fish which belongs to the sea cage in Geomun-do, Yeosu-si, Jeolla-namdo province, and we have watched them. The average of breeding water temperature was $21.5{\sim}24.5^{\circ}C$ (average $23.0{\pm}1.5^{\circ}C$). In yolk-sac larvae just after hatching, ossification was not observed at all, and when the average total length reached 2.66 mm, the premaxillary and maxillary forming the upper jaw, dentary forming the lower jaw, and parasphenoid forming the skull base were ossified. In addition, the preopercle and opercle forming the opercular region began to be ossified. The caudal skeleton supporting the caudal fin is built with the caudal complex consisting of the malleus and incus. Until the average total length reached 2.59 mm, ossification did not occur at all and the urostyle bent at 45-degree angle. As to the ossification of the pterygiophore, when the average total length was 6.12 mm, the first interhemal spine began to be ossified in articulation with two spines of the anal fin, and the synapse also began to be ossified in articulation with two spines of the dorsal fin. When the average total length was 21.9 mm, individuals with completed ossification of the interhemal spines and synaptic clefts began to appear.

Establishments of the seedlings inside the natural forest from adjacent artificial forests would be an important factor in forest stand dynamics. This study was conducted to see the invasion of Korean pine (Pinus koraiensis) seedlings which is not native in this region, into the natural deciduous broad-leaved forest in Gwangneung, Korea. There is no mother tree at the I ha study site while the number of naturally regenerated P. koraiensis seedlings was 345 trees and 56% of them were clumped with more than two seedlings at each point. Applying the image segmentation method to IKONOS satellite image of January, 2003, the distance from the center of 1 ha study site to the nearest mother tree and plantation of Korean pine were 200 m and 270 m, respectively. The average height and root-collar diameter of the seedlings were 34 em and 7 mm, respectively and the age of 207 seedlings (60%) were below 5 years old. Most abundant range of soil moisture gradient and LAl (leaf area index) were from 16 to 20% and those of LAI were from 3.1 to 3.5. To understand the dynamics and seed dispersal pattern of Korean pine in the Gwangneung natural deciduous broad-leaved forests, additional studies not only long-term monitoring of growth and mortality of naturally regenerated Korean pine seedlings but also application of stable isotope analysis and molecular genetic techniques was recommended.
