Effect of Particular Breed on the Chemical Composition, Texture, Color, and Sensorial Characteristics of Dry-cured Ham

  • The present study demonstrates the impact of specific breed on the characteristics of dry-cured ham. Eighty thighs from Korean native pig (KNP), crossbreed (Landrace${\times}$YorkshireLandrace${\times}$Yorkshire)♀${\times}$Duroc♂ (LYD), Berkshire (Ber), and Duroc (Du) pig breeds (n = 10 for each breed) were used for processing of dry-cured ham. The thighs were salted with 6% NaCl (w/w) and 100 ppm $NaNO_2$, and total processing time was 413 days. The effects of breed on the physicochemical composition, texture, color and sensory characteristics were assessed on the biceps femoris muscle of the hams. The results revealed that the highest weight loss was found in the dry-cured ham of LYD breed and the lowest weight loss was found in Ber dry-cured ham. The KNP dry-cured ham contain higher intramuscular fat level than other breed hams (p<0.05). It was observed that the dry-cured ham made from KNP breed had the lowest water activity value and highest salt content, while the LYD dry-cure ham had higher total volatile basic nitrogen content than the Ber and Du hams (p<0.05). Zinc, iron and total monounsaturated fatty acids levels were higher in KNP ham while polyunsaturated fatty acids levels were higher in Du ham when compared to other breed hams (p<0.05). Additionally, the KNP dry-cured ham possessed higher Commission International de l'Eclairage (CIE) $a^*$ value, while the Du dry-cured ham had higher $L^*$, CIE $b^*$ and hue angle values (p<0.05). Furthermore, breed significantly affected the sensory attributes of dry-cured hams with higher scores for color, aroma and taste found in KNP dry-cured ham as compared to other breed hams (p<0.05). The overall outcome of the study is that the breed has a potential effect on the specific chemical composition, texture, color and sensorial properties of dry-cured hams. These data could be useful for meat processors to select the suitable breeds for economical manufacturing of high quality dry-cured hams.

한국재래닭 및 토착화 품종간의 유연 관계 및 유전 특성 분석 (Genetic Relationship between Populations and Analysis of Genetic Structure in the Korean Native Chicken and the Endemic Chicken Breeds)

  • 본 연구는 한국재래닭의 유전적 특성과 차별성을 검증하기 위하여 microsatellite(MS) marker를 이용한 타품종들과의 유전적 유연 관계를 분석하였다. 분석을 위해 7개 계통의 닭 317수(백색레그혼: 79, 로드아일랜드레드: 40, 코니쉬: 37, 적갈재래닭: 44, 황갈재래닭: 39, 흑색재래닭: 39, 오골계: 39)를 대상으로 7개의 MS marker들을 이용해 대립 유전자 및 유전자형을 분석하였다. 7개의 집단간의 유전적 유연 관계를 알아보기 위해 각 MS marker별 대립 유전자의 빈도를 산출하여 이를 근거로 집단간의 유전적 거리에 대한 추정 결과 KY와 KL간의 유전적 거리는 0.074로 가장 가까운 것으로 나타났으며, KR과 KY(0.101), KR과 KL(0.173) 역시 가까운 유전적 거리를 나타내고 있음을 확인하였다. 로드아일랜드의 경우, 한국재래닭 3계통과의 유전적 거리가 평균 0.233으로 타품종에 비해 비교적 가까운 것으로 나타났다. 레그혼은 다른 모든 품종들과의 거리가 가장 먼 것으로 확인되었다. 또한, 산란종인 백색레그혼과 육용종인 코니쉬 간의 유전적 거리는 가장 먼 것으로 확인되었다. 분석된 집단간의 유전적 구조에 따라 각 개체들이 어떻게 분포되어 있는가를 확인하기 위하여 각 개체들간의 유전적 거리를 분석하였다. 분석 결과, 백색레그혼의 경우 하나의 큰 그룹으로 분포하고 있음을 확인하였다. 또한, 로드아일랜드레드, 코니쉬 및 오골계 역시 각각 그룹을 형성하여 분포하고 있음을 확인하였다. 한국 재래닭 3계통은 각각 그룹을 이루지 못하였으며, 3계통이 합쳐져 넓게 분포하고 있음을 확인하였다. 재래닭의 경우 넓게 분포되어 있기는 하지만 다른 품종들과의 분포의 차이가 있음을 확인할 수 있었다.

Genomic diversity and admixture patterns among six Chinese indigenous cattle breeds in Yunnan

  • Objective: Yunnan is not only a frontier zone that connects China with South and Southeast Asia, but also represents an admixture zone between taurine (Bos taurus) and zebu (Bos indicus) cattle. The purpose of this study is to understand the level of genomic diversity and the extent of admixture in each Yunnan native cattle breed. Methods: All 120 individuals were genotyped using Illumina BovineHD BeadChip (777,962 single nucleotide polymorphisms [SNPs]). Quality control and genomic diversity indexes were calculated using PLINK software. The principal component analysis (PCA) was assessed using SMARTPCA program implemented in EIGENSOFT software. The ADMIXTURE software was used to reveal admixture patterns among breeds. Results: A total of 604,630 SNPs was obtained after quality control procedures. Among six breeds, the highest level of mean heterozygosity was found in Zhaotong cattle from Northeastern Yunnan, whereas the lowest level of heterozygosity was detected in Dehong humped cattle from Western Yunnan. The PCA based on a pruned dataset of 233,788 SNPs clearly separated Dehong humped cattle (supposed to be a pure zebu breed) from other five breeds. The admixture analysis further revealed two clusters (K = 2 with the lowest cross validation error), corresponding to taurine and zebu cattle lineages. All six breeds except for Dehong humped cattle showed different degrees of admixture between taurine and zebu cattle. As expected, Dehong humped cattle showed no signature of taurine cattle influence. Conclusion: Overall, considerable genomic diversity was found in six Yunnan native cattle breeds except for Dehong humped cattle from Western Yunnan. Dehong humped cattle is a pure zebu breed, while other five breeds had admixed origins with different extents of admixture between taurine and zebu cattle. Such admixture by crossbreeding between zebu and taurine cattle facilitated the spread of zebu cattle from tropical and subtropical regions to other highland regions in Yunnan.

Complete mitochondrial genome sequence of Tosa-Jidori sheds light on the origin and evolution of Japanese native chickens

  • Objective: In Japan, approximately 50 breeds of indigenous domestic chicken, called Japanese native chickens (JNCs), have been developed. JNCs gradually became established based on three major original groups, "Jidori", "Shoukoku", and "Shamo". Tosa-Jidori is a breed of Jidori, and archival records as well as its morphologically primitive characters suggest an ancient origin. Although Jidori is thought to have been introduced from East Asia, a previous study based on mitochondrial D-loop sequences demonstrated that Tosa-Jidori belongs to haplogroup D, which is abundant in Southeast Asia but rare in other regions, and a Southeast Asian origin for Tosa-Jidori was therefore suggested. The relatively small size of the D-loop region offers limited resolution in comparison with mitogenome phylogeny. This study was conducted to determine the phylogenetic position of the Tosa-Jidori breed based on complete mitochondrial D-loop and mitogenome sequences, and to clarify its evolutionary relationships, possible maternal origin and routes of introduction into Japan. Methods: Maximum likelihood and parsimony trees were based on 133 chickens and consisted of 86 mitogenome sequences as well as 47 D-loop sequences. Results: This is the first report of the complete mitogenome not only for the Tosa-Jidori breed, but also for a member of one of the three major original groups of JNCs. Our phylogenetic analysis based on D-loop and mitogenome sequences suggests that Tosa-Jidori individuals characterized in this study belong to the haplogroup D as well as the sub-haplogroup E1. Conclusion: The sub-haplogroup E1 is relatively common in East Asia, and so although the Southeast Asian origin hypothesis cannot be rejected, East Asia is another possible origin of Tosa-Jidori. This study highlights the complicated origin and breeding history of Tosa-Jidori and other JNC breeds.

  • Sixteen primary free amino acids were determined for each 10 longissimus muscles of Yorkshir, Landrace and Korean native black pig. Eight amino acids at 7 d classified three breeds at ca. 80% confidence. The difference in free amino acids for aged meats could be one of factors responsible for more desirable palatability of Korean native black pig.

The current status of Korean native pig production

  • Korean native pigs (KNPs) have been one of the traditional livestock primarily raised in rural areas of Korea for centuries. KNPs have adapted to the climate and geography of the Korean Peninsula for a long time, exhibiting excellent adaptability even in challenging environments. For these reasons, the preservation and purification of KNPs are crucial in securing unique genetic resources. Therefore, this review covers the characteristics, production status, commercial value and potential breeding directions of KNPs. Unfortunately, there is still a long way to go for the improvement of KNPs. It is crucial to acknowledge the current challenges, identify the issues, and dedicate efforts to the breed's improvement. Each section of this comprehensive review will play an important role in integrating related research and data into the overall findings. In-depth discussions on the genetic diversity, productivity, genetic conservation, ecological roles, and sustainability of KNPs will be crucial components in the future of KNP business.

한국 토종닭의 전장 유전체 복제수변이(CNV) 발굴 (Genome-wide Copy Number Variation in a Korean Native Chicken Breed)

  • 복제수변이(Copy number variation, CNV)는 DNA 다양한 구조적 변화의 한 형태이다. 복제수변이는 인간의 질병 및 농업의 생산성에 영향을 미치는 것으로 알려져 있다. 이전 우리나라의 닭의 품종은 유럽에서 유입되어진 품종을 기반으로 구축되어져 있었다. 따라서 농촌진흥청 국립축산과학원에서는 20년 동안 재래품종을 복원하려고 노력하였고, 5품종 12계통으로 복원하였다. 최근 염기서열분석 기술의 발달로, 해상도가 좋은 게놈 전체의 복제수변이를 발굴할 수 있게 되었다. 그러나 한국 재래닭 품종에 대해서는 체계적인 연구가 이루어지지 않고 있다. 본 연구에서는 한국 재래 닭(계통 L)에 대해서 게놈 전체의 염기서열을 분석하고 닭의 참고서열과 비교하여 재래닭에서 확인된 복제수 변이를 보고하였다. 닭의 28개 염색체에서 총 501개의 복제수 변이를 확인하였고, 이를 Gain과 Loss로 나누어서 표시하였다. 또한 우리는 501개의 복제수 변이를 포함하고 있는 유전자의 기능을 분류하였다. 그 결과, 전사 및 유전자 조절에 관련된 유전자들이 많이 분류되었다. 본 연구의 결과는 복제수 변이와 한국 재래닭의 경제형질 간의 연관성을 설명할 수 있는 기초자료로 활용될 것으로 사료된다.

품종 특이성을 이용한 제주마 판별 표지인자 재발 (Development of Sequence Characterized Amplified Regions (SCAR) Showing for Cheju Native Horse)

  • 본 연구는 RAPD 기법을 이용한 종 특이 marker 개발 및 이 marker의 SCAR marker로의 개발을 목표로 수행되었다. Random primer 700개에 대하여 PCR 수행결과, 품종간, 개체간에 많은 다형성이 관찰되었으며 품종특이적인 양상을 나타내는 MG30, MG53의 primer는 각각 2.0kb, 2.3kb의 위치에서 제주말과 더러브렛종의 특이적인 RAPD 단편을 나타내었다. 이들 단편들 중 품종 특이적인 단편을 클로닝한 후 random primer가 포함된 부분의 염기서 열을 결정하였다. 10 bp의 RAPD random primer에 10bp의 염기를 추가하여 SCAR primer를 제작하였다. SCAR marker의 수행결과 RAPD marker와 같은 2.3kb, 2.0kb의 크기에서 제주마와 더러브렛종에 특이적인 하나의 밴드가 증폭되었다. 따라서 이 Cnh-SCAR marker는 보다 안정적이고 재현성 있는 marker로서 사용이 가능하여 제주말의 판별에 유용하게 사용될 수 있을 것이다.

Physicochemical properties of M. longissimus dorsi of Korean native pigs

  • Background: The meat quality of Korean native pigs (KNP) and crossbred pigs (LYD; Landrace ${\times}$ Yorkshire ${\times}$ Duroc) was examined to generate data useful for selecting native pigs for improved pork production. Methods: Fifty Korean native pigs (KNP) and 50 crossbred pigs (LYD) were tested. Loin samples (M. longissimus dorsi) of the two breeds were analyzed to determine meat quality and sensory properties. Result: KNP had a higher moisture content than LYD (p < 0.05); however, it had significantly lower crude fat and ash content than that of LYD (p < 0.001). KNP had significantly higher shear force than LYD (p < 0.01). KNP also showed significantly higher cooking loss than LYD (p < 0.05). KNP had a lower $L^*$ value than LYD (p < 0.05); however, it had a markedly higher $a^*$ and $b^*$ value than LYD (p < 0.001). KNP showed significantly higher linoleic acid, linolenic acid, and arachidonic acid content than LYD (p < 0.05). Although KNP had significantly better flavor and overall palatability than LYD, it was less tender than LYD (p < 0.01). Conclusion: KNP had a markedly higher $a^*$ value than LYD. KNP had significantly higher shear force than LYD. The total unsaturated fatty acid content was higher in KNP than in LYD.

Characteristics of Seven Japanese Native Chicken Breeds Based on Egg White Protein Polymorphisms

  • In this study, to examine genetic variability within a breed and genetic relationships between populations/breeds, we genotyped 606 birds from seven Japanese native chicken breeds at seven polymorphic loci of egg white proteins and compared those with Asian native chicken populations and commercial breeds. Genotyping of the Japanese native breeds showed that ovalbumin, two ovoglobulins and ovotransferrin were polymorphic, but ovomacroglobulin, ovoflavoprotein and lysozyme were monomorphic. The proportion of polymorphic loci ($P_{poly}$) and average heterozygosity ($\bar{H}$) within a population ranged from 0.286 to 0.429 and from 0.085 to 0.158, respectively. The coefficient of gene differentiation ($G_{ST}$) was 0.250 in the Japanese native chicken breeds. This estimate was higher than that of Asian native chicken populations ($G_{ST}$ = 0.083) and of commercial breeds ($G_{ST}$ = 0.169). Dendrogram and PCA plot showed that Satsuma-dori, Jitokko, Amakusa-daio and Hinai-dori were closely related to each other and grouped into Asian native chickens and that Tsushima-jidori, Nagoya and Chan (Utaichan) were ramified far from other Japanese native chicken breeds. The egg white protein polymorphisms demonstrated that the population differentiation of the seven Japanese native chicken breeds was relatively large.
