The Current Status of BCG Vaccination in Young Children in South Korea

  • Background: Delivery of Bacille Calmette-Gur$\acute{e}$in (BCG) Tokyo vaccine, with the multipuncture device, has been much preferred over BCG Pasteur, with the intradermal method, possibly due to the easier manner of administration, a desire to avoid any trouble with scars, as well as side effects and higher profits to providers in South Korea. Methods: To determine BCG scar status in 0~6 year old children vaccinated with two BCG vaccines (Pasteur BCG vaccine with intradermal method and BCG Tokyo vaccine with percutaneous method), the data from the national BCG scar survey in 2006 was analyzed. Results: Based on the national survey, the high proportion that were vaccinated with BCG Tokyo vaccines with the multipuncture method (64.5%) was noted in 0~6 year old Korean children. From inspection of scar formation, as an indicator of vaccination, the median number of the visible pin scars from the percutaneous method was 16 (interquartile range, 12~18) in the Korean children, and pin scars decreased as the age of the children increased (p<0.001). Conclusion: The findings in this survey clearly showed a growing preference of parents for the BCG Tokyo vaccines by the multipuncture method in South Korea.

Continuous Automated Determination of Urea Using a New Enzyme Reactor (새로운 효소반응기를 이용한 요소의 연속·자동화 정량)

  • The response properties of continuous automated system using an enzyme reactor for determination of urea were investigated. The enzyme reactor was constructed to packed-bed form which filled with nylon-6 beads (42∼48 mesh), which immobilized urease with glutaraldehyde, in teflon tube (2 mm I.D., 20 cm length). The system was composed of the enzyme reactor, gas dialyzer, and tublar PVC-nonactin membrane ammonium ion-selective electrode as an indicator electrode in serial order. The response characteristics of this system were as follows. That is, the concentration range of linear response, slope of linear response, detection limit, and conversion percentage were $5.5{\times}10^{-6}$$2.4{\times}10^{-3}M$, 57.8 mV/decade, $1.5{\times}10^{-6}$, and 80.8%, respectively. The optimum buffer and life time of urease reactor were 0.01M Tris-HCl buffer solution (pH 7.0∼7.8) and 0.01M phosphate buffer solution (pH 6.9∼7.5) and about 150 days, respectively. And the urease reactor had no interferences of the other physiological materials.

Antibiosis and Bacteriocin Production of Lactic Acid Bacteria Isolated from Kimchi

  • In order to elucidate roles of lactic acid bacteria(LAB) for the antibiosis occurring in th fermenting environment of Kimchi, 2.052 strains of LAB were isolated from Kimchi. Fifty tow strains which showed antagonistic effect against 4 indicator strains were finally selected and investigated. Based upon responses to protease treatment, antibiosis of the 52 strains of LAB were classified into 3 types. Type A antibiosis resulted from action of antibiotic-like substances which were not affected by protease treatment and which had broad action spectra against even natural inhabitants of Kimchi. Type B antibiosis was due to bacteriocin-like substances which were very sensitive to treatment of protease and more effective against foreign bacteria than original inhabitant microflora. Type C antibiosis was owing to proteinaceous compounds which were activated or induced by the presence of protease and then exerted antibacterial activities. Therefore, lactic acid bacteria appeared to contribute to antibiosis of Kimchi by the concerted action of these three different types of antibacterial compounds. As one of model system for type B bacteriocin, the antagonistic compound produced by LAB31-9 as well as th producer strain itself was further charaacterized. Strain LAB31-9 was identified as L. casei. Bacteriocin produced by LAB31-9 was proteinaceous and stable over wide range of pH and to various solvents, but very labile to heat treatment. Its mode of action was bactericidal. Based upon these data, bacteriocin produced by LAB31-9 was named as 'caseicin K319'. Genetic determinant for the bacteriocin production of LAB31-9 was located in the chromosome.

Cultural Conditions of Lactobacillus sp. GM7311 for the Production of Bacteriocin (Lactobacillus sp. GM7311에 의한 박테리오신의 생산 조건)

  • A lactic acid bacteria which showed antimicrobial activity was isolated from dairy products and identified as Lactobacillus sp. according to the morphological, physiological and biochemical properties, which was named Lactobacillus sp. SH 7311. The bacteriocin of Lactobacillus sp. GM 7311 showed a broad range of inhibitory spectrum against some gram positive and negative bacteria. Especially, Proteus mirabilis was highly sensitive to bacteriocin and used as indicator strain for further investigation. The optimal condition for the production of bacteriocin was showed on MRS broth at $37^{\circ}C$ and pM 6.0. Bacteriocin production of this strain cultured under optimal condition was increased late logarithmic phase to early stationary phase. This bacteriocin was fully active at the pH range $2.0\~5.0$, also was stable at $100^{\circ}C$ for 60 min. at pH 5.0, But about $40\%$ of bacteriocin activity was diminished by the treatment of acetone, ethanol, iso-butanol and ethyl ether during 2 hours at $4^{\circ}C$.

Effect of Wildlife Access on Microbial Safety of Irrigation Water Used in the Cultivation of Chinese Cabbage in Goesan (야생동물 출입이 괴산 지역 배추 재배 농업용수의 미생물 안전성에 미치는 영향)

  • Water is an important component in the production of fresh produce. It is mainly used for irrigation and application of pesticides and fertilizers. Several outbreaks cases related to fresh produce have been reported and water has been identified as the most likely source. On the other hand, wildlife has been identified as a possible source of the waterborne pathogens. The purpose of this study was to investigate the effect of wildlife access on irrigation water used in the cultivation of Chinese cabbage in Goesan. The frequency of wild animals access to upstream water source and the contamination level of bacteria such as Escherichia coli and Enterococci of irrigation water used in Chinese cabbage farm was examined. A total of 37 wildlife including the wild bear and water deer were observed in upstream of water source during the cultivation of Chinese cabbage. The result indicated the presence of hygienic indicator bacteria from the upstream where there is no human access. The contamination range of coliforms, E. coli, and Enterococcus spp. Detected in the irrigation water were 2.13~4.32 log MPN / 100 mL, 0.26~2.03 log MPN / 100 mL, and 1.43~3.49 log MPN / 100 mL, respectively. Due to low water temperatures, the contamination levels of coliform bacteria and E. coli in the irrigation water during harvesting time was lower compared to those recorded during transplanting of Chinese cabbage. However, no significant difference was detected in the number of Enterococci during the cultivation of Chinese cabbage. The results indicated the need to manage the microbial risk in irrigation water to enhance safety in cultivation of Chinese cabbage.

Evaluation of the Bacteriological and Toxicological Safety for the Shellfish Growing Area in the Kamakman Area, Korea (가막만해역 패류의 세균학적·독물학적 안전성 평가)

  • To evaluate bacteriological and toxicological safety, the hygienic indicator bacterium and paralytic and diarrhetic shellfish toxins in the shellfish produced in the Kamakman Area from 2012 to 2016 were investigated. Fecal coliforms and E. coli of all 194 oyster samples tested did not exceed 230 MPN/100 g. The geometric mean of the fecal coliform analyzed with the oyster samples of harvesting period was 19.6 MPN/100 g, which was more stable than the non-harvesting period (26.5 MPN/100 g). For the toxicological evaluation of the Kamakman Area, 77 oyster samples and 350 mussel samples as an indicator were analyzed. Paralytic shellfish toxins were detected very low in the range of $40{\sim}46{\mu}g$/100 g in 13 mussel samples during late April and early June, but not in oyster samples. Diarrhetic shellfish toxin was detected in 2 of 180 samples, but it was found to be below the regulation value (0.16 mg OA equ./kg). Based on the bacteriological studies, it was confirmed that the shellfish produced in Kamakman area meets the standard of shellfish hygiene of the Food Sanitation Act and meets the Grade A of the shellfish production area of EU. As the results of the paralytic and diarrhetic shellfish toxin evaluation, it was confirmed that the Kamakman Area is also toxicologically safe for shellfish production.

First Report on Tomato bushy stunt virus Infecting Tomato in Korea

  • A new virus-like disease of tomato showing chlorotic spots, malformation and necrosis on leaves, and chlorotic blotching, rings, and necrosis on fruits was observed around Sacheon, Gyeongsangnam-do, Korea in 2004. Host range analysis could not differentiate 4 field isolates collected from tomatoes showing different symptoms but identified them as Tomato bushy stunt virus (TBSV). TBSV-tsf2 isolate induced symptoms in indicator plants similar to those caused by the TBSV-C, -S and -Nf. As the isolate could not systemically infect Chenopodium quinoa, the isolate might belong to the previously described TBSV-S isolate. TBSV-tsf2 isolate caused similar cytological alterations that were similar to that generally caused by previously reported TBSV isolates. TBSV-tsf2 isolate, however, could be considered to belong to new strain of TBSV because masses of small electron-dense patches that were not observed from the previously described TBSV. The complete nucleotide sequences of the genomic RNA of 4739 nt excluding non-translated sequences at both termini have been determined and compared to sequences of other TBSV strains. The complete nucleotide sequence identity among TBSV isolates was 98.9% to 99.7%, and to the other tombusviruses ranged from 80.8% to 94.9%. Comparison of the amino acid sequences all five ORFs with those of other TBSV strains shows a similar genomic organization, and high percentage of amino acid sequence homology with TBSV-Nf than TBSV-S isolate. Since the TBSV symptoms were only observed in Sacheon fields where imported seeds from Japan were planted, the TBSV incidence probably caused by the planting contaminated tomato seeds and thus require more through quarantine procedure to prevent settlement of TBSV in Korea. Altogether, these results support that the Korean isolate of TBSV infecting tomato might be new strain.

Factors Affecting Chemical Disinfection of Drinking Water

  • This research sought to compare chlorine, chlorine dioxide and ozone as chemical disinfectants of drinking water, with inactivation of total coliform as the indicator. The inactivation of total coliform was tested against several variables, including the dose of disinfectant, contact time, pH, temperature and DOC. A series of batch processes were performed on water samples taken from the outlet of a settling basin in a conventional surface water treatment system that is provided with the raw water drawn from the mid-stream of the Han River. Injection of 1 mg/L of chlorine, chlorine dioxide and ozone resulted in nearly 2.4, 3.0 and 3.9 log inactivation, respectively, of total coliform at 5 min. To achieve 99.9 % the inactivation, the disinfectants were required in concentrations of 1.70, 1.00 and 0.60 mg/L for chlorine, chlorine dioxide and ozone, respectively. Bactericidal effects generally decreased as pH increased in the range of pH 6 to 9. The influence of pH change on the killing effect of chlorine dioxide was not strong, but that on ozone and free chlorine was sensitive. The activation energies of chlorine, chlorine dioxide and ozone were 36,053, 29,822 and 24,906 J/mol for coliforms with inactivation effects being shown in the lowest orders of these.

Effect of processed foods on serum levels of eosinophil cationic protein among children with atopic dermatitis

  • The prevalence of atopic dermatitis (AD) in school-age children has increased in industrialized countries. As diet is one of the main factors provoking AD, some studies have suggested that food additives in processed foods could function as pseudoallergens, which comprise the non-immunoglobulin E-mediated reaction. Eosinophil cationic protein (ECP) is an eosinophil granule protein released during allergic reactions to food allergens in patients with AD. Thus, serum ECP levels may be a useful indicator of ongoing inflammatory processes in patients with AD. The purpose of this study was to investigate the effect of consuming MSG in processed foods on serum ECP levels among children with AD. This study was performed with 13 patients with AD (age, 7-11 years) who had a normal range of total IgE levels (< 300 IU/ml). All participants ate normal diets during the first week. Then, six patients were allocated to a processed food-restricted group (PRDG) and seven patients were in a general diet group (GDG). During the second week, children in the PRDG and their parents were asked to avoid eating all processed foods. On the third week, children in the PRDG were allowed all foods, as were the children in the GDG throughout the 3-week period. The subjects were asked to complete a dietary record during the trial period. Children with AD who received the dietary restriction showed decreased consumption of MSG and decreased serum ECP levels and an improved SCORing score on the atopic dermatitis index (P < 0.05). No differences in serum ECP levels or MSG consumption were observed in the GDG. Serum total IgE levels were not changed in either group. In conclusion, a reduction in MSG intake by restricting processed food consumption may lead to a decrease in serum ECP levels in children with AD and improve AD symptoms.

Design Optimization of M8 Blind Rivet Nut Geometry using Finite Element Analysis (유한요소해석을 이용한 M8 블라인드 리벳 너트 형상 최적 설계)

  • Blind rivet nuts are increasingly used in automotive for the joining of sheets. Their application, however, requires appropriate design guides to prevent catastrophic events arising from the failure of joints. In this study, the shaft shape of a frequently used M8 blind rivet nut is optimized based on 3D numerical analysis of the blind rivet nut considering the characteristics of thread. The thread needs to be modeled to suitably consider the fastening of the M8 bolt after the crimping process. FE analysis showed that while the friction in the contact between crimp flange and plate has no significant effect on the crimp geometry, shaft thickness (t) and shaft height (h) are the most significant design variables. The parameter study including various combinations of t and h reveals that they affect the gap (the distance between the crimped flange and the plate that develops through riveting) and the load acting on the plate. The gap is an indicator of the tightening force. It is found that t is inversely proportional to the gap, and proportional to the load, whereas h is proportional to the gap and inversely proportional to the load. Based on our FE analysis results, we propose the range 0.062 < t/h < 0.1 to ensure sufficient fastening (high clamping load, small gap) of the M8 blind rivet nut. The design guide for determining the t/h ratio proposed in this study can be used for general quantitative analysis of the size and the t/h ratio of blind rivet nuts.
