  • Gene-editing is currently one of the most popular technologies in recent years. Development of the new crop using the gene editing have advantage of improved accuracy and efficiency compared with conventional breeding. Soybean (Glycine max L.) is one of the most important crops worldwide used as food and forage. We tried to establish a system for breeding improvement of soybean through gene-editing technology. For the gene-editing system of soybean, i) selection of efficiency gRNA of targeted gene, ii) efficient genetic transformation of the selected gRNA, iii) selection of trans-clean mutant is essential. First of all, we investigated the selection conditions of gRNA with high editing efficiency of targeted gene using isolated protoplast of soybean. Furthermore, we performed the Agrobacterium-mediated genetic transformation of various soybean cultivars. We identified the tissue culture ability in 23 soybean cultivars for genetic transformation of soybean. The six cultivars with high tissue culture ability were selected and confirmed the transgenic plants in four cultivars. Finally, we established a speed-breeding system as a powerful tool for the fast selection of trans-clean mutants from transgenic plants. Our laboratory will provide the valuable system for improvement of soybean by the gene-editing technology.

  • Environmental changes, especially global climate change, are creating new challenges to the development of the Arctic regions, which have substantial energy resources. And attention to offshore structures has increased with oil and gas development. The structural impact response of an explosion-resistant profiled blast walls normally changes when it operates in low temperatures. The main objectives of this study are to investigate the structural response of blast walls in low temperature and suggest useful guidelines for understanding the characteristics of the structural impact response of blast walls subjected to hydrocarbon explosions in Arctic conditions. The target temperatures were based on the average summer temperature ($-20^{\circ}C$), the average winter temperature ($-40^{\circ}C$) and the coldest temperature recorded (approximately $-68^{\circ}C$) in the Arctic. The nonlinear finite element analysis was performed to design an explosion-resistant profiled blast wall for use in Arctic conditions based on the behaviour of material properties at low temperatures established by performing a tensile test. The conclusions and implications of the findings are discussed.

  • Potato (Solanum tubersosum L.) is susceptible to various environmental stresses such as salt, high temperature, and drought. Especially, potato tuber growth is greatly affected by drought that causes not only yield reduction but also loss of tuber quality. Since unpredictable global weather changes cause more severe and frequent water limiting conditions, improvement of potato drought tolerance can minimize such adverse effects under drought and can impact on sustainable potato production. Genetic engineering can be utilized to improve potato drought tolerance, but such approaches using endogenous potato genes have rarely been applied. We were obtained AtbZIP28 gene transgenic potato plants. It is identified transcript levels at various stress conditions, polyethylene glycol (PEG), NaCl, (ABA). Also, For identification to regulate ER stress response genes in AtbZIP28 gene transgenic potato plant, we screened seven potato genes from RNA-seq analysis under TM treatment. Five and two genes were up- and down-regulated by TM, respectively. Their expression patterns were re-examined at stress agents known to elicit TM, DTT, DMSO and salt stress.

  • In offshore structures, fire is one of the most important hazardous events. The concern of fires has recently been reflected in the rules and quantified risk assessment based design practice. Within the framework of quantified risk assessment and the management of offshore installations, therefore, more refined computations of the consequences or hazardous action effects due to fire are required. To mitigate fire risk, passive fire protection(PFP) is widely used on offshore structures. This study presents methods for a nonlinear structural response analysis considering the PFP effects under fires. It is found that a structural response analysis is most likely to use valuable technology for the optimization and design of offshore structures with PFP. Thermal and structural response analyses have been performed using LS-DYNA and FAHTS/USFOS. The results of these structural response analyses are compared with each other.
