Transformation of Terpene Synthase from Polyporus brumalis in Pichia pastoris for Recombinant Enzyme Production

  • Terpenoids have a wide range of biological functions and have extensive applications in the pharmaceutical, cosmetic, and flavoring industry. The white-rot fungus, Polyporus brumalis, is able to synthesize terpenoids via terpene synthase, which catalyzes an important step that forms a large variety of sesquiterpene products from farnesyl pyrophosphate (FPP). To improve the production of sesquiterpenes, the terpene synthase gene was isolated from Polyporus brumalis and was heterologously transformed into a Pichia pastoris strain. The open reading frame of the isolated gene (approximately 1.2 kb) was inserted into Pichia pastoris to obtain a recombinant enzyme. Five transformants were obtained and the expression of terpene synthase was analyzed at the transcript level by reverse transcription PCR (polymerase chain reaction) and at the protein level by SDS-PAGE (sodium dodecyl sulfate polyacrylamide gel electrophoresis). Expression of the terpene synthase gene product was elevated in the transformants and as expected the molecular weight of the protein was approximately 45 kDa. These recombinant enzymes have potential practical applications and future studies should focus on their functional characterization.

Effect of Sodium Chloride (NaCl) on the Mycelial Growth and Ascospore Germination of Rhizina undulata, the Root Rot Fungus of Coniferous Trees

  • Rhizina undulata, the causal agent of Rhizina root rot, is a soil-borne fungus occurring on coniferous trees. The destruction of coastal forests caused by R. undulata infection has been mainly associated with bonfires at camping sites. However, Rhizina root rot was observed in the western coastal forests without fire. It was hypothesized that Rhizina root rot in this area might be closely related to the soil salinity, which can facilitate the growth and survival of R. undulata. So, the variation in sodium chloride (NaCl) resistance among isolates of Rhizina undulata was compared using liquid media containing different concentrations of NaCl ranging from 0 mM to 300 mM. Our results showed that, albeit of no growth at a higher concentration of NaCl (300 mM), most of R. undulata isolates were capable of germinating and grew at up to 100 mM, indicating that NaCl resistance varies among R. undulata isolates. It was further found that isolates from coastal areas seemed to be more tolerant to NaCl than those further away the coast. We demonstrated that R. undulata could be possible to survive in coastal areas, but was lower NaCl tolerance than other fungi.

미이용 산림바이오매스 공급에 있어 수확벌채의 원목 혼입량 추정 (Estimation of the Amount of Round Wood in Unused Forest Biomass Reporting in Forest Clearing)

  • To respond to global warming, there is an increasing interest in eco-friendly alternative energy sources. Therefore, unused forest biomass that has been neglected due to a lack of marketability is attracting attention. With the introduction of the "unused forest biomass certification system" in 2019, ways of determining quantity of unused forest biomass have steadily increased. However, there have been reported cases whereby unused forest biomass weighed more than the amount of harvested trees. It was found that it was possible that forest resources that can be used as round wood were mixed with unused forest biomass. In this context, this study aimed to estimate the amount of mixed round wood in the unused forest biomass supply. The relative expression of growing stock/ha versus the amount of final clearing/ha collected was modeled (y=1.490x-94.341, R2=0.861). As a result, it was found that round wood was mixed into the unused forest biomass, contributing to the disparity observed between the weighted forest biomass and the amount of trees harvested. In conclusion, proper declaration and certification procedures should be carried out for the use of forest resources and promoting unused forest biomass usage.

산림재해 방지와 산림관리를 위한 산악기상정보 (Mountain Meteorology Data for Forest Disaster Prevention and Forest Management)

  • 우리나라의 산림은 지형적으로 복잡한 특성을 가지고 있기 때문에 효율적인 산림관리를 위해서는 산림지역에 특화된 기상관측이 중요하다. 특히, 최근 기후변화에 따른 건조와 집중호우 등 이상기상 현상으로 산림재해가 발생할 수 있기 때문에, 이를 효과적으로 예방하고 관리하기 위한 대책 마련이 필요하다. 이를 위해 산림청에서는 산악지역에 대한 기상 자료를 수집하기 위해 2012년부터 산악기상관측망 구축을 시작했고, 현재 464개소의 산악기상관측망을 운영하고 있다. 산악기상관측망에서는 기온, 상대습도, 풍향과 풍속, 강수량, 지면온도, 대기압 등 7개 기상요소를 관측한다. 기상 자료는 1분 간격의 실시간 자료를 수집하며, 자료의 신뢰도 확보를 위한 품질관리 시스템을 운영하고 있다. 산악기상관측 자료의 품질관리는 물리한계검사, 단계검사, 내적 일치성검사, 지속성검사, 기후범위검사, 중앙값 필터검사 등 6가지 품질검사를 수행한다. 고품질 산악기상정보는 공동활용을 위해 산악기상정보시스템과 공공데이터 포털을 통해 자료를 공개하고 있다. 산악지역에 특화된 산악기상정보는 산림관리 및 산림재해 방지뿐만 아니라 국민 생활안전과 산림휴양⋅레저 등 다양한 분야에 활용될 수 있을 것으로 기대된다.

  • 본 연구에서는 우리나라 대나무 자원의 평가를 위하여 GIS 기반의 인벤토리를 구축하고 활용 방안을 제안하였다. 대나무 자원의 일반현황, 임황 및 지황 정보, 입지토양 정보 등의 인자를 추출하고 현지표본점 조사를 통해 획득한 자료를 활용하여 면적 및 축적량을 산출하여 통합하였다. 구축된 인벤토리는 방치되어 있는 대나무 자원의 건강성 및 지속성 확보는 물론 대나무의 기능 증진을 위한 장기적인 목표와 방향을 설정할 수 있는 의사결정 도구로 활용할 수 있을 것으로 판단된다.

Forest Certification Scheme; Perceptions and Willingness-to-pay of Consumers and Manufacturers in South Korea

  • This study was carried out to examine the perceptions of consumers and forest product manufacturers in South Korea about forest certification and to identify their willingness-to-pay for certified products by personal interviews. Sixteen percent of interviewees knew and heard about forest certification. However, fifty-six percent of interviewees had an intention to participate in forest certification system. The consumers' group can be described in relative terms as the male of 30 age bracket. The average price premium charged to consumers was higher than one paid to manufacturers, and thus manufacturers are willing to include the additional costs for certified products.

Biodegradability and Risk Assessment of Biomass-based Polymeric Materials

  • With the intention to solve environmental problems caused by synthetic plastics from petroleum resources, biodegradable polyurethane foams and thermosetting moldings were prepared from biomass, such as wood and wheat bran by liquefaction method. Biodegradability of these biomass-based polymeric materials was investigated. In activated sludge, polyurethane foams from liquefied wheat bran and thermosetting molding from phenolated wood were decomposed approximately 14% and 29% for 20 days, respectively. One of the wood fungi, Coriolus versicolor was able to grow without supplemental nutrition, only with distilled water and polyurethane foam as a nutrition source. Risk assessments were also conducted and results showed that estrogenicity, mutagenicity, and carcinogenicity were not observed in the extractives of biomass- based polymeric materials.

일본잎갈나무림과 침활혼효림의 입지환경이 산양삼 종묘의 초기 생육에 미치는 영향 (Effect of Location Environments on Early Growth of Wild-simulated Ginseng (Panax ginseng C. A. Meyer) Seedlings in Larix kaempferi and Mixed Forest)

  • 본 연구는 일본잎갈나무림과 침활혼효림의 입지환경이 산양삼 종묘의 초기 생육에 미치는 영향을 구명하기 위해 수행하였다. 일본잎갈나무림과 침활혼효림에 시험포지를 조성하고, 입지환경과 산양삼 종묘의 생육특성을 조사하였다. 입지환경 중에서 토양 유기물, 전질소, 양이온치환용량과 같은 토양특성은 일본잎갈나무림에 비해 침활혼효림에서 높게 나타났고, 산양삼 종묘의 생육특성 또한 일본잎갈나무림에 비해 침활혼효림에서 유의적으로 높게 나타났다. 시험포지 내 수종 비율, 토양특성, 산양삼 종묘 생육특성 간의 상관관계를 분석한 결과, 토양특성과 생육특성은 활엽수림의 비율과 유의적인 정의 상관관계를 보였고, 토양특성 중에서 유기물, 전질소, 양이온치환용량이 산양삼의 생육과 유의적인 정의 상관관계를 보였다. 본 연구에서는 일본잎갈나무림과 침활혼효림의 입지환경 중에서 임상과 토양특성이 산양삼 종묘 생육특성과 유의미한 상관관계를 보이는 것을 구명할 수 있었고, 본 연구의 결과를 바탕으로 향후 산양삼의 최적 재배지를 선정하는데 기초적인 자료로서 도움을 줄 수 있을 것으로 사료된다.

진주지역 우산고로쇠나무 인공림의 수액 출수 특성 (Characteristics of Sap Exudation from Acer okamotoanum (Nakai) Plantation Forest in Jinju Region)

  • 본 연구는 우산고로쇠나무 조림지에서 수액 출수 특성을 구명하고자 수행하였다. 그 결과, 우산고로쇠나무 수액 출수량은 일 평균기온 $3.4^{\circ}C$ 이상 오를 시 출수량이 급격히 감소하였으며, 이를 고려해볼 때, 2월 하순 이전에 수액채취를 하는 것이 효율적일 것으로 판단되었다. 하루 중 수액출수는 약 9시경부터 출수를 시작하여 11시경에 최대량이 출수되었고 16시경에 출수가 종료되었다. 수액 당도는 시간이 흐름에 따라 감소하는 경향이 나타났으며, Na를 제외한 다른 무기성분들은 시기별 차이는 없었다.

Ergothioneine Contents of Shiitake (Lentinula edodes) Fruiting Bodies on Sawdust Media with Different Nitrogen Sources

  • Ergothioneine is a natural compound with strong antioxidant properties. In this study, the effects of different nitrogen sources including ammonium nitrate, ammonium sulfate, sodium nitrate, and histidine in sawdust media were investigated to enhance ergothioneine contents in Shiitake (Lentinula edodes) fruit bodies. The addition of 0.2% ammonium sulfate in the growth media showed the highest enhancement of ergothioneine content in shiitake fruit bodies which was 1.7-fold higher than the control. On the other hand, histidine, a building block of ergothioneine decreased the concentration of ergothioneine significantly. Our results demonstrate that the cultivation of shiitake in sawdust media with suitable nutrients was effective to enhance its ergothioneine contents.
