Effects of Some Chemials on Ethylene Evolution and Abscission of Fruits and Leaves in Oriental Pera (수종(數種)의 약제(藥劑)가 배나무의 낙과(落果) 및 에틸렌 발생(發生)에 미치는 영향(影響))

  • In order to examine the relationship of fruit abscission and ethylene evolution in 'Jojuro' and 'Imamuraaki' pear trees, the thinning chemicals, ${\beta}$-naphthalene acetic acid (NAA), O, O-dimethyl-O-(3-methyl-4 nitrophenyl) posphorothioate (MEP), 1-naphthyl N-methyl carbamate (carbaryl), and 2-chloroethyl phosphonic acid (ethephon) were applied. Concurrently the effect of calcium acetate on the control of fruit abscission were examined when calcium acetate was added to ethephon and carbaryl. 1. All the applied chemicals (MEP, NAA, carbaryl and ethephon) was effective to abscise fruits in 'Jojuro', and MEP and carbaryl in 'Imamuraaki's. 2. The application of ethephon increased the ethylene evolution but other chemicals did not increase it. 3. There were no significant differences in total sugar contents of fruits by MEP, NAA, carbaryl and ethephon treatments. 4. When the calcium acetate was added to the ethephon and carbaryl, the thinning effects were offsetted. 5. The rate of defoliation due to ethephon treatment was controlled by addition of calcium acetate at 0.1~0.25 M. It was clarified that fruit abscission induced by thinning chemicals except ethephon was not directly related to ethylene production and that degree of fruit thinning can be regulated by addition of calcium acetate to thinning chemicals.

In Vitro Propagation of Wild Cynanchum wilfordii Through Axillary Bud Culture (액아배양을 통한 야생 백하수오(Cynanchum wilfordii)의 기내증식)

  • This study was conducted to establish the optimal condition for in vitro propagation of wild Cynanchum wilfordii. The highest in vitro seed germination rate of 91.6% was obtained from the seed treated with gibberellic acid ($GA_{3}$) (100 ppm) for 24 hours followed by cultured on Woody Plant Medium (WPM) media supplemented with 1.0 mg/L 6-benzyl adenine (BA). The best shoot height obtained (5.2 cm) in medium with 1.0 mg/L $GA_{3}$, but the plants was grown abnormally and eventually died. The highest number (2.4) was shown when shoot were in cultured on the media including 0.1 mg/L BA after 4 weeks. Root induction from shoot obtained in vitro culture was effective on ventilation and without plant growth regulators (PGRs) and root length was highly developed (2 cm) at 0.1 mg/L naphthalene acetic acid (NAA). The highest survival rate (70%) was when plantlet grew pre-ventilation of in vitro condition.

Development of Odor-Reduced Korean Traditional Cheonggukjang Added with Job's Tears (냄새저감형 율무청국장 제조에 관한 연구)

  • This study was performed to develop an odor-reduced Korean traditional Cheonggukjang (fermented soybean paste) by adding Job's tears to Cheonggukjang made with soybean (control) only. The study included the determination of best addition percentage of Job's tears versus soybean, sensory evaluation, and aroma patterns and its components as well as proximate composition analyses of Cheonggukjang. Job's tears Cheonggukjang (CAJT) was prepared by inoculating Bacillus subtilis and fermented at $40^{\circ}C$ for 48 hours. Ratios of soybean : Job's tears for Job's tears Cheonggukjang were 1:1, 2:1, 3:1, and 4:1, respectively. In comparison of proximal composition, Job's tears Cheonggukjang was high in moisture and carbohydrates, but low in calorie, fat, protein, ash and amino nitrogen. The pH of CAJT was lower than those of control and conventional Cheonggukjang (CC) sold in the market, and the acidity was the lowest in CC. The color of Job's tears Cheonggukjang appeared the whiter and more yellowish, but less reddish in proportion to contents of Job's tears. The contents of viscous substances were higher in CAJT compared with those of control and CC. The contents of glutamic acid, the major components of viscous substances were lower in CAJT than in control and CC, and the contents of fructose higher in CAJT. The scores of sensory evaluation were the highest in Job's tears Cheonggukjang mixed with soybean : Job's tears=4:1. The aroma pattern of CC analyzed was conspicuously dissimilar to the control and CAJT and also discriminated by electronic nose examination. The pyrazines, volatile compounds peculiar to Cheonggukjang, were found to be lower in CAJT by SPME-GC/MS assay. Additionally, the acetic acid, butanoic acid, and naphthalene causing off-flavor were identified in CC, but not in Job's tears Cheonggukjang. The strength of odor through sensory evaluation was by far the lowest in CAJT among the groups. From the findings, it had shown that Job's tears Cheonggukjang would be produced successfully when the ratio of 4:1 (soybean : Job's tears) is employed. Also, it was proved that appropriate ratio of Job's tears addition makes Cheonggukjang odor-reduced and well accepted by people.

Micropropagation of Oak Seedlings from 37 Plus Half-Sib Families (참나무류(類) 수형목(秀型木) 37가계(家系)의 기내증식(器內增殖))

  • In vitro shoot proliferation and rooting were tested for 2-0 seedlings of half-sib families of 4 plus oaks trees. Nodal segments having axillary buds from 37 families(16 of Quercus acutissima, 10 of Q. variabilis, 7 of Q. serrata, and 4 of Q. mongolica) were cultured on WPM(Woody Plant Medium) supplemented with 0.5 mg/l BA (6-benzyladenine) and 0.01 mg/l NAA(${\alpha}$-naphthalene acetic acid) and subcultured at 2-3 weeks of intervals fur 6 months. In vitro rooting was carried out on GD(Gresshoff and Doy) medium supplemented with 0.5mg/l IBA(indole butyric acid). The capacity for shoot proliferation and rooting was highly varied with families. Generally, white oaks(Q. serrata and Q. mongolica) showed poor response than black oaks(Q. acutissima and Q, variabilis) in shoot proliferation and rooting. Among the total of 37 families, 7 of Q. acutissima, each 2 of Q. variabilis, Q. serrata, and Q. mongolica revealed abilities for continuous shoot proliferation, and the others failed to proliferate. Rooting of the selected oak trees also greatly varied among the families. In Q. acutissima, rooting ratio ranged from 10.0%(CB 25. KG 4) to 89.8%(CB 18). Although 26.7% of KG 16 in Q. variabilis, 3.3% of JN 15 in Q. serrata were rooted, Q. mongolica was not rooted at all in this experimental conditions. No relationship between shoot growth and the rooting ability was observed. Present results suggest the possibility of large-scale micropropagation, but further studies on family differences, shoot-tip necrosis, and callusing of rooting junction are still required to develop reliable micropropagation systems.

Effects of Cytokinin and Auxin on Organ Formation in Leaf Scale Tissue of Allium Sativum L. (Cytokinin과 Auxin이 Allium Sativum L.의 인엽배양(鱗葉培養)에서 기관(器官)의 분화(分化)에 미치는 영향(影響))

  • In order to determine the effects of cytokinin and auxin on organ formation from tissue of garlic cloves, leaf blades and basal tissues contained meristem of garlic (Allium sativum L.) cloves harvested in 1979 (old cloves) and 1980 (new cloves) were explanted on a MS medium contained various levels of BA ($N^6$-benzyl amino purine), NAA (naphthalene acetic acid), and 2, 4-D (2, 4-dichlorophenoxyacetic acid). And some of the new cloves were explanted on a media contained BA and NAA after chilling treatment at $4^{\circ}C$ for 10, 20, 30 and 40 days. 1. In a culture of leaf blades of old cloves, shoots were differentiated on a medium supplemented with 2mg/l of BA and NAA. 2. Callus was grown as a quite straw-coloured globular mass on a medium contained 0.2 or 2mg/l 2.4-D. 3. As subcultures of globular calli, shoots and roots were differentiated on a medium contained 2mg/l BA and 0.5 or 1 mg/l NAA, whereas no shoots was shown on a conterol. 4. Shoots were differentiated in a culture of leaf blades of new cloves, but they were not in an old cloves in control, and better effect was shown on a medium contained 2mg/l BA and 1mg/l NAA. However shoots were no differentiated from leaf blades chilled at $4^{\circ}C$ for 30 or 40 days at the same condition. 5. Large numbers of adventitious shoots could be obtained from basal region of garlic cultured on a medium contained 1mg/l BA and 4mg/l NAA, or 2mg/l BA and 2mg/l NAA.
