알파-피넨(alpha-pinene)이 함유된 스카티카증 치료용 나노복합체 개발 및 Miamiensis avidus에 대한 살충 효과 (Development of a Layered Nanocomposite Containing α-pinene to Treat Scuticociliatosis Caused by Miamiensis avidus)

  • Alpha-pinene is an organic compound that possesses antibiotic and anti-parasitic activities. In this study, we developed a layered nanocomposite to combat against Miamiensis avidus which causes scuticociliatosis in the olive flounder Paralichthys olivaceus. We used a solid-solid reaction to develop the layered nanocomposite, incorporating-pinene ($C_{10}H_{16}$) into organically modified montmorillonite. We used cetyltrimethylammonium cations as the interlayered modifier for the adsorption of hydrophobic pinene molecules. The X-ray diffraction patterns of the nanocomposite structure showed that the basal spacing increased from $9.6{\AA}$ to $30.4{\AA}$. Interestingly, the fraction of ${\alpha}$-pinene released remained constant for a long period of time (228 h) due to the layered nature of the nanocomposite. Additionally, optical microscopic images of the treated scuticociliatids revealed that their cells were lysed, and this effect increased with the increasing concertration of ${\alpha}$-pinene. Histopathological assessment of ${\alpha}$-pinene nanocomposite-treated olive flounder gills revealed no significant morphological changes, even at the highest concentration of the ${\alpha}$-pinene. The nanocomposite has several advantages, including easy handling, high solubility, low toxicity, and the easy formulation of granules or powder, which improve the pesticidal activity of ${\alpha}$-pinene. Collectively, our results suggest that ${\alpha}$-pinene nanocomposite may be a useful treatment against scuticociliatosis.

The Role of Vibrational Coherency in Ultrafast Reaction Dynamics of PYP

  • Coherent oscillations in is fluorescence dynamics of W.-t. PYP and its site-directed mutants have been observed. Two oscillatory modes coupled with the ultrafast fluorescence due to the twisting of the excited chromophore were identified, a high ftequency mode (∼135 cm$\^$-1/) with ∼550 is damping time and a low frequency overdamped mode (-45 cm$\^$-1/) with ∼250 is damping time, respectively. Both modes disappear in the fluorescence dynamics of denatured PYP emphasizing the important role of the protein nanospace as the environment for photoreaction. The qualitative picture of fluorescence dynamics in site-directed mutants was rather similar to that in W.-t. PYP, i.e., similar oscillatory modes (∼130-140 cm$\^$-1/ and ∼40-70 cm$\^$-1/) have been observed. This indicates that the vibrational modes and electron-vibration couplings do not change dramatically due to the mutation though the damping time of low frequency mode a little decreases as the protein nanospace structure becomes looser and more disordered by mutation. On the other hand, in the case of some PYP analogues, the qualitative picture of fluorescence dynamics changes, showing the familiar picture of solvation effect whereas the oscillations are almost damped. Comparative analyses of these observations are presented.

Nanospace Confinement of Conducting Polymers using Mesoporous Silica and Organosilica

  • Conducting polymers (e.g. poly[2-methoxy-5-(2'-ethyl-hexyloxy)-1,4-phenylen vinylene] (MEH-PPV)) confined in one-dimensional nanoscale channels of mesoporous materials, are expected to lead the novel applications for electroconductive and optoelectronic devices. We investigated the adsorption behavior of MEH-PPV on organically surface-modified mesoporous silica (FSM-16) and mesoporous organosilica. The amount of the confined MEH-PPV was found to strongly depend on the surface modifications of the mesoporous materials. The optical absorption edge of the confined MHE-PPV was clearly blue-shifted when compared to that of a free MHE-PPV.

황산 제1철을 이용한 방추형 괴타이트 나노 입자의 합성 (Synthesis of Spindle Shape α-FeOOH Nanoparticle from Ferrous(II) Sulfate Salt)

  • A wet-chemical route was utilized to obtain nanosized crystalline goethite ($\alpha$-FeOOH) particle, which was known as an oxidation catalyst in reducing carbon monoxide (CO) and dioxine during incineration. A cost-effective $FeSO_4{\cdot}7H_2O$ was used as starting raw material and a successive process of hydrolysis-oxidation was utilized as synthetic method. The effects of the initial $Fe^{2+}$ concentration, hydrolysis time and oxidation period on the crystalline phase and particle characteristics were systematically investigated by X-Ray Diffraction (XRD), Scanning Electron Microscopy (SEM) and BET analyses. It was found that the spindle-shaped crystalline $\alpha$-FeOOH particle with the width of 70 nm and the length of 200 nm could be obtained successfully when the initial concentration of 1.5 M, hydrolysis time of 4h, and oxidation period of 10 h, respectively. In addition, it was observed that the spindle-shaped $\alpha$-FeOOH particle consisted of nano-sized primary crystallites of $30\~50\;nm$, which were de-agglomerated into individual particle and successively re­agglomerated into spherical or irregular-shaped agglomerates beyond certain periods in the hydrolysis and oxidation process.

  • The importance of nanopore structures of carbons is shown in terms of interaction potential for various molecules including supercritical gases such as $H_2$ and $CH_4$. The key factors for adsorption of supercritical $H_2$ and $CH_4$ are shown for single wall carbon nanohorn, single wall carbon nanotube, and double wall carbon nanotube. The cluster formation of molecules is a key process for water adsorption on hydrophobic carbon nanopores. The X-ray absorption spectroscopic examination elucidates an explicit dehydration structure of ions confined in carbon nanopores.

건강기능식품에서 HPLC-UVD를 이용한 자일로올리고당 시험법의 표준화 연구 (Studies on Xylooligosaccharide Analysis Method Standardization using HPLC-UVD in Health Functional Food)

  • 본 연구는 자일로올리고당의 과학적이고 체계적인 표준화된 시험법을 마련하여 다양한 제형의 제품에 적용하고자 하였다. 최적화된 시험법을 마련하기 위해 초음파 처리 시간, 산 가수분해 시간 및 농도를 검토하여 전처리 방법을 비교 평가하였으며, HPLC-UVD를 이용하여 시료 중의 자일로올리고당을 분석하였다. 분석 시 1-phenyl-3-methyl-5-pyrazoline (PMP)으로 유도체화하고, photo diode array (PDA)가 장착된 high performance liquid chromatography (HPLC) (Nanospace SI-2)를 사용하였으며, 칼럼은 Cadenza C18 (4.6 × 250 mm, 3 ㎛)이었으며, 이동상은 pH를 6.0으로 맞춘 20 mM 인산완충용액과 아세토니트릴을 78:22 비율로 혼합하여 사용하였고, 0.5 mL/min 유속으로 254 nm로 하여 분석하였다. 건강기능식품 등 시험법 마련 표준절차에 관한 가이드라인에 따라 밸리데이션을 수행하고, 표준화된 시험법을 이용하여 유통 중인 건강기능식품 대상 품목에 대해 시험법 적용 여부를 확인하였다. 표준화된 시험법은 자일로올리고당을 함유한 건강기능식품 품질관리에 대한 신뢰성을 더 높일 것으로 본다.
