The Changes of $Vit.D_2$ and $Vit.B_2$ Contents according to Ultraviolet rays and Cooking Methods of Mushrooms (버섯의 자외선조사와 조리조건에 따른 $Vit.D_2$$Vit.B_2$ 함량의 변화)

  • This study was to investigate the effect of ultraviolet rays, soaking, boiling and baking to ergocalciferol ($Vit.D_2$) and riboflavin($Vit.B_2$) contents of mushrooms, Lentinus edodes, Pleurotus ostreatus and Agaricus bisporus. The results were as follow: 1. Mushrooms were exposed to ultraviolet rays Lentinus edodes : $10J/cm^2$, Pleurotus ostratus : $2J/cm^2$ and Agaricus bisporus : $2J/cm^2$. 2. Before exposing to ultraviolet rays, the ergocalciferol contents of mushrooms were all $0{\mu}g/g$ dry base, but after exposing to it , those of Lentinus edodes, Pleurotus ostreatus and Agaricus bisporus were $222.50{\pm}5.30{\mu}g/g$ dry base, $150.90{\pm}6.60{\mu}g/g$ dry base and $23.98{\pm}1.20{\mu}g/g$ dry base, respectively 3. Before and after exposing to ultraviolet rays, the riboflavin contents of Lentinus edoes, Pleurotus ostreatus and Agaricus bisporus were $18.22{\pm}0.71{\mu}g/g$ dry base and $11.72{\pm}0.50{\mu}g/g$ dry base, $4.57{\pm}0.20{\mu}g/g$ dery base and $3.26{\pm}0.15{\mu}g/g$ dry base, and $37.42{\pm}1.20{\mu}g/g$ dry base and $27.33{\pm}2.10{\mu}g/g$ dry base, respectively. 4. The ergocalciferol contents of mushrooms according to boiling time were not significantly different but the riboflavin contents of them were decreased according to the increase of boiling time. 5. The ergocalciferol and riboflavin contents of mushrooms were decreased according to the increase of NaCl concentration and baking temperature. 6. The ergocalciferol content of Lentinus edodes after a short time soaking at $80^{\circ}C$ was higher than a long time soaking at $20^{\circ}C,\;40^{\circ}C$ and $60^{\circ}C$.

Establishment of Elution and Concentration Procedure for Rapid and Sensitive Detection of Norovirus in Foods of Diverse Matrices (다양한 매트릭스가 혼합된 식품을 대상으로 노로바이러스 신속검출을 위한 탈리 및 농축방법 확립)

  • This article reports the development of an effective test procedure for detection of norovirus (NoV) in foods of diverse matrices. In this study, target foods included fermented milk, soybean paste, powders made from uncooked grains and vegetables, sesame leaves preserved in soy sauce, pickled mooli, and mooli. Viral recovery varied depending on the food matrices or elution buffers tested. Buffers were compared to determine effective elution buffers from artificially virus-contaminated foods. The conventional test procedure for concentrating viruses from food (elution-polyethylene glycol(PEG) precipitation-chloroform-PEG precipitation) was modified to save time by eliminating one PEG precipitation step. The modified procedure (elution-chloroform-PEG precipitation) was able to concentrate viruses more effectively than the conventional procedure. It also removed RT-PCR inhibitors effectively. The modified procedure was applied to target food for genogroup II NoV detection. NoV RNA was detected at the initial inoculum levels 3.125-12.5 RT-PCR units per 10-25 g tested food. The use of this newly established procedure should facilitate detection of low levels of norovirus in diverse foods.

  • Polyamine type polymer flocculants were synthesized and their characterization, viscosity and flocculation studies were conducted. In order to increase the molecular weight of polyamine flocculant which was prepared by polycondensation reaction from dimethylamine and epichlorohydrin, a small amount of 1,6-hexanediamine was employed. The incorporation of 1,6-hexanediamine up to 5.5 mole % replacing corresponding part of dimethylamine gave a branched type polyamine sample with increased intrinsic viscosity ([${\eta}$]=0.46 in 1 wt % aqueous NaCl solution). The amount of 1,6-hexanediamine above 5.5 mole %, however, resulted in gelation during polymerization. Utilizing raw water from Maegok potable water treatment plant, it was found that the addition of polyamine flocculant at a concentration of 1 mg/L level could reduce the amount of polyaluminum chloride (PAC) inorganic flocculant by half (15 mg/L). It was also observed that the incoporation of polyamine flocculant at 1 mg/L level was effective in the higher pH raw water, while PAC inorganic flocculating agent alone was not effective.

A Study on the Characteristics of Soil in the Asian Dust Source Regions of Mongolia (황사발원지 (몽골) 토양에 대한 특성 분석)

  • This study aims to identify the characteristics of soil in Mongolia, one of the major Asian dust sources that influence the Korean Peninsula. Soil particle size was analyzed and the result shows that sand (57.5~97.3%) was identified prominently in most regions, followed by silt (2.5~34.7%) and clay (0.0~7.8%). Soil pH of the covered regions were in the range 7.1~10.1, either weak alkaline or strong alkaline. Analysis of ion species in the soil samples exhibited that $Na^+$ ($91.9\;mg\;kg^{-1}$), $Cl^-$ ($65.9\;mg\;kg^{-1}$), and $Ca^{2+}$ ($53.5\;mg\;kg^{-1}$) were detected more in the soil than other species such as ${SO_4}^{2-}$ ($19.2\;mg\;kg^{-1}$), ${NO_3}^-$ ($46.6\;mg\;kg^{-1}$), ${NH_4}^+$ ($3.9\;mg\;kg^{-1}$), $K^+$ ($22.0\;mg\;kg^{-1}$), and $Mg^{2+}$ ($10.2\;mg\;kg^{-1}$). As for heavy metal content in the soil, concentrations of soil-borne metals including Fe, Al, Ca, Mg, and K tended to be high, while metals that come from manmade sources Pb, Cd, Cr, V, and Ni were remarkably low. The concentration of organic carbon (OC) was relatively high at $15.9\;{\mu}g\;mg^{-1}$, while elemental carbon (EC), directly released in the process of fossil fuel combustion, was not detected at all or found in very small amounts. The result indicates that pollution from manmade sources scarcely occurred. The analysis results from this study may contribute to improving modeling accuracy by providing input data for Asian dust prediction models, and be used as base data for determining the process of physiochemical transformation of Asian dust during long-range transport.

Development of the Functional Beverage Containing the Prunus mume Extracts (매실 추출물을 함유한 기능성 음료 개발)

  • This study was performed to investigate the cytotoxic effect of the Prunus mume extracts containing beverages on the growth of SNU-16 gastric cancer cell, SNU-C2A colon cancer cell, SNU-449 liver cancer cell and HeLa cervical cancer cell. The inhibitory effect on the growth of the cancer cell lines was examined by MTT(3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay, cytotoxicity test and microscopy. Also this study aimed to compare the changes of blood electrolytes and osmolarity during submaximal exercise for the intake of the Prunus mume extracts containing beverages. 20% Prunus mume extracts containing beverage exhibited the greatest inhibitory effect on the growth of SNU-16 and significantly inhibited at the concentration of $1000\;{\mu}g/mL$ in the MTT assay. Morphological changes in SNU-16 which treated with the same beverage were observed under inverted microscope. The change of blood electrolytes and osmolarity during submaximal exercise showed no significant differences between before and after intake of the beverage in both groups.

Effect of Organic Acids on Suppression of Fishy Odor in Salted Clam Pickle (유기산에 의한 조개젓 비린내 억제효과에 관한 연구)

  • To investigate the effect of organic acids on suppression of fishy odor, the change of the odor and the remaining amount of trimethyamine (TMA), selected as the representative component of fishy odor in clam pickle (15.5% NaCl) were examined by sensory evaluation and gas chromatography. The fishy odor linearly decreased as the amount of vinegar increased at ${\alpha}=0.01$, but there was no significant difference of fishness score for the changes of storage period and kinds of acid. Overall acceptability score was found to be the best when 8 ml vinegar added to 100g clam pickle. The remaining amount of TMA showed the tendency to decrease as the amount of acetic acid and storage time increased and kinds of acid were changed in order of acetic, malic and citric acid. However when $4\;{\mu}l/ml$ acetic acid (=8 ml vinegar/100g clam pickle) was added, the percentage of the remaining amount of TMA showed little decrease from about 95% for 72 hours of storage time. From the results of sensory evaluation and gas chromatography, it was considered that acids have the effect on suppression of fishy odor by producing non-volatile salts by combining with TMA chemically and that the effect of acids on suppression of fishy odor is mainly due to the masking effect at the concentration commonly used.

  • In order to investigate the feasibility of 30K protein from silkworm (Bombyx mori) hemolymph (SH) on the proliferation of human cells, a simple separating procedure by anion-exchange chromatography system with Q-Sepharose fast flow gel was established. The 30K protein was eluted with an optimized condition of 0.16 M sodium chloride in 20 mM tris buffer (pH 9.0). The separated 30K protein at three concentrations of 0.04, 0.12, and 0.4 mg/ml was added to the culture medium with various human cells, such as chondrocytes, periosteum-derived cells, and MRC-5 cells, and their growth rates were measured. The cell growth rate at protein concentration of 0.4 mg/ml was always higher than that without 30K protein in all human cells tested, suggesting that the 30K protein has positive effect on the increase of the life span of human cells.

Gibberellin A7 production by Aspergillus tubingensis YH103 and cultural characteristics of endophytic fungi isolated from Tetragonia tetragonoides in Dokdo islands (독도 번행초에서 분리된 내생균류의 배양적 특성과 Aspergillus tubingensis YH103의 gibberellin A7의 생산)

  • Coastal plant species Tetragonia tetragonoides (Pall.) Kuntze native to the Dokdo islands was sampled and then 17 endophytic fungi were purely isolated based on morphological differences. The fungal isolates were characterized by their growth properties under NaCl concentration or pH gradient. Culture filtrates of the 17 fungal isolates were treated to Waito-c rice (WR) seedlings for verifying plant growth-promoting activity. As the results, YH103 strain showed the highest plant growth-promoting activity among them. Phylogenetic analysis of the isolates was done by the maximum likelihood method based on partial internal transcribed spacer region (ITS region: contaning ITS1, 5.8S, and ITS2), beta-tubulin (BenA), and calmodulin (CaM) gene sequences. Chromatographic analysis of the strain YH103 culture filtrate showed the existence of gibberellins ($GA_4$, $GA_7$, $GA_8$, and $GA_{19}$). Finally, the strain YH103 was identified as Aspergillus tubingensis by microscopic observation and molecular analysis and, to our knowledge, this is the first report of GAs producing A. tubingensis.

Characteristics of Protease Produced by Rhizopus stolonifer, Rhizopus oryzae and Absidia corymbifera from Korean Traditional Meju (한국 전통 메주 유래의 Rhizopus stolonifer, Rhizopus oryzae 및 Absidia corymbifera가 생성하는 Protease의 특성)

  • Pretense production and its characteristics were investigated for Rhizopus stolonifer, Rhizopus oryzae and Absidia corymbifera which were isolated from Korean traditional meju. The optimum culture conditions of the strains for the production of protease in basic medium [wheat bran : 1% glucose solution=1 : 1 (w/v)] were 3$0^{\circ}C$ and 4 days. The optimum pH and temperature for the enzyme activity of crude enzymes produced by Rhizopus sto-lonifer, Rhizopus oryzae and Absidia corymbifera were pH 6.0 and 5$0^{\circ}C$, respectively. The enzymes were relatively stable at pH 4.0~7.0, at temperature below 4$0^{\circ}C$, and at NaCl concentration lower than 16%. The $K_{m}$ value for Hammastein casein was 3.3$\times$10$^{-4}$ , 0.75$\times$10$^{-4}$ and 1.3$\times$10$^{-4}$ M, and $V_{max}$ value was 17.2$\mu\textrm{g}$/min, 9.4$\mu\textrm{g}$/min and 7.8$\mu\textrm{g}$/min, respectively.y.

Changes of Enzyme Activity and Physiological Functionality of Traditional Doenjang during Fermentation Using Bacillus sp. SP-KSW3 (Bacillus sp. SP-KSW3를 이용하여 제조한 된장 발효 과정중의 효소 활성 및 기능성의 변화)

  • Bacillus sp. SP-KSW3 is an auxothroph bacteria that is being used for starter in fermentation. Physico-chemical characteristics, enzyme activities, ACE inhibitor and antimutagenicity in fermented soybean inoculated with Bacillus sp. SP-KSW3 starter was investigated for the ripening duration of fermentation. Tyrosinase and ACE showed 10% higher activity degree on test field than control. for antimutagenicity using S. enterica serovar Typhimurium TA100 against MNNG and NPD showed 86.24% and 75.63% Similarly, S. enterica serovar Typhimurium TA98 was used against NPD and NQO showed 60.28% and 50.92% respectively. Hydrogen donating ability increased compared to the control having 81.7% and 80.1% respectively. Daidzin of isoflavone in fermented soybean showed higher concentration in control than in the test field. Genistein from two years of ripening test field contained 11.67 mg/kg compared to the test field. The initial test field for daidzin contained 389.96 mg/kg which increased to 453.67 mg/kg after two years and the initial genistein contained 402.68 mg/kg which also increased to 556.86 mg/kg.
