To explore the action mechanism of Ijintang in the brain, the authors investigated the effects of Ijintang fractions on MgNaK ATPase and MgCa ATPase in rat brain synaptosomes prepared from cerebral cortex. The activities of MgNaK ATPase and MgCa ATPase were assayed by the level of inorganic phosphate liberated from the hydrolysis of ATP. Fraction WH-95-7 at the concentration of $10^{-2}%$ decreased the activity of MgNaK ATPase about 34.1% and also reduced the activity of MgCa ATPase about 49.3% But, other fractions (WB-95-7, WC-95-7, MB-95-7, MC-95-7, MH-95-7) did not significantly changed the activities of the MgNaK ATPase and MgCa ATPase The decreased activity of MgNaK ATPase by WH-95-7 will decrease the rate of $Ca^{2+}$ efflux, probably via an Na-Ca exchange mechanism and will increase the rate of $Ca^{2+}$ entry by the depolarization of nerve terminals. The reduced activity of MgCa ATPase by WH-95-7 will result in the decreased efflux of $Ca^{2+}$. As a conclusion, it can be speculated that lithium elevates the intrasynaptosomal $Ca^{2+}$ concentration via inhibition of the activities of MgNaK ATPase and MgCa ATPase. and this increased $[Ca^{2+}]i$ will cause the release of neurotransmitters.
Using BHK cells with stable expression of cardiac $Na^+/Ca^{2+}$ exchanger(BHK-NCX1), reverse mode(i.e. $Ca^{2+}$ influx mode) of NCX1 current was recorded by whole-cell patch clamp. Repeated stimulation of reverse NCX1 produced a cytosolic $Ca^{2+}$-dependent long-term inactivation of the exchanger activity. The degrees of inactivation correlated with NCX1 densities of the cells and were attenuated by reduced $Ca^{2+}$ influx via the reverse exchanger. The inactivation of NCX1 was attenuated by(i) inhibition of $Ca^{2+}$ influx with reduced extracellular $Ca^{2+}$, (ii) treatment with NCX1 blocker($Na^{2+}$), and (iii) increase of cytoplasmic $Ca^{2+}$ buffer(EGTA). In BHK-NCX1 cells transiently expressing TRPV1 channels, $Ca^{2+}$ influx elicited by capsaicin produced a marked inactivation of NCX1. We suggest that cytoplasmic $Ca^{2+}$ has a dual effect on NCX1 activities, and that allosteric $Ca^{2+}$ activation of NCX1 can be opposed by the $Ca^{2+}$-dependent long-term inactivation in intact cells.
$[Ca^{2+}]_i$ transients by reverse mode of cardiac $Na^+/Ca^{2+}$ exchanger (NCX1) were recorded in fura-2 loaded BHK cells with stable expression of NCX1. Repeated stimulation of reverse NCX1 produced a long-lasting decrease of $Ca^{2+}$ transients ('rundown'). Rundown of NCX1 was independent of membrane $PIP_2$ depletion. Although the activation of protein kinase C (PKC) was observed during the $Ca^{2+}$ transients, neither a selective PKC inhibitor (calphostin C) nor a PKC activator (PMA) changed the degrees of rundown. By comparison, a non-specific PKC inhibitor, staurosporine (STS), reversed rundown in a dose-dependent and reversible manner. The action of STS was unaffected by pretreatment of the cells with calphostin C, PMA, or forskolin. Taken together, the results suggest that the stimulation of reverse NCX1 by STS is independent of PKC and/or PKA inhibition.
Ca-type and Na-type bentonites show the great difference of some physicochemical properties. Na exchanged bentonite is mainly used for the foundry and construction materials in domestic utilization. This study tries to identify in detail the differences of some physicochemical properties and thermal properties between Ca-type and Na-type bentonites. Also the adsorption behavior and interlayer expansion for the HDTMA (Hexadecyltrimethylammonium) exchanged and CP (Cetylprydinium) exchanged Ca-type and Na-type bentonites were compared. Na-type bentonite shows the strong alkaline property, high viscosity and swelling compared to Ca-type bentonite. However, two types are very similar for the cation exchange capacity and MB (Methylene Blue) adsorption. The decomposition of adsorbed and interlayer water of Na-type bentonite is caused in the lower temperature than Ca-type bentonite. And Ca-type bentonite shows the decomposition of structural water in the lower temperature than Na-type bentonite. The interlayer expansion of montmorillonite resulted to the intercalation of HDTMA and CP into bentonite is so strongly caused from 12~15 $\AA$ to $40\AA$ (basal spacing). HDTMA-bentonite is almost expanded to $37~38\AA$ when 200% CEC equivalent amount of HDTMA is added, and CP-bentonite is fullly expanded to 40 $\AA$ in the 140% CEC equivalent amount of CP It means that CP causes the stronger interlayer expansion of montmorillonite and easier adsorption than HDTMA. Adsorption behaviors of CP into bentonite is so stable and continuously sorbed in the proportion to the treatment of amount until 200% of the CEC equivalents. CP-bentonite shows the same adsorption behavior regardless of Ca-type or Na-type montmorillonite.
The $Ca^{++}$ and $Mg^{++}$ released during Ca, Mg-Na exchange on Kampo 78 montmorillonite which was treated with various concentrations of NaCl solution, were measured with EDTA titration metbod in the leaching solutions. Lanthanide montmorillonite was prepared with various neutral lanthanide ions from sodium montmorillonite in which the exchangeable ions are displaced from the exchanger, such as the displacement of $Na^+$ by $Ln^{3+}$ ions, Cation exchange capacity (CEC) is determined on remaining lanthanides in the leaching solutions with E. D. T. A titration method. As a results of this study, there were no difference of C. E. C in series of lanthanide contraction, but C. E. C depends on charge density of montmorillonite. When we conformed the structure of Ln-montmorillonite by X-ray diffraction. It was found that there was much difference of pattern between Na-montmorillonite and Ln-montmorillonite.
Thyroid function is mainly regulated through cAMP and phophatidylinositol, and it is well known that TSH-stimulated thyroxine ($T_4$) release is inhibited by catecholamine from mouse thyroids via the ${\alpha}_1$-adrenoceptor stimulation. Previous study has established that the inhibition of $T_4$ release by ${\alpha}_1$-adrenoceptor stimulation results in activated protein kinase C (PKC). The purpose of this study was to determine if ion transport systems are involved in the inhibition of $T_4$ release elicited by ${\alpha}_1$-adrenergic agonist in mouse thyroids. TSH-, IBMX- and cAMP analogue-stimulated $T_4$ release were significantly inhibited by methoxamine, R59022 (diacylglycerol kinase inhibitor), and MDL (adenylate cyclase inhibitor). TSH-stimulated $T_4$ release could be inhibited by Bay K 8644 and cyclopiazoic acid, but not by verapamil and tetrodotoxin. The addition of nifedipine ($Ca^{2+}$ channel blocker), tetrodotoxin and lidocaine ($Na^+$ channel blockers), but not amiloride (EIPA) and ryanodine, completely blocked the inhibitory effects of methoxamine on $T_4$ release. TSH-stimulated $T_4$ release was also inhibited by benzamil ($Na^+-Ca^{2+}$ exchange inhibitor). TSH-, IBMX- and cAMP-stimulated $T_4$ release were inhibited by methoxamine or R59022, these effects were reversed by nifedipine. but not by verapamil. Furthermore, nifedipine reversed the inhibitory effects of benzamil and R59022 on TSH-stimulated $T_4$ release. These data suggest that the observed ${\alpha}_1$-adrenoceptor-mediated inhibition of $T_4$ release in mouse thyroids is the result of an increase in intracellular $Na^+$ or $Ca^{2+}$ effected via activation of fast $Na^+$ or nifedipine-sensitive $Ca^{2+}$ channels, and that $Na^+-Ca^{2+}$ exchange may play an important role in reducing thyroid hormone by increasing intracellular $Ca^{2+}$.
Two dolomite samples mined from the different mines were calcined using a batch-type microwave kiln ($950/60min^{\circ}C$) to produce $CaO{\cdot}MgO$. The hydration of the $CaO{\cdot}MgO$ samples shows different reactivity. MgO was separated by reacting with a strong acid cation exchange resin using the reactivity of the hydration properties of light dolomite ($CaO{\cdot}MgO$). Calcium ($Ca-(R-SO_3)_2$) was separated from the prepared $CaO{\cdot}MgO$ by the cation exchange resin ($CaO{\cdot}MgO:R-SO_3H=1:12mass%$). High purity MgO (higher than 94 mass %) with unburned $CaCO_3$ (1~2 mass %) was obtained by the separation process. The separated MgO was heated at $950^{\circ}C$ for 60 minutes to afford high purity MgO with MgO content higher than 96%. And High-grade $CaCO_3$ was prepared from the reaction with calcium adsorbed resin ($Ca-(R-SO_3)_2$) and NaOH, $CO_2$ gas.
The underlying mechanism commonly applicable for both the positive and negative force-frequency relationships (FFR) was pursued in left atria (LA) of rat and rabbit. The species differences in the roles of $Na^+/Ca^{2+}$ exchanger and sarcoplasmic reticulum (SR), which are major intracellular $Ca^{2+}$ regulatory mechanisms in the heart, were examined in the amplitude accommodation to the frequency that changed from 3 Hz to the variable test frequencies for 5 minutes in the electrically field stimulated left atria (LA) of rat and rabbit. Norepinephrine strongly increased the frequency-related amplitude accommodation in both of rat and rabbit LA, while monensin, oubain or the reduced $Na^+$ and 0 mM $Ca^{2+}$ containing Tyrode solution increased the frequency-related amplitude accommodation only in the rabbit LA. Monenisn was also able to increase the frequency-related amplitude accommodation only in 1-day old rat LA but not in 4-week old rat LA that had 75% less $Na^+/Ca^{2+}$ exchanger with 97% higher SR than 1-day old rat LA. Taken together, it is concluded that the differences in the prevalence between myocardial $Na^+/Ca^{2+}$ exchanger and SR in the amplitude accommodation to the frequency-change determine the difference in the FFR between rat and rabbit heart.
The $Na^+-and\;K^+-induced\;Ca^{++}$ release was measured isotopically by millipore filter technique in pig heart mitochondria. With EGTA-quenching technique, the characteristics of mitochondrial $Ca^{++}-pool$ and the sources of $Ca^{++}$ released from mitochondria by $Na^+\;or\;K^+$ were analyzed. The mitochondrial $Ca^{++}-pool$ could be distinctly divided into two components: internal and external ones which were represented either by uptake through inner membrane, or by energy independent passive binding to external surface of mitochondria, respectively. In energized mitochondria, a large portion of $Ca^{++}$was transported into internal pool with little external binding, while in de-enerigzed state, a large portion of transported $Ca^{++}$ existed in the external pool with limited amount of $Ca^{++}$ in the internal pool which was possibly transported through the $Ca^{++}-carrier$ present in the inner membrane. $Na^+$ induced the $Ca^{++}$ release from both internal pool and external pool and external binding pool of mitochondria. In contrast, $K^+$ did not affect $Ca^{++}$ of the internal pool, but, displaced $Ca^{++}$ bound to external surface of the mitochondria. When the $Ca^{++}-reuptake$ was blocked by EGTA, the $Ca^{++}$ release from the internal pool by $Na^+$ was rapid; the rate of $Ca^{++}-efflux$ appeared to be a function of $[Na^+]^2$ and about 8mM $Na^+$ was required to elicit half-maximal velocity of $Ca^{++}-efflux$. So it was revealed that $Ca^{++}-efflux$ velocity was particulary sensitive to small changes of the $Na^+$ concentration in physiological range. Energy independent $Ca^{++}-binding$ sites of mitochondrial external surface showed unique characteristics. The total number of external $Ca^{++}-binding$ sites of pig heart mitochondria was 29 nmoles per mg protein and the dissociation constant(Kd) was $34{\mu}M$. The $Ca^{++}-binding$ to the external sites seemed to be competitively inhibited by $Na^+\;and\;K^+$; the inhibition constant(Ki) were 9.7 mM and 7.1 mM respectively. Considering the intracellular ion concentrations and large proportion of $Ca^{++}$ uptake in energized mitochondria, the external $Ca^{++}-binding$ pool of the mitochondria did not seem to play a significant role on the regulation of intracellular free $Ca^{++}$ concentration. From this experiment, it was suggested that a small change of intracellular free $Na^+$ concentration might play a role on regulation of free $Ca^{++}$ concentration in cardiac cell by influencing $Ca^{++}-efflux$ from the internal pool of mitochondria.
In this study, the heterogeneous ion exchange membranes prepared by the combination of the carbon electrode and mixed the cation and anion exchange polymers and polyvinylidene fluoride as the basic polymer together were made to recognize the efficiency of the salt removal for the application of the membrane capacitive deionization process. The mixing weight ratio of the solvent, basic polymer and ion exchange resin was 7 : 2 : 1 and this mixed solution was directly cast on the electrode. As for the operating conditions of the adsorption voltage and time, feed flow rate, desorption voltage and time of the feed solution NaCl 100 mg/L, the salt removal efficiencies (SRE) were measured. Apart from this NaCl, the $CaCl_2$ and $MgSO_4$ solutions were investigated in terms of SRE as well. Typically, SRE for NaCl 100 mg/L solution under the conditions of adsorption voltage/time, 1.5 V/3 min, desorption voltage/time -0.1 V/3 min, was shown 98%. And for the $CaCl_2$ and $MgSO_4$ solutions, the SREs of 70 and 59% were measured under the conditions of adsorption voltage/time, 1.2 V/3 min, desorption voltage/time -0.5 V/5 min, respectively.
