Effect of Sam Hwa San Extract on Renal Function in Rabbit (삼화산(三和散)이 가토(家兎) 신장기능(腎臟機能)에 미치는 영향(影響))

  • In order to examine that the effect of Sam Hwa San, circulating the vital energy of Sam Cho and controlling body fluid metabolism, gives any influence on renal function, changes in the urine flow, eletrolytes excretion, plasma aldosterone concentration and renin activity were observed after intravenous infusion of the Sam Hwa San extract in rabbit. Also in vitro effect of the herb extract on oxygen consumption in renal cortical slices and ATPase activity in kidney microsomes was measured. The following results were obtained : 1. The urine flow was markedly increased at 10 min after intravenous infusion of the Sam Hwa San extract($0.134{\pm}0.015$ vs. $0.433{\pm}0.046ml/min.kg$), but return ed to normal value after 40 min of infusion. 2. The glomerular filtration rate was significantly increased at 10 min after in travenous infusion of the Sam Hwa San extract, and the renal plasma flow at 10 and 20 min after infusion of the Sam Hwa San extract, following return to normal value. 3. $Na^+$ excretion was significantly increased during 10-40 min after intravenous infusion of the Sam Hwa San extract, although showed the maximal rate at 10-20 min. The fractional $Na^+$ excretion was also increased during 10-40 min. $K^+$ excretion was rapidly increased at 10 min after the intravenous Infusion of the Sam Hwa San extract and then gradually decreased to normal level at 40 min. The fractional $K^+$ excretion was significantly increased during 10-40 min after the intravenous infusion of the Sam Hwa San extract. 4. The plasma aldosterone concentration and renin activity were not altered by the infusion of the Sam Hwa San extract. 5. The ouabain-sensitive oxygen consumption of renal cortical slices was significantly reduced by the Sam Hwa San extract(0.5 and 1.0 vol.%). 6. The Na-K-ATPase activity of renal microsomes was strongly inhibited by the Sam Hwa San extract(0.5 and 1.0 vol.%). These results suggest that the Sam Hwa San causes a strong diuretic effect which results from reduction of Na reabsorption in renal tubule by a direct inhibition of Na-pump and, in part, from all increase in renal blood flow. In clinic, it is considered to obtain the therapeutic effect in body fluid metabolism disharmony to cause the circular disorder of vital energy.

A study on the mechanism for reduction of lead-induced toxicity in nervous system by aloe vera (Aloe vera의 연 유도성 신경계 독성 저감 기전에 관한 연구)

  • Effects of water extract of aloe vera on lead-induced neurotoxicity were investigated in sciatic nerve isolated from rat. The mechanism on toxicity reduction by measuring activities of axonal enzymes, metabolism of myo-inositol in nerve, lead concentration in several organs and so on were further examimed. In the lead-treated rats, the transport rate of axonal enzyme, such as acetyl cholinesterase and choline acetyltransferase, was reduced by from 50% to 30% respectively. Reduction in myo-inositol concentration and $Na^+/K^+$ ATPase activity were also observed in sciatic nerve from lead-treated rat. However, the aloe extract administration significantly eliminated the impairment and maintained myo-inositol concentration to about 85% of normal level. Also aloe extract promoted the excretion rate of lead which is accumulated in blood, sciatic nerve and kidney. These results suggest that lead-induced neurotoxicity was significantly reduced by administration of aloe extract and the mechanism might be partly increase in kidney excretion rate of lead and parity normalization of $Na^+/K^+$ ATPase activity which is critical factor in order to keep nerve maintaining normal myo-inositol level.

Effect of Salviae Radix herb-acupuncture on rabbits with Hg-induced acute renal failure (단삼약침(丹蔘藥鍼)이 급성신부전(急性腎不全) 가토(家兎)의 신세뇨관(腎細尿管)에 미치는 영향(影響))

  • Objectives ; This study was undertaken to determine if Salviae Radix herb-acupuncture (SRA) exerts protective effect against alterations in membrane transport function in rabbits with mercury chloride (Hg)-induced acute renal failure. Methods and Results ; The administration of Hg at a subcutaneous single dose of 10mg/kg caused a reduction in GFR to 9.4% of the basal value and an increase in fractional Na+ excretion to 10-fold, indicating generation of acute renal failure. When animals were acupunctured with $0.5m{\ell}$ of SRA extract (0.1%) in both sides of Shinsu(BL23) for 7 days prod to Hg administration, such changes were significantly attenuated. The fractional excretion of glucose and phosphate was increased to approximately 132-fold and 7-fold, respectively, in rabbits treated with Hg alone, but the fractional excretion of glucose was increased to 26-fold and that of phosphate was not different from the basal value in SRA-pretreated rabbits. Uptakes of glucose and phosphate in purified isolated brush-border membrane and $Na^+-K^+$-ATPase activity in microsomal fraction were inhibited in rabbits treated with Hg alone, suggesting that impairment in proximal reabsorption of glucose and phosphate is resulted from a direct damage of membrane transport carriers and disruption of the normal $Na^+$ gradient. Conclusions ; Such changes were prevented by SRA. Uptakes of organic ions, PAH and TEA, in renal cortical slices were inhibited by the administration of Hg, which was prevented by SRA. Pretreatment of an antioxidant DPPD attenuated the increase in the fractional excretion of glucose and phosphate induced by the administration of Hg.

Association between Sodium Excretion and Obesity of Adults in Gwangju (광주지역 성인의 나트륨 배설량과 비만의 관계)

  • Purpose: The aim of this study was to analyze the association between sodium excretion and obesity for healthy adults in the Gwangju area. Methods: The participants included 80 healthy adults aged 19 to 69 years in Gwangju. The dietary intake and sodium excretion were obtained using the 24-hour recall method and 24 hour urine collection. The participants were classified into two groups according to the amount of urinary sodium excretion: (${\leq}141.75mmol/dL$, > 141.75 mmol/dL). Results: After adjusting for sex, age, smoking history, and income, the high excretion of sodium group was significantly higher for weight, body mass index, body fat mass, percent body fat, visceral fat area (VFA), waist circumference, hip circumference, and WHR. The energy and nutrients intake were significant after adjusting for sex, age, smoking history, and income. The LSE group had a significantly higher fat intake and Na/K intake ratio. The HSE group had significantly higher fiber intake, and K intake. As the amount of urinary sodium excretion increased, the risk of obesity before correction was 3.57 (95% CI: 1.13-11.25) times greater, and the risk of obesity of T3 increased significantly by 3.33 times (95% CI: 1.05-10.59). After correcting for sex and age, the obesity risk of T2 increased significantly by 4.23 times (95% CI: 1.11-16.06), and after correcting for sex, age, smoking history, and income, the obesity risk of T2 increased significantly by 6.81 times (95% CI: 1.44-32.19) the risk of obesity. Conclusions: An association exists between sodium excretion and obesity in Korean adults. In this study, the high excretion of sodium group was obese and the risk of obesity was higher than the low excretion of sodium group.

A Comparative Study on Urinary Ca Excretion in Young and Middle-Aged Korean Women (한국 젊은 성인 여성과 중년 여성의 소변중 Ca 배설과 이에 영향을 미치는 요인분석)

  • A study was conducted to compare urinary Ca excretion and factors influencing urinary Ca excretion in 30 young and 62 middle-aged Korean women. Mean daily intake levels of total protein and P were significantly higher in middle-aged women but Ca and animal protein intake levels of the two groups were similar. The average percentage of daily Ca intake from milk and milk products in young women was 45% while in middle-aged women it was about 24% The frequency of milk consumption was inversely correlated with blood pressures of the subjects. Mean 24-hour urinary calcium excretion in young and middle-aged women were 163.7mg and 174.9mg respectively. The difference was not significant. Menopause of the mid-dle-aged women did not affect urinary calcium levels. However the proportion of subjects with more than 250mg of Ca in 24-hour urine tended to be higher in middle-aged group Factors significnatly correlated with urinary Ca excretion of subjects were systolic and diastolic The study verifies the need for more systematic studies on Ca requirements and the interrelation-ship among Ca and na metabolism blood pressures and bone loss in the middle-aged Korean.

  • In order to evalulate the cause of polyuric acute tubular necrosis, we measured electrolytes, $Na^{+}\;and\;K^{+}$ excreted in urine, and activities of $Na^{+}-K^{+}$adenosine triphosphatase ($Na^{+}-K^{+}$ATPase) Excretion of $Na^{+}\;and\;K^{+}$ significantly increased in 24hr exposure on the uranyl nitrate and then decreased below the normal level after 3 days. The concentration of $Na^{+}\;and\;K^{+}$ in urines of the rats treated uranyl nitrate was less than that of the normal rats. The activities of $Na^{+}-K^{+}$ATPase decreased only in the group treated with high dose group of uranyl nitrate (30mg/kg BW) on the 3rd day but were not changed in the low dose groups(5 mg/kg BW and 15mg/kg BW).

Response of broiler chickens to diets containing different levels of sodium with or without microbial phytase supplementation

  • Phytate induced excessive mineral excretion through poultry litter leads to poor performance and environmental pollution. Exogenous microbial phytase supplementation to poultry diets reduce the environmental excretion of nutrient and improve bird's performance. However, excessive dietary sodium (Na) level may hinder the phytase-mediated phytate hydrolysis and negate the beneficial effects of phytase. Therefore, this experiment was conducted to investigate the effects of different concentration dietary Na on phytase activity and subsequent impact on broiler performance, bone mineralisation and nutrient utilisation. In this study, six experimental diets, consisting of three different levels of Na (1.5, 2.5, or 3.5 g/kg) and two levels of microbial phytase (0 or 500 U/kg) were formulated by using $3{\times}2$ factorial design. The six experimental diets were offered to 360 day-old Ross 306 male chicks for 35 days, where, each experimental diet consisted of 6 replicates groups with 10 birds. Along with growth performance, nutrient utilization, intestinal enzyme activity, dry matter (DM) content of litter and mineral status in bone were analysed. Dietary Na and phytase had no effect on bode weight gain and feed intake. Birds on the low Na diet showed higher (p < 0.05) feed conversion ratio (FCR) than the mid-Na diets. High dietary Na adversely affected (p < 0.001) excreta DM content. Phytase supplementation to the high-Na diet increased (p < 0.01) the litter ammonia content. High dietary Na with phytase supplementation improved ($Na{\times}phytase$, p < 0.05) the AME value and ileal digestibility of Ca and Mg. The total tract retention of Ca, P, and Mg was reduced with high Na diet, which was counteracted by phytase supplementation ($Na{\times}phytase$, p < 0.001). The diets containing mid-level of Na improved (p < 0.001) the function of Na-K-ATPase and Mg-ATPase in the jejunum. The overall results indicate that high dietary Na did not affect phytase activity but influenced the nutrient utilization of birds, which was not reflected in bird overall performance.

Effect of Seaweed Intake on the Absorption of Sodium, Calcium, Potassium and Hypolipidemic Mechanism in Healthy Male Subjects (해조류 섭취가 성인 남자의 Na, Ca, K 흡수와 지질대사에 미치는 영향)

  • This study was conducted to determine the effect of seaweed ingestion on Na, Ca and K balances in healthy male subjects and to elucidate possible hypolipidemic mechanisms. Six subjects were given a nutritionally controlled diet based on their usual intake for 5-days followed by 5 days in which seaweed was added to the basal diet. Based on the results of the study, the relation between the levels of intake and excretions in stool, urine and serum concentrations was analyzed. In this study, the ingested sodium and potassium were mostly excreted in urine. The highest the rate of urinary excretion to intake for K at 64%, followed by 56% for Na, Conversely, Ca was excreted in stool at higher rates. Analysis of the relation between the intake and excretion in stool revealed that Na and K showed a positive relation with statistical significance. There was no statistically significant relation between the intake and serum concentration of any these minerals. The levels of serum cholesterol and triglyceride were remarkably reduced (p<0.05) by seaweed intake. Thus, the results suggest a beneficial effect of seaweed on Na, Ca, K metabolic control and hypolipidemic mechanism of the healthy male subjects.

Comparision of Sodium Intakes Pattern in the Family Members of Normal and Stomach Cancer Patients (위암 환자 가족과 정상인 가족간의 Na섭취 패턴 비교)

  • Purpose of this study was to investigate intake pattern of sodium in the family members of normal and stomach cancer patients, excluding patients themselves. Every food samples that they consumed for 3 days, drinking water, hot pepper paste, soybean paste and soy sauce from the each family were collected for Na analysis. Three days of morning urine from the each subjects was collected for determination of urinary Na excretion. Sodium contents of hot pepper paste, pickles, soups and meats in stomach cancer families were significantly higher than those in normal families. However, urinary sodium excretion between the two groups was not different. This suggests that sodium metabolism in human may be altered with a long-term intake of sodium=rich foods.

  • Author have already reported that urinary aldosterone excretion of the Korean who usually eat high sodium diet is significantly lower comparing with the American, although the plasma aldosterone concentration is identical in the former with that of the latter. Measurement of urinary aldosterone excretion and Plasma concentration only is insufficient to establish the pressence and/or mode of evolution of the Korean. In this experiments, aldosterone secretion rate(ASR) was measured in normotensive Korean during high and low dietary sodium intake with or without additional potassium supply. Results were as follows; 1) In normal Korean, dietary sodium restriction resulted in appreciable increase in ASR, and a sustained increase in urinary aldosterone excretion with an increase in plasma level. 2) Oral potassium loading easily stimulated the adrenal cortex of the Korean who already adapted to a high sodium diet when dietary sodium is still identical with not·mal American. 3) Quantitative relationships between aldosterone secretion rate, plasma concentration and urinary excretion of aldosterone were altered by potassium loading. 4) Urinary aldosterone excretion didn't reflect concurrent increase aldosterone secretion in subjects with Potassium intake. It was discussed that the changes of tile relationships and of adrenal hyper response on Potassium Beading in the Korean will be elucidated by measuring the metabolic clearance rate.
