20(S)-protopanaxadiol promotes the migration, proliferation, and differentiation of neural stem cells by targeting GSK-3β in the Wnt/GSK-3β/β-catenin pathway

  • Background: Active natural ingredients, especially small molecules, have recently received wide attention as modifiers used to treat neurodegenerative disease by promoting neurogenic regeneration of neural stem cell (NSC) in situ. 20(S)-protopanaxadiol (PPD), one of the bioactive ingredients in ginseng, possesses neuroprotective properties. However, the effect of PPD on NSC proliferation and differentiation and its mechanism of action are incompletely understood. Methods: In this study, we investigated the impact of PPD on NSC proliferation and neuronal lineage differentiation through activation of the Wnt/glycogen synthase kinase (GSK)-3β/β-catenin pathway. NSC migration and proliferation were investigated by neurosphere assay, Cell Counting Kit-8 assay, and EdU assay. NSC differentiation was analyzed by Western blot and immunofluorescence staining. Involvement of the Wnt/GSK3β/β-catenin pathway was examined by molecular simulation and Western blot and verified using gene transfection. Results: PPD significantly promoted neural migration and induced a significant increase in NSC proliferation in a time- and dose-dependent manner. Furthermore, a remarkable increase in anti-microtubule-associated protein 2 expression and decrease in nestin protein expression were induced by PPD. During the differentiation process, PPD targeted and stimulated the phosphorylation of GSK-3β at Ser9 and the active forms of β-catenin, resulting in activation of the Wnt/GSK-3β/β-catenin pathway. Transfection of NSCs with a constitutively active GSK-3β mutant at S9A significantly hampered the proliferation and neural differentiation mediated by PPD. Conclusion: PPD promotes NSC proliferation and neural differentiation in vitro via activation of the Wnt/GSK-3β/β-catenin pathway by targeting GSK-3β, potentially having great significance for the treatment of neurodegenerative diseases.

Modeling shear capacity of RC slender beams without stirrups using genetic algorithms

  • High-strength concrete (HSC) is becoming increasingly attractive for various construction projects since it offers a multitude of benefits over normal-strength concrete (NSC). Unfortunately, current design provisions for shear capacity of RC slender beams are generally based on data developed for NSC members having a compressive strength of up to 50 MPa, with limited recommendations on the use of HSC. The failure of HSC beams is noticeably different than that of NSC beams since the transition zone between the cement paste and aggregates is much denser in HSC. Thus, unlike NSC beams in which micro-cracks propagate around aggregates, providing significant aggregate interlock, micro-cracks in HSC are trans-granular, resulting in relatively smoother fracture surfaces, thereby inhibiting aggregate interlock as a shear transfer mechanism and reducing the influence of compressive strength on the ultimate shear strength of HSC beams. In this study, a new approach based on genetic algorithms (GAs) was used to predict the shear capacity of both NSC and HSC slender beams without shear reinforcement. Shear capacity predictions of the GA model were compared to calculations of four other commonly used methods: the ACI method, CSA method, Eurocode-2, and Zsutty's equation. A parametric study was conducted to evaluate the ability of the GA model to capture the effect of basic shear design parameters on the behaviour of reinforced concrete (RC) beams under shear loading. The parameters investigated include compressivestrength, amount of longitudinal reinforcement, and beam's depth. It was found that the GA model provided more accurate evaluation of shear capacity compared to that of the other common methods and better captured the influence of the significant shear design parameters. Therefore, the GA model offers an attractive user-friendly alternative to conventional shear design methods.

  • With the rapid shift to a knowledge-based economy, science and technology (S&T) is considered as the principle drivers in a nation's competitiveness. All countries are focused on upgrading the level of their S&T of nations and strengthening their national competitiveness. With these factors in mind, Korea has increased its support of national research and development (R&D). As a result of this added support, Korea S&T has made great strides. This improvement can be seen in the drastic increase in the number of Korean researchers who have been published in Nature, Science and Cell (NSC), three top journals in the fields of S&T. This study is a quantitative analysis on the NSC Publications of Korean researchers. The purpose of this study is to derive research policy suggestions to improve Korea's national competitiveness in S&T.

Chemical resistance of Non-Sintered Cement Mortar using Inorganic Industrial Wastes as activator (무기계 산업폐기물을 자극제로 이용한 비소성 시멘트 모르타르의 내화학성)

  • If cement can be manufactured with industrial byproducts such as granulated blast furnace slag(GBFS), phosphogypsum(PG), and waste lime(WL) instead of clinker as its counterproposal, there would be many advantages, including maximum use of these industrial byproducts for high value-added resources, conservation of natural resources and energy by omitting the use of clinker, minimized environmental pollution problems caused by $CO_2$ discharge, and reduction of the production cost. This research investigates the chemical resistance of NSC mortar added PG and WL to GBFS as sulfate and alkali activators. The result of experiment of chemical resistance, showed that NSC is very excellent in acid resistance and seawater resistanc. Such a reasons are that the hydrate like CSH gel and ettringite formed dense pore structure of NSC matrix.

  • This study is to specify the properties of solidification/stabilization of heavy metals in connection with looking over the hydration features of non-sintering cement using industrial by-products. In this study, we added Cr and Pb to non-sintering cement(NSC), ordinary portland cement (OPC), and Blast-furnace slag cement(BSC) to specify the solidification process. Heavy metal leaching test was carried out to evaluate solidification degree of various cement. Follow result, marking no higher than 0.7% of un-solidified ratio of BSC was the most predominant result when we mixed the materials with Cr. 5.8% for NSCI and 6.2% for NSC2. On the contrary, in case of adding Pb, NSCl and NSC2 made better solidification results than those of OPC(below 0.2%) and BSC(below 0.05%), marking nearly 0%.

  • The present study aims to derive optimal interface treatment conditions for emulating a monolithic construction. The joints in this construction are formed through the bonding shear evaluation method during the placement of ultra-high-performance concrete (UHPC) and normal strength concrete (NSC). The evaluation items include push-off tests for homogeneous UHPC + UHPC and heterogeneous NSC + UHPC. The experimental samples comprised a monolithic placement as the baseline, two levels for the separated placement according to the compression strength of concrete, and five levels for the interface treatment. The increase in the number of grooves and their cross-sectional areas only slightly influenced the bonding shear performance. The optimal interface treatment method for the homogeneous UHPC + UHPC construction grooves was at least 30mm. The heterogeneous NSC + UHPC construction should utilize waterjet roughening to expose the aggregate for the increased roughness.

Preparation and Characterization of Durable Softener for Cotton Fiber (면섬유용 내구성유연제의 제조 및 유연특성)

  • Organic acid salt of fatty polyamide (DDDT) and acrylate of fatty carbamide (DDTCA) were synthesized as a main component for the softener. O/W type non-ionic softener (NSC) was prepared by blending DDDT and DDTCA with beef tallow, lanolin anhydride. polyoxyethylene(7) stearyl ether, and polyoxyethylene(50) oleyl ether. After treatment of NSC to all cotton fabrics, the physical properties such as tear strength, crease recovery, and flexing abrasion resistance were measured. As a result of the measurement, NSC was proved to be durable non-ionic softener with good softness.
