The Analgesic Effect and Its Opioidergic Mechanism of Electroacupuncture on Inflammatory Pain in the Type II Collagen-induced Arthritis Rats (전침(電鍼)의 collagen 유발(誘發) 관절염(關節炎)에 대한 진통(鎭痛) 효과(效果) 및 그 기전(機轉)에 관한 연구(硏究) - opioidergic mechanism을 중심으로 -)

  • Objectives : The aim of this study is to evaluate the analgesic effect of electroacupuncture on Jogsamni (ST36) in the collagen-induced arthritis rats and investigate the role played by opioid receptor subtypes $({\mu},\;{\delta},\;{\kappa})$ in the antinociceptive effect of electroacupuncture (EA) In the thermal hyper algesia test. Methods : Immunization of male Sprague-Dawley rats with bovine type H collagen emulsified in incomplete Freund's adjuvant, followed by booster injection 2 weeks later induced collagen-induced arthritis (CIA). The thermal hyperalgesia was evaluated weekly with tail flick latency (TFL). In the fourth week after first immunization, EA stimulation (2 Hz, 0.07 mA, 0.3 ms) was delivered into Jogsamni (5736) for 20 minutes. Analgesic effect was evaluated by using the tail flick latency (TFL) after intraperitoneal injection of normal saline, naloxone, naltrindole and nor-binaltorphimine respectively to CIA rats. Results : The results were as follows; 1. The TFL were gradually decreased in CIA as time elapsed after e immunization of arthrogenic collagen and the maximum value was reached between the third to fifth week. 2. EA stimulation on 5736 inhibited chronic inflammatory pain induced by CIA. 3. The analgesic effect of EA was inhibited by pretreatment of ${\mu}-receptor$ antagonist (naloxone),${\delta}-receptor$ antagonist (naltrindole) and ${\kappa}-receptor$ antagonist (nor-binaltorphimine) respectively. Conclusion : Electroacupuncture has an analgesic effect on the CIA rat and has an antinociception mediated by 8, 5, H receptors.

Fabrication of Organic IC based on Pentacene TFTs on Plastic Substrate (플라스틱 기판에 펜타센 유기박막트랜지스터를 이용한 집적회로 제작)

  • In this article, the organic integrated circuits such as inverter, ring oscillator, NAND and NOR gates, and rectifier have been fabricated on PEN substrate by using pentacene TFTs, The OTFTs used bottom contact structure and produced the average mobility of 0.26 $cm^2/V.sec$ and on/off current ratio of $10^5$. All circuits successfully worked as the simulation results. Especially, the rectifier was able to operate up to 1 MHz input AC signals, and ring oscillator exhibited oscillation frequency of 1MHz at 40 V. Based on the results of organic integrated circuits we could confirm the possibility of the low cost RFID tags and flexible display with OTFTs.

Taxonomy of Yellow koji mold (Aspergillus flavus/oryzae) in Korea

  • Koji molds are comprised of yellow, black and white. Black and white koji molds were recently re-visited by this author and it is concluded that they consists of Aspergillus luchuesnsis, A. niger and A. tubingensis, and the most important species for alcoholic beverage production is A. luchuensis. In the case of yellow koji mold, it is comprised of Aspergillus oryzae, A. sojae and A. tamari. In the case of A. sojae, the species is scarcely isolated from nature and rarely used for industry in Korea. Aspergillus tamari is often isolated from traditional Korean Meju, a fermented soybean product, and the classification of the species is clear. However, in the case of A. oryzae, differentiation between A. oryzae and A. flavus is still in controversy. In this study, we collected 415 strains of Aspergillus flavus/oryzae complex from air, rice straw, soybean, corn, peanut, arable soil and Meju in Korea and we examined the aflatoxin producing capacity of the strains. The norB-cypA, omtA and aflR genes in the aflatoxin biosynthesis gene cluster were analyzed. We found that 367 strains (88.4%) belonged to non-aflatoxigenic group (Type I of norB-cypA, IB-L-B-, IC-AO, or IA-L-B- of omtA, and AO type of aflR), and only 48 strains (11.6%) belonged to aflatoxin-producible group (Type II of norB-cypA, IC-L-B+/B- or IC-L-B+ of omtA, and AF type of aflR). In the case of A. flavus/oryzae strains from Meju, almost strains (178/192, 92.7%) belonged to non-aflatoxigenic group and only 14 strains (7.3 %) belonged to aflatoxin-producible group. It is proposed in this study that non-aflatoxigenic strain from Meju is classified as A. oryzae, considering that Meju is food material.

Characteristics of Metallic and Ionic Concentration in Fine Particle during Haze Days in Busan (부산 지역 연무 발생일의 미세먼지 중 금속과 이온 성분 농도 특성)

  • This research investigates the characteristics of metallic and ionic elements in $PM_{10}$ and $PM_{2.5}$ on haze day and non-haze day in Busan. $PM_{10}$ concentration on haze day and non-haze day were 85.75 and $33.52{\mu}g/m^3$, respectively, and $PM_{2.5}$ on haze day and non-haze day were 68.24 and $23.86{\mu}g/m^3$, respectively. Contribution rate of total inorganic water-soluble ion to $PM_{10}$ mass on haze day and non haze day were 58.2% and 61.5%, respectively, and contribution rate of total water-soluble ion to $PM_{2.5}$ mass on haze day and non haze day were 58.7% and 64.7%, respectively. Also, contribution rate of secondary ion to $PM_{10}$ mass on haze day and non haze day were 52.1% and 47.5%, respectively, and contribution rate of secondary ion to $PM_{2.5}$ mass on haze day and non haze day were 54.4% and 53.6%, respectively. AC (anion equivalents)/CE (cation equivalents) ratio of $PM_{10}$ mass on haze day and non haze day were 1.09 and 1.0, respectively, and AC/CE ratios of $PM_{2.5}$ mass on haze day and non haze day were 1.12 and 1.04, respectively. Also, SOR (Sulfur Oxidation Ratio) of $PM_{10}$ mass on haze day and non haze day were 0.32 and 0.17, respectively, and SOR of $PM_{2.5}$ on haze day and non haze day were 0.30 and 0.15, respectively. Lastly, NOR (Nitrogen Oxidation Ratio) of $PM_{10}$ on haze day and non haze day were 0.17 and 0.08, respectively, and NOR of $PM_{2.5}$ on haze day and non haze day were 0.13 and 0.06, respectively.

Effectiveness of Bioremediation on Oil-Contaminated Sand in Intertidal Zone

  • Bioremediation technologies were applied to experimental microcosms, simulating an oil spill in a lower intertidal area. Three treatments (oil only, oil plus nutrients, and oil plus nutrients and microbial inocula) were applied, and each microcosm was repeatedly filled and eluted with seawater every 12 h to simulate tidal cycles. To minimize washing-out of the inoculum by the tidal cycles, microbial cells were primarily immobilized on diatomaceous earth before they were applied to the oiled sand. Oil degradation was monitored by gravimetric measurements, thin layer chromatography/flame ionization detector (TLC/FID) analysis, and gas chromatography (GC) analysis, and the loss of oil content was normalized to sand mass or nor-hopane. When the data were normalized to sand mass, no consistent differences were detected between nutrient-amended and nutrient/inoculum-amended microcosms, although both differed from the oil-only microcosm in respect of oil removal rate by a factor of 4 to 14. However, the data relative to nor-hopane showed a significant treatment difference between the nutrient-amended and nutrient/inoculum-treated microcosms, especially in the early phase of the treatment. The accelerating effect of inoculum treatment has hardly been reported in studies of oil bioremediation in the Tower intertidal area. The inoculum immobilized on diatomaceous earth seemed to be a very effective formulation for retaining microbial cells in association with the sand. Results of this study also suggest that interpretation of the effectiveness of bioremediation could be dependent on the selection of monitoring methods, and consequently the application of various analytical methods in combination could be a solution to overcome the limitations of oil bioremediation monitoring.

The Effects of Pre-emptive Administration of Ketamine and norBNI on Pain Behavior, c-Fos, and Prodynorphin Protein Expression in the Rat Spinal Cord after Formalin-induced Pain Is Modulated by the DREAM Protein

  • Background: We investigated the effects of pre-emptive administration of ketamine and norBNI on pain behavior and the expression of DREAM, c-Fos, and prodynorphin proteins on the ipsilateral side of the rat spinal cord at 2 and 4 hours after formalin injection. Methods: Eighty-four male Sprague Dawley rats were divided into 4 major groups consisting of control rats (C) (n = 12), rats given only formalin injections (F) (n = 24), and rats treated with pre-emptive administration of either ketamine (K+F) (n = 24) or norBNI (N+F) (n = 24). The non-control groups were further divided into subgroups consisting of rats that were sacrificed at 2 and 4 hours (n = 12 for each group) after formalin injection. Pain behavior was recorded for 1 hour. After 2 and 4 hours, the rats were sacrificed and the spinal cords (L4-L5 sections) were removed for immunohistochemistry and Western blot analysis. Results: The pain behavior response was reduced in the K+F group compared to the other groups during the second phase of the formalin pain response. We detected an increase in the nuclear DREAM protein level in the K+F group at 2 and 4 hours and a transient decrease in the N+F group at 2 hours; however, it increased at 4 hours after injection. Fos-like immunoreactivity (FLI) and Prodynorphin-like immunoreactivity (PLI) neurons decreased in the K+F group but increased in the N+F group at 2 hours after injection. While FLI decreased, PLI increased in all groups at 4 hours after injection. Conclusions: We suggest that NMDA and kappa opioid receptors can modulate DREAM protein expression, which can affect pain behavior and protein transcriptional processes at 2 hours and bring about either harmful or protective effects at 4 hours after formalin injection.

Development of Quantification Method and Stability of ${\Delta}^9$-Tetrahydrocannabinol and 11-Nor-9-carboxy-${\Delta}^9$-Tetrahydrocannabinol in Oral Fluid (타액 중 ${\Delta}^9$-Tetrahydrocannabinol 및 11-Nor-9-carboxy-${\Delta}^9$-Tetrahydrocannabinol의 분석법 확립 및 안정성 검토)

  • Oral fluid has become increasingly popular as an alternative specimen in the field of driving under the influence of drugs (DUID) and work place drug testing. In this study, an analytical method for the detection and quantification of ${\Delta}^9$-tetrahydrocannabinol (THC) and its metabolite, 11-nor-9-carboxy-${\Delta}^9$-tetrahydrocannabinol (THC-COOH) in oral fluid by SPE and GC-MS was established and fully validated. The stability of THC and THC-COOH in oral fluid during storage was also determined by examining the THC and THC-COOH concentration changes depending on time and container materials. Oral fluid samples were kept over 21 days at room temperature, $-4^{\circ}C$ and $-20^{\circ}C$ in two different specimen collection tubes; glass and polypropylene tubes. Three replicates for each condition with different temperature and types of a container were analyzed at five different time points over 21 days. When oral fluid samples were stored in glass tubes, the loss of both THC and THC-COOH was less than 10% at all room temperature, $-4^{\circ}C$ and $-20^{\circ}C$. However, in polypropylene tubes, the loss of both THC and THC-COOH increased significantly over the study period. In particular, the concentration of THC decreased more rapidly than that of THC-COOH at room temperature and the maximal percentage of THC lost was 90.3% after 21 days. The result indicates that it would be necessary to collect oral fluid samples in glass containers and cool the samples until analysis in order to prevent the degradation of analytes.

Study on analytical method of fluoroquinolone residues in eggs by LC/MS/MS (LC/MS/MS를 이용한 식용란 중 fluoroquinolone계 항균물질의 분석법에 관한 연구)

  • An atmospheric pressure chemical ionization (APcI) LC/MS/MS method was developed for the simultaneous analysis of fluoroquinolones (norfloxacin, ciprofloxacin, enrofloxacin, danofloxacin) residues in eggs. The spiked and blank samples were extracted from whole eggs using 50mM phosphate buffer (pH 7.4). The extract was cleaned up by passage though $Oasis^{(R)}$ MAX extraction cartridge for solid-phase extraction followed by elution with 4% formic acid in methanol. The extract of sample was separated on a Waters $Atlantis^{TM}$ $dC_{18}$ reversed-phase column ($4.6{\times}150mm,\;5{\mu}m$) and analyzed by APcI positive mode mass spectrometry. The mobile phase consists of aqueous 0.2% nonafluoropentanoic acid (NFPA) and methanol. Multiple reaction monitoring (MRM) using the precursor to product ion combinations of m/z $320\;{\dashrightarrow}\;302,\;332\;{\dashrightarrow}\;314,\;360\;{\dashrightarrow}\;342$ and m/z $358\;{\dashrightarrow}\;340$ were used to quantify norfloxacin (NOR), ciprofloxacin (CIP), enrofloxacin (ENR) and danofloxacin (DAN), respectively. The limits of quantification (LOQ) were 7.8ppb for NOR, 8.5ppb for CIP, 8.9ppb for ENR, and 4.8ppb for DAN. Average recoveries of fortified sample at levels of 0.025 to 0.1 ppm were estimated 71.29% for NOR, 75.27% for CIP, 85.51% for ENR and 81.22% for DAN. These results could be applied for the confirmation and quantification in eggs.

Anti-proliferative and Apoptotic Effects of Basella rubra (L.) Against 1, 2-Dimethyl Hydrazine-induced Colon Carcinogenesis in Rats

  • Colorectal cancer is a very prevalent diagnosed cancer. The current study was performed in order to examine the role of BRAE (Basella rubra aqueous extract) in regulating aberrant crypt foci (ACF) formation, cell proliferation and inhibition of apoptosis in a colon carcinogenesis model in male Wistar rats. Rats were randomly allocated into six groups. Group I served as control, and group II acted as a drug control administered BRAE (250mg/kg b.w.) orally for 30 weeks. Rats in group III-VI were given subcutaneous injections of DMH (25mg/kg b.w. weekly) for 15 weeks to initiate colon carcinogenesis. Those in group IV and VI were administered BRAE along with DMH injections. Rats in group V were administered with BRAE after cessation of DMH injection. After 30 weeks of experimental period colons were obtained from experimental groups and analyzed for ACF incidence, argyrophilic nucleolar organizing region-associated proteins (AgNOR) count, histopathological and immunohistochemical changes. Only in DMH exposed groups were ACF and AgNOR numbers increased. Administration of BRAE appreciably decreased the numbers of ACF and AgNOR in BRAE treated groups. Histopathological findings revealed a high level of dysplastic changes with decreased number of goblet cells found only in only DMH injected rats. Administration of BRAE in treated group rats reversed these changes. Expression markers for cell proliferation (PCNA and Ki67) were elevated in DMH treated rats, but reduced with BRAE treatement. This expression was reversed with apoptosis markers (p53 and Caspase-3). Thus the results results of the present study were found to be significant and confirmed the potential efficacy of BRAE against colon carcinogenesis.

Survival Rate of Breast Cancer Patients In Malaysia: A Population-based Study

  • Breast cancer is the most common cancer among Malaysian women. Other than hospital-based results, there are no documented population-based survival rates of Malaysian women for breast cancers. This populationbased retrospective cohort study was therefore conducted. Data were obtained from Health Informatics Centre, Ministry of Health Malaysia, National Cancer Registry and National Registration Department for the period from $1^{st}$ January 2000 to $31^{st}$ December 2005. Cases were captured by ICD-10 and linked to death certificates to identify the status. Only complete data were analysed. Survival time was calculated from the estimated date of diagnosis to the date of death or date of loss to follow-up. Observed survival rates were estimated by Kaplan-Meier method using SPSS Statistical Software version 17. A total of 10,230 complete data sets were analysed. The mean age at diagnosis was 50.6 years old. The overall 5-year survival rate was 49% with median survival time of 68.1 months. Indian women had a higher survival rate of 54% compared to Chinese women (49%) and Malays (45%). The overall 5-year survival rate of breast cancer patient among Malaysian women was still low for the cohort of 2000 to 2005 as compared to survival rates in developed nations. Therefore, it is necessary to enhance the strategies for early detection and intervention.
