Effect of Kangdangboeumbang on the Anti-diabetic Activity in NOD Mice (강당보음방이 자가면역 당뇨모델인 NOD mice의 혈당강하에 미치는 영향)

  • This study was aimed to verify the anti-diabetic activity of Kangdangboeumbang(KBB) in NOD mice which is Insulin Dependent Diabetes Mellitus(IDDM). The reduction of blood glucose after oral administration between 14 weeks by 2 weeks period to a NOD mice in KBB extract treatment group was showed from 7 day after comparing with control group. KBB extract treatment group increasd insulin secretion amount of serum than control group and decreased IFN­γ production. The pancreatic β-cells is destroyed by Th1-dependent autoimmune disease in NOD mice. KBB extract treatment group intercepted the progress of edematous islet controlling inflammatory mononuclear cells of infiltration that also destruction of pancreatic β-cells electively in a NOD mice.

  • 현재 인터넷을 통한 뉴스의 서비스는 크게 전자신문의 형태와 NOD(News on Demand)의 형태가 있다. 전자신문은 사용자가 원하는 정보를 스스로 브라우징을 해야하므로 번거롭지만 NOD는 사용자가 원하는 정보를 자동으로 분배해 준다. 향후 네트워크의 속도가 향상이 되면 멀티미디어 형태의 NOD 뉴스가 일반적인 현상을 차지할 것이다. 본 논문에서는 이러한 추세에 대비하여 부산일보사의 CTS(Computerized Typesetting System)를 기반으로한 인터넷 전자신문의 구조와 향후 일반 사용자에게 서비스될 NODtltmxpa의 구조를 설명한다.

Nucleotide-binding oligomerization domain protein 2 attenuates ER stress-induced cell death in vascular smooth muscle cells

  • Nucleotide-binding oligomerization domain protein 2 (NOD2), an intracellular pattern recognition receptor, plays important roles in inflammation and cell death. Previously, we have shown that NOD2 is expressed in vascular smooth muscle cells (VSMCs) and that NOD2 deficiency promotes VSMC proliferation, migration, and neointimal formation after vascular injury. However, its role in endoplasmic reticulum (ER) stress-induced cell death in VSMCs remains unclear. Thus, the objective of this study was to evaluate ER stress-induced viability of mouse primary VSMCs. NOD2 deficiency increased ER stress-induced cell death and expression levels of apoptosis mediators (cleaved caspase-3, Bax, and Bak) in VSMCs in the presence of tunicamycin (TM), an ER stress inducer. In contrast, ER stress-induced cell death and expression levels of apoptosis mediators (cleaved caspase-3, Bax, and Bak) were decreased in NOD2-overexpressed VSMCs. We found that the $IRE-1{\alpha}-XBP1$ pathway, one of unfolded protein response branches, was decreased in NOD2-deficient VSMCs and reversed in NOD2-overexpressed VSMCs in the presence of TM. Furthermore, NOD2 deficiency reduced the expression of XBP1 target genes such as GRP78, PDI-1, and Herpud1, thus improving cell survival. Taken together, these data suggest that the induction of ER stress through NOD2 expression can protect against TM-induced cell death in VSMCs. These results may contribute to a new paradigm in vascular homeostasis.

The Physical Interaction between Nucleotide-Binding Oligomerization Domain Containing 2 and Leucine-Rich Repeat Kinase 2

  • Recently, decades of robust researches on degenerative brain disorder have been highlighted on the interactive connection of gut and brain. In terms of inflammatory cytokine production, others have shown that Nucleotide-Binding Oligomerization Domain Containing 2 (NOD2) is involved with Leucine-Rich Repeat Kinase 2 (LRRK2). HEK293T cells were transiently co-transfected with Myc-tagged LRRK2 and Flag-tagged NOD2 and then followed by co-immunoprecipitation assay. In this study, we provide the novel finding of physical protein-protein interaction between NOD2 and LRRK2. G2019S variant has shown stronger interactions against NOD2 than those of wild type LRRK2. In an axis of NOD2-LRRK2 communication, it is believed to pave a new way in the understanding of the bidirectional molecular mechanism of brain disorder, including Parkinson's disease into gut inflammatory disease, including Crohn's disease.

Expression of c-Jun in pancreatic islet $\alpha$-cells of nonobese diabetic(NOD) mice

  • This is the first report of c-Jun protein expression and mRNA in a pancreatic islet in a nonobese diabetic(NOD) state mice. In this experiment NOD mice with insulin-dependent diabetes mellitus type I at age 16 weeks(n=7) just before death(n=4) were used. The control group consist of prediabetic NOD(8 weeks n=7) and ICR(8 weeks n=7 and 16 weeks n=7) mice. c-Jun positive cells in the pancreatic islet of NOD mice were localized in the same positions as a-glucagon producing cells. immunoreactivity was negative in the prediabetic NOD(8 weeks) and ICR(8 weeks and 16 weeks) mice. The number of c-Jun positive cells in mice with severe diabetic state just before death were significantly decreased when compared to NOD(16 weeks) mice. Expression of c-Jun in mRNA level was assessed by RT-PCR method. The levels of mRNA in NOD(16 weeks) mice group were elevated in total pancreatic tissues. The present results suggest that the induction of proto-oncogene protein may be of significance in assessing cell specific injury and may play a functional role between pancretic islet $\alpha$-cells and $\beta$-cells in the diabetic state.

The Fractionation Characteristics of BOD in Streams (하천에서 BOD 존재형태별 분포 특성)

  • In this study, the distribution characteristics according to the type of BOD and the effect of nitrogenous oxygen demand (NOD) and algal oxygen demand on BOD in three streams (Bokhacheon, Byeongseongcheon, and Gulpocheon) were evaluated. Although the BOD and NOD concentrations demonstrated a difference in the three streams, the carbonaceous BOD(CBOD)/BOD ratio was 0.75 (p=0.053, one-way ANOVA), and there was no significant difference in the three streams (r2≥0.92, p<0.0001). The NOD concentration of the Bokhacheon with high NH3-N was 1.7±1.3 mg/L, which was the highest among the three streams and showed a significant correlation with BOD. Seasonal variations in NOD in the three streams did not show a significant correlation with changes in NH3-N concentration (r2<0.28, p≥0.1789), and there was no significant difference in NOD even though NH3-N concentration in Gulpocheon was about twice that of Byeongseongcheon (p=0.870, one way ANOVA). The particulate CBOD(PCBOD)/CBOD ratio of the three streams was 0.55~0.64, and about 60% of the biodegradable organic matter was present in the particulate form. When the Chl.a concentration in the stream was more than 7 ㎍/L, the PCBOD tended to increase with the Chl.a concentration (r2=0.61, p=0.003). In the three streams, particulate NOD accounted for 81% of NOD; however, despite the large variation in NH3-N concentration (0.075~3.182 mg/L), there was no significant difference in soluble NOD(SNOD) concentration that ranged from 0.1 to 0.3 mg/L. In this study, the low contribution rate of SNOD to NOD is considered as a result of the removal of nitrifying bacteria along with the particles during the filtration process.

Distribution of Organic Matter and Nitrogenous Oxygen Demand in Effluent of Sewage and Wastewater Treatment Plants (하·폐수처리시설 방류수내 유기물질 및 NOD 분포 특성)

  • In this study, an analysis of the characteristics of organic matter and nitrogenous oxygen demand (NOD) of 17 sewage effluent and wastewater treatments was conducted. High CODMn and carbonaceous biological oxygen demand (CBOD) concentrations were observed in the livestock treatment plants (LTP), wastewater treatment plants(WTP), and night soil treatment plants (NTP), but the highest NOD concentration and contribution rates of NOD to BOD5 were found in sewage treatment plants (STP). There was no significant difference in the CBOD/CODMn ratio for each of the six pollution source groups, but the LTPs, WTPs, and NTPs all showed relatively high CODMn concentrations in their effluent samples, indicating that they are facilities which discharge large amounts of refractory organic matter. The seasonal change of NOD in all facilities' effluent was found to be larger than the seasonal change of CBOD, and data results also revealed an elevation of NOD and NH3-N concentration from December to February, when the water temperature was low. There was no significant difference in NH3-N concentration in relation to pollution source group (p=0.08, one-way ANOVA), but the STP, which had a high NOD contribution rate to BOD5 of 48%, showed a high correlation between BOD5 and NOD (r2=0.95, p<0.0001). These results suggest that the effect of NOD on BOD5 is an important factor to be considered when analyzing STP effluent.

The Genetic Variations of NOD2 Are Associated With White Blood Cell Counts

  • The cytoplasmic elicitor, nucleotide-binding domain and leucine-rich repeat containing domain receptors (NLRs) is well established molecules in its role in inflammatory response. Among 22 NLR receptors, NOD2 is one of the intensively studied genes of elucidating for the inflammatory bowel disease and Crohn's disease as well. Recent research have accumulated that common genetic mutations in Parkinson's disease (PD) are increasingly related to the susceptibility to Crohn's disease. In this study, with the Korean Genome and Epidemiology Study, we aimed to perform the association between NOD2 polymorphisms and blood cell counts [WBC (white blood cell) count, RBC (red blood cell) count, platelet count], which linked supposedly to cytoplasmic inflammatory responses with clinical specialty. Linear regression analyses were performed, controlling for residential area, sex, and age as covariates. As a results, 12 SNPs from NOD2 gene were significantly associated with WBC counts (Bonferroni correction P-value criteria < 0.05/23=0.00218). In this study, we could ensure an association with NOD2 gene and WBC counts. This is the first report to have relationship between SNPs of NOD2 gene and WBC counts.

Differentiation and Proliferation of Porcine T Lymphocytes in NOD/SCID Mice (NOD/SCID 모델 마우스 생체 내 돼지 T 면역세포의 증식 및 분화)

  • The nonobese diabetic / severe combined immune deficiency (NOD/SCID) has been used for determination of proliferation and differentiation of hematopoietic stem cells as xenotransplantation animal model. In this study, we transplanted porcine hematopoietic cells from bone marrow into NOD/SCID mice via intravenous injection to confirm the activity of differentiation and proliferation for porcine hematopoietic cells in vivo. Interestingly, we observed the result of high efficiency with pig T lymphocytes in hematopoietic organs, liver, spleen lymph node, and bone marrow in NOD/SCID mice. The porcine $CD3^{+}$ T cells were detected with $5.4{\pm}1.9%$ in bone marrow, $15.4{\pm}7.3%$ in spleen, $21.3{\pm}1.4%$ in liver, and $33.5{\pm}32.8%$ in lymph node of NOD/SCID mice at 6 weeks after trans-plantation Furthermore, immunohistochemical analysis showed the high engraftment of porcine T lymphocytes in spleen of NOD/SCID mice. Our data suggest that NOD/SCID mice are excellent animal model to determinate the generation md function of pig T lymphocytes.

Microcystins and Nodularin in Agricultural Products: Toxicity, Analytical Methods, Contamination Pathway, Occurrence, and Safety Management (농산물 내 마이크로시스틴과 노둘라린: 독성, 분석법, 오염 경로, 오염 현황 및 관리 동향)

  • The peptide-type hepatotoxins microcystins (MCs) and nodularin (NOD) are secondary metabolites produced by cyanobacteria. MCs and NOD can bioaccumulate in agricultural products through toxin-contaminated water, soil, and manure and can cause human health risks through the consumption of agricultural products. As interest in the contamination of agricultural products by MCs or NOD has recently emerged, occurrence studies based on various analysis methods for agricultural products have been conducted. However, studies on agricultural products are still insufficient compared to research on drinking water and seafood. In addition, research is primarily conducted on agricultural products grown in areas where green algae occur, but not on marketed products. In the present study, we review the physicochemical properties, toxicity, analysis methods, occurrence studies, and management status of MCs and NOD in agricultural products to build a foundation for systematic monitoring and safety management.
