The Effects of the Next-Generation System in the Banking Industry on the Simplification of Business Processes and the Development of New Products (금융기관의 차세대시스템이 금융상품 개발 및 비즈니스 프로세스 단순화에 미친 효과: 기업은행 사례를 중심으로)

  • Since the IMF crisis, the competition among banks in Korea has fiercely intensified. To survive in the severe competition, several leading banks including Industrial Bank of Korea (IBK) have recently developed their own proprietary systems called 'the Next-Generation Systems(NGS).' This case study was to answer several important issues as follows; 1); the reason why a bank make huge investment in the IT infrastructure; 2) the development methodology of the NGS; 3) the new architecture of the NGS; 4) the governance structure of the NGS investment; and 5) the effects of the NGS on the development of new products. To achieve such goals, a systematic tandem interview method and many archival reports from IBK were utilized. The conclusions are summarized as follows: First, the NGS has contributed to the integration and simplification of business processes. Second, new products as demanded in the market can be developed much faster through the NGS. Third, the change management of IT infrastructure is critically important. Fourth, the systems architecture can be different according to the IT governance. Fifth, the alignment of the NGS and business processes is most important to obtain the organizational goals.

A Comparison of Mathematically Gifted and Non-gifted Elementary Fifth Grade Students Based on Probability Judgments (초등학교 5학년 수학영재와 일반아의 확률판단 비교)

  • The purpose of this study was to discover differences between mathematically gifted students (MGS) and non-gifted students (NGS) when making probability judgments. For this purpose, the following research questions were selected: 1. How do MGS differ from NGS when making probability judgments(answer correctness, answer confidence)? 2. When tackling probability problems, what effect do differences in probability judgment factors have? To solve these research questions, this study employed a survey and interview type investigation. A probability test program was developed to investigate the first research question, and the second research question was addressed by interviews regarding the Program. Analysis of collected data revealed the following results. First, both MGS and NGS justified their answers using six probability judgment factors: mathematical knowledge, use of logical reasoning, experience, phenomenon of chance, intuition, and problem understanding ability. Second, MGS produced more correct answers than NGS, and MGS also had higher confidence that answers were right. Third, in case of MGS, mathematical knowledge and logical reasoning usage were the main factors of probability judgment, but the main factors for NGS were use of logical reasoning, phenomenon of chance and intuition. From findings the following conclusions were obtained. First, MGS employ different factors from NGS when making probability judgments. This suggests that MGS may be more intellectual than NGS, because MGS could easily adopt probability subject matter, something not learnt until later in school, into their mathematical schemata. Second, probability learning could be taught earlier than the current elementary curriculum requires. Lastly, NGS need reassurance from educators that they can understand and accumulate mathematical reasoning.

Quality Changes and Shelf-life of Grill Sausages Re-pasteurized after Packaging during Chilled Storage (이차살균한 그릴소시지의 냉장 저장 중 품질 변화와 저장 수명)

  • Changes in quality characteristics and shelf-life of two types of grill sausage manufactured by using pork hind leg and re-pasteurized after packaging were investigated during storage at 5$^{\circ}C$ and 10$^{\circ}C$ up to 30 days. The total aerobic plate counts(APC) of N rnberger style grill sausage(NGS) and vegetable added grill sausage(VGS) were initially 3.08 and 3.57 log$\sub$10/CFU/$\textrm{cm}^2$, respectively, and gradually increased during storage time. After 30 days, the APC of NGS stored at 5$^{\circ}C$ and 10$^{\circ}C$ maintained relatively low at the levels of 5.32 and 6.58 log$\sub$10/CFU/$\textrm{cm}^2$, respectively. However, the APC of VGS exceeded the level of 8 log$\sub$10/CFU/$\textrm{cm}^2$ after 30 days at 5$^{\circ}C$ and after 20 days at 10$^{\circ}C$. pH was decreased with storage period, however it tended to increase after 30 days. The values of TBA and VBN for both sausages tended to increase with storage time, and this trend was pronounced at 5$^{\circ}C$ rather than at 10$^{\circ}C$. The purge loss was apparently increased in the VGS compared to the NGS sample as the storage period extended. According to the sensory evaluation, the marketing values of grill sausages stored at 5$^{\circ}C$ and 10$^{\circ}C$ were maintained until day 20 for NGS and day 14 for VGS, respectively.

Analyzing Vomit of Platalea minor (Black-faced Spoonbill) to Identify Food Components using Next-Generation Sequencing and Microscopy (차세대염기서열 및 현미경 분석을 통한 저어새의 토사물 내 먹이생물 분석)

  • We sampled vomit of black-faced spoonbills(Platalea minor) during the brood-rearing season (from June 2011 to June 2014) at the Namdong reservoir in Incheon and analyzed the food components in the vomit using microscopy and next-generation sequencing (NGS). Microscopic observations primarily helped in identifying osteichthyes (bony fishes), crustaceans, and polychaetes. In particular, species belonging to the families Mugilidae and Gobiidae among the fish, and Macrophthalmus japonicas among the crustaceans, were observed at high frequency. Results of NGS analysis revealed the predominant presence of bony fish (42.58% of total reads) and crustaceans (40.75% of total reads), whereas others, such as polychaetes (12.66%), insects (0.24%), and unidentified species (3.78%), occurred in lower proportions. At the species level, results of NGS analysis revealed that Macrophthalmus abbreviates and Macrobrachium sp. among the crustaceans, and Acanthogobius hasta, Tridentiger obscurus, and Pterogobius zacalles among the bony fish, made up a high proportion of the total reads. These food species are frequently found at tidal flats in the Songdo and Sihwa lakes, emphasizing the importance of these areas as potential feeding sites of the black-faced spoonbill. Feed composition of the black-faced spoonbill, as evaluated by analyzing its vomit, differed when the evaluations were done by microscopic observation or by NGS analysis. Evaluation by microscopic observation is difficult and not error free, owing to the degradation of the samples to be analyzed; however, NGS analysis is more accurate, because it makes use of genetic information. Therefore, accurately analyzing food components from morphologically indistinguishable samples is possible by using genetic analysis.

Effect of Water Temperature on the Expression of Stress Related Genes in Atlantic Salmon (Salmo salar) Fry (수온이 대서양 연어(Salmo salar) 치어의 체내 스트레스 관련 유전자 발현에 미치는 영향)

  • The warming of water as a result of climate change affects fish habitat. Variations in water temperature affect fish physiology almost totally. The rise in water temperature due to climate change leads to hypoxia following decreased oxygen solubility and decreased binding capacity of oxygen-carrying hemoglobin. This study was conducted to evaluate the health status of Atlantic salmon (Salmo salar) fry at elevated water temperatures($20^{\circ}C$) compared with optimum water temperature ($15^{\circ}C$). The method facilitated the detection of biomarker genes using NGS RNAseq analysis and evaluation of their expression pattern using RT-qPCR analysis. The biomarker genes included interferon alpha-inducible protein 27-like protein 2A transcript variant X3, protein L-Myc-1b-like, placenta growth factor-like transcript variant X1, fibroblast growth factor receptor-like 1 transcript variant X1, transferrin, intelectin, thioredoxin-like, c-type lectin lectoxin-Thr1-like, ladderlectin-like and calponin-1. The selected biomarker genes were sensitive to changes in water temperature based on NGS RNAseq analysis. The expression patterns of these genes based on RT-qPCR were similar to those of NGS RNAseq analysis.

  • Hexaploid wheat (common wheat/bread wheat) is one of the most important cereal crops in the world and a model for research of an allopolyploid plant with a large, highly repetitive genome. In the heritability of agronomic traits, variation in gene presence/absence plays an important role. However, there have been relatively few studies on the variation in gene presence/absence in crop species, including common wheat. Recently, a reference genome sequence of common wheat has been fully annotated and published. In addition, advanced next-generation sequencing (NGS) technology provides high quality genome sequences with continually decreasing NGS prices, thereby dawning full-scale wheat functional genomic studies in other crops as well as common wheat, in spite of their large and complex genomes. In this review, we provide information about the available tools and methodologies for wheat functional genomics research supported by NGS technology. The use of the NGS and functional genomics technology is expected to be a powerful strategy to select elite lines for a number of germplasms.

  • Highly pathogenic avian influenza (HPAI) viruses have caused severe respiratory disease and death in poultry and human beings. Although most of the avian influenza viruses (AIVs) are of low pathogenicity and cause mild infections in birds, some subtypes including hemagglutinin H5 and H7 subtype cause HPAI. Therefore, sensitive and accurate subtyping of AIV is important to prepare and prevent for the spread of HPAI. Next-generation sequencing (NGS) can analyze the full-length sequence information of entire AIV genome at once, so this technology is becoming a more common in detecting AIVs and predicting subtypes. However, an analysis pipeline of NGS-based AIV sequencing data, including AIV subtyping, has not yet been established. Here, in order to support the pre-processing of NGS data and its interpretation, we developed a user-friendly tool, named prediction of avian influenza virus subtype (PAIVS). PAIVS has multiple functions that support the pre-processing of NGS data, reference-guided AIV subtyping, de novo assembly, variant calling and identifying the closest full-length sequences by BLAST, and provide the graphical summary to the end users.

  • 최근에 등장한 Next Generation Sequencing(NGS)은 전통적인 방법에 비해 빠르고 저비용으로 대용량의 시퀀스 데이터를 이용한 차세대 시퀀싱 기술을 말한다. 이렇게 얻은 NGS 데이터를 분석하는 단계 중에서 alignment 단계는 시퀀서에서 얻은 대량의 read를 참조 염기서열에 맵핑하는 단계로 NGS 데이터 분석의 가장 기본이면서 핵심인 단계이다. alignment 도구는 긴 참조 염기서열을 색인화해서 짧은 read를 빠르게 맵핑하는 용도로 사용된다. 현재 많이 사용되고 있는 일반적인 alignment 도구들은 입력데이터에 대한 별도의 전처리 과정이 없으며 나열된 read를 순차적으로 맵핑하는 단순한 구조를 가지고 있다. 본 논문은 NGS 데이터의 특징 중에 특히 read간의 중복성이 존재하고 이를 이용한 read의 효율적 공통부분 서열을 찾는다. 중복이 가능한 read의 공통부분서열과 read의 관계를 그래프 이론의 Hitting Set 문제로 모델링하고 여러 read가 포함하는 공통 부분서열을 사용해서 alignment 단계의 효율을 높일 수 방법을 제안한다.

Analysis of Microbial Composition Associated with Freshwater and Seawater

  • Knowledge of the distribution and biodiversity of environmental bacteria and the ecosystem that influences them is crucial for predicting an ecosystem. However, bacterial culture methods can only analyze approximately 0.1% of the existing microorganisms, those that are readily cultured under laboratory conditions. By contrast, next-generation sequencing (NGS) has generally been known to obtain more diverse profiling of bacterial composition. We compared the bacterial communities using both a culture-dependent (MALDI-TOF) and culture-independent (NGS) methods. Environmental specimens were obtained from both freshwater and seawater. Water samples were also analyzed by both pyrosequencing and MiSeq sequencing, in order to select one NGS platform which could analyze comparatively more diverse microbiota. Bacterial distribution analyzed with MALDI-TOF showed no difference between the microbiota of freshwater and seawater, whereas the results analyzed with NGS distinguished between the two. The diversity indexes of MiSeq sequencing were higher than for Pyrosequencing. This indicated that MiSeq sequencing is capable of analyzing a comparatively wider diversity of bacteria. The genus of Flavobacterium and Planktophila were identified as being unique to freshwater, whereas EU801223 and OM43 were found in the seawater. Difference between the bacterial composition of the freshwater and seawater environments was identified by MiSeq sequencing analysis.

Whole genome sequencing of foot-and-mouth disease virus using benchtop next generation sequencing (NGS) system

  • In countries with FMD vaccination, as in Korea, typical clinical signs do not appear, and even in FMD positive cases, it is difficult to isolate the FMDV or obtain whole genome sequence. To overcome this problem, more rapid and simple NGS system is required to control FMD in Korea. FMDV (O/Boeun/ SKR/2017) RNA was extracted and sequenced using Ion Torrent's bench-top sequencer with amplicon panel with optimized bioinformatics pipelines. The whole genome sequencing of raw data generated data of 1,839,864 (mean read length 283 bp) reads comprising a total of 521,641,058 (≥Q20 475,327,721). Compared with FMDV (GenBank accession No. MG983730), the FMDV sequences in this study showed 99.83% nucleotide identity. Further study is needed to identify these differences. In this study, fast and robust methods for benchtop next generation sequencing (NGS) system was developed for analysis of Foot-and-mouth disease virus (FMDV) whole genome sequences.
