Influence of Supplemental Dietary Yeast Culture on the Noxious Gas Emission in Broiler Houses and Performance of Broiler Chicks (효모배양물 첨가 사료가 계사내 유해가스 발생 및 육계의 생산성에 미치는 영향)

  • Two experiments were conducted to investigate the effect of dietary supplementation of yeast culture on the performance of broiler chicks and noxious gas emission in broiler houses. Two hundred forty and three hundred and twenty, one day old Cobb male broiler chicks in Expt 1 and Expt 2, respectively were alloted to four treatment levels of yeast culture (0, 0.1, 0.2, 0.4%). To each treatment, 60 birds were assigned in Expt 1 and 5 replicates of 16 birds each were assigned in Expt 2. Basal diets contained 21.5% and 19.0% CP, and 3,100kcal/kg ME for the starting and finishing periods, respectively. Ammonia and $CO_2$ gas emission were detected twice a day for seven days during the five week period of Expt 1. Weight gain, feed intake and feed efficiency were measured for five weeks in Expt 2. Intestinal microbes, blood cholesterol and ND antibody titer were examined at the end of Expt 2. In Expt 1, the concentration of $NH_3$ in the house of birds fed yeast culture tended to be lower than the control. It was significantly lower in the 0.4% yeast culture treatment than the control (P<0.05). $CO_2$ concentration was significantly lower in all yeast culture treatments regardless of its dietary supplemental level than the control (P<0.05). different from others. Feed efficiency (feed/gain), however, was significantly improved in all yeast culture treatments relative to that of the control for starting period (P<0.05) and that of 0.2% yeast culture treatment was significantly lower than those of the control and 0.4% for the overall period. Total number of E. coli in the ileum of birds fed yeast culture at 0.1 and 0.2% was significantly lower than those of the control and 0.4% in the ileum. The CFU of Lactobacillus spp. of birds fed yeast culture at 0.1% was higher in the cecum compared to other treatments (P<0.05). Total cholesterol level of chicks fed 0.1% yeast culture seemed to be lower compared to that of other treatments, whereas LDL-cholesterol level was significantly lower than those of the control and 0.4% treatment. ND antibody titer tended to be higher in the yeast culture treatments than the control, but was not significantly different. The results of these experiments indicated that 0.2% yeast culture may have a potential to reduce the noxious gas emission in broiler houses and maximize the performance of broiler chicks.

Immunization of broiler chicks deprived food and water with live Newcastle disease vaccine(LaSota strain) by drinking water

  • To confirm the effect of food and water deprivation prior to Newcastle disease(ND) virus vaccination, three hundred chicks were divided into five groups with three replications. ND vaccine were sprayed to at 1 -day old chicks at commercial hatchery. Secondary and third vaccination was conducted at 2-week old and 24-day old chicks by LaSota strain. Control was conventionally vaccinated without withdrawing the food and water before or after vaccination. In group 2(G2) and 3(G3), LaSota strain was vaccinated to chicks before and after fasting the food and water for 3 and 2 hours, respectively. Group 4(G4) has the same fasting time of group 2, but supplemented the skim milk in vaccin dilution water. In group 5(G5), skim milk was added into group 3. Weight gain, feed intake and feed conversion were weekly measured for 5 weeks. Blood was collected from wing vein at 24 and 35 days of age. Each serum antibody level were measured by hemagglutination inhibition(HI) test. The average weight gain, feed intake, feed conversion of all group were not significantly different. Weight gain of each groups was 1910.30(control), 1875.28(G2), 1952.12(G4) and 1896.05(G5), respectively. Feed intake of all group was recorded at 3160.67(control), 3167.07(G2), 3189.48(G3), 3157.85(G4) and 3178.16(G5), respectively. The feed conversion of each groups was 1.655(control), 1.688(G2), 1.633(G3), 1.699(G4) and 1.676(G5), respectively. The HI titer of G4 was $ 5.50{\Pm}$1.40 and significantly higher than the other groups (p<0.05)(control : $4.36{\Pm}$1.87 , G2 : $5.18{\Pm}$2.14, G3 : $4.51{\Pm}$2.19, G : $5.28{\Pm}$1.58 at 35 days old. The results of this experiment indicated that two or three hours of fasting time before or after vaccination would be able to show the higher antibody level against ND virus.

  • These experiments were undertaken as a basic study to develop immunocontraceptive vaccine and to understand the role of zona pellucidae in early fertilization process by identifying the monoclonal and polyclonal antibody to porcine zona pellucidae and polyclonal antibody to mouse zona pellucidae by indirect ELISA and indirect immunofluorescence test. The results obtained in these experiments were summarized as follows : 1. The titer of the antibodies to zona was determined by indirect ELISA using solubilized porcine zona coated plates. Both monoclonal and rabbit polyclonal antibodies showed very high titers ; O.D at 1 : 12,800 dilution of antibodies was still significantly higher than that of non-immunized control serum. Rabbit anti-mouse zona pellucidae sera also reacted with porcine zona pellucidae. 2. By indirect immunofluorescence test strong fluorescences were observed on the egg treated with homologous and heterologous rabbit polyclonal antibodies and FITC lablled 2nd antibodies and found to crossreact strongly with the eggs from the pig and mouse. While weaken fluorescences were observed on the eggs treated with monoclonal antibodies.

Serological Survey of Major Avian Viral Diseases Related with Egg Production in Commercial Chicken Flocks in Korea

  • While use of mass rearing systems improved poultry production, chances of exposing to contagious diseases have been increased, making flocks more vulnerable to diseases. Diseases of interest which affects egg production adversely include Low pathogenic avian influenza (LPAI), Infectious bronchitis (IB), Avian meta-pneumoviral infection (aMPV) and Egg drop syndrome'76 (EDS'76). This report collected and analyzed 5,385 serum samples, which were collected from 1,330 different chicken flock, provided by Chungbuk National University, Avian Disease Laboratory at 2009. Serums were analyzed based on rearing stages; 0~1.3weeks (wks) (maternal antibody period), >1.3~3 wks (starting period), >3~10 wks (growing period), >10~22 wks (developing period), >22~40 wks (peak laying period), >40~60 wks (late laying period) and over 60 wks (post-molting period). Results showed the 99.7% of the tested flocks were immunized against ND and73.8%, 97.1%, 78,2% and 78% of the flocks were immunized against other 4 agents (LPAI, IB, EDS'76, aMPV). Maternal antibody was transferred to enough quantity for NDV. Generally, antibody titers which were developed at 22 weeks were stabilized permanently for life. In case of IB and aMPV, infection titer emerged as early as 10 weeks and the titer was increased from 99.4% to 100% for life. EDS76 showed increase in titers, reflecting decreased frequency of vaccination programs. Overall, this study displayed general trends of major viral disease in layers, but considering the trend of development of preventive measures and evolution of pathogens, conducting serological surveys on a regular basis is important.

Serological Survey for the Major Viral Diseases in the Layers (국내 산란계의 주요 바이러스성 질병에 대한 혈청학적 모니터링 결과 및 분석)

  • Serological evaluation for the poultry is important for various reasons, such as designing and assessing the vaccination program and diagnosing diseases and for this reason, serologic tests for the layer flocks have been conducted on a regular basis. Moreover, the nationwide serological survey and analysis are essential to understand the epidemiological status of national poultry industry. In this sense, the study was conducted to evaluate the immune status of the layer flocks with the sera submitted to Avian Disease Laboratory, Chungbuk National University in 2009, and several important viral diseases were selected for evaluation including low pathogenic avian influenza (LPAI), Newcastle disease (ND), infectious bronchitis (IB) and avian metapneumovirus (aMPV). For LPAI and ND, the age-related patterns of geometric mean titer (GMT) changes were similar but there were differences in the flock positive rate and the level of GMT due to the different vaccination policy. In the case of IB, the values of GMT showed that the field infection was more prevalent than expected. For aMPV, positive birds in a flock increased as the layers got older, which reflected the course of field infection because vaccination against aMPV was not allowed in 2009. From this study, the immune status for the main viral diseases in layers became more clarified but this information was limited because of only one year study. Therefore, serological survey needs to be conducted on a yearly basis and furthermore include broilers and breeders for a better understanding of the health status in the national poultry industry.

Histopathological observation on the piglets experimentally infected with Aujeszky's disease virus isolated in Korea (국내분리(國內分離) Aujeszky's disease virus의 실험적(實驗的) 감염자돈(感染仔豚)의 병리조직학적(病理組織學的) 관찰(觀察))

  • Thirty-day-old piglets were intranasally or subcutaneously inoculated with 2ml of Aujeszky's disease virus, NYJ-1 strain, at the titer of $10^{6.75}$ $TCID_{50}/0.1ml$, that was isolated from the diseased piglets in Korea, and histopathological studies were performed to elucidate the pathognomonic characters of the isolate. Results obtained through the experiments were as follows: 1. Major clinical signs on the 2nd and 3rd days post inoculation (p.i.) were fever, anorexia and dyspnea. On the 6th and 7th days p.i., nervous signs, severe dyspnea and salivation were observed in the group of intranasal inoculation, and one out of 3 piglets in this group died on the 7th day p.i.. General signs were more severe in the group of intranasal inoculation than the group of subcntaneous injection. Between the 8th and l0th days p.i., the signs subsided and the piglets were completely recovered from the illness. 2. Hematologically, most of the inoculated pigs showed a mild lymphocytopenia on the 5th and 6th days p.i.. 3. By necropsy, swelling and hemorrhagic lesions were observed in tonsil, central nervous system and lung. No specific changes were grossly found in other parenchymatous organs. 4. In histopathological study, degeneration and necrosis of nervous cells, non-suppurative meningoencephalitis, diffuse or focal gliosis, perivascular cutting and degeneration of ganglion cells were observed in central nervous system, and swelling and hemorrhagic changes were shown in the tissues of liver, lung and lymph nodes. 5. By indirect immunofluorescence antibody assay using ADV-monoclonal antibody, specific ADV antigens were detected in the tissues of tonsil, brain and spleen of the succumbed piglet. However, in the experimentally slaughtered piglets, the specific reactions were noted only in the tonsils.

Influence of Dietary Supplemental Bio-Plus on Performance and Intestinal Microflora of Broiler Chicks (바이오 플러스의 급여가 육계의 생산성과 장내 미생물에 미치는 영향)

  • Two experiments were conducted to evaluate the dietary supplemental fruit extract(Bio-plus) on performance and intestinal microflora of broiler chicks. Seven treatments were allocated into control, 0.05, 0.10, 0.20, 0.10, 0.20, 0.40, 0.80, 1.60% with four replications for five weeks, in both experiments. Diets contained 21.5% CP, 3, 100 kcal/kg ME for the rest two weeks. Weight gain, feed intake, feed conversion ratio(FCR) were weekly measured. Birds were sacrificed to examine the E. coli and Salmonella from cecum and ileum at the end of experiment. In experiment 1, Weight gain of Bio plus treatments tended to increase compared to that of control, but was not consistency between them. Chicks fed Bio-plus showed significantly lower feed conversion ratio(FCR) than control(P〈0.05). In experiment 2, Weight gain of Bio plus treatments was significantly different from control(P〈0.05) for starting period and linearly increased upto 0.80% for the rest of two weeks, but was not significantly different. Feed intake of chicks fed Bio-plus tended to be higher than control. Bio-plus treatments was not statistically different FCR from control. ND titer showed no significance, but tended to be higher in Bio-plus treatments than control. The number of ileum salmonella was significantly decreased in Bio-plus treatments compared to that of control in expt 2 (P〈0.05). Total yeast and Lactobacillus spp. of illeum tended to increase in expt 1 and were significantly higher in expt 2 than control(P〈0.05). The results of these experiment indicated that dietary Bio-plus supplements increased weight gain, the number of ileum yeast and Lactobacillus spp. of broiler chicks.

Detection of Helicobacter pylori Infection Rate in Resected Gastric Cancer Patients (위절제술을 받은 위암 환자의 Helicobacter pylori 감염률 측정에 관한 연구)

  • Purpose: This study aims to determine the infection rate of Helicobacter pylori (H. pylori) in gastric cancer patients who received gastrectomies, and to compare the rates of H.pylori infection detected by serological test and that of histopathological test, and to evaluate its clinical meaning. Materials and Methods: Fifty two patients were selected from those who underwent gastrectomies at the Department of Surgery, Samsung Medical Center, from March 1997 to May 1997. The control group consisted of healthy 103 persons visited the center for health promotion in Samsung Medical Center. In both groups, we quantitatively checked serum level of IgG anti H. pylori antibody titer by ELISA using GAP IgG test kit (BioRad, USA) for the serological test, and we microscopically examined the surgical specimen stained by Warthin-Starry silver staining method for the histopathological test. Results: The seropositive rate of H. pylori in the patients' group was $71.2\%$ (37/52), and the control group was $65.0\%$ (67/103). The difference between two groups was statistically significant. However the histopathological study showed that the overall detection rate of H. pylori was $61.5\%$ (32/52) in the patients' group and $61.2\%$ (63/103) in the control group; nd this difference was not statistically significant Conclusion: We could confirm that H.pylori infection rate in the gastric cancer resected patients was statistically higher than in the normal healthy persons even in small population. And the detection method for the H. pylori infection by serological test was presumed to be better than that of histopathological test using surgical specimen. Further study for the larger population by well-organized multicenters will be needed.

The effect of Acacia nilotica bark extract on growth performance, carcass characteristics, immune response, and intestinal morphology in broilers as an alternative to antibiotic growth promoter

  • Objective: Present study was designed to evaluate the efficacy of Acacia nilotica bark extract as an alternative to antibiotic growth promoters in broilers. Methods: Six hundred, day-old broiler chicks were randomly divided into six groups (NC, without any supplementation; AB, NC+Zinc Bacitracin; PB, NC+Safmannan; ANBE1, NC+A. nilotica bark extract 0.1%; ANBE3, NC+A. nilotica bark extract 0.3%; ANBE5, NC+A. nilotica bark extract 0.5%), with ten replicates per group (10 chicks/replicate) and feeding trial was lasted for 35 days. Results: Results showed that weight gain (1,296.63 g) and feed conversion ratio (FCR, 1.59) of AB was better than NC, during the finisher phase. Overall FCR of AB (1.53), PB (1.54), and ANBE5 (1.54) was significantly (p<0.05) better than NC. From carcass parameters relative weight of wing and heart were highest in ANBE3 (2.5% and 1.51%, respectively). Significantly (p<0.05) highest blood glucose level was observed in NC (264.5 mg/dL) and highest albumin concentration was found in AB (1.46 mg/dL). In addition, antibody titer levels against ND and IBD were higher in ANBE5 than NC, while higher relative weight of bursa was observed in ANBE3 than NC. The villus height to crypt depth ratio in all experimental groups was better than NC. Conclusion: Acacia nilotica bark extract could be a suitable alternative to antibiotic growth promoters to support the growth in broilers.

Immunogenicity and Safety of Recombinant Hepatitis B Vaccine(HG-IIR) in Healthy Infants and Children (유전자 재조합 B형간염 백신의 10세이하 소아에서의 면역원성 및 안전성)

  • Objective : To evaluate the immunogenicity and safety afforded by the HG-II$^{(R)}$ recombinant hepatitis B vaccine given to healthy neonates and children and to find the influence of preceding BCG vaccination on immunogenicity. Methods : Three doses of recombinant hepatitis B vaccine with a dose of $10{\mu}g$ were given at birth, 1 and 6 months of age. This study was conducted in three hospitals (Gyeongsang National University Hospital(Group A), Kangnam General Hospital(Group B) and Younsei University Hospital(Group C)) from April, 1995 to June, 1996. Group A and Group B received 2nd dose of hepatitis B vaccine at 1 week after and before BCG vaccine, respectively. Antibidy levels, at 1 month after the 3rd dose of hepatitis B vaccine were determined by a radioimmunoassay. Results : 1) One hundred four infants and ten children were enrolled : 55 infants and 43 infants received 2nd dose of hepatitis B vaccine at 1 week after( After BCG Group) and before BCG vaccine(Before BCG Group), respectively. 2) The seropositive rate was 99.1%, and geometric mean anti-HBs titer was 131.2mIU/ml. 3) The geometric mean titers were 105.5mIU/ml and 162.8mIU/ml in After BCG and Before BCG Group, respectively(p<0.025). 4) Among 359 episodes of vaccination, the occurrence of systemic and local side reaction were reported in 7.8% and 1.4%, respectively. Conclusion : Recombinant hepatitis B vaccine(HG-II$^{(R)}$))was highly immunogenic and safe. The significantly lower geometric mean antibody titer in the BCG preceding group was observed. Well-designed controlled study with the large number of sample size will be required to show the influence of preceding BCG vaccination.
