Induction of lipid peroxidation and melanoma cell death by turmeric oleoresin through its photosensitizing properties (심황색소의 감광활성과 빛 조사에 의한 지질산화 및 세포독성 유도 효과)

  • Curcuminoids in turmeric oleoresin (TO) are known to be effective antioxidants; they exhibit photosensitizing properties under light. In this study, the photoreactive properties of TO and its consequent induction of lipid peroxidation and cytotoxicity were evaluated. TO exhibited photosensitizing activities as evidenced by the reduction of nitro blue tetrazolium and by the decolorization of formazan under light, whereas light-irradiated TO did not enhance the levels of reactive oxygen species. The levels of hydroperoxide and thiobarbituric acid reactive substance (TBARS) were significantly elevated under a fluorescent light (10 W/m2) in corn, canola, and soybean oils containing 10-40 and 20-80 ㎍/mL of TO (p<0.05) but not in olive oil. Canola oil was the most sensitive to photo-oxidation induced by TO. The level of TBARS from linoleic acid in the oil in water system was, however, decreased by TO under light. The cytotoxicity effect of TO on melanoma cells was also substantially enhanced under light.

Cytosolic Phospholipase A2 Activity in Neutrophilic Oxidative Stress of Platelet-activating Factor-induced Acute Lung Injury (Platelet-activating Factor에 의한 급성폐손상에서 호중구성 산화성 스트레스에 관여하는 Cytosolic Phospholipase A2 활성도의 변화)

  • Background: The present investigation was performed in rats and isolated human neutrophils in order to confirm the presumptive role of the positive feedback loop of cytosolic phospholipase $A_2$ ($cPLA_2$) activation by plateletactivating factor (PAF). Methods: The possible formation of the positive feedback loop of the $cPLA_2$ activation and neutrophilic respiratory burst was investigated in vivo and in vitro by measurement of the parameters denoting acute lung injury. In addition, morphological examinations and electron microscopic cytochemistry were performed for the detection of free radicals in the lung. Results: Five hours after intratracheal instillation of PAF ($5{\mu}g/rat$), the lung leak index, lung myeloperoxidase (MPO) activity, the number of neutrophils and the concentration of cytokine-induced neutrophil chemoattractant (CINC) in bronchoalveolar lavage fluid were increased by PAF as compared with those of control rats. The NBT assay and cytochrome-c reduction assay revealed an increased neutrophilic respiratory burst in isolated human neutrophils following exposure to PAF. Lung and neutrophilic $cPLA_2$ activity were increased following PAF exposure and exposure to hydrogen peroxide increased $cPLA_2$ activity in the lung. Histologically, inflammatory findings of the lung were observed after PAF treatment. Remarkably, as determined by $CeCl_3$ cytochemical electron microscopy, increased production of hydrogen peroxide was identified in the lung after PAF treatment. Conclusion: PAF mediates acute oxidative lung injury by the activation of $cPLA_2$, which may provoke the generation of free radicals in neutrophils.

Effects of the Culture Broth of Lactic Acid Bacteria Cultured in Herb Extracts on Growth Promotion and Nonspecific Immune Responses of Aquacultured Fish (한약재 추출물에 배양된 유산균 배양액이 양식어류의 사료첨가제로서 성장과 비 특이적 면역에 미치는 영향)

  • We have investigated the effects of the culture broth of lactic acid bacteria (LAB) cultured in herb extract on growth, hematological parameter, nonspecific immune responses and disease resistance of olive flounder (Paralichthys olivacells) and parrot fish (Oplegnathus fasciatus) for 12 weeks. Weight gain of olive flounder fed diet with mixture was not significant among the control group. But In parrot fish, was significantly higher 20g than control group. The feed efficiency of olive flounder were 25% higher in the experimental groups than in the control. There were no significant differences in feed efficiency among each group on parrot fish. Treatment of olive flounder contents of GOT and GPT in serum decreased after 8 weeks. But there were no significant differences in GLU and TP among each group. Also, there was no significant of NBT reduction. The activities of lysozyme were higher in experimental group of olive flounder than in the control after 8 weeks. On the other hand, activities of lysozyme were triple higher in the experimental group of parrot fish than in the control after 12 weeks. In the oliver flounder case, the survival rate (%) after an artificial challenge with $10^7$ CFU/ml of Vibrio anguillarum and Streptococcus iniae per fish, was 18% higher in the experimental groups than the control. The higher survival rate of parrot fish were 17% and 16% in the experimental groups than the control respectively.

Effects of Supplemental Undaria, Obosan and Wasabi in the Experimental Diets on Growth, Body Composition, Blood Chemistry and Non-specific Immune Response of Juvenile Flounder, Paralichthys olivaceus (배합사료에 미역, 어보산 및 고추냉이 첨가가 넙치 치어의 성장, 체성분, 혈액성상 및 비특이적 면역능에 미치는 효과)

  • When fed on one of the six isonitrogenous (45%) and isolipic (8%) feed containing 5 or 10% Undaria, 2% wasabi leaf, 2% wasabi stem and 0.5% herb (Obosan) for a period of 8 weeks, 95-98% juvenile flounder survived. Growth, feed efficiency and condition factor of the flounder fed on diet containing 0.5% herb were significantly (p<0.05) higher than those fed on diet supplemented with 10% Undaria. There was no significant (p>0.05) differences in moisture, crude protein, lipid and ash of the flounder receiving the different diets. The flounder fed on diet supplemented with 10% Undaria had the highest moisture but the lowest lipid in liver. Hematological parameters such as red blood cell, hematocrit and hemoglobin and serum constituents such as glucose, total cholesterol and glutamic-oxaloacetic transaminase of the flounder fed on the diets varied but no specific trend became apparent. Lysozyme activity in the serum of the flounder fed on diet supplemented with 5% Undaria and the herb, as well as nitroblue tetrazolium (NBT) reduction of macrophage in the head kidney of the flounder fed on diet containing the herb and 2% wasabi stem were significantly (p<0.05) higher than those receiving control diet. Briefly, the herb supplementation promoted growth and that of Undaria and wasabi stem enhanced non-specific immune response.

Korean Mistletoe Viscum album coloratum Induces Specific and Non-Specific Immune Responses in Japanese Eel Anguilla japonica

  • Effects of Korean mistletoe extracts (KM-110), Viscum album coloratum on the specific and non-specific immune responses of Japanese eel Anguilla japonica were examined. The optimal concentration not showing toxicity of KM-110 was determined to $30-40{\mu}g/ml$ in vitro and $100{\mu}g$/100 g of fish in vivo. Even $1000{\mu}g$ of KM-110/100 g of fish did not show any clinical problem in fish though the levels of toxic parameters were slightly increased. In terms of antibody production, KM-110 significantly elicited more antibody production than FCA or $\beta$-glucan. $\beta$-glucan plus KM-110 group synergistically enhanced antibody production. There was no significant difference between KM-110 and KM-110 plus $\beta$-glucan group. The ROI production by head kidney (HK) leucocytes of eel injected with 500 or $1000{\mu}g$ KM-110 was significantly (P<0.05) enhanced than the control and FCA-treated group. Maximum increase in the NBT reduction value was observed in $1000{\mu}g$ KM-110 group but no significant difference was found between 500 and $1000{\mu}g$ KM group. The level of serum lysozyme activity was significantly (P<0.05) higher in the 500 and $1000{\mu}g$ KM-110- or FCA-treated group than in the control and $200{\mu}g$ KM-110 group. The phagocytic activities of HK leucocytes isolated from eel injected with 500 and $1000{\mu}g$ KM-110 were significantly (P<0.05) higher than $200{\mu}g$ KM-110 and PBS-injected control group. Korean mistletoe appeared to be a good activator of the specific and non-specific immune responses of Japanese eel.

Test of Superoxide Dismutase Characteristics and Antioxidant Activity in Perilla Leaves (들깨잎에 함유된 Superoxide Dismutase의 특성 및 항산화 활성 검정)

  • This study was conducted to investigate the superoxide dismutase (SOD) characteristics and antioxidant activity by nonenzymatic(Fe$^{2+}$/Ascorbate) and Fe$^{3+}$-ADP/NADPH method in perilla(Perilla frutescens var. japonica Hara.) and jaso(Perilla frutescens Briton var. acuta Kudo.) leaves. The characteristics were evaluated by the nitro blue tetrazolium reduction method. Perilla leaves contained three or four major SODs depending on the varieties. The inhibitor test indicated that the Perilla leaves contained two Cu /ZnSODs and one or two FeSODs, but Jaso leaves have only Cu/ZnSOD. However, no varietal differences were detected in the Cu /ZnSOD isozyme patterns. FeSODs, however, showed different varietal isozyme patterns through the different combinations of the two FeSOD isozymes. Among MeOH extractes, "mil yang 2" showed very strong antioxidant activity. Relatively large differences in the levels of SOD and antioxidant activity detected in the Perilla varietites.
