High Glucose Potentiates the Alloxan-induced Cytotoxicity in Cultured Rat Insulinoma Cells (흰쥐 인슐린종세포에서 고농도 포도당의 Alloxan 독성 증강 효과)

  • Reactive oxygen species are produced under diabetic conditions and possibly cause various forms of tissue damage in patients with diabetes mellitus. The aim of this study was to examine the effects of high glucose on the alloxan-induced beta cell injury. The insulinoma (RINm5F) cells were clutured either with high glucose (22.2 mM) or normoglucose (5.6 mM) in RPMI 1460 media for 3 days. The SOD activities were determined by spectrophotometric assay and nitroblue tetrazolium (NBT) stain. The effects of high glucose on the cytotoxicity of alloxan were also investigated in RINm5F cells and the cells viability were determined by 3-(4,5-dimethylthiazolyl)-2,5-diphenyltetrazolium bromide (MTT) methods. Results showed that the CuZn-SOD activity was decreased but Mn-SOD activity was increased significantly in RINm5F cells cultured with high glucose (22.2 mM) media. The cytotoxicity of alloxan was increased by high glucose compared with normoglucose in RINm5F cells. Diethyl-dithiocarbarmate (DDC), as inhibitor of CuZn-SOC, also potentiate the alloxan-induced cytotoxocity in RINm5F cells. These results suggest that, in RINm5F cells, short term culture with high glucose media decreases Cu-Zn-SOD activity and the decreased activity of CuZn-SOD many one of the causative factors of beta-cell injury induced by high glucose.

Synthesis of Shape Controlled Pd Nanoparticles and Surface-Induced Photoreduction of 4-Nitrobenzenethiol on Pd (모양이 조절된 팔라듐 나노입자의 합성과 4-나이트로벤젠 사이올의 광환원 반응)

  • The facile synthesis of shape-controlled Pd nanoparticles (PdNPs) with ascorbic acid as a reducing agent and cetyltrimethylammonium bromide (CTAB) as a capping agent is presented in this study. The synthesized PdNPs were characterized by UV-vis spectroscopy, transmission electron microscopy (TEM), scanning electron microscopy (SEM), X-ray diffraction (XRD), and Raman Spectroscopy. The prepared PdNPs show efficient surface-enhanced Raman scattering (SERS) properties. SERS studies on the adsorption characteristics of 1,4-phenylene diisocyanide (1,4-PDI) on colloidal PdNPs have revealed that the relative peak intensity of the $(NC)_{free}$ and $(NC)_{bound}$ modes distinctly depends on the 1,4-PDI concentration as well as the shape of the PdNPs. Furthermore, we found that the PdNPs are also efficient photoelectron emitters such that the SERS spectrum of 4-nitrobenzenethiol (4-NBT) on PdNPs is readily converted to that of 4-aminobenzenethiol (4-ABT) under 632.8 nm radiation.

Taxonomic Revision of the Genus Lactifluus (Russulales, Basidiomycota) of South Korea

  • Lactifluus (Pers.) Roussel is an ectomycorrhizal genus that was recently recognized to be distinct from the genus Lactarius. To date, 226 Lactifluus species have been reported worldwide. Misidentification of Lactifluus species is common because of intraspecific morphological variation, cryptic diversity, and the limited number of taxonomic keys available. Molecular data are indispensable for species delimitation; a multilocus phylogenetic analysis showed that most Asian Lactifluus species are not conspecific with morphologically similar species present on other continents. In particular, Korea has misused European and North American Lactifluus names. In this study, we evaluated the taxonomy of Lactifluus in Korea using both morphological and multilocus molecular (ITS, nrLSU, rpb1, and rpb2) data. We examined 199 Lactifluus specimens collected between 1980 and 2016, and a total of 24 species across the four Lactifluus subgenera were identified. All Korean species are distinct and clearly separated from European and North American species. Five taxa corresponded to previously described species from Asia and the remaining 19 taxa are confirmed as new species. Herein, we provide keys to the Korean Lactifluus species within their subgenera, molecular phylogenies, a summary of diversity, and detailed description of the new species.

  • "Eating cosmetic" market comes to prominence worldwide. Inner Beauty has established itself as the top trend nowadays and is leading the health functional food market. Another noticeable trend other than beauty-from-within is that products using "plant-based" natural botanical raw material are increasing. Based on these trends above, we have developed a plant extract named Agatri® for skin health using Agastache rugosa. By ingesting Agatri®, it is expected that the formation of skin wrinkles is suppressed, and skin elasticity is increased by suppressing the decrease in collagen content caused by ultraviolet rays. At the same time, it is possible to prevent skin aging by improving skin hydration and trans-epidermal water loss (TEWL). The studies show that it significantly increased production of hyaluronic acid and collagen, and decreased production of matrix metalloproteinases (MMPs). And Agatri® has been approved as an "Individually recognized functional ingredient for skin health by the Ministry of Food and Drug Safety, Korea in 2020. As a result, Agatri® is a good solution for those who are in need of skin protection from exposure to UV and aging.

Effect of Dietary Lipid Sources on Growth, Enzyme Activities and Immuno-hematological Parameters in Catla catla Fingerlings

  • Ninety advanced Catla catla fingerlings (av. wt. 16 g) were randomly distributed in six treatment groups with three replicates each for an experimental period of 60 days to study the effect of dietary lipid source on growth, enzyme activities and immuno-hematological parameters. Six isoprotein (40.0-41.9%) and isocaloric (4,260 kcal $kg^{-1}$) semi-purified diets were prepared with varying levels of soybean oil (SBO) and cod liver oil (CLO) within a total of 8% lipid viz., $D_1$ (Control), $D_2$ (8% SBO), $D_3$ (6% SBO and 2% CLO), $D_4$ (4% SBO and 4% CLO), $D_5$ (2% SBO and 6% CLO) and $D_6$ (8% CLO). Highest SGR was noted in $D_5$ (0.73${\pm}$0.03) group, which was similar with $D_3$ (0.71${\pm}$0.02) and $D_4$ (0.69${\pm}$0.01) groups. Activity of intestinal lipase, hepatic glucose-6-phosphate dehydrogenase (G6PDH) and aspartate amino transferase (AST) of the lipid treatment groups were significantly higher (p<0.05) than the control group. The respiratory burst activity of the phagocytes (Nitroblue tetrazolium (NBT)) was highest in $D_2$ (1.95${\pm}$0.21) followed by $D_3$ (1.19${\pm}$0.15) group, which were significantly (p<0.05) higher than the other groups. Globulin level was significantly higher in $D_3$ (1.29${\pm}$0.08) than in the other groups expect $D_4$. Hemoglobin content and total erythrocyte count did not show any significant difference. From this study, it is concluded that a diet containing 6% soybean oil and 2% cod liver oil ($D_3$) yields higher growth and immune response in Catla catla fingerlings and would be cost effective.

Changes in Antioxidant Activity during Growth of Artemisia iwayomogi (더위지기 생육 중 항산화 활성 변화)

  • The aim of this study was to investigate the effect of plant growth at several different growing periods on antioxidant activities and zeatin and ABA contents of Artemisia iwayomogi. Measurements of antioxidant activities, lipid peroxidation inhibition, and superoxide radical scavenging activity were done using PMS, NBT and lipid auto-oxidation analysis, respectively. The results show that activities of antioxidants from Artemisia iwayomogi had much higher than BHT. DPPH free radical scavenging effect of Artemisia leaf extract was increased from $71.06{\pm}4.36%$ in April to $90.06{\pm}4.41%$ in October. Activities of superoxide radical scavenging and lipid peroxidation inhibition were $33.83{\pm}3.45%$ and $45.60{\pm}3.10$ in April and then increased to $84.40{\pm}4.00%$ and $75.86{\pm}3.50%$ in October, respectively. An ELISA technique has been developed for the determination of zeatin and ABA in Artemisia leaves. By this method, the content changes of zeatin and ABA from Artemisia during the growth were investigated. The zeatin content in leaf was measured to be $186.86{\pm}1.40$ pmol/g dry weight in April, however, decreased to $117.93{\pm}5.83$ pmol in October. The ABA content in leaf increased from $19.00{\pm}1.26$ nmol in April to $68.20{\pm}2.52$ nmol in October. Relationship between antioxidant activities and plant hormone contents was indicated that antioxidant activity may depend on decreasing zeatin content or increasing ABA content.

Radical Scavenging and Anti-Obesity Effects of 50% Ethanol Extract from Fermented Curcuma longa L. (발효울금 50% 에탄올 추출물의 라디칼 소거능 및 지방형성 억제 효과)

  • In this study, free radical scavenging activities (ABTS, DPPH, NBT, TBARS, and ORAC) and anti-obesity potential were evaluated using 50% ethanol extract from fermented Curcuma longa L. (FCE50). FCE50 showed free radical scavenging activities and anti-oxidant potential. Lipid accumulation and intracellular TG content were significantly reduced by 25.8% and 28.6%, respectively, by $250{\mu}g/mL$ of FCE50 compared to adipocytes. Glucose uptake was significantly reduced by 12.0%. FCE50 significantly reduced mRNA expression of acetyl-CoA carboxylase in 3T3-L1 cells. These results indicate that the anti-adipogenic effect of FCE50 might be due to its radical scavenging activity and anti-oxidant potential.

Effect of Phellinus linteus on Differentiation and Cell Proliferation in Human Leukemia HL-60 cells (상황버섯이 인간 백혈병 세포주인 HL-60 세포의 분화유도 및 증식에 미치는 영향)

  • We have examined the effect of water extract of Phellinus linteus, a raw material of Korean traditional herbal medicine, on the induction of HL-60 cell differentiation. The proliferation of HL-60 cell was inhibited dose-dependently by treatment with various doses of P. linteus extract. It also caused a significant change in NBT reduction (7.5 times). The expression of CD11b and CD14 was increased in the cells treated with the extract, especially in those arrested at G0/G1 stage, which suggested that some components in P. Linteus extract induced HL-60 cell differentiation to granulocytic and monocyte lineages. Moreover, the expression levels of $p21^{WAF1/CIP}$ and $p27^{KIP}$ were up-regulated during HL-60 cell differentiation induced by P. Linteus extract. These results together suggest that P. Linteus extract contains potential HL-60 cell differentiation agents.

Effects of Different Dietary Vitamin E Levels on Growth Performance, Non-specific Immune Responses, and Disease Resistance against Vibrio anguillarum in Parrot Fish (Oplegnathus fasciatus)

  • We report nutritional physiology and non-specific immune responses of vitamin E in parrot fish for the first time. This study aimed to investigate the essentiality and requirements in diets based on growth performances, non-specific immune responses and a challenge test against Vibrio angullarum. Six casein-gelatin based semi-purified diets were formulated to contain six graded levels of DL-${\alpha}$-tocopheryl acetate (${\alpha}$-TA) at 0, 25, 50, 75, 100 and 500 mg/kg diet (designated as E0, E25, E50, E75, E100 and E500, respectively) and fed to triplicate groups of juvenile parrot fish for 12 weeks. The analyzed dietary concentrations of vitamin E were 0, 38, 53, 87, 119 and 538 mg/kg diet for E0, E25, E50, E75, E100 and E500, respectively. At the end of the feeding trial, growth performance and feed utilization of fish fed the E25 were significantly higher compared to that of fish fed the other diets. Liver ${\alpha}$-tocopherol concentration was significantly increased with an increase in dietary ${\alpha}$-TA in a dose dependent manner. No apparent clinical signs of vitamin E deficiency and mortality were observed in fish fed the basal diet for 12 weeks. Among the immune responses assayed, phagocytic (NBT assay) and myeloperoxidase activities were significantly increased with increment of dietary ${\alpha}$-TA levels. During the challenge test with V. anguillarum, E75, E100, and E500 diets resulted in higher survivals than E0, E25 and E50 diets. The findings of this study suggest that parrot fish require exogenous vitamin E and the optimum dietary level could be approximately 38 mg ${\alpha}$-TA/kg diet for normal growth and physiology. Dietary ${\alpha}$-TA concentration over 500 mg/kg could be required to enhance the nonspecific immune responses and improve the resistance of juvenile parrot fish against V. anguillarum.

The Extracting Methods of the Seeds of Yew(Taxus cuspidata Sieb) by Solvent Extraction and Its Efficacy (주목(朱木)씨앗 추출물의 제조방법 및 효능효과)

  • Yew (Taxus cuspidata Sibe) selected cultivation as drug, food and decorative plant in Gyong-gi province in Korea. To extract the water soluble active ingredients, as a extracting method, there was extracted with 20g of dried seeds with each 20g of butylene glycol(BG) and propylene glycol(PG), and 40 mL of water mixing 72 hours at $40{\pm}5^{\circ}C$, and then they were filtrated by 400 mesh. Appearance of extract of seeds was pale brown, $pH=4.5{\pm}0.5$, $gravity=1.013{\pm}0.05$, a reflective $index=1.373{\pm}0.05$, and yield=75%. Also, to extract the high purity oil from seeds, it minutely pulverized the dried seeds and added the hexane, mixing 2 hours at $20{\pm}57^{\circ}C$. And then, this filtrated it with 400-mesh. It got the purified oil through evaporating them at $55^{\circ}C$ during under vacuum. As the results, appearance was slightly brown, gravity=0.922 acid value=0.12, saponification value=192, and it should be obtained the $40{\pm}5%$ of yield. As the efficacy evalution of cosmetic field, the antioxidative activities by NBT method were stronger 86.0% from extract of talus seeds than 52.0% from green tea extract and 35.0% from skullcap extract as well as the antioxidative activities by DPPH method were stronger 93.7% from extract of seed than 60.3% from extract of green tea and 27.1% from extract of skullcap. These are more effective than other plant extracts. The collagen synthesis rate on the activating fibroblast for Taxus cuspidata Sibe extract showed 35.43%. As the activity of the skin elasticity, PPE(porcine pancreatic elastase)-inhibitory activities of talus extract was 50.8%. Anti-inflammatory activity was more effective to be taken 41.1% of taxus seed oil than 24.2% of steady glycyrrhetinate (SG) as a control.
