N-methyl-D-aspartate 수용기의 다양한 조절이 일차 배양된 정상사람구강각화세포의 생존에 미치는 영향 (THE EFFECT OF DIFFERENTIAL MODULATION OF N-METHYL-D-ASPART ATE RECEPTOR ON THE VIABILITY OF PRIMARY CULTURED NORMAL HUMAN ORAL KERATINOCYTES)

  • In the present study, I investigated the effects of N-methyl-D-aspartate (NMDA), arachidonic acid (AA), and Nitric Oxide Synthase Inhibitor (NOS-I), alone or in combination, on the viability of cultured primary normal human oral keratinocytes (NHOK). Specifically, we examined whether AA and NOS-I could protect primary NHOK from glutamate cytotoxicity. The purpose of this study was therefore the preliminary study for the examination of the interaction between these agents and NHOK in order to elucidate the mechanisms by which epithelial growth and regeneration are regulated. NHOK were obtained from gingival tissue of 20 individuals aged 20 to 29, and third passage (P3) cells were used for this study. Cell viability was measured by the MTT assay. NMDA and NNA, a calcium dependent NOS inhibitor, induced an initial increase in cell number, which subsequently decreased by the $7^{th}$ day. Low concentration of AA ($0.5\;{\mu}M$ & $1\;{\mu}M$) induced an increase in cell number while high concentrations of AA ($5\;{\mu}M$ & $10\;{\mu}M$) induced a decrease in cell number. The decrease in cell number induced by NMDA at the $7^{th}$ day was abolished by the addition of low concentrations of AA ($0.5\;{\mu}M$ & $1\;{\mu}M$) or NOS inhibitors. Low concentrations of AA ($0.5\;{\mu}M$ & $1\;{\mu}M$) or NOS inhibitors may protect the NHOK from NMDA induced cytotoxicity. These reactions might be related to the NMDA receptor in the cell and the change of the intracellular calcium ion concentration.

N-methyl-D-aspartate 수용체 길항제가 몰핀 신체의존성 및 진통내성에 미치는 영향 (Comparison of the Effects of MK-801 and Dextromethorphan on Opioid Physical Dependence and Analgesic Tolerance)

  • N-methyl-D-aspartate(NMDA) receptor has been well known as an important mediator of several forms of neural and behavioral plasticity. But different results were reported about the effect of MK-801 or dextromethorphan on opioid dependence. The present studies examined whether NMDA receptor antagonists can alter the opioid dependence and tolerance in rodents. Naloxone precipitated withdrawal symptoms and changes of locomotor activities were observed in MK-801 or dextromethorphan pretreated morphine-dependent rats. Tail-flick assay was used for morphine analgesia and tolerance was found after 4 day's consecutive injections (10 mg/kg, s.c., twice/day) of morphine in mice. Locomotor activity was increased and the withdrawal symptoms were decreased by the pretreatment of MK-801 in morphine-dependent rats. But 0.3 mg/kg i.p. of MK-801 intensified the body weight loss and produced severe ataxia and rotation although some withdrawal signs were attenuated. Morphine induced analgesic tolerance was inhibited by the pretreatment of MK-801 and dextromethorphan. Dextromethorphan was more potent than MK-801 in inhibiting the development of the analgesic tolerance in mice. These results suggest that NMDA system may be involved in opioid withdrawal and analgesic tolerance but appropriate caution should be requested when MK-801 is used in combination with opioid because of untoward neurologic signs.

The Antinociceptive Effect of Sigma-1 Receptor Antagonist, BD1047, in a Capsaicin Induced Headache Model in Rats

  • Intracranial headaches, including migraines, are mediated by nociceptive activation of the trigeminal nucleus caudalis (TNC), but the precise mechanisms are poorly understood. We previously demonstrated that selective blockage of spinal sigma-1 receptors (Sig-1R) produces a prominent antinociceptive effect in several types of pain models. This study evaluates whether the Sig-1R antagonist (BD1047) has an antinociceptive effect on capsaicin (a potent C-fiber activator) induced headache models in rats. Intracisternal infusion of capsaicin evoked pain behavior (face grooming), which was significantly attenuated by BD1047 pretreatment. BD1047 consistently reduced capsaicin-induced Fos-like immunoreactivity (Fos-LI), a neuronal activator, in the TNC in a dose-dependent manner. Moreover, capsaicininduced phosphorylation of N-methyl-D-aspartate receptor subunit 1 was reversed by BD1047 pretreatment in the TNC. These results indicate that the Sig-1R antagonist has an inhibitory effect on nociceptive activation of the TNC in the capsaicin-induced headache animal model.

Effects of Placing Micro-Implants of Melatonin in Striatum on Oxidiative Stress and Neuronal Damage Mediated by N-Methyl-D-Aspartate (NMDA) and Non-NMDA Receptors

  • Overstimulation of both kainate (KA) and N-methyl-D-aspartate (NMDA) receptors has been reported to induce excitatoxicity which can be characterized by neuronal damage and formation of reactive oxygen free radicals. Neuroprotective effect of melatonin against KA-induced excitotoxicity have been documented in vitro and in vivo. It is, however, not clear whether melationin is also neuroportective against excitotoxicity mediated by NMDA receptors. In the present work, we tested the in vivo protective effects of striatally infused melatonin against the oxidative stress and neuronal damage induced by the injection of KA and NMDA receptors into the rat striatum. Melatonin implants consisting of 22-gauge stainless-steel cannule with melatonin fused inside the tip were placed bilaterally in the rat brain one week prior to intrastriatal injection of glutamate receptor subtype agonists. Melatonin showed protective effects against the elevation of lipid peroxidation induced by either KA or NMDA and recovered Cu, Zn-superoxide dismutase activities reduced by both KA and NMDA into the control level. Melatonin also clearly blocked both KA- and NMDA-receptor mediated neuronal damage assessed by the determination of choline acetyltransferase activity in striatal monogenages and by microscopic observation of rat brain section stained with cresyl violet. The protective effects of melatonin are comparable to those of DNQX and MK801 which are the KA- and NMDA-receptor antagonist, respectively. It is suggested that melatonin could protect against striatal oxidative damages mediated by glutamate receptors, both non-NMDA and NMDA receptors.

Kainate-induced Elevations of Intracellular $Ca^{2+}$ and Extracellular Glutamate are Partially Decreased by NMDA Receptor Antagonists in Cultured Cerebellar Granule Neurons

  • Several lines of evidence indicate that physiological activity of N-methyl-D-aspartate (NMDA) receptor was blocked by physiological concentration of $Mg^{2+}$ (1.2 mM). However, the activity of NMDA receptor may not be blocked totally with this concentration of $Mg^{2+}$ under elevated membrane potential by kainate. Here, we described the effect of $Mg^{2+}$ on NMDA receptor and how much of NMDA receptor functions could be activated by kainate. Effects of NMDA receptor antagonist on kainate-induced elevation of intracellualr $Ca^{2+}$ levels $([Ca^{2+}]_i)$ and extracellular glutamate level were examined in cultured rat cerebellar granule neurons. kainate-induced elevation of $([Ca^{2+}]_i)$ was not affected by physiological concentration of $Mg^{2+}$. Kainate-induced NMDA-induced elevation was blocked by the same concentration of $MG^{2+}$Kainate-induced elevation of [$([Ca^{2+}]_i)$ was decreased by 32% in the presence of NMDA antagonists, MK-801 and CPP (3-[2-carboxypiperazine-4-yl]propyl-1-phosphonic acid), in $Mg^{2+}$ free buffer. Kainate receptor-activated gluamate release was also decreased (30%) by MK-801 or CPP. These resuts show that certain extent of elevations of intracellular $Ca^{2+}$ and extracellular glutamate by kainate is due to coativation of NMDA receptors.

Distinct $[^3H]$MK-801 Binding Profiles with the Agonist, Partial Agonist, and Antagonist Acting at the Glycine Binding Site of the N-Methyl-D-Aspartate Receptor

  • The N-methyl-D-aspartate (NMDA) receptor-ion channel complex is activated by the simultaneous presence of L-glutamate and glycine, allowing the binding of MK-801 to the phencyclidine (PCP) site of the receptor. The $[^3H]$MK-801 binding assay system was established for determination of pharmacological functions of test compounds acting at the glycine site of the receptor. The binding in the presence of 0.1 $\mu$M L-glutamate was increased by an agonist (glycine) in a dose-dependent fashion, while decreased by either partial agonist (R-(+)-HA-966) or antagonist (5,7-dichlorokynurenic acid: 5,7-DCKA). To distinguish partial agonism from antagonism, various concentrations of 7-chlorokynurenic acid (7-CKA) were added in the assay to eliminate the interference of the endogenous glycine present in the membrane preparations. The bindings in the presence of L-glutamate (0.1$\muM$) and 7-CKA (1, 5, or 10$\muM$) were increased by R-(+)-HA-966. Being a weak partial agonist, the extent of potentiation was much less than that by the agonist. These binding profiles were clearly distinguishable from those by the antagonist, 5,7-DCKA, which exhibited no intrinsic activity. The binding assays established in the present study are a useful system to classify ligands acting at the glycine site of the NMDA receptor by their pharmacological functions.

Modulation of Sarcodon Aspratus on lon Currents-induced by Excitatory Neurotransmitters in Rat Periaqueductal Gray Neurons

  • Sarcodon aspratus is the mushroom of Telephoracea which was been classified into Alphllophorales. The aqueous extract of Sarcodon aspratus in known to have anti-tumor activity, immune modulatory effect, and anti-oxidative action. The descending pain control system consists of three major components: the periaqueductal gray (PAG) of the midbrain, the rostroventral medulla including the nucleus raphe magnus, and the spinal dorsal horn. Glutamate is the primary excitatory neurotransmitter in the brain. Glutamate ionotropic receptors are classified as N-methyl-D-aspartate (NMDA) receptor, ${\alpha}$-amino-3-hydroxy-5-methyl-4-isoxazolepropionate (AMPA) receptor, and kainate receptor. In the present study, the modulation of Sarcodon aspratus on the ion currents activated by glutamate, NMDA, AMPA, and kainate in the acutely dissociated PAG neurons was investigated by nystatin-perforated patch-clamp technique under boltage-clamp condition. Sarcodon aspratus increased glutamate- and NMDA-induced ion currents were not increased by Sarcodon aspratus. The present results show that Sarcodon aspratus may activate the descending pain control system in rat PAG neurons through NMDA receptor.

신생 백서의 저산소성 허혈성 뇌손상에서 NMDA receptor 조절을 통한 유전자 재조합 인 에리스로포이에틴의 신경보호 (Neuroprotection of Recombinant Human Erythropoietin Via Modulation of N-methyl-D-aspartate Receptors in Neonatal Rats with Hypoxic-ischemic Brain Injury)

  • 목 적 : 신장에서 분비되어 적혈구를 생산하는 빈혈제로 알려진 에리스로포이에틴(Erythropoietin, EPO)은 단순히 피를 만드는 조혈기능 뿐 아니라 최근 신경계 보호 및 신경강화 효과가 있다고 발표되고 있지만 주산기 가사로 인한 저산소성 허혈성 뇌병증의 치료제로서 그 기전이 명확하게 밝혀지지 않았다. 저자들은 유전자 재조합 인 에리스로포이에틴(recombinant Human EPO, rHuEPO)을 이용하여 주산기 저산소성 허혈성 뇌병증의 치료제로서 N-methyl-D-aspartate (NMDA) 수용체와 관련된 흥분성 독성작용을 통한 조절 등 그 기전을 알아보고자 하였다. 방 법 : 재태기간 19일된 태아 백서의 대뇌피질 세포를 배양하여 정상산소군은 5% $CO_2$ 배양기(95% air, 5% $CO_2$)에 두었고, 저산소군과 농도별 뇌손상 전 rHuEPO 투여군(1, 10, 100 IU/mL)은 1% $O_2$ 배양기(94% $N_2$, 5% $CO_2$)에서 6시간 동안 뇌세포손상을 유도하였다. 세포성장과 생존력을 평가하기 위해 MTT 실험을 시행하였다. 동물 모델에서는 생후 7일된 신생백서의 좌측 총 경동맥을 결찰한 후 6개 군; 정상산소군, sham-operated군, 저산소-허헐성군, 저산소-허헐성+vehicle군, 저산소-허헐성 손상 전 rHuEPO 투여군, 저산소-허헐성 손상 후 rHuEPO 투여군으로 나누었고, 저산소-허헐성 손상은 특별히 제작한 통속에서 2시간 동안 8% $O_2$ (8% $O_2$, 92% $N_2$)에 노출시켰다. rHuEPO은 뇌손상 전후 30분에 체중 kg당 1000 IU를 투여하였고, 저산소-허헐성 손상 후 7일째 조직을 실험하였다. 적출한 조직으로 H&E 염색을 하여 뇌손상을 형태학적으로 관찰하였다. 세포배양 및 동물실험에서 NMDA 수용체의 아단위인 NR1, NR2A, NR2B, NR2C, NR2D를 이용하여 실시간 중합효소연쇄반응을 실시하였다. 결 과 : 저산소군에서 세포 생존률은 정상산소군보다 60% 감소하였으며, rHuEPO 투여군(1, 10 IU/mL)은 80% 증가하였다. rHuEPO 투여군(100 IU/mL)은 회복되지 않았다. 우측 반구 대비 좌측 반구의 범위는 정상산소군 98.9%, sham-operated군 99.1%, 저산소-허헐성군 57.1%, 저산소-허헐성+vehicle군 57.0%, 저산소-허헐성 손상 전 rHuEPO 투여군 87.6%, 저산소-허헐성 손상 후 rHuEPO 투여군 91.6%으로 나타났다. NMDA 수용체의 아단위 생체외 실험에서 실시간 중합효소연쇄반응의 결과 NMDA 수용체 아단위 mRNA의 발현은 rHuEPO 투여군에서 저산소군보다 모두 증가하였다. 결 론 : 본 연구에서 rHuEPO은 흥분성 독성작용과 관련되어 NMDA 수용체를 조절하면서 저산소성 허헐성 뇌손상을 보호하는 것을 알 수 있었다.

NMDA 수용체 아단위 2B의 Tyrosine 인산화 위치의 동정 (Identification of a Potential Tyrosine Phosphorylation Site on the NR2B Subunit of the N-methyl-D-aspartate Receptor)

  • NR2B는 연접후 치밀질의 주요 tyrosine 인산화 단백질이다. 본 연구에서는 mass spectrometry 방법을 적용하여 NR2B의 tyrosine 인산화 위치를 동정하였다. NR2B를 N-octyl glucoside (NOG)에 용해되지 않는 PSD 분획으로부터 SDS-PAGE와 electroelution방법으로 분리하였다. 분리한 단백질을 NR2B와 phos-photyrosine에 특이한 항체로 조사한 결과 이들은 phosphotyrosine을 유지하고 있는 NR2B임이 확인되었다. 이 단백질을 trypsin 혹은 endolys-C 처리하고, matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry 방법으로 조사한 결과 Tyr-1304이 인산화됨을 확인하였다.

인삼 품종별 뿌리 추출물의 NMDA 수용체 길항 효과 및 진세노사이드 함량 (N-Methyl-D-Aspartate (NMDA) Receptor Antagonistic Effect and Ginsenoside Content of Panax ginseng C. A. Meyer Cultivar Root Extracts)

  • Background: Although a number of Panax ginseng cultivars have been developed by Korean researchers in recent years, there has been insufficient analysis of their beneficial properties. In this study, we sought to identify useful ginseng varieties as functional materials. Methods and Results: We evaluated effects of root extracts of 10 ginseng cultivars (Cheongsun; CS, Chunpoong; CP, Gopoong; GP, Gumpoong; GMP, K1, Sunhyang; SH, Sunone; SO, Sunpoong; SP, Sunun; SU and Yunpoong; YP) against the inhibitory effects of nitric oxide (NO) and reactive oxygen species (ROS) production in mouse brain microglial BV2 cells, as well as the binding of N-methyl-D-aspartate receptor (NMDAR), a marker related to memory. Ginsenosides, such as 20 (S)-protopanaxadiols (PPDs), including ginsenoside-Rb1, -Rb2, -Rb3, -Rc, -Rd, and - Rg3 and 20 (S)-protopanaxatriols (PPTs) including -Re, -Rg1, and -Rg2 were analyzed by HPLC. We observed that the cultivar GMP showed the highest inhibitory effect (60.8%) against NO production at 20 ㎍/㎖. Those cultivars showing the significantly highest inhibition effects against ROS at 20 ㎍/㎖ were K1 (57.3%), SP (54.5%), YP (53.1%), CP (51.7%), CS (50.9%) and SH (49.6%). At 50 ㎍/㎖, K1 showed the most potent inhibitory effect (51.2%) on NMDAR binding. The total phenol content of SH (1.89 mg/g) and K1 (1.73 mg/g) were higher than those of the other cultivars, whereas in terms of PD/PT ratios, the values of CP (0.98), K1 (1.05) and SO (1.05) were lower than those of the other cultivars. On the basis of correlation coefficient (0.7064) between NMDAR inhibition and ONOO- scavenging activity. Conclusions: The findings of this study indicate that the cultivars K1 and SH could be useful ginseng resources as functional materials with favorable cognition-improving and antioxidative properties.
