전리방사선과 Cisplatin이 신경아세포종세포와 섬유모세포에서 Peroxiredoxin I과 II 발현 및 세포생존율에 미치는 영향 (Effects of Ionizing Radiation and Cisplatin on Peroxiredoxin I & II Expression and Survival Rate in Human Neuroblastoma and Rat Fibroblast Cells)

  • 목 적: 항산화 효소인 peroxiredoxin (Prx) I과 II가 유해자극에 의해서 유발되는 세포내 반응성 산소족(reactive oxygen species, ROS)에 대한 방어기전에 관여하는지 알아보고자 이 연구를 시행하였다. 대상 및 방법: SK-N-BE2C (신경아세포종세포)와 Rat2 (섬유모세포)에서 PrxI과 PrxII 발현을 보기 위하여 방사선조사, cisplatin 단독투여 및 cisplatin-방사선조사병합투여 후에 PrxI과 PrxII에 대한 western blot을 시행하였다. 또한 N-acetyl-L-cysteine (NAC)에 의하여 PrxI과 PrxII 발현에 미치는 영향과 세포생존율을 함께 조사하였다. 두 종류 세포에 방사선조사, 다양한 농도의 cisplatin을 단독투여 및 방사선조사와 병합투여 시 생존율을 각각 분석하였고 SK-N-BE2C의 각 군에서 시간별 생존율을 관찰하였다. 결 과: PrxI의 발현은 SK-N-BE2C에서 방사선조사 후 60분까지 증가하였으나 NAC 전처치한 경우 방사선조사 후 60분에서는 대조군에 비하여 약간 증가하였다. Rat2에서는 방사선조사 후 NAC 전처치 여부에 관계없이 증가하지 않았다. PrxII의 발현은 두 가지 세포 모두에서 방사선조사와 NAC 전처치 여부에 관계없이 증가하지 않았다. SK-N-BE2C와 Rat2에서 다양한 농도의 cisplatin 단독투여나 cisplatin-방사선조사병합시에는 PrxI 및 PrxII의 발현은 증가하지 않았다. SK-N-BE2C와 Rat2에서 NAC 전처치 여부에 따른 방사선조사군 및 cisplatin-방사선조사병합군의 생존율을 각각 비교한 결과 PrxI의 발현이 증가되었고, NAC 전처치하였으며 cisplatin의 농도가 낮을수록 유의하게 생존율이 높았다. SK-N-BE2C에서 NAC를 전처치한 방사선조사군의 생존율이 NAC 전처치 안한 방사선조사군에 비하여 높은 경향만 보였으나, Rat2에서는 유의한 차이로 NAC를 전처치한 방사선조사군의 생존율이 높았다. SK-N-BE2C에서 시간별 생존율을 측정한 결과는 방사선조사군과 cisplatin-방사선조사병합군을 비교하면 방사선조사군이 빠른 세포생존율의 감소를 보였으며 12시간 때에 최대의 차이를 보였으나 48시간에서는 cisplatin-방사선조사병합군의 세포생존율이 유의하게 낮았다. 결 론: 방사선조사로 반응성 산소족이 증가되면 PrxI 발현이 증가되었으며 반응성 산소족 청소제인 NAC의 전처치에 의하여 PrxI의 발현이 감소되었다. Cisplatin은 PrxI의 발현을 억제하여 반응성 산소족에 의한 세포손상을 증가시키며 방사선으로 인한 세포치사효과를 증가시켰다고 판단된다. 이상의 결과로 PrxI의 발현여부가 방사선조사나 cisplatin의 세포치사기전에 부분적으로 관여하고 있을 것이라고 생각한다.

화학적 허혈에 의해 손상된 마우스 간세포에 대한 hydrogen sulfide의 간세포 보호 효과 (Protection of Primary Cultured Mouse Hepatocytes from Chemical Hypoxia-induced Injury by Hydrogen Sulfide)

  • 본 연구는 화학적 허혈에 의해 손상된 마우스 간세포에서 hydrogen sulfide ($H_2S$)의 효과를 규명하기 위해 수행되었다. 본 연구에서 허혈 모방 화합물로 알려져 있는 cobalt chloride ($CoCl_2$)는 간세포 손상을 시간 및 농도 의존적으로 유의성 있게 증가 시켰다. $CoCl_2$에 의한 간세포 손상은 Sodium sulfide (NaHS, $H_2S$ 공여제)의 전처리에 의해 유의적으로 감소 되었다. $CoCl_2$는 세포 내 활성산소(reactive oxygen species, ROS)의 농도를 증가시켰으며, 이는 NaHS 및 N-acetyl-cysteine (NAC, a ROS 제거제)에 의해 감소하였다. 또한, $CoCl_2$에 의해 증가된 p38 MAPK 인산화가 NaHS 및 NAC에 의해 억제되었다. $CoCl_2$에 의해 증가된 Bax/Bcl-2 비율은 NaHS, NAC 및 SB 203580 (p38 MAPK 저해제)에 의해 차단되었으며, $CoCl_2$에 의해 유발된 간세포의 손상 또한 NaHS, NAC 및 SB 203580의 전처리에 의해 억제되었다. NaHS는 $CoCl_2$에 의해 증가된 COX-2의 발현을 억제하였다. 또한, NaHS의 효과와 유사하게 $CoCl_2$에 의해 증가된 COX-2의 발현이 NAC에 의해 억제되었다. 더욱이, NS-398 (COX-2 선택적 억제제)는 $CoCl_2$에 의한 Bax/Bcl-2 비율의 증가를 억제하였을 뿐 아니라, 간세포의 세포 손상 또한 억제하였다. 결론적으로, $H_2S$는 초대배양 된 마우스 간세포에서 $CoCl_2$에 의해 유발된 간세포의 손상을 ROS에 의해 유발된 p38 MAPK 및 COX-2 경로의 활성화를 억제함으로써 세포보호효과를 수행하는 것을 알 수 있었다.

인후두 역류질환에서 N-Acetyl Cysteine의 증상 개선 효과에 대한 다기관 평가 (Multicenter Evaluation on the Efficacy of N-Acetyl Cystine in Relieving the Symptoms of Laryngopharyngeal Reflux Disease)

  • Background and Objectives : Laryngopharyngeal reflux disease (LPRD) is relatively common disease. N-acetyl cysteine (NAC) has both mucolytic and antioxidant effect, also may be beneficial in inflammatory airway diseases. The purpose of this study was to evaluate the efficacy and safety of inhaled NAC therapy in LPRD. Materials and Method : We retrospectively reviewed the medical records of 525 LPRD patients at 12 medical centers. Finally 401 patients subjected to inhaled NAC therapy for 2 months were enrolled in the study. We analyzed the change of Reflux Symptom Index (RSI) and Reflux Finding Score (RFS) after use of NAC for 4 weeks and 8 weeks in addition to the patient's compliance of the treatment. Results : The RSI score significantly decreased from $19.87{\pm}6.34$ to $12.78{\pm}6.93$ after 4 weeks and to $10.65{\pm}7.47$ after 8 weeks. The RFS score also significantly decreased from $9.29{\pm}3.4$ to $7.17{\pm}3.41$ after 4 weeks and to $6.1{\pm}3.73$ after 8 weeks (p<0.05). During the treatment periods, 42 patients (10.4%) reported to have 80 episodes of discomfort. Throat discomfort (33%) and nausea (28%) were most common complaints, but the duration of discomfort was usually less than 4 weeks. Conclusion : Inhaled NAC treatment is highly effective for the reduction of both subjective and objective findings in LPRD patients. This study will provide the evidence of new treatment option for patients with LPRD. However, further studies will be needs to assess the real effect of inhaled NAC therapy as a standard treatment regimen of LPRD.

Toxicological Studies on the Essential Oil of Eugenia caryophyllata Buds

  • The essential oil (EC-oil) obtained from the buds of Eugenia caryophyllata (Myrtaceae) was examined for its free radical-scavenging activity, cytotoxicity, and in vivo toxicity. To find the xenobiotic properties of EC-oil, serum thiobarbituric acid reactive substances (TBARS) level and hepatic drug-metabolizing enzyme activities were measured. It was found that EC-oil displayed xenobiotic properties like bromobenzene. The cytotoxicities of eugenol and of the EC-oil were greatly attenuated by the sulfhydryl-containing N-acetyl-L-cysteine (NAC), suggesting that eugenol was susceptible to nucleophilic sulfhydryl. In addition, eugenol also showed potent free radical-scavenging activity in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Moreover, methyleugenol considerably exhibited less cytotoxicity and less potent free radical-scavenging activity than eugenol, and the cell viability of the methyleugenol was more increased with NAC treatment than the eugenol. These results indicate that the phenolic OH in eugenol may play a crucial role in both cytotoxicity and free radical-scavenging activity. The fashion on oxidative stress and hepatic drug-metabolizing enzyme activities of eugenol resembled those of bromobenznene.

Transition Metal Induces Apoptosis in MC3T3E1 Osteoblast: Evidence of Free Radical Release

  • Transition metal ions including $Se^{2+},\;Cd^{2+},\;Hg^{2+}\;or\;Mn^{2+}$ have been thought to disturb the bone metabolism directly. However, the mechanism for the bone lesion is unknown. In this study, we demonstrated that MC3T3E1 osteoblasts, exposed to various transition metal ions; selenium, cadmium, mercury or manganese, generated massive amounts of reactive oxygen species (ROS). The released ROS were completely quenched by free radical scavengers-N-acetyl cysteine (NAC), reduced glutathione (GSH), or superoxide dismutase (SOD). First, we have observed that selenium $(10\;{\mu}M),$ cadmium $(100\;{\mu}M),$ mercury $(100\;{\mu}M)$ or manganese (1 mM) treatment induced apoptotic phenomena like DNA fragmentation, chromatin condensation and caspase-3-like cysteine protease activation in MC3T3E1 osteoblasts. Concomitant treatment of antioxidant; N-acetyl-L-cysteine (NAC), reduced-form glutathione (GSH), or superoxide dismutase (SOD), prevented apoptosis induced by each of the transition metal ions. Catalase or dimethylsulfoxide (DMSO) has less potent inhibitory effect on the apoptosis, compared with NAC, GSH or SOD. In line with the results, nitroblue tetrazolium (NBT) stain shows that each of the transition metals is a potent source of free radicals in MC3T3E1 osteoblast. Our data show that oxidative damage is associated with the induction of apoptosis in MC3T3E1 osteoblasts following $Se^{2+},\;Cd^{2+},\;Hg^{2+}\;or\;Mn^{2+}$ treatment.

생강 페이스트의 저장 안정성 (Storage Stability of Ginger(Zingiber officinale Roscoe) Paste)

  • Storage stability of ginger paste product was investigated from the standpoint of the inhibition of nonenzymatic browning and loss of gingerol contents. For the experimentations, control, 0.04% of N-acetyl-L-cysteine in ginger paste(NAcCys), and combination treatment of NAcCys, 0.92 of water activity and 6.30 of pH in ginger paste (mixed treatment) were stored at 3$0^{\circ}C$ for 40 days and analyzed for browning and gingerol contents. In addition the changes in sugars, organic acids, ascorbic acids, amino acids, and sensory quality were determined. The results revealed that the mixed treatment agent was effective in preventing both nonenzymatic browning and loss of gingerol contents. The inhibition by combination treatment might be resulted from the control of radical formations by sulfhydryl groups of NAcCys and the increase of diffusion resistance in lower water activity. Browning development and total gingerol contents were found to be correlated to some physicochemical characteristics of ginger paste; that is, browning development to amino acid and color value in sensory evaluation, and total gingerol contents to flavor in sensory evaluation.

A Cell-Based Assay System for Monitoring NF-$\kappa$B Activity in Human Epidermal Keratinocytes: A Screening Tool of the Antioxidants and Anti-inflammatories for Dermatological Purpose

  • A cell-based assay system for monitoring NF-$textsc{k}$B activity was developed to determine the influence of activated NF-$textsc{k}$B in human HaCaT cells. The pNF-$textsc{k}$B-SEAP-NPT plasmid that permits expression of the secreted alkaline phosphatase (SEAP) reported gene in response to the NF-$textsc{k}$B activity and contains neomycin phosphotransferase (NPT) gene for the geneticin resistance in host cells was constructed and transfected into human keratinocyte cell line HaCaT. Human HaCaT transfectant cells secreted the SEAP enzyme into the culture medium in a time-dependent manner until 72h. NF-$textsc{k}$B activities were measured in the SEAP reporter gene assay using a fluorescent detection method. The treatment of HaCaT cell transfectants with known antioxidants [e.g., N-acetyl-L-cysteine and vitamin C] showed inhibition of NF-$textsc{k}$B activity in a time-and concentration-dependent manner. The phorbol 12-myristate 13-acetate (PMA) known as a stimulator of NF-$textsc{k}$B expression demonstrated that it increased NF-$textsc{k}$B activity in a time- and concentration-dependent manner. This assay system could be used to determine the quantitative measurement of NF-$textsc{k}$B activity in the human skin and allow the screening of anti-inflammatory agents from various synthetic chemicals and natural products for dermatological purpose. In Vitro Antioxidant Activity Profiles of ${\beta}$-Glucans Isolated from Yeast Saccharomyces cerevisiae and Mutant Saccharomyces cerevisiae IS2

  • To explore the possible usefulness of ${\beta}$-glucans as natural antioxidants, the antioxidant profiles of ${\beta}$-glucan, extracted from Saccharomyces cerevisiae KCTC 7911, and water soluble and insoluble mutant ${\beta}$-glucan, isolated from yeast mutant S. cerevisiae IS2, were examined by five different in vitro evaluation methods: lipid peroxidation value (POV), nitric oxide (NO), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, reducing power, and ${\beta}$-carotene diffusion assay. The antioxidant activities of all ${\beta}$-glucans evaluated in POV test were comparable to or better than that of the known antioxidant, vitamin C. Remarkably, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan possessed 2.5-fold more potent activity than vitamin C at a dosage of 2 mg. Although vitamin C showed 100-fold greater activity than all ${\beta}$-glucans in NO and DPPH tests for measuring the radical scavenging capacity, all ${\beta}$-glucans revealed higher radical scavenging activity than the known radical scavenger, N-acetyl-L-cysteine (NAC), in DPPH test. The water insoluble mutant ${\beta}$-glucan had 2.6- and 5-fold greater antioxidative activity than water soluble ${\beta}$-glucan in NO and DPPH tests, respectively, showing that all ${\beta}$-glucans were able to scavenge radicals such as NO or DPPH. While all ${\beta}$-glucans revealed lower antioxidant profiles than vitamin C in both reducing power activity and ${\beta}$-carotene agar diffusion assay, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan did show a marginal reducing power activity as well as a considerable ${\beta}$-carotene agar diffusion activity. These results confirmed the potential usefulness of these ${\beta}$-glucans as natural antioxidants.

Antiproliferative effect of gold(I) compound auranofin through inhibition of STAT3 and telomerase activity in MDA-MB 231 human breast cancer cells

  • Signal transducer and activator of transcription 3 (STAT3) and telomerase are considered attractive targets for anticancer therapy. The in vitro anticancer activity of the gold(I) compound auranofin was investigated using MDA-MB 231 human breast cancer cells, in which STAT3 is constitutively active. In cell culture, auranofin inhibited growth in a dose-dependent manner, and N-acetyl-L-cysteine (NAC), a scavenger of reactive oxygen species (ROS), markedly blocked the effect of auranofin. Incorporation of 5-bromo-2'-deoxyuridine into DNA and anchorage-independent cell growth on soft agar were decreased by auranofin treatment. STAT3 phosphorylation and telomerase activity were also attenuated in cells exposed to auranofin, but NAC pretreatment restored STAT3 phosphorylation and telomerase activity in these cells. These findings indicate that auranofin exerts in vitro antitumor effects in MDA-MB 231 cells and its activity involves inhibition of STAT3 and telomerase. Thus, auranofin shows potential as a novel anticancer drug that targets STAT3 and telomerase.

Constituents of the Essential Oil of the Cinnamomum cassia Stem Bark and the Biological Properties

  • CC-MS analysis on the essential oil (CC-oil) of Cinnamomum cassia stem bark led to the identification of cinnamaldehyde (CNA, 1), 2-hydroxycinnamaldehyde (2-CNA), coumarin (2), and cinnamyl acetate. The major volatile flavor in CC-oil was found to be 2-CNA. Coumarin was first isolated from this plant by photochemical isolation and spectroscopic analysis. CNA and CC-oil showed potent cytotoxicity, which was effectively prevented by N-acetyl-L-cysteine (NAC) treatment. Intraperitoneal administration with CNA considerably decreased malondialdehyde (MDA) formation and glutathione S-transferase activity in rats. These results suggest that CC-oil and CNA can regulate the triggering of hepatic drugmetabolizing enzymes by the formation of a glutathione-conjugate.
