This study was conducted to evaluate the effects of different levels of crude protein (CP) and protease on nitrogen (N) utilization, nutrient digestibility, and growth performance in growing pigs. A total of six crossbred ([Landrace × Yorkshire] × Duroc) barrows were individually accepted in 1.2 m × 0.7 m × 0.96 m stainless steel metabolism cages. The pigs (average initial body weight of 27.91 ± 1.84 kg) randomly assigned to six diets with six weeks (6 × 6 Latin square design). The experiment was carried out in an environment with a temperature of 23 ± 1.5℃, a relative humidity of 83 ± 2.3% and a wind speed of 0.25 ± 0.03 m/s. The dietary treatments were arranged in a 2 × 3 factorial design with two levels of CP (15.3% or 17.1%) and three levels of protease (0 ppm, 150 ppm, or 300 ppm). The average daily gain and gain to feed ratio (G:F) tended to increase (p = 0.074) with increasing amounts of protease. The low CP level diet reduced (p < 0.050) urinary and fecal N concentrations, the total N excretion in feces, and increased (p < 0.050) N retention. Different protease levels in the diet did not affect (p > 0.05) at N intake, but supplementation of the diets with 300 ppm protease decreased (p < 0.050) the N concentration in urine and feces and tended to increase (p = 0.061) the percentage of N retention retained of the total N intake. The dietary CP level did not affect (p > 0.050) the apparent total tract digestibility (ATTD) of dry matter, digestible energy (DE), and metabolic energy (ME), but diet supplementation with 300 ppm protease showed higher (p < 0.050) ATTD of DE and ME than in the protease-free diet. Therefore, a low protein diet with protease could improve the utilization of nitrogen, thereby reducing the negative effect of N excretion into the environment while maintaining or increasing growth performance compared to a high protein diet.
A growth experiment was conducted to evaluate the nutritivie values of supplemental lysine and methionine in broiler chicks. Two types of L-lysine, liquid and powder type, and DL-methionine were added to the diets at different levels of dietary protein with two growth phases, 0-3 weeks and 4-6 weeks named starter and grower, respectively. Six hundred seventy two chicks were allotted in 14 treatments; 3 controls by dietary CP level (starter-grower) with CP 23-21%, CP 21-19% and CP 20-18, 8 groups of liquid and powder lysine supplementation of 0.1, 0.2, 0.3 and 0.4%, and 3 groups of lysine and methionine supplementation. Body weight, feed intake, and excreta were measured and analyzed to determine growth performance, amino acid digestibilities, and the quantity of excreted nitrogen in feces. Chicks fed CP 23-20 with 3,200 ME kcal showed significantly better growth performance than those fed CP 21-18 for 6 weeks. The supplementation of 0.2% of either type of lysine to CP 21-19 diet improved weight gain and feed efficiecy to the extent that CP 23-21 diet was fed. Physical type of lysine did not affect chick's growth and amino acid digestibilities of the diets. The level of CP in the diet significantly affected nitrogen excretion in feces. Supplementation of lysine and methionine to CP 21-18 diet reduced fecal nitrogen by 10% compared to CP 23-21 diet. It was confirmed that 0.2% of supplemental lysine to the broiler diet spared the dietary protein by 3%, and also reduced nitrogen excretion in feces by 10%.
An experiment was conducted to measure the effect of crude phytase supplementation on the growing performance, blood concentrations of some minerals and tibia characteristics of broiler chickens. Day-old 240 male broiler chickens (Avian) were randomly allotted to four treatments. There were six replicates per treatment, and ten chicks per replicate. Treatments consisted of two levels of crude phytase (0 and 600U/kg) made from Aspergillus ficuum and two levels of non-phytate P (0.45 and 0.35% NPP for the starter period, and 0.35 and 0.25% NPP for the grower period), making the experiment 2$\times$2 factorial. The starter period was from hatch to 21 d of age, and grower period was from 22 to 35 d of age. Feed intake and weight gain of chicks fed diet containing phytase were higher(P〈0.05) than those of chicks fed diets without phytase, however, no differences was found in feed/gain. mortality, and nutrient availabilities regarding the phytase supplementation. Chickens fed diets with low NPP and phytase excreted lower P than did birds fed diets containing normal NPP without phytase. The level of NPP and phytase did not affect N excretion. The Ca availability was increased by feeding low NPP diet. Dietary phytase increased the availabilities of P and Mg, but decreased those of Fe and Zn. There was interactions between dietary NPP level and phytase addition on mineral availability. Tibia was lighter and shorter in low NPP groups, and heavier in phytase treated groups. The tibial contents of Ca, P and Mg decreased in low NPP treated groups, but increased in phytase treated groups. The ash content of tibia of chickens fed diet with phytase was higher than that of birds fed diets without phytase. These data suggest that the crude phytase supplementation to broiler diets containing low NPP level improves growth performance and mineral availability and, reduces fecal P excretion.
This study was done for the determination and excretion profile of adrenosterone and its metabolites in human urine using both liquid chromatography with atmospheric pressure chemical ionization mass spectrometry and gas chromatography with mass spectrometry. Adrenosterone and its two metabolites were detected in human urine after administration a healthy volunteer with 75 mg of adrenosterone. We found that adrenosterone-M1 ($C_{19}H_{26}O_3$) was a reduction and adrenosterone-M2 ($C_{19}H_{26}O_4$) was a hydroxylation at C-ring, which did not know the exact position of the C-ring. The adrenosterone parent was detected by GC/TOF-MS, but not detected by LC/APCI/MS because of low intensity. Adrenosterone and its two metabolites were excreted as their glucuronided fractions. The recovery of this method ranged from 100.7 to 118.4% and the reproducibility and accuracy test were 85.5 to 112.0% and 1.1 to 8.4%, respectively. The excretion studies showed that adrenosterone and its metabolites were detectable in human urine during a 48 h period after oral administration, with maximum level of excretion at 4.1 h. The glucuro-/sulfaconjugated ratio of adrenosterone, M1 and M2 was 0.73 ${\pm}$ 0.03, 0.96 ${\pm}$ 0.06 and 0.89 ${\pm}$ 0.03 (n = 6), respectively. The amounts of adrenosterone excreted in urine were 14.75 ng for 48 h. Also, the maximum level of androsterone and 11$\beta$-hydroxy androsterone, which were endogenous steroids, were reached 4.1 h after the oral administration of adrenosterone.
Dimethylamine (DMA) is the immediate precursor of carcinogenic N-nitrosodimethylamine (NDMA). In vitro and in vivo experiments using whole strawberries, and garlic and kale juices were conducted to determine concentrations of DMA and trimethylamine (TMA) in foods and urine. Experimental diets [an amino-rich diet as nitrosatable precursors in combination with added nitrate-containing drinking water without (TD1) or with whole strawberries or garlic or kale juices (TD2, TD3 and TD4, respectively), or a diet of low in nitrate and amino (TD5) were incubated in simulated saliva and gastric juices at 37$^{\circ}C$ for 1 hour. We also studied the urinary excretion of DMA and TMA after consumption of the experimental diets (TD1~TD5). Urine samples were obtained for 18 hrs after consumption of experimental diets and concentrations of DMA and TMA were measured in the digested diet and urine. The DMA concentration after incubation in experimental diets (TD1~TD5) was 4.7$\pm$0.3, 6.7 $\pm$0.2, 7.9$\pm$0.2, 7.1$\pm$0.2 and 0.3$\pm$0.1 mg/kg, respectively. Urinary excretion of DMA (TD1~TD5) was 22.0$\pm$5.0, 28.3$\pm$4.3, 29.2$\pm$4.1, 27.4$\pm$4.5 and 20.4$\pm$3.1 mg/18 hr, respectively. Consumption diets with added strawberries or juices of kale or garlic increased urinary TMA and DMA, suggesting that those precursors were excreted and not converted to the carcinogen, NMDA.
The sea squirt Halocynthia roretzi is mainly cultured in Tongyeong, Southern coastal area of Korea. This study presents the physiological rates of respiration, excretion, feeding and assimilation efficiency of the sea squirt Halocynthia roretzi to analyze the SFG(scope for growth) and net growth efficiency, determined during 2007. Oxygen consumption and nitrogen excretion rates increased with a rise in temperature during the summer period whereas feeding rates decreased. The O:N ratio was high during winter(October to February). Assimilation efficiency showed an annual average of 75.4% during the experimental period, except during a period of elevated temperature in July to September(average $25^{\circ}C$). Net growth efficiency($K_2$) was 8.7 to 64.2% except for May to September, when temperature increased at the aquaculture farm. SFG was negative from May to September, reflecting high temperatures and low feeding rates during this period; its highest positive values occurred during winter.
This study was conducted to investigate the effects of chitosan, chitosan oligomer and beef tallow of different levels on lipid metabolism in rats, Seventy male rats of the Sprague-Dawley strain were blocked into 10 groups according to body weight , and were raised for 4 weeks. Dietary fat levels were 20% en and 40% en, and chitosan and chitosan oligomer were given at levels of 05, 3%, and 5%(wt/wt) of diet. The results are summarized as follows, chitosan oligomer supplement decreased serum total lipid, as chitosan 양, and exhibited a tendency to decrease serum total cholesterol. Chitosan oligomer supplement tended to increase the HDL cholesterol ; total cholesterol ration as chitosan did. Liver total lipid and triglyceride concentration were lower in high fat groups than in low fat groups. Liver total lipid concentrations was decreased slightly by chitosan and sigfnificantly by chitosna ligomer. Epididymal fat pad total lipid. total cholesterol and triglyceride concentration were slightly decreased by chitosan and chitosan oligomer supplement, especially in high fat groups. fat absorptivity was decreased by low fat level, chitisan and chitosan oligamer supplement. Fecal excretion of total lipid and triglyceride were increased by high fat level, chitosan and chi샌무 oligomer supplement. However, fecal excretion of total cholesterol was increased by high fat level and chitosan supplement. This indicated that chitosan and chitosan oligomer were effective in interfering with lipid and triglyceride absorption, In conclusion, chitosan oligomer at levels of 3% and 5% has more effective lipid, cholesterol and triglyceride lowering activity than chitosan.
Each parching Anemarrhenae Rhizoma with 25% ethanol and normal saline has been used for anti-inflammatory, expectorant, antipyretic, sedative and diuretic, and so on. In order to investigate the differences of referential efficacy about each fractionated part of the aqueous extracts, pharmacological studies were carried out. The results of studies were summerized as follows : 1. By the administration of 25% ethanol treated preparation (F-I-1), the increase in urinary volume and $Na^+$ extraction was significantly recognized. 2. The excretion of electrolyte $Na^+$ by saline solution treated preparation (F-II-1) was significantly recognised in normal rats. 3. The increase of urinary volume and urinary $Na^+$ and $Cl^-$ excretion by F-I-1 and n-butanol insoluble parts of F-I-1 (F-I-3) was significantly recognized in $HgCl_2-induced$ acute renal failure of rats. 4. The increase of plasma $Na^+$ by F-I-1 and urea nitrogen by F-I-1 and F-I-3 in acute renal failure of rats was significantly recognized. 5. The increase of urinary volume by F-I-1 and F-I-3 was recognised in mice.
This study was to investigate the effects of chitin, chitosan , NOCC and cellulose on cadmium toxicity and lipid metabolism in rats. Fifty male rats of Sprague-Dawley strain weighing 155$\pm$17g were divided into 10 groups according to body weight, and were raised for 5 weeks. Levels of 0 and 400 ppm of cadmium chloride were in the diets. And chitin, chitosan , NOCC and cellulose were given at the level of 0 and 4%(w/w) of the diets. Chitosan and NOCC decreased Cd concentration of liver and kidney, and increased the fecal excretion of Cd. Although cellulose was less effective, cellulose also decreased Cd concentration of liver and increased fecal Cd excretion . However chitin had little effect on alleviating Cd toxicity. In addition, chitosan and NOCC lowered total lipid, cholesterol and triglyceride contents in serum and liver by decreasing the lipid absorption. The lowering of cholesterol especially had a remarkable effect. Although it was less effective than chitosan and NOCC, chitin decreased the lipid absorption ratio and lowered cholesterol contents in serum and liver. In conclusion , chitosan and NOCC showed effects of decreasing the absorption of Cd and lipid.
Objective: This trial was performed to examine the effects of ruminally degradable starch (RDS) levels in total mixed ration (TMR) with low corn-based starch on the milk production, whole-tract nutrient digestibility and nitrogen balance in dairy cows. Methods: Eight multiparous Holstein cows (body weight [BW]: $717{\pm}63kg$; days in milk [DIM]: $169{\pm}29$) were assigned to a crossover design with two dietary treatments: a diet containing 62.3% ruminally degradable starch (% of total starch, low RDS) or 72.1% ruminally degradable starch (% of total starch, high RDS). Changes to the ruminally degradable levels were conducted by using either finely ground corn or steam-flaked corn as the starch component. Results: The results showed that dry matter intake, milk yield and composition in dairy cows were not affected by dietary treatments. The concentration of milk urea nitrogen was lower for cows fed high RDS TMR than low RDS TMR. The whole-tract apparent digestibility of neutral detergent fiber, acid detergent fiber and crude protein decreased, and that of starch increased for cows fed high RDS TMR over those fed low RDS TMR, with no dietary effect on the whole-tract apparent digestibility of dry matter and organic matter. The proportion of urinary N excretion in N intake was lower and that of fecal N excretion in N intake was higher for cows fed high RDS TMR than those fed low RDS TMR. The N secretion in milk and the retention of N were not influenced by the dietary treatments. Total purine derivative was similar in cows fed high RDS TMR and low RDS TMR. Consequently, estimated microbial N flow to the duodenum was similar in cows fed high RDS TMR and low RDS TMR. Conclusion: Results of this study show that ruminally degradable starch levels can influence whole-tract nutrient digestibility and nitrogen balance in dairy cows fed low corn-based starch diets, with no influence on performance.
