A Study on the Dry Matter Yield and Nutritive Values of Wild Korean Lespedeza (Lespedeza stipulacea Maxim.)

  • This study was conducted to evaluate the dry matter (DM) yield, quality and utilization of nutrients in Korean lespedeza (Lespedeza stipulacea Maxim.) swards as a roughage source. DM yield in various stages, chemical composition, and DM digestibility were observed, and DM intake, digestibility and utilization of nitrogen and energy by Korean native goat were determined. Experimental diets include three treatments: mixture hay (MH) 100%, MH (80%)+wild Korean lespedeza hay (LH) 20% and MH (60%)+LH (40%). As growth stage advanced, the DM yield of Korean lespedeza tended to increase significantly (p<0.05). The mean DM yield of all growing stages was observed as 7,336 kg/ha. Crude protein (CP) content of Korean lespedeza tended to decrease, but fibrous contents tended to increase when the growth stage advanced. The tannin content of Korean lespedeza was the highest at the bud stage (64.7 mg/g). However, it showed a tendency to decrease as the growth stage advanced, and thus it was the lowest at the ripe seed stage (26.8 mg/g) (p<0.05). With increasing level of LH, voluntary DM intake by Korean native goats slightly increased, but no differences were observed between diets. The digestibility of DM, cellular constituents and NDF was slightly higher in LH containing diets than that of MH 100% diet (p<0.05). There was no significant difference in apparently digested N% among all diets, but retained N% and retained N% of the absorbed were higher in MH (60%)+LH (40%) than those of other diets (p<0.05). A difference in the utilization of energy was not detected. In conclusion, Juvenile LH improved the digestibility of nutrients, presumably due to its high CP content and low fibrous compound. Especially, tannin in LH did not affect in DM intake, but increased the nitrogen utilization of Korean native goats. Accordingly, it could be suggested that Korean lespedeza has a potential to be provided as a roughage source for Korean native goats.

  • N-nitrosodimethylamine (NDMA) is a typical nitrogen disinfection by-product, which has posed a potential threat to human health during drinking water disinfection. Because of the well-known effects of mutagenesis, carcinogenesis and teratogenesis, the high detection rate in water engineering systems (such as coagulation, membrane filtration and biological systems), and difficulty to remove, it has received wide concern in the field of water engineering systems. The NDMA is a low molecular weight hydrophilic organic substance, which is difficult to remove. Also, the mechanism for NDMA formation is also recognized to be complex, and many steps still needed to be further evaluated. Therefore, the mechanistic knowledge on NDMA formation potential and their removal processes is of particularly interest. Few papers summarize the occurrence and control of NDMA in water engineering systems. It is for this reason that the content of this paper is particularly important for us to understand and control the amount of NDMA thus reducing the threat of disinfection by-products to drinking water. Four parts including the mechanisms for the NDMA formation potential, the factors affecting the NDMA formation potential, the technologies for removal of NDMA are summarized. Finally, some definite suggestions are given.

Evaluation of the Nutritional Composition and Quality Traits of Rabbit Meat (토끼고기의 영양성분 및 품질특성 평가)

  • This study evaluated the nutritional composition and quality traits of rabbit meat as compared to chicken meat. Samples of loin (M. longissimus dorsi) and breast meats were collected from rabbit and chicken carcasses, respectively. The meats were then analyzed for the proximate composition, collagen and energy contents, fatty acid composition, myoglobin and heme iron contents, pH value, water-holding capacity (WHC), cooking loss, meat color, Warner-Bratzler shear force (WBSF) value, and texture profile. Compared to chicken breast meat, lower (p<0.05) protein content and higher (p<0.05) ash and collagen contents were obtained in rabbit loin meat. Rabbit meat remarkably had higher (p<0.05) total polyunsaturated fatty acids (PUFA) and linolenic acid contents and lower (p<0.05) n-6/n-3 PUFA ratio as compared to chicken meat. The pH value and WHC were lower (p<0.05) in rabbit meat than in chicken meat (p<0.05). Rabbit meat exhibited lower (p<0.05) L* value and higher (p<0.05) a* and b* values compared to chicken meat (p<0.05). The WBSF value, hardness, and gumminess were higher (p<0.05) in rabbit meat than in chicken meat (p<0.05). These findings suggest that rabbit meat has higher essential n-3 PUFA, darker color, and firmer texture as compared to chicken meat.

Effects of Feeding Different Protein Supplements on Digestibility, Nitrogen Balance and Calcium and Phosphorus Utilization in Sheep

  • Two metabolism trials were conducted with 24 wether lambs to investigate the effects of feeding crab meal and other protein supplements on N utilization, digestibility and Ca and P balance in sheep. The lambs (avg. BW, 25 kg) were randomly allotted to eight diets in each of two trials. The supplements were: i) none, negative control (NC); ii) soybean meal (SBM), control; iii) supplement based on industrial byproducts of both plant and animal origin (IPA); iv) experimental supplement based on byproducts of animal origin (ESA); v) hydrolyzed supplement No 4. (HESA); vi) commercial supplement based on animal protein (CS), $Pro-Lak^{(R)}$ vii) crab meal (CM); and viii) urea (U). The supplements supplied 33% of the total dietary N (CP, 9.8%; DM basis). Lambs fed the NC diet had lower (p<0.05) DM and OM digestibility. Lower (p<0.05) apparent absorption of N was recorded for the lambs fed the HESA and NC diets. Sheep fed CM had lower Ca absorption compared to SBM. Highest (p<0.05) P absorption was observed for lambs fed CS and CM and lowest for U and NC diets. Sheep fed CM had higher (p<0.05) total VFA concentration (65.7 ${\mu}mol/ml$), compared to those fed ESA, CS, and NC diets (47.3, 49.8, and 49.5 ${\mu}mol/ml$, respectively). Highest (p<0.05) ruminal $NH_3$ N (29.6 mg/dl) was observed in lambs fed the U diet, while those fed the NC diet had the lowest (p<0.05) average value (7.66 mg/dl). Lambs fed the U diet had the highest (p<0.05) blood urea N (10.67 mg/dl). The present study showed that N utilization of diets supplemented with experimental supplements based on feather meal and blood meal; commercial supplement based on animal protein, $Prolak^{(R)}$ supplement based on plant protein and blood meal; and crab meal are comparable with that of soybean meal.

EFFECT OF URINARY NITROGENOUS COMPOUNDS ON THE ENERGY METABOLISM OF BIRD

  • In order to study the relationships between the contents of urinary nitrogenous compounds and energy utilization of bird, the sum of nitrogen contents of uric acid, ammonia, creatine and urea voided in excreta was estimated as the urinary nitrogen (UN) in 13-33 day-old fed or fasted White Leghorn male chicks. Energy retention and heat production of birds were determined by comparative slaughter studies. 2.75 mg of endogenous urinary nitrogen (EUn) and 2.19 mg of uric acid was excreted constantly per kJ heat production in fasted bird. One mg of UN was proportionated to 32.26 J (r = 0.999, n = 8) of the urinary energy (UE) in fed and 32.97 J (r = 0.9998, n = 8) of the endogenous urinary energy (UEn) in the fasted bird. Also relationships between 1 mg of uric acid and 38.95 J of UE (r = 0.998, n = 8) or 38.97 J of UEe (r = 0.996, n = 8) were significant (p<0.01). The EUn (r = 0.997, n = 4), uric acid (r = 0.995, n = 4) and metabolic fecal energy (FEm) plus UEe (r = 0.961, n = 4) were increased with the increase of body weight (g/bird). Metabolic fecal nitrogen (MFn) or energy (FEm), EUn and UEe per unit diet were not influenced by the age of day or body weight. The results indicated that energy and protein utilization of bird can be approximated by the relationships among urinary nitrogen, urinary energy, uric acid content in excreta and body weight of bird.

Effect of Replacing Rolled Corn with Potato Pulp Silage in Grass Silage-based Diets on Nitrogen Utilization by Steers

  • Three Holstein steers fitted with ruminal and duodenal cannulae were fed grass silage-based diets supplemented with potato pulp silage as a substitute for rolled corn at levels of 0%, 50% and 100% on a DM basis in a $3{\times}3$ Latin square design to investigate the effect of potato pulp silage on nitrogen (N) utilization in ruminants. Organic matter (OM) intake, and rumen and total tract digestibilities did not differ among treatment diets. Rumen and post-rumen starch digestibilities were similar among treatments, although starch intake decreased (p<0.01) with potato pulp supplementation. There were no significant differences (p>0.05) in ruminal N utilization and non-ammonia N supply to the duodenum of steers fed grass silage supplemented with potato pulp silage as a substitute for rolled corn. There were no treatment differences (p>0.05) in rumen pH, volatile fatty acid (VFA) concentration or the molar percentages of acetate and propionate. The ammonia-N concentration in rumen fluid tended to decrease (p<0.1) when rolled corn was substituted with potato pulp silage. Ether extract intake and post-ruminal digestibility significantly (p<0.01) decreased in steers fed diets containing potato pulp silage. Concentrations of total cholesterol and phospholipids in serum markedly decreased (p<0.01) with potato pulp silage supplementation without adversely affecting liver function. These data suggested that potato pulp silage has a similar value as rolled corn as an energy source for rumen microorganisms.

Calorie, Cholesterol, Collagen, Free Amino Acids, Nucleotide-related Compounds and Fatty Acid Composition of Hanwoo Steer Beef with 1++ Quality Grade (1++ 등급 거세한우의 부위별 칼로리, 콜레스테롤, 콜라겐, 유리아미노산, 핵산관련물질 및 지방산조성)

  • This study was conducted to investigate the compositional properties by different cut of Hanwoo steer beef. 10 cuts [Abjin (short plate), Bosup (top sirloin), Cheggt (striploin), Dngsim (Ioin), Guri (chuck tender), Hongduke (eye of round), Moksim (chuck roll), Sulgit (bottom round), Udoon (top round), Yangji (brisket)] were prepared from 10 Hanwoo steers ($24{\sim}28$ mon old) were used for the experiment. Calorie contents were $2,205{\sim}3713.20\;cal/g$ and loin, striploin, short plate were significantly higher than the other cuts (p<0.05). Loin was highest in cholesterol content, whereas top round and eye of round were lowest among 10 cuts (p<0.05). There were no significant differences in soluble and insoluble collagen contents although chuck tender, chuck roll and bottom round contained significantly higher total collagen and loin and striploin contained significantly lower total collagen contents than the other cuts (p<0.05). The result from the analysis of free amino acids showed that glutamate and alanine contents were $275.99{\sim}536.04\;mg/100g$, $130.63{\sim}175.23\;mg/100g$, respectively and followed by arginine ($7.00{\sim}10.49\;mg/100g$), phenylalanine ($4.70{\sim}13.88\;mg/100g$), and leucine ($5.98{\sim}10.26\;mg/100g$) in 10 cuts. $1^{++}$ grade Hanwoo steer beef had significantly higher IMP (Inosine monophosphate) and inosine in eye of round (30.04% and 6.25%) and top round (33.44% and 6.28%), while hypoxanthine contents were significantly higher in chuck tender than the other cuts (p<0.05). In fatty acid analysis, there were not significantly differences in fatty acid composition among 10 cuts only except for C16:1n7, C20:3n6, and C20:4n6 (p>0.05).

COMPARISON OF UTILIZATION OF CELLULOSE AND CORN DIETARY FIBER AS AN ENERGY SOURCE IN CHICKS

  • An experiment was conducted to investigate effects of fiber source on growth performance, N and neutral detergent fiber (NDF) digestibility, and utilization of energy in chicks fed an isocaloric low-energy diet from 7 to 21 days of age. Two fiber sources, cellulose and corn dietary fiber (CDF), were included in a diet at 10, 20 and 30% at the expense of kaolin, an inert diluent. The CDF contained 76.5% NDF consisting mainly of hemicellulose. The results showed that growth performance, N and NDF digestibility, dietary DE and ME values, energy deposition, and NE for production in birds fed CDF were inferior to those in birds fed cellulose. It can be concluded, from the present study, that chicks can utilize cellulose more efficiently than CDF up to a level of 30%.

Whitening Activities of Ethanol Extract from Polygonum amphibium L. (물여뀌 에탄올 추출물의 미백 효과)

  • The purpose of this study was to investigate the melanogenesis inhibiting activity of the ethanol extract from Polygonum amphibium L. Firstly, the n-hexane (Hx), chloroform ($CHCl_3$), ethyl acetate (EA), n-butanol (BuOH), and water (Water) fractions were isolated from the P. amphibium L. ethanol extract. The efficacy of melanogenesis was found to significantly decrease via the EA and BuOH fractions when compared to the control in B16F10 cells. EA particularly showed the lowest melanin content in B16F10 cells when compared to all the other extracts. Concentration-dependent inhibition of melanin synthesis was also observed in the EA fraction at concentrations below $50{\mu}g/ml$, which did not exhibit cytotoxicity in B16F10 cells. Notably, the expression of three key proteins (tyrosinase, tyrosinase-related protein-1 (TRP-1), and TRP-2), which are involved in melanogenesis, were significantly decreased via the EA fraction. EA also inhibited body pigmentation in vivo in a zebrafish model. Overall, we demonstrated melanogenesis suppression using the EA fraction from P. amphibium L., which could be a potential candidate for an antimelanogenesis agent.

Overexpression of Mutant Galactose Permease (ScGal2_N376F) Effective for Utilization of Glucose/Xylose or Glucose/Galactose Mixture by Engineered Kluyveromyces marxianus

  • Mutant sugar transporter ScGAL2-N376F was overexpressed in Kluyveromyces marxianus for efficient utilization of xylose, which is one of the main components of cellulosic biomass. K. marxianus ScGal2_N376F, the ScGAL2-N376F-overexpressing strain, exhibited 47.04 g/l of xylose consumption and 26.55 g/l of xylitol production, as compared to the parental strain (24.68 g/l and 7.03 g/l, respectively) when xylose was used as the sole carbon source. When a mixture of glucose and xylose was used as the carbon source, xylose consumption and xylitol production rates were improved by 195% and 360%, respectively, by K. marxianus ScGal2_N376F. Moreover, the glucose consumption rate was improved by 27% as compared to that in the parental strain. Overexpression of both wild-type ScGAL2 and mutant ScGAL2-N376F showed 48% and 52% enhanced sugar consumption and ethanol production rates, respectively, when a mixture of glucose and galactose was used as the carbon source, which is the main component of marine biomass. As shown in this study, ScGAL2-N376F overexpression can be applied for the efficient production of biofuels or biochemicals from cellulosic or marine biomass.
