Application Effect of the Controlled Release Fertilizer Applied on Seedling Tray at Seeding Time in Rice (벼 모판 파종동시처리 완효성비료 시용효과)

  • The optimal application rate of a controlled release fertilizer (CRF) on the growth, yield, and seeding time of rice grown on seedling trays was investigated. The experimental field was located at $37^{\circ}22^{\prime}10^{{\prime}{\prime}}N$ latitude and $127^{\circ}03^{\prime}85^{{\prime}{\prime}}E$ longitude in Hwaseong, Gyeonggi-do, Republic of Korea. The soil in the paddy field was a clay loam. The CRF used in the experiment contained $300g\;kg^{-1}$ of nitrogen, $60g\;kg^{-1}$ of phosphate, and $60g\;kg^{-1}$ of potassium, respectively. The CRF was applied at the rate of 0, 200, 300, 400, 500, and 600 grams on rice seedling tray compared with the field application based on soil testing (control), respectively. The CRF can be applied as single application(which can replace basal fertilizer application and two top dressing application) directly to the seedling tray, and showed the minimum release at the seedling period. Considering the plant growth, nitrogen use efficency and yield of rice, the optimal application rate of developed CRF was 500 g per seedling tray and the yield of rice at this application rate was $4.92{\sim}5.04Mg\;ha^{-1}$. The regression formula between the rice yield and application rates of CRF was as follows ; "$Y=0.0002{\chi}^2+0.0963{\chi}+411.6$($R^2$ : 0.9922) in 2010 and $Y=8E-6{\chi}^2+0.2723{\chi}+344.04$($R^2$:0.9864) in 2011, Y : Rice yield ($Mg\;ha^{-1}$), ${\chi}$ : Application rate (grams) of controlled release fertilizer". The optimum application rates of CRF per rice seedling tray by regression formula was 498 grams in 2010 and 513 grams in 2011.

Apoptotic Effect of Proso Millet Grains on Human Breast Cancer MDA-MB-231Cells Is Exerted by Activation of BAK and BAX, and Mitochondrial Damage-mediated Caspase Cascade Activation (기장 종자 유래 추출물의 인간 유방암 MDA-MB-231 세포에 대한 세포독성에 관련된 미토콘드리아 손상-의존적 아폽토시스 유도 효과)

  • To examine the antitumor effect of proso millet grains, whether proso millet grains exert apoptotic activity against human cancer cells was investigated. When the cytotoxicity of 80% ethanol (EtOH) extract of proso millet grains was tested against various cancer cells using MTT assay, more potent cytotoxicity was observed against human breast cancer MDA-MB-231 cells than against other cancer cells. When the EtOH extract was evaporated to dryness, dissolved in water, and then further fractionated by sequential extraction using four organic solvents (n-hexane, methylene chloride, ethyl acetate, and n-butanol), the BuOH fraction exhibited the highest cytotoxicity against MDA-MB-231 cells. Along with the cytotoxicity, TUNEL-positive apoptotic nucleosomal DNA fragmentation and several apoptotic responses including BAK/BAX activation, mitochondria membrane potential (Δψm) loss, mitochondrial cytochrome c release into the cytosol, activation of caspase-8/-9/-3, and degradation of poly (ADP-ribose) polymerase (PARP) were detected. However, human normal mammary epithelial MCF-10A cells exhibited a significantly lesser extent of sensitivity compared to malignant MDA-MB-231 cells. Irrespective of Fas-associated death domain (FADD)-deficiency or caspase-8-deficiency, human T acute lymphoblastic leukemia Jurkat cells displayed similar sensitivities to the cytotoxicity of BuOH fraction, excluding an involvement of extrinsic apoptotic mechanism in the apoptosis induction. These results demonstrate that the cytotoxicity of BuOH fraction from proso millet grains against human breast cancer MDA-MB-231 cells is attributable to intrinsic apoptotic cell death resulting from BAK/BAX activation, and subsequent mediation of mitochondrial damage-dependent activation of caspase cascade.

  • The purpose of the study is to collect basis data for to establish standard administrative processes of liquid fertilizer treatment. From this survey we could make out the key point of each step through a case of effective liquid manure treatment system in pig house. It is divided into six step; 1. piggery slurry management step, 2. Solid-liquid separation step, 3. liquid fertilizer treatment (aeration) step, 4. liquid fertilizer treatment (microorganism, recirculation and internal return) step, 5. liquid fertilizer treatment (completion) step, 6. land application step. From now on, standardization process of liquid manure treatment technologies need to be develop based on the six steps process.

Effect of Thresher Drum-Speed on the Quality of the Milled Rice (탈곡기의 급동 속도가 도정 손실에 미치는 영향)

  • It is understood that drum speed of threshers and the moisture content of paddy grains to be threshed, respectively, have a signific:mt effect upon rice recoveries. Threshing under an increased drum speed would give a high performance rate, which is the general practice in custom work threshing in association with the use of semiauto-t hreshers. In the connection, however, it may result in the promotion of grain cracks and brokens of the rice product after milling. No reference or determination for an opti mum drum speed of the thresher is made available for various grain moisture contents at the time of the threshing operation and for different rice varieties especially for the Tongil rice varieties. This study was Conducted to find out and determine effects of the drum speeds on grain losses. The grain loss was quantified in terms of recovery rates of rice grains after treatments. Samples of each of all treatments were taken from the grain sampling plate placed in the grain conveyor of threshers. The grain sample plate was specially provided for this experiment. The brown-rice, milling, and head-rice recJveries were tes ted in the laboratory mill, respectively. Two rice varieties, Akibare and Suweon 251, each with five levels of different moist?ure contents at harvest and six levels of different drum speeds of threshers, were selected and used for treatments in this experiment. Two conditions of materials were tested in the thresher. One condition was to thresh the experimental material immediately after cutting, referred to as the wet-material thr eshing in this study. The other was to thresh the experimental :material, dried to contain about 15-16 percent of the grain moisture under the shocking operation. This is referred to as the dry-material threshing in this study. In additioon, field measurements for the grain moistures and drum-sdeeds under actual operation practices of the traditional field threshing, were conducted with a view to comparing with results of the experimental treatments. The results of the study may be summarized as follows: 1. For threshing treatments of Japonica-type rice variety (Akibare) , the effect of drum speeds and levels of grain moisture at cutting upon brown-rice, milling, and head-rice recoveries were found statistically significant. No significant difference in these recovery rates was noticed regardless of whether the material was threshed right after cutting or after drying by the shocking operation. 2. For the Tongil-sister rice variety(Suweon 251), milling recovery for the varied drum-speed and the grain~moisture level at cutting was found statististically significant. Th milling recovery was much significant when associated with the wet-material thres?hing compared to the dry-material threshing. 3. The optimum peripheral velocity to be maintained at the edge of teeth on the thr?esher drum was determined and may be recommanded as that of about 12 to 13 meters per second in view of the maximum recovery rate of the milled rice. 4. The effect of the drum speed on the qualitative loss of the milled rice was much greater in the case of the Tongil variety than Japonica. This effect was also greater by the wet-material threshing than by the dry-material threshing. Therefore, to apply the wet-material threshing operation for the Tongil variety, in particular, it should be very important to introduce the kind of threshing technology which would maintain the drum speed at optimum. 5. A field survey for the actual drum speed of threshing operations for 50 threshers indicated that average peripheral velccity was 12.76m/sec., and that the range was from 10.50 to 14.90m/sec. Approximately, more than 30% of the experimented and measured threshers were being operated at speeds which exceeded the optimum speed determined and assessed in this study. Accordingly, it should be highly desirable and important to take counter-measures against these threshing practices of operational overspeed.

Wilted Symptom in Watermelon Plant under Ventilation Systems (환기처리에 의한 수박의 시듦증 발생 기작)

  • Occurrence of wilted symptom in watermelon plant ($Citrullus$ $lanatus$ L.) is known to be caused by physiological disorder. The symptom results in the loss of fruit production and thus the economical loss of watermelon growers. The incidence of symptom is often found from the middle of March to the end of May in the major watermelon crop production areas of Korea (i.e. Uiryeong, Gyeongnam (lat $37^{\circ}$56'64"N, long $126^{\circ}$99'97"E)). Despite of extensive information about the physiological disorder, little study has been conducted to understand a relationship between the wilted symptom and accompanying environment factors (e.g. temperature). This study aimed to investigate effects of environmental conditions amended by a forced-ventilation system on physiological characteristics of watermelon and incidence of the wilted symptom. Watermelon plants were grown from January to May, 2009 with either the forced-or natural-ventilation treatment in a greenhouse located in the Uiryeong. In the result, the forced-ventilation treatment decreased the air, leaf and root-zone temperature approximately $4.5^{\circ}C$, $5^{\circ}C$ and $3^{\circ}C$, respectively, compared to the natural-ventilation. The fruit growth rate was maximized twice during the entire growing period. The higher rate of fruit growth was observed under the natural-ventilation than the forced one. Maximization of the fruit growth rate (approximately 430 g per day) was first observed by 12 days after fruiting under the natural-ventilation treatment, while the second one (approximately 350 g per day) was observed by 24 days after fruiting. The wilted symptom started occurring by 22 days after fruiting under the natural-ventilation, whereas no incidence of the symptom was found under the forced-ventilation treatment. Interestingly, the forced-ventilation lowered the fruit growth rate (approximately 320 g per day) compared to the natural one. Maximization of the fruit growth rate under the forced-ventilation was found at 4 days later than that under the natural one. This result coincided with a slower plant growth under the forced-ventilation treatment. These results suggest that the forced-ventilation slows down extension growth of fruit and plant, which may be associated with lowering leaf temperature and saturation deficit. We suggest the hypothesis that the forced-ventilation may alleviate stress of the wilted symptom by avoiding extreme water evaporation from leaves due to high temperature and thus by reducing competition between leaves and fruits for water. More direct and detailed investigations are needed to confirm the effect of the forced ventilation.

Simultaneous Determination of Carbohydrates and Organic Acids in Various Cultured Dairy Foods by High-Performance Liquid Chromatography: A Preliminary Study (다양한 낙농 발효유제품에서 HPLC를 이용하여 탄수화물과 유기산의 동시 검출에 관한 연구)

  • Various carbohydrates (lactose, glucose, and fructose), lactic acid, uric acid, and acetoin were separated on a ZORBAX Carbohydrate Analysis column using the Agilent 1200 HPLC ChemStation$^{TM}$, and were identified according to retention times with 325 Dual Wavelength UV-Vis Detector and Refractive Index Detector with 0.013 N $H_2SO_4$ at a flow rate of 0.8 mL/min. In addition, the lactase activity of four commercial probiotic lactic acid bacteria during 6-hour incubation was determined using a high-performance liquid chromatography (HPLC) method. Among the tested samples, Bifidobacterium animalis subsp. lactis showed the greatest lactase activity, followed by Lactobacillus rhamnosus and Lactobacillus casei, with Streptococcus salivarius subsp. thermophilus showing the lowest activity. Therefore, this HPLC technique shows potential for evaluating the fermentation processes of probiotic lactic acid bacteria and could simultaneously confirm the degree of ripening in various fermented dairy foods within only a half hour.

Effects of the Additives on the Quality of Alfalfa Silage (첨가제 처리가 알팔파 사일리지의 품질에 미치는 영향)

  • This experiment was carried out to investigate the effects of additives on the quality of alfalfa(Medicago sativa L.) silage at the forage experimental field, National Livestock Research Institute, RDA, Suwon in 1996. The experiment was arranged in a randomized block design with three replications. The treatments consisted of different additives(control, formic acid, molasses, inoculant A and inoculant B). Crude protein(CP) and Nitrogen free extract(NFE) content of alfalfa silage with additives were higher compared with those obtained control, but Crude ash(CA) and Crude fiber(CF) content of control(11.5 and 39.6%) were higher than those of additive treated plots. ADF and NDF contents of control silage were the highest as 36.2 and 48.6%, respectively, and increased by 4% compared with those obtained before ensiling. The acidity(pH) of control was the highest(5.45), but the lowest in inoculant A(4.32). Inoculant teratment significantly reduced acidity of silage compared with control. DM content of control silage was the lowest but DM loss of control silage was higher than that of additive treated silage. Ammonia-N content was the highest at formic acid treatment and level of Lactobacillus was decreased at control and formic acid treated silage. Total organic acid and lactic acid content of inoculant A and inoculant B were higher than those of other treated plots. The results of this study indicate that the treatment of LAB(Lactic acid bacteria) could be recommended as producing high quality of alfalfa silage.

An Evaluation of Condensed Molasses Solubles (CMS) as a Source of Nitrogen for Ruminal Microbes In Vitro (반추위 미생물의 질소공급원으로서 Condensed Molasses Solubles (CMS)의 사료 가치 평가)

  • A series of four in vitro experiments were conducted to evaluate condensed molasses solubles (CMS) as a source of nitrogen for ruminal microbes. In experiment 1, as compared with urea, the value of CMS as a nitrogen source was examined. In experiment 2, to determine the time needed for maximal response of microbial synthesis, the treatments were incubated for increasing times (from 6 h to 16 h). Because a sediment that was assumed to cause nitrogen loss was found after incubation in experiments 1 and 2, it was decided to avoid formation of sediment using sugar instead of molasses or a shorter time incubation (experiments 3 and 4). Furthermore, in experiment 4, because the extent to which ammonia nitrogen is released from CMS and urea before 6 h of incubation was uncertain, it was decided to examine the peaks of concentrations of ammonia nitrogen released from CMS and urea by sampling after 2 h incubation. There was no significant difference in the concentration of microbial-N between molasses/CMS and molasses/ urea treatments in experiment 1, although there were greater decreases in ammonia concentration with the molasses/CMS treatment. The microbial protein synthesis was increased progressively until 10 h for both treatments (experiment 2). Although ingredients that were completely soluble (sucrose, urea) were used in experiment 3, the sediment was still evident suggesting that the sediment was largely of microbial not feed origin. Ammonia release from CMS was much faster than from urea during 2 h incubation. In conclusion, the results of the present studies suggest that the feed value of CMS as a source of nitrogen for ruminal bacteria was similar to that of urea when it was estimated in vitro.

Changes of Physico-chemical Characteristic on Swine Manure Using Different Suction Strength in Composting System (돈분 퇴비화 시 공기 흡입 강도에 따른 이화학적 특성변화)

  • The aim of this experiment was to investigate the effect of air suction rate (SR) during the composting process of swine manure mixed with sawdust used as a bulking agent. In the 25 L composting reactors, the suction rate (SR) was at four different treatment levels (100%, 200%, 300%, 400%), and were fixed on the based on constant aeration rate into the composting mixtures. The temperature reached to thermophilic phase within 2 days and it was maintained up to the $5^{th}$ day of the composting process in all reactors and then gradually decreased to room temperature at the end of the composting process. The moisture content (MC, %) of the initial mixtures was 64.27%, and it was reduced to 38.4, 33.08, 14.59 and 11.93 in the different suction rate of 100%, 200%, 300%, 400%, respectively in the end process. During the composting, the level of pH was increased from 6.83 to 8.67 and it gradually decreased to 7.56 in 100% and 200%(SR). At the same time, the pH values were reduced only up to 8.19 at 300%, and 8.08 at 400%(SR), showing that suction strengths of 100% and 200% were the better option for composting than those of 300% and 400%. The total Kjeldahl nitrogen (TKN) of initial composts mixtures was 2.3% and were changed in 3.3, 3.1, 2.5, and 2.3% at the end of the composting period from the 100%-400% (SR) variations respectively. These results also indicated that 100% and 200% (SR) were more affected by the dry mass loss as $CO_2$ and water evaporation. The initial value of C/N ratio was 25.17 and were significantly reduced to 11.88, 11.97, 14.31, and 14.72 at the end of the experiment, respectively from the 100%-400% (SR) variations. These results suggest that the suction rate (SR) of 100% and 200% relative to constant air supply would be the optimal conditions to produce high-quality compost.

Studies on the Quality and Utilization of Pumpkin Silages (호박 Silage의 품질(品質) 및 이용성(利用性)에 관(關)한 연구(硏究))

  • The preservation efficiency, quality and utilization of silage from 3 species of pumpkins (Mammoth pumpkin, Queensland blue pumpkin, Korean pumpkin) without and with 10, and 20% wheat bran additive were studied in this experiment. Silages were analysised and tested the chemical composition, pH and quality of silages between at 40-60 days and egg performance were carried out with mammoth pumpkin silage without additive. The results were summaried as follows. 1. The losses of all silage ware lower and similar as about 15% at 6 monthes following after silage-making but all raw pumpkins were spoilaged during the winter storaging. 2. The moisture content of silages were higher as about 97% in mammoth pumpkin silage, 94% in Queensland pumpkin silage and 91% in Korean pumpkin silage without additive and all nutrient content of silage without and with additive were depended on its content of raw silage material of pumpkins and wheat bran. The contents of moisture and N-free extract were slightly decrease but not significantly difference during the silaging and other contents were not so much changed. 3. Good quality of silage were made from all pumpkins with and without additive. Organic acid contents were 2.09-2.93% of lactic acid, 0.68-1.71% of acetic acid and 0% of butyric acid and it was pH 3.8-4.0 in silages. 4. Feed intakes, egg production and quality of egg were showed good result in 5.0 and 7.5% silage feeding group as D.M. base for egg performance. (P<0.01) 5. It was concluded that good quality of silage were made from pumpkins with and without wheat bran additive and it was suggested that poor quality feedstuff may be improved it feeding value by extended palatability with pumpkin additive silage.
