Penicillin sensor was manufactured by immobillizing penicillinase with glutaraldehyde on the $H^+$-selective membrane based on PVC-COOH-TDDA. This membrane was not inter fered by $K^+$ ion in Pc-G potassium salt. When enzyme was immobilized with glutaraldehyde, the PVC-COOH matrix was more effective than PVC matrix. Calibration curve calculated from Nernst equation was not linear. But potential was relative to concentration of Pc-G. And maximal potentiometric velocity was also relative to concentration of Pc-G. Therefore, it may be applied to Michaelis-Menten equation. The penicillin sensor was useful for determination of Pc-G at concentration of 0.1~10mM level.
The purpose of this study was to investigate whether D-myo-inositol-l,2,6-trisphosphate (PP56) can effectively antagonize vasoconstriction caused by neuropeptide Y in the dental pulp, and to understand involvement of neuropeptide Y in the regulation of microcirculation in the dental pulp with the aim of elucidating neurogenic inflammation. Experiments were performed on 7 cats anesthetised with sodium pentobarbital, and neuropeptide Y and a neuropeptide Y antagonist PP56 were injected close intra-arterially into the dental pulp. The probe of laser Doppler flowmeter was placed on the buccal surface of ipsilateral canine teeth to the drug administration and pulpal blood flow was measured. Intra-arterial injection of neuropeptide Y (1.3-$2.0\;{\mu}g$/kg) resulted in pulpal blood flow decrease of $37.73{\pm}5.73%$(mean${\pm}$SEM) (n=9). Intra-arterial injection of PP56(0.3 mg/kg) alone changed pulpal blood flow little by 1.03 % reduction. The effect of neuropeptide Y in the presence of PP56 resulted in significantly less decreases in pulpal blood flow ranging from $27.17{\pm}5.37$ to $16.63{\pm}3.48%$ from control as compared with neuropeptide Y alone(n = 13). In effect, PP56 attenuated pulpal blood flow caused by neuropeptide Y. Results of the present study have provided evidences that a non-peptide PP56 is capable of antagonizing vasoconstriction caused by neuropeptide Y in the feline dental pulp. In addition, they show functional evidences that neuropeptide Y plays an active role in modulating the microcirculation of the dental pulp.
Actin dynamics play an essential role in myogenesis through multiple mechanisms, such as mechanotransduction, cell proliferation, and myogenic differentiation. Twinfilin-1 (TWF1), an actin-depolymerizing protein, is known to be required for the myogenic differentiation of progenitor cells. However, the mechanisms by which they epigenetically regulate TWF1 by microRNAs under muscle wasting conditions related to obesity are almost unknown. Here, we investigated the role of miR-103-3p in TWF1 expression, actin filament modulation, proliferation, and myogenic differentiation of progenitor cells. Palmitic acid, the most abundant saturated fatty acid (SFA) in the diet, reduced TWF1 expression and impeded myogenic differentiation of C2C12 myoblasts, while elevating miR-103-3p levels in myoblasts. Interestingly, miR-103-3p inhibited TWF1 expression by directly targeting its 3'UTR. Furthermore, ectopic expression of miR-103-3p reduced the expression of myogenic factors, i.e., MyoD and MyoG, and subsequently impaired myoblast differentiation. We demonstrated that miR-103-3p induction increased filamentous actin (F-actin) and facilitated the nuclear translocation of Yes-associated protein 1 (YAP1), thereby stimulating cell cycle progression and cell proliferation. Hence, this study suggests that epigenetic suppression of TWF1 by SFA-inducible miR-103-3p impairs myogenesis by enhancing the cell proliferation triggered by F-actin/YAP1.
Objectives : This experimental study was designed to investigate the effect of KangwhalSokdan-tang(Jianghuoxuduan-tang) on the muscle regeneration of atrophied rat muscle by hindlimb suspension. Materials and methods : In this study, Sprague-Dawley rats weighing about 250g were subjected to hindlimb suspension and divided into total four groups: Normal group(n=6), Control group(n=6), Hindlimb non-treatment group(n=6), Hindlimb treatment group(n=6). Experiments were seperately tried two times. The first trial was studied by the following two groups; The first was normal group(n=6). The second was group(n=18) for hindlimb suspension during 2 weeks (control I group). The second trial after 2 weeks hindlimb suspension was studied by the following three groups; The third group(n=6) was expired immediately after 2 weeks hindlimb suspension. The forth group(n=6) was given free activity during 2 weeks after 2 weeks hindlimb suspension. The fifth group(n=6) was administrated of KST during 2 weeks after 2 weeks hindlimb suspension. In order to investigate degree of muscle atrophy, body weight and gastrocnemius muscle mass were compared. To analyze muscle regeneration factors(expression of IGF-1, Myogenin, MyoD), Western blot was used. Results : The results were analyzed by statistical process as follows, 1. In body weight, all hindlimb suspension groups were lower than normal group, but tendency of increase was shown in KST group compared to non-treatment group after 2 weeks hindlimb suspension. 2. In gastrocnemius muscle mass, KST group on both side was significantly higher than non-treatment group after 2 weeks hindlimb suspension. 3. In case of IGF-I, Type I of KST group was significantly increased than non-treatment group, but Type II was not shown significance. 4. There was no significantly difference in Myogenin. 5. In MyoD, Type I of KST group was significantly increased than control group, and Type II of KST group was significantly increased than non-treatment group. Conclusions : In summary, this study demonstrates that KST administration has an effect to prevent muscle atrophy and contribute muscle regeneration and proliferation. And also it is suggested that IGF-I and MyoD is major factors of myogenesis expression to KST adminstration after hindlimb suspension.
This study was investigated to establish a regeneration system of plant via adventitious bud formation from leaf explant cultures of strawberry (Fragaria ananassa Duch). Effects of plant growth regulators (2,4-D, BAP), agar sucrose and myo-inositol on adventitious bud formation were investigated. When the leaf explants were cultured on MS medium supplemented with 0.1 mg/L 2,4-D and 3 mg/L BAP, the adventitious bud formation was most promoted. The adventitious bud formation was not induced from leaf explants cultured on MS medium containing 2,4-D alone. Adventitious bud formation was enhanced to almost 3 times on medium with low level of agar concentration (0.4%) in comparison with those on the medium with high level of agar (1%), but almost of shoot was vitrificated on the medium. Therefore, the normal adventitious bud formation from leaf explants was most effective on the medium containing 0.05 mg/L 2,4-D,1 mg/L BAP, 0.8% agar, 30 g/L sucrose and 100 mg/L myo-inositol. Thus, the mass propagation of healthy strawberry could be established using leaf explants.
Oyster mushroom is one of the popular mushrooms for Korean people and it is thus one of the mushrooms that were mostly cultivated in Korea in addition to winter and king oyster mushrooms. To promote the consumption in terms of its nutritional value and determine the absorption mechanism in its physiological and metabolic aspects, we investigated the sugar composition of its fruit bodies according to the amount of sugar component in the medium. As a result from the treatment of 4 different sugar components to the sawdust medium of oyster mushroom varieties (i.g., Suhan 1 ho, Yeorum, Chunchu 2 ho), we detected fructose, glucose, ribose, xylose as a monosaccharide with ${\alpha}$-lactose and trehalose as a disaccharide. The sugar alcohols were also detected including glycerol, mannitol, myo-inositol, and sorbitol. The sugar components that were detected in all treatments were trehalose, mannitol, ${\alpha}$-lactose, fructose, glucose, ribose, and myo-inositol and the rest of the components were found in some treatments with their different quantity included in the fruit bodies. The amount of monosaccharides included in fruit bodies is comparatively low and Suhan 1 ho contained them more than other varieties. There was no remarkable difference in the amount of sugar components in fruit bodies depending on treated sugars and their amounts in the treatment. With the increase of lactose treatment, the sugar components were markedly increased and the amount of trehalose in the fruit bodies was different according to the varieties. Therefore, it is not possible to detect the trend of sugar components according to the medium used in the cultivation. The sugar alcohols in the fruit bodies were comparatively contained more in Suhan 1 ho and Chunchu 1 ho. The amount of mannitol in the fruit bodies was not possible to be detected in terms of its trend according to the treatment. For myo-inositol, its quantity is gradually increasing in Suhan 1 ho and Chunchu 1 ho and it is not possible to detent the trend in Yeorum. In conclusion, we only detected ${\alpha}$-lactose as a sugar component that showed a correlation between the amount of treatment and the amount of the fruit bodies. Therefore, the sugar components detected in the fruit bodies were transformed rather than direct absorption into fruit bodies and further studies are needed to address this question.

A compact Wilkinson power divider with meta-material transmission line(MM-TL) is proposed. The divider is designed by adding the MM-TL with $+90^{\circ}$ phases shifting instead of the ${\lambda}_g/4$ with $-90^{\circ}$ phases shifting at a simple Wilkinson power divider. The MM-TL consists of three phase shifter unit cells and each cell has the characteristics of the $30^{\circ}$ phases shilling and 6 mm length. Therefore, the length of ${\lambda}_g/4(210 mm)$ TL with $-90^{\circ}$ phases shifting at a simple Wilkinson power divider can be reduced to 18 mm and the Wilkinson divider is very compact.

The $H^+$-selective membrane consists of tridodecylamine, 2-nitrophenyl octyl ether and a little amount of additive in carboxylated PVC matrix, where penicillinase(Pcase ) and glutaraldehyde may be covalently attached to the matrix. When the $H^+$-selective electrode was used as a detector of biosensor, calibration curve calculated from Nernst equation was not linear. So we investigated the characteristic responses of the $H^+$-selective electrode to the product H+ from hydrolysis of penicillin-G(Pc-G) and plotted calibration curve correlating potential to concentration of Pc-G linearly. The optimal concentration of buffer solution was theoretically calculated and was also experimented. We tried to explain the linear curve of potential to concentration of Pc-G by using Henderson-Hasselbach equation. This method is more effective in calibration curve plotting than any other previous methods. The results obtained may help in further developing pH electrodes with improved analytical preformance.
This study investigated the effects of dietary rumen-protected L-tryptophan (TRP) supplementation (43.4 mg of L-tryptophan kg-1 body weigt [BW]) for 65 days in Hanwoo steers on muscle development related to gene expressions and adipose tissue catabolism and fatty acid transportation in longissimus dorsi muscles. Eight Hanwoo steers (initial BW = 424.6 kg [SD 42.3]; 477 days old [SD 4.8]) were randomly allocated to two groups (n = 4) of control and treatment and were supplied with total mixed ration (TMR). The treatment group was fed with 15 g of rumen-protected TRP (0.1% of TMR as-fed basis equal to 43.4 mg of TRP kg-1 BW) once a day at 0800 h as top-dressed to TMR. Blood samples were collected 3 times, at 0, 5, and 10 weeks of the experiment, for assessment of hematological and biochemical parameters. For gene study, the longissimus dorsi muscle samples (12 to 13 ribs, approximately 2 g) were collected from each individual by biopsy at end of the study (10 weeks). Growth performance parameters including final BW, average daily gain, and gain to feed ratio, were not different (p > 0.05) between the two groups. Hematological parameters including granulocyte, lymphocyte, monocyte, platelet, red blood cell, hematocrit, and white blood cell showed no difference (p > 0.05) between the two groups except for hemoglobin (p = 0.025), which was higher in the treatment than in the control group. Serum biochemical parameters including total protein, albumin, globulin, blood urea nitrogen, creatinine phosphokinase, glucose, nonesterified fatty acids, and triglyceride also showed no differences between the two groups (p > 0.05). Gene expression related to muscle development (Myogenic factor 6 [MYF6], myogenine [MyoG]), adipose tissue catabolism (lipoprotein lipase [LPL]), and fatty acid transformation indicator (fatty acid binding protein 4 [FABP4]) were increased in the treatment group compared to the control group (p < 0.05). Collectively, supplementation of TRP (65 days in this study) promotes muscle development and increases the ability of the animals to catabolize and transport fat in muscles due to an increase in expressions of MYF6, MyoG, FABP4, and LPL gene.
